The kind of this research is quantitative research. This research use correlation methode and submited the data use questionnaire and test.     Gene expression of bacterial collagenolytic proteases in root caries

    Objective: It is unknown whether bacteria play a role in the collagen matrix degradation that occurs during caries progression. Our aim was to characterize the expression level of genes involved in bacterial collagenolytic proteases in root biofilms with and without caries. Method: we collected samples from active cavitated root caries lesions (RC, n = 30) and from sound root surfaces (SRS, n = 10). Total microbial RNA was isolated and cDNA sequenced on the Illumina Hi-Seq2500. Reads were mapped to 162 oral bacterial reference genomes. Genes encoding putative bacterial collagenolytic proteases were identified. Normalization and differential expression analysis was performed on all metatranscriptomes (FDR8) but none in SRS were Pseudoramibacter alactolyticus [HMPREF0721_RS02020; HMPREF0721_RS04640], Scardovia inopinata [SCIP_RS02440] and Olsenella uli DSM7084 [OLSU_RS02990]. Conclusion: Our findings suggest that the U32 proteases may be related to carious dentine. The contribution of a small number of species to dentine degradation should be further investigated. These proteases may have potential in future biotechnological and medical applications, serving as targets for the development of therapeutic agents

    Functional Remineralization of Dentin Lesions Using Polymer-Induced Liquid-Precursor Process

    It was hypothesized that applying the polymer-induced liquid-precursor (PILP) system to artificial lesions would result in time-dependent functional remineralization of carious dentin lesions that restores the mechanical properties of demineralized dentin matrix. 140 µm deep artificial caries lesions were remineralized via the PILP process for 7–28 days at 37°C to determine temporal remineralization characteristics. Poly-L-aspartic acid (27 KDa) was used as the polymeric process-directing agent and was added to the remineralization solution at a calcium-to-phosphate ratio of 2.14 (mol/mol). Nanomechanical properties of hydrated artificial lesions had a low reduced elastic modulus (ER = 0.2 GPa) region extending about 70 μm into the lesion, with a sloped region to about 140 μm where values reached normal dentin (18–20 GPa). After 7 days specimens recovered mechanical properties in the sloped region by 51% compared to the artificial lesion. Between 7–14 days, recovery of the outer portion of the lesion continued to a level of about 10 GPa with 74% improvement. 28 days of PILP mineralization resulted in 91% improvement of ER compared to the artificial lesion. These differences were statistically significant as determined from change-point diagrams. Mineral profiles determined by micro x-ray computed tomography were shallower than those determined by nanoindentation, and showed similar changes over time, but full mineral recovery occurred after 14 days in both the outer and sloped portions of the lesion. Scanning electron microscopy and energy dispersive x-ray analysis showed similar morphologies that were distinct from normal dentin with a clear line of demarcation between the outer and sloped portions of the lesion. Transmission electron microscopy and selected area electron diffraction showed that the starting lesions contained some residual mineral in the outer portions, which exhibited poor crystallinity. During remineralization, intrafibrillar mineral increased and crystallinity improved with intrafibrillar mineral exhibiting the orientation found in normal dentin or bone

    Review of Matrix Metalloproteinases’ Effect on the Hybrid Dentin Bond Layer Stability and Chlorhexidine Clinical Use to Prevent Bond Failure

    This review describes the relationship between dentin collagen hybrid bond layer degradation and the Matrix Metalloproteinases (MMPs) after their release by acid etch and rinse adhesives and self etching bonding adhesives that can reduce the bond stability over time. MMP-2, MMP-8 and MMP-9 are indicated as the active proteases that breakdown the collagen fibrils in the hybrid bond layer. Phosphoric acid in the acid etch and rinse bonding process and acid primers in the self etch process are implicated in the release of these proteases and their activation by several non-collagen proteins also released from dentin by the etching. MMPs are released in saliva by salivary glands, by cells in the gingival crevices to crevicular fluid and by pulpal odontoblasts cells to the dentinal fluids. These sources may affect the hybrid layer also. Evidence of the bond strength deterioration over time and the ability of Chlorhexidine to prevent bond deterioration by inhibiting MMP action are discussed. Dentin Bonding procedure utilizing Chlorhexidine for different application times and concentrations are being developed. The application of 2% Chlorhexidine to the phosphoric acid etch surface after rinsing off the acid is the only procedure that has been clinically tested for a longer period of time and shown to prevent bond strength degradation so far. The adoption of this procedure is recommended as means of improving bond stability at this time

    Matrix metalloproteinases (MMPs) in the dentin-pulp complex of healthy and carious teeth

    Abstract The dentin-pulp complex comprises mineralized dentin and the vital soft tissues encased inside dentin, i.e. odontoblasts and pulp tissue. During caries progression, the dentinal minerals are dissolved and eventually the collagenous organic matrix is degraded. However, the exact mechanisms and enzymes responsible for the organic matrix breakdown remain unknown. Matrix metalloproteinases (MMPs), a family of endopeptidases capable of degrading in concert virtually all extracellular matrix components, are expressed during normal dentin-pulp complex formation and maintenance. MMP activity has also been suggested to contribute to the organic matrix degradation during dentin caries progression and to the repair and defense reactions elicited by caries in the dentin-pulp complex cells. The aim of the study was to further elucidate the role of host MMPs in dentin caries progression and the origin of MMPs in carious dentin as well as the possible changes in MMP expression in the cells of the dentin-pulp complex in response to caries. MMP inhibitors decreased the area of dentin caries lesions in vivo, suggesting the involvement of host MMPs in dentin caries pathogenesis. When the overall MMP gene expression was examined by cDNA microarray, pooled pulp samples demonstrated a high level of MMP-13 expression, but failed to show any unequivocal changes in MMP expression due to caries. MMP-13 expression is rare among normal human adult tissues. Real-time quantitative PCR of individual pulp and odontoblast samples demonstrated a rather large variation in relative MMP-13 mRNA expression between samples, especially pulp samples. Protein expression of MMP-13 was detected in pulp and odontoblasts without any major differences between the tissues of sound and carious teeth. This was also the case with the MMP-20 (enamelysin) protein, which was demonstrated in odontoblasts and the pulp tissue of fully developed human teeth. MMP-20, MMP-8, and gelatinases (especially MMP-2) were demonstrated in human dentin, and dentinal MMPs exhibited activity against native and denatured type I collagen when released. The study demonstrates the presence of MMPs in the soft and hard tissue compartments of the dentin-pulp complex. These enzymes may also contribute to dentin caries progression and response reactions to caries

The articles in this publication are based on pro gradu theses on Finnish as a second and foreign language (S2). They focus on the communication strategies used by the learners of Finnish, the teaching and learning of spoken Finnish, the learning of the structure and lexicon of Finnish, and the order of teaching in textbooks. The articles have been reviewed by experts in the field, and edited and improved based on their recommendations. The last section of the book contains a historical review of the development of Finnish as a second and foreign language as a subject in the university of Oulu and a list of the S2 student assignments
