4-phenyl butyric acid improves hepatic ischemia/reperfusion and affects gene expression of ABC transporter Abcc5 in rats
Aim To assess the effects of 4-phenyl butyric acid (PBA) on
oxidative stress, inflammation, liver histology, endoplasmic
(ER) reticulum stress, and the expression levels of ATP-binding cassette transporter family members in a hepatic ischemia-reperfusion (IR) model.
Methods Thirty-five rats were randomly divided into five
groups: sham, IR, IR + 100 mg kg−1 PBA, IR + 200 mg kg−1
PBA, and IR + placebo. After sacrifice, we assessed serum
biochemical variables, myeloperoxidase (MPO), malondialdehyde (MDA), total antioxidant status (TAS), and total oxidant status (TOS). The expression levels of Abcc (2 and 5),
Abcg2, Abcf2, Ire1-α, and Perk genes were measured with a
quantitative real-time polymerase chain reaction.
Results Serum biochemical variables, MPO, MDA, TAS,
and TOS levels of the PBA groups (especially in the low
dose group) were lower than in the IR and placebo group

A novel mutation in FK506 binding protein-like (FKBPL) causes male infertility
Aim To perform a mutation analysis of FK506 binding protein-like (FKBPL) in patients with azoospermia.
Methods DNA samples were isolated from the peripheral
blood of 30 azoospermic male patients with normal 46 XY
karyotype and 10 healthy controls. Multiplex polymerase
chain reaction assays were used to evaluate Y microdeletions, and the patients without deletions were further analyzed. Sanger sequencing was used for mutation analysis.
Results A heterozygous adenine to guanine substitution
was observed at position c.28 (c.28A>G) (one patient), guanine to adenine substitution at c.90 (c.90G>A) (three patients), and a novel insertion mutation of TCTCATAAGTCT
at c. 229_240dup (two patients), all in FKBPL exon 2. Furthermore, four different benign variants were observed in
the same gene.
Conclusion Our study supports the literature on the etiologic effects of changes on autosomal chromosomes
and highlights the importance of molecular analysis of
all known and unknown genes that could be involved in
male sexual development and functio
Red Blood cell distribution width: an emerging diagnostic factor of acute appendicitis?
Acute appendicitis is the most common surgical abdominal emergency. Immidiate diagnosis of this disease is crucial, because this condition can lead to appendiceal perforation, potential peritonitis, and even death. We read with great interest the article ‘The role of red cell distribution width (RDW) in the diagnosis of acute appendicitis: a retrospective case-controlled study’ by Narci et al. and wanted to discuss whether RDW alone provide certain information about the inflammatory status of the patient with acute appendicitis
Thermal Expansion Behavior of Nonoriented Polypropylene/Clay Composites
Linear thermal expansion coefficient (LTEC) was measured for compression molding samples of polypropylene (PP)/clay composites with clay loading of 0 to 7 wt%. Composites were prepared by internal batch mixer and specimens were prepared by compression molding. These processing methods are not anticipated to have a preference for orientation; therefore effect of anisotropy was minimal. The LTEC was measured along three different faces of the compression molding sheets, parallel to compression direction S1 and perpendicular to compression directions S2 and S3. The LTEC for neat PP measured by current research, 1×10-4 mm/mm/°C, was not found to be altered by direction of the measurements. Similar behavior was observed with composites having very moderate clay content, that is, 5 wt%. An interesting finding by current study was that incorporating clay particles into the PP matrix led to better shrinkage or contraction behavior of the samples prepared by compression molding.</jats:p
An Extremely Uncommon Case of Parasitic Infection Presenting as Eosinophilic Ascites in a Young Patient
We report the case of a 24-year-old male patient admitted for recent ascites and splenomegaly of unknown origin. The patient was referred to our institution with complaints of diarrhea, epigastric pain, abdominal cramping and weight loss over the past three weeks. The acute onset presented with colicky abdominal pain and peritoneal effusion. History revealed reduced appetite and weight gain of 7 kg over the last one month. His past medical history and family history was negative. He had no history of alcohol abuse or viral hepatitis infection. Laboratory data revealed normal transaminases and bilirubin levels, and alkaline phosphatase and gammaglutamyltransferase were within normal range. A diagnostic laparoscopy was performed which showed free peritoneal fluid and normal abdominal viscera. Upper gastrointestinal system endoscopy performed a few days later revealed diffuse severe erythematous pangastritis and gastroduodenal gastric reflux. Duodenal biopsies showed chronic nonspecific duodenitis. Antrum and corpus biopsies showed chronic gastritis. The ascitic fluid was straw-colored and sterile with 80% eosinophils. Stool exam was negative for parasitic infection. Treatment with albendazole 400 mg twice daily for 5 days led to the disappearance of ascites and other signs and symptoms. Three months after albendazole treatment the eosinophilic cell count was normal. The final diagnosis was consistent with parasitic infection while the clinical, sonographic and histological findings suggested an eosinophilic ascites. We emphasize the importance of excluding parasitic infection in all patients with eosinophilic ascites. We chose an alternative way (albendazole treatment) to resolve this clinical picture. With our alternative way for excluding this parasitic infection, we treated the patient and then found the cause
Evaluation of endothelial dysfunction in patients with familial Mediterranean fever: the relationship between the levels of asymmetric dimethylarginine and endocan with carotid intima-media thickness and endothelium-dependent vasodilation
It has been suggested that there is an ongoing subclinical inflammation in familial Mediterranean fever (FMF) patients also in attack-free periods as well. Due to this ongoing inflammation, endothelial dysfunction (ED) may develop. Previously, ED has been suggested to increase the risk of the atherosclerosis and cardiovascular disease (CVD). Endocan is recognized as a specific molecule of the endothelium and has been shown to increase in some cases associated with inflammation. However, there is not sufficient data whether those with FMF could develop ED in the early period of life. In this study, we aimed to investigate ED and its relation with endocan in young FMF patients. A total of 57 male patients diagnosed with FMF according to the Tel Hashomer criteria and a total of 33 healthy males with similar characteristics to the patient group were included in this research. Complete blood count, erythrocyte sedimentation rate (ESR), fibrinogen, serum glucose, serum LDL cholesterol (LDL-C) and triglyceride (TG), asymmetric dimethylarginine (ADMA), and endocan levels were tested from fasting blood samples. Moreover, carotid intima-media thickness (CIMT) and flow-mediated dilatation (FMD) were measured. The endocan levels of the FMF patients during an attack-free period were significantly higher than those of the control group (p 0.05). CIMT values were similar among FMF patients and healthy controls (p > 0.05). These results have suggested that ED may develop in the patients with FMF who have no additional CVD risk, even during young adulthood, and endocan may be a favorable biomarker at demonstration of ED than ADMA among FMF patients
Midkine level may be used as a noninvasive biomarker in Crohn's disease
Background/aim: Crohn's disease (CD) is a kind of inflammatory bowel disease. Midkine (MDK) is an endogenous inflammatory marker. We aimed to investigate the relationship between MDK levels and inflammation and hence determine whether MDK can be used as a noninvasive biomarker in active CD. Materials and methods: Sixty-five consecutive patients over the age of 18 with CD and 36 healthy controls were included in this study. CD patients' venous blood samples were taken before treatment. Serum MDK levels were determined in human plasma samples by enzyme-linked immunosorbent assay (ELISA) method. Results: The mean age of the study patients was 44.8 +/- 12.5 years, 35 patients were female, and 30 were male. Of these 65 patients, 37 had active CD and 28 were in the remission phase. MDK levels were significantly higher in active and remission CD than in healthy controls (P = 0.01, P = 0.038, respectively). Conclusion: We report that there is an association between MDK levels and CD activation, and therefore with enhanced inflammation. MDK levels were significantly correlated with inflammatory indices. In line with our findings, we suggest the theory that MDK inhibitors may be useful in treating Crohn's disease
Soft body impact resistance of composite foam core sandwich panels with unidirectional corrugated and tubular reinforcements
Bird strikes represent a major hazard in the lifecycle of composite aircraft components, due to the low impact resistance of composites. The research presented in this paper investigates soft body impact performance of composite sandwich panels with corrugated and tubular core reinforcements. This type of panel with augmented strength and stiffness in one direction is of high importance for specific aerospace applications. The panels were subjected to high velocity impact with soft gelatine projectile as used in bird strike tests. In addition, the experimental part included non-destructive inspections of the sandwich panel samples. Panel performance was also analysed with the non-linear transient analysis software LS-DYNA with finite element and SPH capability. The panel with corrugated reinforcement showed good impact resistance with damage restricted to the impacted face sheet, foam core and corrugated reinforcement. The panel with tubular reinforcement, of the same thickness, did not suffer any damage at the same impact velocity of 115 m/s, but was damaged at a higher impact velocity of 235 m/s. The numerical studies helped to understand the experimental data, enabling comparison of impact performance of the reinforced sandwich panels and a benchmark conventional sandwich panel. The proposed reinforced sandwich panels, with the desired augmented strength and stiffness in one direction, showed improved impact resistance in comparison to conventional sandwich panels and therefore have potential for application in aerospace structures where these properties are desirable.
