    Assessing the conservation status of the Sinai Baton Blue butterfly (Pseudophilotes sinaicus)

    Arid environments are resource-limited, with scarcity of water the key limiting factor for plants and their associated fauna. Consequentially bottom-up forces often control food webs, influencing the whole system through high levels of competition. The Sinai Baton Blue butterfly, Pseudophilotes sinaicus, is Critically Endangered, with a tiny endemic distribution in the St Katherine Protectorate, South Sinai, an arid environment. Its range is restricted to that of its sole host plant, the near-endemic endangered Sinai Thyme, Thymus decussatus, leaving the butterfly in a highly fragmented distribution. This study looks into the spatio-temporal variations in quality and abundance of the host plant and its implications for the Sinai Baton Blue. Over the past decade the butterfly has exhibited severe population cycles, with the causes still unclear; it could be due to the fluctuating resource levels with large temporal variation in the quality of thyme and density of inflorescences. The number of flowers significantly influences the larval distribution, indicating that resources play a key role in offspring survivorship. Population viability analysis has also highlighted the importance of management techniques aimed at increasing the butterfly's survivorship. The butterfly population sizes are positively correlated with the total resource area and the number of host plants but not the distance between habitat patches. Population viability analysis also suggests that habitat area is more influential than connectivity in this system driving current dynamics. Overall this project has highlighted the urgent need for conservation focused on improving plant quality within patches in order to save this fragile species

    Alaska native studies unit for fourth grade using place-based education, project-based learning, cooperative learning and indigenous knowledge

University of Alaska Fairbanks, 2019As a fourth-grade educator who was responsible for teaching social studies, specifically meeting the Alaska Standards that focus on Alaska’s history of Indigenous peoples it is incredibly important that I teach accurately and genuinely. This has been a weak area of mine, since I am not from the state of Alaska. Therefore, it is an area that I wanted to further develop in my teaching practices. I developed an integrated social studies quarter long (nine weeks) unit for fourth grade that focuses on the history of Alaska from the Indigenous viewpoint. There are countless atrocities that occurred to the Indigenous population of Alaska that often get brushed under the rug. It is a disservice to not educate my students on these things. Another issue when teaching Alaska history and culture is that stereotypes and biases are often unintentionally taught as well. It is necessary that I understand my own perceptions and beliefs as a White female with little exposure and understanding of Alaska Native culture and education. Being a white female puts me in a position where I am not able to share my own experiences and knowledge as someone who is Alaska Native and grew up with the culture and language, because of this I needed to seek resources outside of myself to be able to accurately and relevantly teach a unit on Alaska Native history, knowledge and culture. I included place-based learning, cooperative learning and project-based lessons into my unit which allows students to explore the local environment and incorporate Alaska Native knowledge. This unit goes beyond social studies, because teaching must be open and welcoming to diversity and differences. Classrooms must be accepting and understanding so that students feel safe to share their own knowledge and stories with one another, and listen with respect and kindnes

    Is pain perception altered in people with depression? A systematic review and meta-analysis of experimental pain research

    While clinical studies suggest depressed patients may be more vulnerable to pain, experimental research is equivocal. This meta-analysis aimed to clarify whether depression is associated with altered pain perception in response to noxious stimulation and to identify factors that might influence this association. A search of major electronic databases was conducted to identify experimental studies investigating pain response in depressed participants vs. healthy controls using established pain outcome measures. Random effects meta-analysis of standardized mean differences was conducted on data from 32 studies (N=1,317). For high-intensity noxious stimulation, overall pain tolerance was similar across depressed and control groups (Hedge's g=0.09, p=0.71, studies=10). For low-intensity stimulation, a small, but statistically significant higher mean sensory threshold (g=0.35, p=0.01, studies=9) and pain threshold (g=0.32, p=0.02, studies=25) was observed in depressed participants, suggesting diminished pain. However, considerable heterogeneity in the direction and magnitude of effects was observed, indicating a likely condition-specific impact of depression on pain. Subgroup analysis found that pain threshold/tolerance was increased in depression for exteroceptive (cutaneous) stimulation but decreased for interoceptive (ischemic) stimulation, but that substantial heterogeneity remained. Overall, results provide some support for altered pain processing in depression, but suggest this link is dependent upon modality and additional, unidentified factors

    A Nationwide Survey of the Quality of Antimalarials in Retail Outlets in Tanzania

    INTRODUCTION\ud \ud Retail pharmaceutical products are commonly used to treat fever and malaria in sub-Saharan African countries. Small scale studies have suggested that poor quality antimalarials are widespread throughout the region, but nationwide data are not available that could lead to generalizable conclusions about the extent to which poor quality drugs are available in African communities. This study aimed to assess the quality of antimalarials available from retail outlets across mainland Tanzania.\ud \ud METHODS AND FINDINGS\ud \ud We systematically purchased samples of oral antimalarial tablets from retail outlets across 21 districts in mainland Tanzania in 2005. A total of 1080 antimalarial formulations were collected including 679 antifol antimalarial samples (394 sulfadoxine/pyrimethamine and 285 sulfamethoxypyrazine/pyrimethamine), 260 amodiaquine samples, 63 quinine samples, and 51 artemisinin derivative samples. A systematic subsample of 304 products was assessed for quality by laboratory based analysis to determine the amount of the active ingredient and dissolution profile by following the published United States Pharmacopoeia (USP) monogram for the particular tablet being tested. Products for which a published analytical monogram did not exist were assessed on amount of active ingredient alone. Overall 38 or 12.2% of the samples were found to be of poor quality. Of the antifolate antimalarial drugs tested 13.4% were found to be of poor quality by dissolution and content analysis using high-performance liquid chromatography (HPLC). Nearly one quarter (23.8%) of quinine tablets did not comply within the tolerance limits of the dissolution and quantification analysis. Quality of amodiaquine drugs was relatively better but still unacceptable as 7.5% did not comply within the tolerance limits of the dissolution analysis. Formulations of the artemisinin derivatives all contained the stated amount of active ingredient when analysed using HPLC alone.\ud \ud CONCLUSIONS\ud \ud Substandard antimalarial formulations were widely available in Tanzania at the time of this study. No products were detected that did not contain any amount of the stated active ingredient. Quinine and sulfadoxine/pyrimethamine products were the most widely available and also the most likely to be of poor quality. Substandard products were identified in all parts of the country and were labeled as made by both domestic and international manufacturers. With the expansion of the retail pharmaceutical sector as a delivery channel for antimalarial formulations the need for regular nationwide monitoring of their quality will become increasingly important

    Wellness and resilience have been at the epicenter of attention amongst many law enforcement researchers, clinicians, and professionals in recent years. Both resilience and wellness aim to provide law enforcement officers with knowledge and effective tools that can be employed during both professional and personal challenges. The current manuscript presents wellness within a context of prevalent conditions and/or situations (i.e., what is called “Good” during the “Bad” and “Ugly”) that law enforcement officers experience as part of their duties as well as in their personal lives. The authors aim to raise awareness of police wellness that needs to be viewed within the context of police work and not in a vacuum. Considering that, tangible actions and recommendations are also discussed

    Metabarcoding assays for the detection of freshwater mussels (Unionida) with environmental DNA

    Freshwater mussels of the order Unionida are a widely distributed taxon that are important in maintaining freshwater ecosystems and are also highly imperiled throughout the world. Monitoring of mussel populations with environmental DNA (eDNA) is an attractive alternative to traditional methods because it is noninvasive and requires less labor and taxonomic knowledge from field personnel. We developed eDNA metabarcoding assays specific to freshwater mussels and tested them at six sites in the Clinch River, located in the southeastern United States. Our objective was to determine the utility of eDNA metabarcoding for future monitoring of mussel populations and restoration efforts in this watershed. Two metabarcoding assays that target the mitochondrial DNA regions of the cytochrome c oxidase subunit I (COI) and NADH dehydrogenase subunit (ND1) genes were developed and tested. Our assays appear to be order specific, amplifying members from the two families found in North America, Unionidae and Margaritiferidae, while not amplifying nontarget fish or other bivalve species. From the field collected samples, our assays together detected 19 species, eight of which are listed as federally endangered. The assays also detected 42%, 58%, and 54% of the species identified by recent quantitative visual mussel surveys at three sampling sites. Increased sampling effort by processing a greater water volume or number of samples will likely increase species detections. These eDNA metabarcoding assays may enable enhanced monitoring of freshwater mussel assemblages and subsequently inform conservation efforts

    ExpiSf™: A chemically-defined baculovirus-based expression system for enhanced Protein production in Sf9 cells

    The Baculovirus Expression Vector System (BEVS) is one of the major platforms for recombinant protein production and the last decade has become a preferred platform for vaccine development. Unlike mammalian expression systems that have long since transitioned to serum-free, chemically-defined culture media, relatively little innovation has taken place in insect expression systems, with insect cells continuing to rely on undefined, yeastolate-containing culture media that can exhibit significant lot-to-lot variability in terms of both cell growth and protein expression. Here, we present the development of a novel Sf9-based Baculovirus expression system based on a high-density, chemically-defined culture medium, a high-expressing Sf9 cell line, improved transfection reagent to faster generation of baculoviruses and expression enhancer that allow for consistent production of recombinant proteins with two-fold or greater improvements in protein titers compared to traditional BEVS workflows

    Approaches to Understanding and Measuring Interdisciplinary Scientific Research (IDR): A Review of the Literature

    Interdisciplinary scientific research (IDR) challenges the study of science from a number of fronts, including one of creating output science and engineering (S&E) indicators. This literature review began with a narrow focus on quantitative measures of the output of IDR, but expanded the scope as it became clear that differing definitions, assessment tools, evaluation processes, and measures all shed light on aspects of IDR. Key among the broader aspects are (a) characterizing the concept of knowledge integration, and (b) recognizing that it can occur within a single mind or as the result of team dynamics. Output measures alone cannot adequately capture this process. Among the quantitative measures considered, bibliometrics (co-authorships, collaborations, references, citations and co-citations) are the most developed, but leave considerable gaps in understanding. Emerging measures in diversity, entropy, and network dynamics are promising, but require sophisticated interpretations and thus would not serve well as S&E indicators. Combinations of quantitative and qualitative assessments coming from evaluation studies appear to reveal S&E processes but carry burdens of expense, intrusion, and lack of reproducibility. This review is a first step toward providing a more holistic view of measuring IDR; several avenues for future research highlight the need for metrics to reflect the actual practice of IDR
