A theoretical look at ensemble-based optimization in reservoir management
Ensemble-based optimization has recently received great attention as a potentially powerful technique for life-cycle production optimization, which is a crucial element of reservoir management. Recent publications have increased both the number of applications and the theoretical understanding of the algorithm. However, there is still ample room for further development since most of the theory is based on strong assumptions. Here, the mathematics (or statistics) of Ensemble Optimization is studied, and it is shown that the algorithm is a special case of an already well defined natural evolution strategy known as Gaussian Mutation. A natural description of uncer-tainty in reservoir management arises from the use of an ensemble of history-matched geological realizations. A logical step is therefore to incorporate this uncertainty description in robust life-cycle production optimization through the expected objective function value. The expected value is approximated with the mean over all geological realizations. It is shown that the frequently advocated strategy of applying a different control sample to each reservoir realization delivers an unbiased estimate of the gradi-ent of the expected objective function. However, this procedure is more variance prone than the deterministic strategy of applying the entire ensemble of perturbed control samples to each reservoir model realization. In order to reduce the variance of the gradient estimate, an importance sampling algorithm is proposed and tested on a toy problem with increasing dimensionality.acceptedVersio
Study of 2 beta-decay of Mo-100 and Se-82 using the NEMO3 detector
After analysis of 5797 h of data from the detector NEMO3, new limits on neutrinoless double beta decay of Mo-100 (T-1/2 > 3.1 x 10(23) y, 90% CL) and Se-82 (T-1/2 > 1.4 x 10(23) y, 90% CL) have been obtained. The corresponding limits on the effective majorana neutrino mass are: 1.4 x 10(22) y (90% CL) for Mo-100 and T-1/2 > 1.2 x 10(22) y (90% CL) for Se-82. Corresponding bounds on the Majoron-neutrino coupling constant are < (0.5-0.9) x 10(- 4) and <(0.7-1.6) x 10(- 4). Two-neutrino 2beta-decay half-lives have been measured with a high accuracy, (T1/2Mo)-Mo-100 = [7.68 +/- 0.02(stat) +/- 0.54(syst)] x 10(18) y and (T1/2Se)-Se-82 = [10.3 +/- 0.3(stat) +/- 0.7(syst)] x 10(19) y. Measurement of double beta decay of ¹⁰⁰Mo to excited states in the NEMO 3 experiment
The double beta decay of ¹⁰⁰Mo to the 0_{1}^{+} and 2_{1}^{+} excited states of ¹⁰⁰Ru is studied using the NEMO 3 data. After the analysis of 8024 h of data the half-life for the two-neutrino double beta decay of ¹⁰⁰Mo to the excited 0_{1}^{+} state is measured to be T_{1/2}^{2v} = [5.7_{-0.9}^{+1.3} (stat.) ± 0.8 (syst.)] x 10²⁰ y. The signal-to-background ratio is equal to 3. Information about energy and angular distributions of emitted electrons is also obtained. No evidence for neutrinoless double beta decay to the excited 0_{1}^{+} state has been found. The corresponding half-life limit is T_{1/2}^{0v} (0⁺→0_{1}^{+}) > 8.9 x 10²² y (at 90% C.L.). The search for the double beta decay to the 2_{1}^{+} excited state has allowed the determination of limits on the half-life for the two neutrino mode T_{1/2}^{0v} (0⁺→2_{1}^{+}) > 1.1 x 10²¹ y (at 90% C.L.) and for the neutrinoless mode T_{1/2}^{0v} (0⁺→2_{1}^{+}) > 1.6 x 10²³ y (at 90% C.L.)
Technical design and performance of the NEMO3 detector
The development of the NEMO3 detector, which is now running in the Frejus
Underground Laboratory (L.S.M. Laboratoire Souterrain de Modane), was begun
more than ten years ago. The NEMO3 detector uses a tracking-calorimeter
technique in order to investigate double beta decay processes for several
isotopes. The technical description of the detector is followed by the
presentation of its performance.Comment: Preprint submitted to Nucl. Instrum. Methods A Corresponding author:

Structure-Function Analysis of Mammalian CYP2B Enzymes Using 7-Substituted Coumarin Derivatives as Probes: Utility of Crystal Structures and Molecular Modeling in Understanding Xenobiotic Metabolism s
ABSTRACT Crystal structures of CYP2B35 and CYP2B37 from the desert woodrat were solved in complex with 4-(4-chlorophenyl)imidazole (4-CPI). The closed conformation of CYP2B35 contained two molecules of 4-CPI within the active site, whereas the CYP2B37 structure demonstrated an open conformation with three 4-CPI molecules, one within the active site and the other two in the substrate access channel. To probe structurefunction relationships of CYP2B35, CYP2B37, and the related CYP2B36, we tested the O-dealkylation of three series of related substrates-namely, 7-alkoxycoumarins, 7-alkoxy-4-(trifluoromethyl)coumarins, and 7-alkoxy-4-methylcoumarinswith a C1-C7 side chain. CYP2B35 showed the highest catalytic efficiency (k cat /K M ) with 7-heptoxycoumarin as a substrate, followed by 7-hexoxycoumarin. In contrast, CYP2B37 showed the highest catalytic efficiency with 7-ethoxy-4-(trifluoromethyl) coumarin (7-EFC), followed by 7-methoxy-4-(trifluoromethyl) coumarin (7-MFC). CYP2B35 had no dealkylation activity with 7-MFC or 7-EFC. Furthermore, the new CYP2B-4-CPI-bound structures were used as templates for docking the 7-substituted coumarin derivatives, which revealed orientations consistent with the functional studies. In addition, the observation of multiple -Cl and -NH-p interactions of 4-CPI with the aromatic side chains in the CYP2B35 and CYP2B37 structures provides insight into the influence of such functional groups on CYP2B ligand binding affinity and specificity. To conclude, structural, computational, and functional analysis revealed striking differences between the active sites of CYP2B35 and CYP2B37 that will aid in the elucidation of new structure-activity relationships
Results of the BiPo-1 prototype for radiopurity measurements for the SuperNEMO double beta decay source foils
The development of BiPo detectors is dedicated to the measurement of
extremely high radiopurity in Tl and Bi for the SuperNEMO
double beta decay source foils. A modular prototype, called BiPo-1, with 0.8
of sensitive surface area, has been running in the Modane Underground
Laboratory since February, 2008. The goal of BiPo-1 is to measure the different
components of the background and in particular the surface radiopurity of the
plastic scintillators that make up the detector. The first phase of data
collection has been dedicated to the measurement of the radiopurity in
Tl. After more than one year of background measurement, a surface
activity of the scintillators of (Tl) 1.5
Bq/m is reported here. Given this level of background, a larger BiPo
detector having 12 m of active surface area, is able to qualify the
radiopurity of the SuperNEMO selenium double beta decay foils with the required
sensitivity of (Tl) 2 Bq/kg (90% C.L.) with a six
month measurement.Comment: 24 pages, submitted to N.I.M.
Spectral modeling of scintillator for the NEMO-3 and SuperNEMO detectors
We have constructed a GEANT4-based detailed software model of photon
transport in plastic scintillator blocks and have used it to study the NEMO-3
and SuperNEMO calorimeters employed in experiments designed to search for
neutrinoless double beta decay. We compare our simulations to measurements
using conversion electrons from a calibration source of and show
that the agreement is improved if wavelength-dependent properties of the
calorimeter are taken into account. In this article, we briefly describe our
modeling approach and results of our studies.Comment: 16 pages, 10 figure
Plasmodium falciparum Hep1 is required to prevent the self aggregation of PfHsp70-3
The majority of mitochondrial proteins are encoded in the nucleus and need to be imported from the cytosol into the mitochondria, and molecular chaperones play a key role in the efficient translocation and proper folding of these proteins in the matrix. One such molecular chaperone is the eukaryotic mitochondrial heat shock protein 70 (Hsp70); however, it is prone to self-aggregation and requires the presence of an essential zinc-finger protein, Hsp70-escort protein 1 (Hep1), to maintain its structure and function. PfHsp70-3, the only Hsp70 predicted to localize in the mitochondria of P. falciparum, may also rely on a Hep1 orthologue to prevent self-aggregation. In this study, we identified a putative Hep1 orthologue in P. falciparum and co-expression of PfHsp70-3 and PfHep1 enhanced the solubility of PfHsp70-3. PfHep1 suppressed the thermally induced aggregation of PfHsp70-3 but not the aggregation of malate dehydrogenase or citrate synthase, thus showing specificity for PfHsp70-3. Zinc ions were indeed essential for maintaining the function of PfHep1, as EDTA chelation abrogated its abilities to suppress the aggregation of PfHsp70-3. Soluble and functional PfHsp70-3, acquired by co-expression with PfHep-1, will facilitate the biochemical characterisation of this particular Hsp70 protein and its evaluation as a drug target for the treatment of malaria

Measurement of the 2νββ decay half-life of 150Nd and a search for 0νββ decay processes with the full exposure from the NEMO-3 detector
We present results from a search for neutrinoless double-β (0νββ) decay using 36.6 g of the isotope
150Nd with data corresponding to a live time of 5.25 y recorded with the NEMO-3 detector. We construct a
complete background model for this isotope, including a measurement of the two-neutrino double-β decay
half-life of T2ν
1=2 ¼ ½9.34 0.22ðstatÞ þ0.62 −0.60 ðsystÞ × 1018 y for the ground state transition, which represents
the most precise result to date for this isotope. We perform a multivariate analysis to search for 0νββ decays
in order to improve the sensitivity and, in the case of observation, disentangle the possible underlying decay
mechanisms. As no evidence for 0νββ decay is observed, we derive lower limits on half-lives for several mechanisms involving physics beyond the standard model. The observed lower limit, assuming light
Majorana neutrino exchange mediates the decay, is T0ν
1=2 > 2.0 × 1022 y at the 90% C.L., corresponding to
an upper limit on the effective neutrino mass of hmνi < 1.6–5.3 eV
