    Bacterial Toxin Fusion Proteins Elicit Mucosal Immunity against a Foot-and-Mouth Disease Virus Antigen When Administered Intranasally to Guinea Pigs

    Peptides corresponding to the foot-and-mouth disease virus VP1 G-H loop are capable of inducing neutralizing antibodies in some species but are considered relatively poor immunogens, especially at mucosal surfaces. However, intranasal administration of antigens along with the appropriate delivery vehicle/adjuvant has been shown to induce mucosal immune responses, and bacterial enterotoxins have long been known to be effective in this regard. In the current study, two different carrier/adjuvant approaches were used to augment mucosal immunity to the FMDV O1 BFS G-H loop epitope, in which the G-H loop was genetically coupled to the E. coli LT-B subunit and coexpressed with the LTA2 fragment (LTA2B-GH), or the nontoxic pseudomonas exotoxin A (ntPE) was fused to LTA2B-GH at LT-A2 to enhance receptor targeting. Only guinea pigs that were inoculated intranasally with ntPE-LTA2B-GH and LTA2B-GH induced significant anti-G-H loop IgA antibodies in nasal washes at weeks 4 and 6 when compared to ovalbumin or G-H loop immunized animals. These were also the only groups that exhibited G-H loop-specific antigen-secreting cells in the nasal mucosa. These data demonstrate that fusion of nonreplicating antigens to LTA2B and ntPE-LTA2B has the potential to be used as carriers/adjuvants to induce mucosal immune responses against infectious diseases

    Bromelain Inhibits Allergic Sensitization and Murine Asthma via Modulation of Dendritic Cells

    The incidence of atopic conditions has increased in industrialized countries. Persisting symptoms and concern for drug side-effects lead patients toward adjunctive treatments such as phytotherapy. Previously, we have shown that Bromelain (sBr), a mixture of cysteine proteases from pineapple, Ananas comosus, inhibits ovalbumin (OVA)-induced murine model of allergic airway disease (AAD). However, sBr’s effect on development of AAD when treatment is administered throughout OVA-alum sensitization was unknown and is the aim of the present study. C57BL/6J mice were sensitized with OVA/alum and challenged with 7 days OVA aerosol. sBr 6 mg/kg/0.5 ml or PBS vehicle were administered throughout sensitization. Lung, bronchoalveolar lavage (BAL), spleen, and lymph nodes were processed for flow cytometry and OVA-specific IgE was determined via ELISA. sBr treatment throughout OVA-alum sensitization significantly reduced the development of AAD (BAL eosinophils and lymphocytes). OVA-specific IgE and OVA TET+ cells were decreased. sBr reduced CD11c+ dendritic cell subsets, and in vitro treatment of DCs significantly reduced CD44, a key receptor in both cell trafficking and activation. sBr was shown to reduce allergic sensitization and the generation of AAD upon antigen challenge. These results provide additional insight into sBr's anti-inflammatory and antiallergic properties and rationale for translation into the clinical arena

    Home parenteral nutrition with an omega-3-fatty-acid-enriched MCT/LCT lipid emulsion in patients with chronic intestinal failure (the HOME study):study protocol for a randomized, controlled, multicenter, international clinical trial

    BACKGROUND: Home parenteral nutrition (HPN) is a life-preserving therapy for patients with chronic intestinal failure (CIF) indicated for patients who cannot achieve their nutritional requirements by enteral intake. Intravenously administered lipid emulsions (ILEs) are an essential component of HPN, providing energy and essential fatty acids, but can become a risk factor for intestinal-failure-associated liver disease (IFALD). In HPN patients, major effort is taken in the prevention of IFALD. Novel ILEs containing a proportion of omega-3 polyunsaturated fatty acids (n-3 PUFA) could be of benefit, but the data on the use of n-3 PUFA in HPN patients are still limited. METHODS/DESIGN: The HOME study is a prospective, randomized, controlled, double-blind, multicenter, international clinical trial conducted in European hospitals that treat HPN patients. A total of 160 patients (80 per group) will be randomly assigned to receive the n-3 PUFA-enriched medium/long-chain triglyceride (MCT/LCT) ILE (Lipidem/Lipoplus¼ 200 mg/ml, B. Braun Melsungen AG) or the MCT/LCT ILE (Lipofundin¼ MCT/LCT/Medialipide¼ 20%, B. Braun Melsungen AG) for a projected period of 8 weeks. The primary endpoint is the combined change of liver function parameters (total bilirubin, aspartate transaminase and alanine transaminase) from baseline to final visit. Secondary objectives are the further evaluation of the safety and tolerability as well as the efficacy of the ILEs. DISCUSSION: Currently, there are only very few randomized controlled trials (RCTs) investigating the use of ILEs in HPN, and there are very few data at all on the use of n-3 PUFAs. The working hypothesis is that n-3 PUFA-enriched ILE is safe and well-tolerated especially with regard to liver function in patients requiring HPN. The expected outcome is to provide reliable data to support this thesis thanks to a considerable number of CIF patients, consequently to broaden the present evidence on the use of ILEs in HPN.     Air spaces of the temporal bone: a morphometric analysis with clinical implications

    Background: The main objective of the present study was to analyze the morphological variations of the air spaces of the temporal bone, that is, the pneumatized and air-filled spaces of the temporal bone cavities. Materials and methods: A total of 99 sides were analyzed. Temporal bone pneumatic spaces (TBPS) were defined as the free spaces inside the cavities of the temporal bone filled with air, excluding the volume of the structures present in the investigated region. Total volumes of TBPS were calculated as the sum of total volumes of mastoid air cells (MAC), tympanic cavity (TC), and external auditory canal (EAC). Analyses were performed considering the general population and the female and male subgroups. Results: The overall results obtained on Polish popublation were set as follows: the median total volume of TBPS was demonstrated at 7882.58 mm3 (LQ: 6200.56 mm3 ; HQ: 10393.16 mm3). The median volume of MAC was set at 5813.05 mm3 (LQ: 4224.94 mm3 ; HQ: 8181.81 mm3). The median of the total volume of the EAC was demonstrated at 1294.36 mm3 (LQ: 1099.68 mm3 HQ: 1627.84 mm3). Conclusions: In the present study, the morphometric properties of the temporal bone cavities were analyzed. The results showed that the total volume of the MAC was, on average, lower in women than in men. This should be taken into account when performing procedures on the mastoid, such as mastoidectomies. It is hoped that the results of this study can help reduce potential surgical complications associated with otological procedures

    Measurement of polarisation observables in Ks0ÎŁ+K^0_s\Sigma^+ photoproduction off the proton

    The reaction γ p→KS0 Σ+\gamma \, p \rightarrow K^0_S\,\Sigma^+ is studied in the photon energy range from threshold. Linearly polarised photon beams from coherent bremsstrahlung enabled the first measurement of photon beam asymmetries in this reaction up to EÎł=2250E_\gamma = 2250 MeV. In addition, the recoil hyperon polarisation was determined through the asymmetry in the weak decay ÎŁ+→pπ0\Sigma^+ \rightarrow p \pi^0 up to EÎł=1650E_\gamma = 1650 MeV. The data are compared to partial wave analyses, and the possible impact on the interpretation of a recently observed cusp-like structure near the K∗K^* thresholds is discussed.Comment: 6 pages, 5 figures.     The exclusive reactions Îłp→K∗0ÎŁ+(1189)\gamma p \to K^{*0} \Sigma^+(1189) and Îłp→K0π0ÎŁ+(1189)\gamma p \to K^{0} \pi^{0}\Sigma^+(1189), leading to the p 4π0\pi^{0} final state, have been measured with a tagged photon beam for incident energies from threshold up to 2.5 GeV. The experiment has been performed at the tagged photon facility of the ELSA accelerator (Bonn). The Crystal Barrel and TAPS detectors were combined to a photon detector system of almost 4π\pi geometrical acceptance. Differential and total cross sections are reported. At energies close to the threshold, a flat angular distribution has been observed for the reaction Îłp→K0π0ÎŁ+\gamma p\to K^{0} \pi^{0}\Sigma^+ suggesting dominant s-channel production. Σ∗(1385)\Sigma^*(1385) and higher lying hyperon states have been observed. An enhancement in the forward direction in the angular distributions of the reaction Îłp→K∗0ÎŁ+\gamma p \to K^{*0}\Sigma^+ indicates a tt-channel exchange contribution to the reaction mechanism. The experimental data are in reasonable agreement with recent theoretical predictions.Comment: 11 pages, 13 figures, submitted to EPJ

    Photoproduction of {\omega} Mesons off the Proton

    The differential cross sections and unpolarized spin-density matrix elements for the reaction Îłp→pω\gamma p\to p\omega were measured using the CBELSA/TAPS experiment for initial photon energies ranging from the reaction threshold to 2.5 GeV. These observables were measured from the radiative decay of the ω\omega meson, ω→π0Îł\omega\to\pi^0\gamma. The cross sections cover the full angular range and show the full extent of the tt-channel forward rise. The overall shape of the angular distributions in the differential cross sections and unpolarized spin-density matrix elements are in fair agreement with previous data. In addition, for the first time, a beam of linearly-polarized tagged photons in the energy range from 1150 MeV to 1650 MeV was used to extract polarized spin-density matrix elements. These data were included in the Bonn-Gatchina partial wave analysis (PWA). The dominant contribution to ω\omega photoproduction near threshold was found to be the 3/2+3/2^+ partial wave, which is primarily due to the sub-threshold N(1720) 3/2+N(1720)\,3/2^+ resonance. At higher energies, pomeron-exchange was found to dominate whereas π\pi-exchange remained small. These tt-channel contributions as well as further contributions from nucleon resonances were necessary to describe the entire dataset: the 1/2−1/2^-, 3/2−3/2^-, and 5/2+5/2^+ partial waves were also found to contribute significantly.Comment: 8 pages, 5 figures, accepted for publication in Physics Letters

    In-medium ω\omega mass from the Îł+Nb→π0Îł+X\gamma + Nb \to \pi^{0}\gamma + X reaction

    Data on the photoproduction of ω\omega mesons on nuclei have been re-analyzed in a search for in-medium modifications. The data were taken with the Crystal Barrel(CB)/TAPS detector system at the ELSA accelerator facility in Bonn. First results from the analysis of the data set were published by D. Trnka et al. in Phys. Rev. Lett 94 (2005) 192303 \cite{david}, claiming a lowering of the ω\omega mass in the nuclear medium by 14% at normal nuclear matter density. The extracted ω\omega line shape was found to be sensitive to the background subtraction. For this reason a re-analysis of the same data set has been initiated and a new method has been developed to reduce the background and to determine the shape and absolute magnitude of the background directly from the data. Details of the re-analysis and of the background determination are described. The ω\omega signal on the NbNb target, extracted in the re-analysis, does not show a deviation from the corresponding line shape on a LH2LH_2 target, measured as reference. The earlier claim of an in-medium mass shift is thus not confirmed. The sensitivity of the ω\omega line shape to different in-medium modification scenarios is discussed.Comment: 13 pages and 11 figures, submitted for publicatio

    Modification of the ω\omega-Meson Lifetime in Nuclear Matter

    The photo production of ω\omega mesons on the nuclei C, Ca, Nb and Pb has been measured using the Crystal Barrel/TAPS detector at the ELSA tagged photon facility in Bonn. The dependence of the ω\omega meson cross section on the nuclear mass number has been compared with three different types of models, a Glauber analysis, a BUU analysis of the Giessen theory group and a calculation by the Valencia theory group. In all three cases, the inelastic ω\omega width is found to be 130−150MeV/c2130-150 \rm{MeV/c^2} at normal nuclear matter density for an average 3-momentum of 1.1 GeV/c. In the restframe of the ω\omega meson, this inelastic ω\omega width corresponds to a reduction of the ω\omega lifetime by a factor ≈30\approx 30. For the first time, the momentum dependent ω\omegaN cross section has been extracted from the experiment and is in the range of 70 mb.Comment: 5 pages, 4 figure
