Increased functional connectivity patterns in mild Alzheimer’s disease: a rsfMRI study
Background: Alzheimer’s disease (AD) is the most common age-related neurodegenerative disorder. In view of our rapidly aging population, there is an urgent need to identify Alzheimer’s disease (AD) at an early stage. A potential way to do so is by assessing the functional connectivity (FC), i.e., the statistical dependency between two or more brain regions, through novel analysis techniques. Methods: In the present study, we assessed the static and dynamic FC using different approaches. A resting state (rs)fMRI dataset from the Alzheimer’s disease neuroimaging initiative (ADNI) was used (n = 128). The blood-oxygen-level-dependent (BOLD) signals from 116 regions of 4 groups of participants, i.e., healthy controls (HC; n = 35), early mild cognitive impairment (EMCI; n = 29), late mild cognitive impairment (LMCI; n = 30), and Alzheimer’s disease (AD; n = 34) were extracted and analyzed. FC and dynamic FC were extracted using Pearson’s correlation, sliding-windows correlation analysis (SWA), and the point process analysis (PPA). Additionally, graph theory measures to explore network segregation and integration were computed. Results: Our results showed a longer characteristic path length and a decreased degree of EMCI in comparison to the other groups. Additionally, an increased FC in several regions in LMCI and AD in contrast to HC and EMCI was detected. These results suggest a maladaptive short-term mechanism to maintain cognition. Conclusion: The increased pattern of FC in several regions in LMCI and AD is observable in all the analyses; however, the PPA enabled us to reduce the computational demands and offered new specific dynamic FC findings.info:eu-repo/semantics/publishedVersio

Empathy at the heart of darkness: empathy deficits that bind the dark triad and those that mediate indirect relational aggression
The dark triad (DT) traits–psychopathy, narcissism and Machiavellianism–have collectively been linked to reduced empathy and increased aggression; however, their association with distinct empathic subtypes remains unclear; and unique links to indirect relational aggression (IRA) have not been delineated. Moreover, whether dark traits should be conceptualized individually, as a dyad or as a triad with a dark core centered around the absence of empathy is debated. The current study examines (i) whether impaired empathy indeed represents a common “dark core” binding Machiavellianism, narcissism, and psychopathy, and (ii) this core explains associations between the dark traits and IRA. Participants (N = 301, 262 F/39 M) completed measures of the DT traits, cognitive and affective empathy components and IRA (Social Exclusion, Malicious Humor and Guilt Induction). The individual traits model without links between narcissism and IRA showed the best fit, suggesting that, at least in the context of IRA, the DT traits are best viewed as three independent personality traits. Distinct cognitive and affective empathy deficits and capacities are seen in the DT. Peripheral responsivity was the only type of empathy deficit associated with all dark traits, but unrelated to IRA. Psychopathy was the strongest indicator of impaired empathy and all IRAs; however, only online simulation, an affect-related cognitive empathy facet, partially mediated the relationships of psychopathy and Machiavellianism with IRA. Whilst the unique pathways for the dark triad traits suggest stronger alignment of psychopathy and Machiavellianism in their empathic deficits and indirect aggression; the data do not support the notion that an unempathic dark core underpinning all three traits drives indirect aggression. This is the first paper delineating the specific empathic deficits involved using a facet approach and their link to indirect forms of aggression. Results therefore inform theoretical models of aggression in the DT and offer some clarity on the debates surrounding the unempathic dark core in the DT

Cognitive effect following a blended (face to face and videoconference-delivered) format mindfulness training
While evidence supports the feasibility of online mindfulness training (MT), the effect of this approach on cognition remains unclear. The present study investigated changes in cognition following a newly developed 6-week videoconference-delivered MT program on cognitive function in two groups. The first group (n = 17) had two baseline assessments prior to MT [3 weeks after group two (n = 15)] to allow for evaluation of practice and learning effects. Four participants from each group were excluded from the final analysis due to missing data. Following MT, there was an improvement in switching of attention, working memory, executive function, and social cognition, but some of these effects were not easily accounted for by learning or practice effects. No significant changes were found on tasks measuring sustained attention, cognitive flexibility and inhibition, information processing, and sensory-motor function. Our findings suggest that domain-specific cognition might be enhanced by a brief videoconference-delivered MT, and larger, controlled studies to delineate the effects of online MT on subdomains of cognition are needed
The Development and Validation of the Successful Psychopathy Scale
The personality construct known as ‘Successful Psychopathy’ has attracted the
interests of researchers and clinicians alike. The concept suggests an individual
who demonstrates the core traits associated with psychopathy but is able to adapt
and function within society to prototypical or superior standards. There has yet to
be a sound theoretical model of this construct by which to base a psychometric
measure. This protocol presents the ethical procedure that will endeavour to create
such a measure and validate it within general population samples

Spatiotemporal EEG Dynamics of Prospective Memory in Ageing and Mild Cognitive Impairment
Prospective memory (PM, the memory of future intentions) is one of the first complaints of those that develop dementia-related disease. Little is known about the neurophysiology of PM in ageing and those with mild cognitive impairment (MCI). By using a novel artificial neural network to investigate the spatial and temporal features of PM related brain activity, new insights can be uncovered. Young adults (n = 30), healthy older adults (n = 39) and older adults with MCI (n = 27) completed a working memory and two PM (perceptual, conceptual) tasks. Time-locked electroencephalographic potentials (ERPs) from 128-electrodes were analysed using a brain-inspired spiking neural network (SNN) architecture. Local and global connectivity from the SNNs was then evaluated. SNNs outperformed other machine learning methods in classification of brain activity between younger, older and older adults with MCI. SNNs trained using PM related brain activity had better classification accuracy than working memory related brain activity. In general, younger adults exhibited greater local cluster connectivity compared to both older adult groups. Older adults with MCI demonstrated decreased global connectivity in response to working memory and perceptual PM tasks but increased connectivity in the conceptual PM models relative to younger and healthy older adults. SNNs can provide a useful method for differentiating between those with and without MCI. Using brain activity related to PM in combination with SNNs may provide a sensitive biomarker for detecting cognitive decline. Cognitively demanding tasks may increase the amount connectivity in older adults with MCI as a means of compensation
Neural changes following cognitive behaviour therapy for psychosis: a longitudinal study
A growing body of evidence demonstrates that persistent positive symptoms, particularly delusions, can be improved by cognitive behaviour therapy for psychosis. Heightened perception and processing of threat are believed to constitute the genesis of delusions. The present study aimed to examine functional brain changes following cognitive behaviour therapy for psychosis. The study involved 56 outpatients with one or more persistent positive distressing symptoms of schizophrenia. Twenty-eight patients receiving cognitive behaviour therapy for psychosis for 6–8 months in addition to their usual treatment were matched with 28 patients receiving treatment as usual. Patients’ symptoms were assessed by a rater blind to treatment group, and they underwent functional magnetic resonance imaging during an affect processing task at baseline and end of treatment follow-up. The two groups were comparable at baseline in terms of clinical and demographic parameters and neural and behavioural responses to facial and control stimuli. The cognitive behaviour therapy for psychosis with treatment-as-usual group (22 subjects) showed significant clinical improvement compared with the treatment-as-usual group (16 subjects), which showed no change at follow-up. The cognitive behaviour therapy for psychosis with treatment-as-usual group, but not the treatment-as-usual group, showed decreased activation of the inferior frontal, insula, thalamus, putamen and occipital areas to fearful and angry expressions at treatment follow-up compared with baseline. Reduction of functional magnetic resonance imaging response during angry expressions correlated directly with symptom improvement. This study provides the first evidence that cognitive behaviour therapy for psychosis attenuates brain responses to threatening stimuli and suggests that cognitive behaviour therapy for psychosis may mediate symptom reduction by promoting processing of threats in a less distressing way
