    Genetic disruption of NRF2 promotes the development of necroinflammation and liver fibrosis in a mouse model of HFE-hereditary hemochromatosis

    In hereditary hemochromatosis, iron deposition in the liver parenchyma may lead to fibrosis, cirrhosis and hepatocellular carcinoma. Most cases are ascribed to a common mutation in the HFE gene, but the extent of clinical expression is greatly influenced by the combined action of yet unidentified genetic and/or environmental modifying factors. In mice, transcription factor NRF2 is a critical determinant of hepatocyte viability during exposure to acute dietary iron overload.     Regional variation in growth status. The Peruvian health and optimist growth study

    Objective: This study aims to (1) investigate differences in weight, body mass index (BMI), and waist circumference (WC) among Peruvian children and ado lescents living in three areas located at different altitudes; (2) compare age and sex-specific height, weight, and BMI within each site with US reference data. Methods: We sampled 8753 subjects (4130 boys), aged 4 to 17 years from sea level, rainforest, and high-altitude. Height, weight, and WC were measured and BMI was calculated. Analysis of variance was used to compare variables across geographic regions, and the Hoff and Blackburn procedure was used to compare the Peruvian results with US reference data. Results: Participants living at sea level were taller, heavier, had greater BMI and WC relative to those living at high-altitude and in the rainforest. Peruvian schoolchildren of both sexes from the three geographical areas were shorter and lighter than their American peers. Boys and girls living in the rainforest and at high-altitude had lower BMI, whereas WC values of American schoolchildren are higher than those of the Peruvian children by age and gender. Conclusions: Peruvians living at different altitudes differ in their growth indi cators (height, weight, BMI, and WC), with significant differences between those living at sea level relative to their peers from other regions.     Populational analysis of Saccharomyces cerevisiae strains from different appellations of origin and grape varieties by microsatellite analysis.

    The objective of the present study was to evaluate populational relationships among Saccharomyces cerevisiae strains isolated from some of the Portuguese most important grapevine varieties in different appellations of origin, using polymorphic microsatellites. 
One hundred ninety two grape samples were collected during the 2006 and 2007 harvest season in the Vinho Verde (grape varieties: Arinto, Alvarinho, Avesso, Loureiro, Touriga Nacional) Bairrada (grape varieties: Arinto, Baga, Castelão Francês, Maria Gomes, Touriga Nacional) Alentejo (grape varieties, Aragonês, Trincadeira, Touriga Nacional), Terras do Sado (grape variety Castelão) Bucelas (grape variety Arinto) and Estremadura (grape varieties: Arinto, Aragonês, Castelão, Trincadeira, Touriga Nacional) appellations of origin. From the final stage of spontaneous fermentations, 2820 yeast isolates were obtained, mainly belonging to the species S. cerevisiae. An initial genetic screen, based on mitochondrial DNA restriction fragment length polymorphism (mtDNA RFLP) and/or interdelta sequence analysis was followed by microsatellite analysis of strains with unique genetic profiles, using 10 highly polymorphic microsatellites. Our results showed that microsatellite analysis revealed a high resolution populational screen, showing that genetic differences and populational structures among S. cerevisiae populations derived from both “diagnostic” vineyard-, specific alleles and the accumulation of small allele-frequency differences across ten microsatellite loci. Heterozygosity was three to four times lower than the expected value, confirming the strong populational substructuring. The presented large-scale approach shows that each vineyard contains differentiated S. cerevisiae populations, showing the occurrence of specific native strains that can be associated with a terroir.

    LEF1-AS1, long non-coding RNA, inhibits proliferation in myeloid malignancy

    LEF1 antisense RNA 1 (LEF1-AS1) is an antisense long non-coding RNA encoded in the lymphoid enhancer-binding factor 1 (LEF1) locus. LEF1-AS1 is a conserved transcript dysregulated in hematopoiesis. This study aimed to functionally characterize the role of this transcript in myeloid malignancy and explore a possible regulatory effect of LEF1-AS1 upon LEF1. We show that LEF1-AS1 is highly expressed in normal hematopoietic stem cells but barely detectable in myeloid malignant cell lines. Additionally, bone marrow cells from myelodysplastic syndrome (n=12) and acute myeloid malignancy patients (n=28) expressed significantly reduced levels of LEF1-AS1 compared to healthy controls (n=15). Artificial LEF1-AS1 over-expression inhibited proliferation in HL60 and led to an upregulation of tumor suppressors p21 and p27, and reduced ERK1/2 activation. Unexpectedly, no underlying modulation of LEF1 was detected. Ectopic expression of LEF1-AS1 also inhibited proliferation in HELA, a cell line lacking endogenous expression of LEF1, supporting a LEF1-independent mechanism. Additionally, transient over-expression of LEF1-AS1 in AML patient cells also led to reduced proliferation and colony formation capacity. We used a mass spectrometry-based proteomics approach.     The clinical courses of polycythemia vera (PV) and essential thrombocythemia (ET) are characterized by thrombohemorrhagic diathesis. Several groups have suggested an association between JAK2V617F mutation and thrombosis. We hypothesized a relationship between JAK2V617F allele burden, cellular activation parameters, and thrombosis. We evaluated a group of PV and ET patients using flow cytometry: platelet CD62P, CD63, and dense granules, platelet-leukocyte aggregates (PLA), leukocyte CD11b and monocyte tissue factor (TF) expression. All patients had increased baseline platelet CD62P and CD63 expression (p 50 % presented higher levels of leukocyte activation. In ET, thrombosis was associated with JAK2V617F mutation (p < 0.05, χ (2) = 5.2), increased monocyte CD11b (p < 0.05) and with platelet-PMN aggregates (p < 0.05). In ET patients, hydroxyurea does not significantly reduce the activation parameters. Our data demonstrate that JAK2V617F allele burden is directly correlated with activation parameters that drive mechanisms that favor thrombosis.info:eu-repo/semantics/publishedVersio

    Dietary whey protein lessens several risk factors for metabolic diseases: a review

    Obesity and type 2 diabetes mellitus (DM) have grown in prevalence around the world, and recently, related diseases have been considered epidemic. Given the high cost of treatment of obesity/DM-associated diseases, strategies such as dietary manipulation have been widely studied; among them, the whey protein diet has reached popularity because it has been suggested as a strategy for the prevention and treatment of obesity and DM in both humans and animals. Among its main actions, the following activities stand out: reduction of serum glucose in healthy individuals, impaired glucose tolerance in DM and obese patients; reduction in body weight; maintenance of muscle mass; increases in the release of anorectic hormones such as cholecystokinin, leptin, and glucagon like-peptide 1 (GLP-1); and a decrease in the orexigenic hormone ghrelin.
