    Speculations on the application of foliar 13C discrimination to reveal groundwater dependency of vegetation and provide estimates of root depth and rates of groundwater use

    © Author(s) 2018. Groundwater-dependent vegetation is globally distributed, having important ecological, social, and economic value. Along with the groundwater resources upon which it depends, this vegetation is under increasing threat through excessive rates of groundwater extraction. In this study we examined one shallow-rooted and two deep-rooted tree species at multiple sites along a naturally occurring gradient in depth-to-groundwater. We measured (i) stable isotope ratios of leaves (δ 13C), xylem, and groundwater (δ 2H and δ 18O); and (ii) leaf-vein density. We established that foliar discrimination of 13C (Δ13C) is a reliable indicator of groundwater use by vegetation and can also be used to estimate rooting depth. Through comparison with a continental-scale assessment of foliar Δ13C, we also estimated the upper limits to annual rates of groundwater use. We conclude that maximum rooting depth for both deep-rooted species ranged between 9.4 and 11.2 m and that annual rates of groundwater use ranged from ca. 1400 to 1700 mm for Eucalyptus camaldulensis and from 600 to 900 mm for Corymbia opaca. Several predictions about hydraulic and leaf traits arising from the conclusion that these two species made extensive use of groundwater were supported by additional independent studies of these species in central Australia

    Hā Kūpuna National Resource Center for Native Hawaiian Elders: Decolonizing Research through Qualitative Methods and Community Partnership

    Housed under the Thompson School of Social Work & Public Health at the University of Hawai‘i at Mānoa, Hā Kūpuna National Resource Center for Native Hawaiian Elders, strives to decolonize Western research as we increase opportunities of Native Hawaiian elders to pass their knowledge and stories to younger generations. One of Hā Kūpuna’s current projects is a five-year qualitative study examining healthcare experiences among Native Hawaiian elders in rural communities to gain advice for medical and social service providers to improve Native Hawaiian health. The project was co-designed by ALU LIKE, Inc.’s Kumu Kahi program (Elderly Services Department), which advised us to conduct a series of three interviews with each elder to build rapport before jumping into questions about healthcare. The first interview focuses on establishing rapport and learning about the kupuna’s family and everyday life. The second interview asks about values they learned from their own kupuna, what they want to pass to their mo`opuna, and other strengths and resiliencies. The third interview hones in on healthcare experiences they had over their lifetime and what advice they would like to share with providers. Results from the first 26 kūpuna have revealed that many kūpuna grew up with limited access to allopathic healthcare (healthcare providers treating diseases and symptoms with drugs and surgery) and that families treated many illnesses and injuries with traditional Hawaiian cultural healing practices, including lāʻau lapaʻau (plant-based medicine), lomilomi (massage), and ho`oponopono (conflict resolution). Even with the increased access and utilization of allopathic medicine, many kūpuna preferred cultural practices or a combination of both. Kūpuna advised that allopathic healthcare providers should take the time to gain knowledge of Native Hawaiian history and culture, allow for use of both Hawaiian and allopathic modes of healing, and interact with patients on both a personal as well as a professional level. They also noted that increasing access to specialty care on Neighbor Islands could improve Native Hawaiian health and life expectancy. Results and experiences from the ALU LIKE interview project helped to inform the creation of a 48-page qualitative interviewing protocol aimed to help researchers avoid extractive practices by increasing their knowledge of Hawaiian history, engaging communities in research, and creating safe and trusting research environments. Although experiences of colonization and discrimination are unique to each Indigenous and minority group, this protocol can apply to other populations as they are at a similar risk for extractive research experiences as well. As elements of the protocols were developed, they were discussed in-depth with researchers, non-profit stakeholders, community-based organization leaders (including ALU LIKE, Inc.), and past research participants. The qualitative protocol includes sections summarizing the history of colonization and instances of poorly-executed research in Hawaiʻi that caused harm. Also included are examples of Native Hawaiian researchers who are changing the face of research, a guide for researcher self-reflection and cultural humility, roles for community members in research, data ownership and management, the need to give more than take from participants, and a step-by-step guide on how to successfully join with community partners to conduct one-on-one interviews. Included are tips on developing research questions, and gathering and reporting data in ways accessible to the community. This qualitative protocol can be used as a guide to decolonizing research.     Identification of Altered Evoked and Non-Evoked Responses in a Heterologous Mouse Model of Endometriosis-Associated Pain

    The aim of this study was to develop and refine a heterologous mouse model of endometriosis-associated pain in which non-evoked responses, more relevant to the patient experience, were evaluated. Immunodeficient female mice (N = 24) were each implanted with four endometriotic human lesions (N = 12) or control tissue fat (N = 12) on the abdominal wall using tissue glue. Evoked pain responses were measured biweekly using von Frey filaments. Non-evoked responses were recorded weekly for 8 weeks using a home cage analysis (HCA). Endpoints were distance traveled, social proximity, time spent in the center vs. outer areas of the cage, drinking, and climbing. Significant differences between groups for von Frey response, climbing, and drinking were detected on days 14, 21, and 35 post implanting surgery, respectively, and sustained for the duration of the experiment. In conclusion, a heterologous mouse model of endometriosis-associated evoked a non-evoked pain was developed to improve the relevance of preclinical models to patient experience as a platform for drug testing

    Maternal Undernutrition Significantly Impacts Ovarian Follicle Number and Increases Ovarian Oxidative Stress in Adult Rat Offspring

    BACKGROUND: We have shown recently that maternal undernutrition (UN) advanced female pubertal onset in a manner that is dependent upon the timing of UN. The long-term consequence of this accelerated puberty on ovarian function is unknown. Recent findings suggest that oxidative stress may be one mechanism whereby early life events impact on later physiological functioning. Therefore, using an established rodent model of maternal UN at critical windows of development, we examined maternal UN-induced changes in offspring ovarian function and determined whether these changes were underpinned by ovarian oxidative stress. METHODOLOGY/PRINCIPAL FINDINGS: Our study is the first to show that maternal UN significantly reduced primordial and secondary follicle number in offspring in a manner that was dependent upon the timing of maternal UN. Specifically, a reduction in these early stage follicles was observed in offspring born to mothers undernourished throughout both pregnancy and lactation. Additionally, antral follicle number was reduced in offspring born to all mothers that were UN regardless of whether the period of UN was restricted to pregnancy or lactation or both. These reductions were associated with decreased mRNA levels of genes critical for follicle maturation and ovulation. Increased ovarian protein carbonyls were observed in offspring born to mothers UN during pregnancy and/or lactation and this was associated with peroxiredoxin 3 hyperoxidation and reduced mRNA levels; suggesting compromised antioxidant defence. This was not observed in offspring of mothers UN during lactation alone. CONCLUSIONS: We propose that maternal UN, particularly at a time-point that includes pregnancy, results in reduced offspring ovarian follicle numbers and mRNA levels of regulatory genes and may be mediated by increased ovarian oxidative stress coupled with a decreased ability to repair the resultant oxidative damage. Together these data are suggestive of maternal UN potentially contributing to premature ovarian ageing in offspring

    Brain metastases from breast cancer: lessons from experimental magnetic resonance imaging studies and clinical implications.

    Breast cancer that has metastasized to the brain presents difficult clinical challenges. This diagnosis comes with high mortality rates, largely due to complexities in early detection and ineffective therapies associated with both dormancy and impermeability of the blood-brain barrier (BBB). Magnetic resonance imaging (MRI) is the current gold standard for diagnosis and assessment of brain tumors. It has been used clinically to investigate metastatic development as well as monitor response to therapy. Here, we describe preclinical imaging strategies that we have used to study the development of brain metastases due to breast cancer. Using this approach, we have identified three subsets of metastatic disease: permeable metastases, nonpermeable metastases, and solitary, dormant cancer cells, which likely have very different biology and responses to therapy. The ability to simultaneously monitor the spatial and temporal distribution of dormant cancer cells, metastatic growth, and associated tumor permeability can provide great insight into factors that contribute to malignant proliferation. Our preclinical findings suggest that standard clinical detection strategies may underestimate the true metastatic burden of breast cancer that has metastasized to the brain. A better understanding of true metastatic burden in brains will be important to assist in the development of more effective chemotherapeutics-particularly those targeted to cross the BBB-as well as detection of small nonpermeable metastases

    Training evaluation: a case study of training Iranian health managers

    <p>Abstract</p> <p>Background</p> <p>The Ministry of Health and Medical Education in the Islamic Republic of Iran has undertaken a reform of its health system, in which-lower level managers are given new roles and responsibilities in a decentralized system. To support these efforts, a United Kingdom-based university was contracted by the World Health Organization to design a series of courses for health managers and trainers. This process was also intended to develop the capacity of the National Public Health Management Centre in Tabriz, Iran, to enable it to organize relevant short courses in health management on a continuing basis. A total of seven short training courses were implemented, three in the United Kingdom and four in Tabriz, with 35 participants. A detailed evaluation of the courses was undertaken to guide future development of the training programmes.</p> <p>Methods</p> <p>The Kirkpatrick framework for evaluation of training was used to measure participants' reactions, learning, application to the job, and to a lesser extent, organizational impact. Particular emphasis was put on application of learning to the participants' job. A structured questionnaire was administered to 23 participants, out of 35, between one and 13 months after they had attended the courses. Respondents, like the training course participants, were predominantly from provincial universities, with both health system and academic responsibilities. Interviews with key informants and ex-trainees provided supplemental information, especially on organizational impact.</p> <p>Results</p> <p>Participants' preferred interactive methods for learning about health planning and management. They found the course content to be relevant, but with an overemphasis on theory compared to practical, locally-specific information. In terms of application of learning to their jobs, participants found specific information and skills to be most useful, such as health systems research and group work/problem solving. The least useful areas were those that dealt with training and leadership. Participants reported little difficulty in applying learning deemed "useful", and had applied it often. In general, a learning area was used less when it was found difficult to apply, with a few exceptions, such as problem-solving. Four fifths of respondents claimed they could perform their jobs better because of new skills and more in-depth understanding of health systems, and one third had been asked to train their colleagues, indicating a potential for impact on their organization. Interviews with key informants indicated that job performance of trainees had improved.</p> <p>Conclusion</p> <p>The health management training programmes in Iran, and the external university involved in capacity building, benefited from following basic principles of good training practice, which incorporated needs assessment, selection of participants and definition of appropriate learning outcomes, course content and methods, along with focused evaluation. Contracts for external assistance should include specific mention of capacity building, and allow for the collaborative development of courses and of evaluation plans, in order to build capacity of local partners throughout the training cycle. This would also help to develop training content that uses material from local health management situations to demonstrate key theories and develop locally required skills. Training evaluations should as a minimum assess participants' reactions and learning for every course. Communication of evaluation results should be designed to ensure that data informs training activities, as well as the health and human resources managers who are investing in the development of their staff.</p

    TRY plant trait database - enhanced coverage and open access

    Plant traits-the morphological, anatomical, physiological, biochemical and phenological characteristics of plants-determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait-based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits-almost complete coverage for 'plant growth form'. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait-environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives

    Valuation of livestock eco-agri-food systems: poultry, beef and dairy

    As input for the TEEBAgriFood study, TEEB asked for a series of studies on livestock, rice, palm oil, inland fisheries and agro-forestry. This report deals with livestock production and aims to improve decision-making in livestock production policies, to enhance its viability, not just economically but also socially and environmentally. Livestock sector is important because they have high externalities and it is expected that livestock consumption will be 76% higher in 2050 compared to 2005 (Alexandratos and Bruisma, 2012) because of population growth, income growth, urbanization and preference shifts. This report aims to provide evidence that will help to identify policy options for the transition towards increased food security with sustainable livestock production systems, with particular emphasis on the role of smallholder farmers
