Analysis of host responses to Mycobacterium tuberculosis antigens in a multi-site study of subjects with different TB and HIV infection states in sub-Saharan Africa.
BACKGROUND: Tuberculosis (TB) remains a global health threat with 9 million new cases and 1.4 million deaths per year. In order to develop a protective vaccine, we need to define the antigens expressed by Mycobacterium tuberculosis (Mtb), which are relevant to protective immunity in high-endemic areas. METHODS: We analysed responses to 23 Mtb antigens in a total of 1247 subjects with different HIV and TB status across 5 geographically diverse sites in Africa (South Africa, The Gambia, Ethiopia, Malawi and Uganda). We used a 7-day whole blood assay followed by IFN-Îł ELISA on the supernatants. Antigens included PPD, ESAT-6 and Ag85B (dominant antigens) together with novel resuscitation-promoting factors (rpf), reactivation proteins, latency (Mtb DosR regulon-encoded) antigens, starvation-induced antigens and secreted antigens. RESULTS: There was variation between sites in responses to the antigens, presumably due to underlying genetic and environmental differences. When results from all sites were combined, HIV- subjects with active TB showed significantly lower responses compared to both TST(-) and TST(+) contacts to latency antigens (Rv0569, Rv1733, Rv1735, Rv1737) and the rpf Rv0867; whilst responses to ESAT-6/CFP-10 fusion protein (EC), PPD, Rv2029, TB10.3, and TB10.4 were significantly higher in TST(+) contacts (LTBI) compared to TB and TST(-) contacts fewer differences were seen in subjects with HIV co-infection, with responses to the mitogen PHA significantly lower in subjects with active TB compared to those with LTBI and no difference with any antigen. CONCLUSIONS: Our multi-site study design for testing novel Mtb antigens revealed promising antigens for future vaccine development. The IFN-Îł ELISA is a cheap and useful tool for screening potential antigenicity in subjects with different ethnic backgrounds and across a spectrum of TB and HIV infection states. Analysis of cytokines other than IFN-Îł is currently on-going to determine correlates of protection, which may be useful for vaccine efficacy trials
ISO Spectroscopy of Young Stellar Objects
Observations of gas-phase and solid-state species toward
young stellar objects (YSOs) with the spectrometers
on board the Infrared Space Observatory
are reviewed. The excitation and abundances of
the atoms and molecules are sensitive to the changing
physical conditions during star-formation. In
the cold outer envelopes around YSOs, interstellar
ices contain a significant fraction of the heavy element
abundances, in particular oxygen. Different ice
phases can be distinguished, and evidence is found for
heating and segregation of the ices in more evolved
objects. The inner warm envelopes around YSOs are
probed through absorption and emission of gas-phase
molecules, including CO, CO_2, CH_4 and H_2O. An
overview of the wealth of observations on gas-phase
H_2O in star-forming regions is presented. Gas/solid
ratios are determined, which provide information on
the importance of gas-grain chemistry and high temperature
gas-phase reactions. The line ratios of molecules
such as H_2, CO and H_2O are powerful probes
to constrain the physical parameters of the gas. Together
with atomic and ionic lines such as [0 I]
63 ”m, [S I] 25 ”m and (Si II] 35 ”m, they can also
be used to distinguish between photon- and shock-heated
gas. Finally, spectroscopic data on circumstellar
disks around young stars are mentioned. The
results are discussed in the context of the physical
and chemical evolution of YSOs
Measurement of Polybrominated Diphenyl Ethers and Metabolites in Mouse Plasma after Exposure to a Commercial Pentabromodiphenyl Ether Mixture
Structure and Vibrations of the Vicinal Copper (211) Surface
We report a first principles theoretical study of the surface relaxation and
lattice dynamics of the Cu(211) surface using the plane wave pseudopotential
method. We find large atomic relaxations for the first several atomic layers
near the step edges on this surface, and a substantial step-induced
renormalization of the surface harmonic force constants. We use the results to
study the harmonic fluctuations around the equilibrium structure and find three
new step-derived features in the zone center vibrational spectrum. Comparison
of these results with previous theoretical work and weith experimental studies

A Review of Controlling Motivational Strategies from a Self-Determination Theory Perspective: Implications for Sports Coaches
The aim of this paper is to present a preliminary taxonomy of six controlling strategies, primarily based on the parental and educational literatures, which we believe are employed by coaches in sport contexts. Research in the sport and physical education literature has primarily focused on coachesâ autonomysupportive behaviours. Surprisingly, there has been very little research on the use of controlling strategies. A brief overview of the research which delineates each proposed strategy is presented, as are examples of the potential manifestation of the behaviours associated with each strategy in the context of sports coaching. In line with self-determination theory (Deci & Ryan, 1985; Ryan & Deci, 2002), we propose that coach behaviours employed to pressure or control athletes have the potential to thwart athletesâ feelings of autonomy, competence,and relatedness, which, in turn, undermine athletesâ self-determined motivation and contribute to the development of controlled motives. When athletes feel pressured to behave in a certain way, a variety of negative consequences are expected to ensue which are to the detriment of the athletesâ well-being. The purpose of this paper is to raise awareness and interest in the darker side of sport participation and to offer suggestions for future research in this area
Pregnant women with bronchial asthma benefit from progressive muscle relaxation: A randomized, prospective, controlled trial
Background: Asthma is a serious medical problem in pregnancy and is often associated with stress, anger and poor quality of life. The aim of this study was to determine the efficacy of progressive muscle relaxation (PMR) on change in blood pressure, lung parameters, heart rate, anger and health-related quality of life in pregnant women with bronchial asthma. Methods: We treated a sample of 64 pregnant women with bronchial asthma from the local population in an 8-week randomized, prospective, controlled trial. Thirty-two were selected for PMR, and 32 received a placebo intervention. The systolic blood pressure, forced expiratory volume in the first second, peak expiratory flow and heart rate were tested, and the State-Trait Anger Expression Inventory and Health Survey (SF-36) were employed. Results: According to the intend-to-treat principle, a significant reduction in systolic blood pressure and a significant increase in both forced expiratory volume in the first second and peak expiratory flow were observed after PMR. The heart rate showed a significant increase in the coefficient of variation, root mean square of successive differences and high frequency ranges, in addition to a significant reduction in low and middle frequency ranges. A significant reduction on three of five State-Trait Anger Expression Inventory scales, and a significant increase on seven of eight SF-36 scales were observed. Conclusions: PMR appears to be an effective method to improve blood pressure, lung parameters and heart rate, and to decrease anger levels, thus enhancing health-related quality of life in pregnant women with bronchial asthma. Statistical mechanics of voting
Decision procedures aggregating the preferences of multiple agents can
produce cycles and hence outcomes which have been described heuristically as
`chaotic'. We make this description precise by constructing an explicit
dynamical system from the agents' preferences and a voting rule. The dynamics
form a one dimensional statistical mechanics model; this suggests the use of
the topological entropy to quantify the complexity of the system. We formulate
natural political/social questions about the expected complexity of a voting
rule and degree of cohesion/diversity among agents in terms of random matrix
models---ensembles of statistical mechanics models---and compute quantitative
