    Classification of Singular Fibres on Rational Elliptic Surfaces in Characteristic Three

    Eyes In The Sky: Linking Satellite Oceanography And Biotelemetry To Explore Habitat Selection By Basking Sharks

    Background: Satellite-based oceanographic data products are a valuable source of information on potential resource availability for marine species. Satellite oceanography data may be particularly useful in biotelemetry studies on marine species that feed at low trophic levels, such as zooplanktivorous whales, sharks, and rays. The basking shark, Cetorhinus maximus, is a well-documented zooplanktivore in the western North Atlantic, yet little is known of its movements and spatial ecology in this region. A combination of satellite tag technologies were used to describe basking shark movements with respect to concurrent satellite-observed oceanographic conditions in order to test for selection of these environmental variables. Results: Satellite-linked ‘smart’ position only transmitting tags (SPOTs, N = 10) were used to assess horizontal movements, activity space, and habitat selection, while pop-up satellite archival tags (PSATs, N = 7) were used to describe depth preferences of basking sharks during summer and fall. The duration of SPOT tracks ranged from 5 to 45 days. Basking sharks used relatively small activity spaces in three focal areas off Massachusetts: Vineyard Sound, the Great South Channel, and Cape Cod Bay. These sharks appeared to select areas with shallow bottom depths, high primary production and chlorophyll concentrations, and steep surface gradients, but significant selection for these variables was only detected between mid-August and mid-October when the sharks were primarily located in Cape Cod Bay. Conclusions: Basking sharks in the southern Gulf of Maine during summer and fall focus their activities in discrete areas likely to support high primary and secondary productivity. Habitat selection may also be influenced by mating and social activity at times, but further research is needed to differentiate these behaviors from foraging activity. Satellite-based biotelemetry and oceanography are powerful tools that together can provide valuable new insights into habitat selection patterns of highly mobile marine species

    Re-evaluation of the Fijianolide/Laulimalide Chemotype Suggests an Alternate Mechanism of Action for C-15/C-20 Analogs.

    Herein, we report on naturally derived microtubule stabilizers with activity against triple negative breast cancer (TNBC) cell lines, including paclitaxel, fijianolide B/laulimalide (3), fijianolide B di-acetate (4), and two new semisynthetic analogs of 3, which include fijianolide J (5) and fijianolide L (6). Similar to paclitaxel, compound 3 demonstrated classic microtubule stabilizing activity with potent (GI50 = 0.7–17 nM) antiproliferative efficacy among the five molecularly distinct TNBC cell lines. Alternatively, compounds 5 or 6, generated from oxidation of C-20 or C-15 and C-20 respectively, resulted in a unique profile with reduced potency (GI50 = 4–9 μM), but improved efficacy in some lines, suggesting a distinct mechanism of action. The C-15, C-20 di-acetate, and dioxo modifications on 4 and 6 resulted in compounds devoid of classic microtubule stabilizing activity in biochemical assays. While 4 also had no detectable effect on cellular microtubules, 6 promoted a reorganization of the cytoskeleton resulting in an accumulation of microtubules at the cell periphery. Compound 5, with a single C-20 oxo substitution, displayed a mixed phenotype, sharing properties of 3 and 6. These results demonstrate the importance of the C-15/C-20 chiral centers, which appear to be required for the potent microtubule stabilizing activity of this chemotype and that oxidation of these sites promotes unanticipated cytoskeletal alterations that are distinct from classic microtubule stabilization, likely through a distinct mechanism of action

    Connecting urban food plans to the countryside: leveraging Denver's food vision to explore meaningful rural-urban linkages

    Includes bibliographical references (pages 14-18).Cities are increasingly turning to food policy plans to support goals related to food access, food security, the environment, and economic development. This paper investigates ways that rural farmers, communities, and economies can both support and be supported by metropolitan food-focused initiatives. Specifically, our research question asked what opportunities and barriers exist to developing food policies that support urban food goals, particularly related to local procurement, as well as rural economic development. To address this question, we described and analyzed a meeting of urban stakeholders and larger-scale rural producers related to Colorado’s Denver Food Vision and Plan. We documented and explored “findings” gleaned from a supply chain diagraming and data compilation process that were then used to inform an event that brought together diverse supply chain partners. Three findings stand out. First, facilitating dialog between urban food policymakers and rural producers to understand potential tensions, mitigate such tensions, and capitalize on opportunities is essential. Second, perceptions and expectations surrounding “good food” are nuanced—a timely finding given the number of preferred procurement programs emerging across the county. Third, critical evaluation is needed across a diverse set of value chain strategies (e.g., conventional and alternative distribution) if food policy intends to support heterogeneous producers, their communities, and urban food policy goals

    Proteomic, biomechanical and functional analyses define neutrophil heterogeneity in systemic lupus erythematosus

Neutrophil trafficking and immune function are intimately linked to cellular biophysical properties. This study used proteomic, biomechanical and functional analyses to further define neutrophil heterogeneity in the context of SLE. Methods: Proteomic/phosphoproteomic analyses were performed in healthy control (HC) normal density neutrophils (NDNs), SLE NDNs and autologous SLE LDGs. The biophysical properties of these neutrophil subsets were analysed by real-time deformability cytometry and lattice light-sheet microscopy. A two-dimensional endothelial flow system and a three-dimensional microfluidic microvasculature mimetic (MMM) were used to decouple the contributions of cell surface mediators and biophysical properties to neutrophil trafficking, respectively. Results: Proteomic and phosphoproteomic differences were detected between HC and SLE neutrophils and between SLE NDNs and LDGs. Increased abundance of type 1 interferon-regulated proteins and differential phosphorylation of proteins associated with cytoskeletal organisation were identified in SLE LDGs relative to SLE NDNs. The cell surface of SLE LDGs was rougher than in SLE and HC NDNs, suggesting membrane perturbances. While SLE LDGs did not display increased binding to endothelial cells in the two-dimensional assay, they were increasingly retained/trapped in the narrow channels of the lung MMM. Conclusions: Modulation of the neutrophil proteome and distinct changes in biophysical properties are observed alongside differences in neutrophil trafficking. SLE LDGs may be increasingly retained in microvasculature networks, which has important pathogenic implications in the context of lupus organ damage and small vessel vasculopathy
