Marketing Website and Extranet Site
This is a hybrid project. As a new Information Systems consultancy the need for a website to market to potential clients was evident. The decision to add an extranet site was made after specific needs for Ace Hardware were made evident to Yar’s founders. Ace Hardware Corporation is a cooperative with over 4,400 members in over 100 nations worldwide. Ace corporate rolled out a B2B pilot program in the fall of 2013 to help Ace independently owned retail stores learn how to sell to other businesses. One of YAR’s founders is currently working with Ace corporate as a program consultant for the B2B group along with four other consultants nationally. The stores have focused on the homeowner and Do It Yourself (DIY) customers for almost 90 years and see the potential in Business to Business sales. However, the stores are not prepared and need training, tools, processes, alignment, and counsel to be successful.
Ace corporate provides much of the IS infrastructure to the stores through Epicor’s ERP suite and other programs. In order for the stores to be successful they need to become competent in account management in sales and customer relationship management and a key tool for that is a Customer Relationship Management (CRM) tool or package. SalesForce and Microsoft’s Dynamics CRM are two of the most popular today. Epicor does offer CRM capabilities in their ERP suite but that is not available to the retail stores and would be extremely cost prohibitive if it was. Before the stores commit to larger and more robust CRM offerings they need a basic tool to help them learn and get the process moving.
The extranet was initiated to provide a simple, cloud based CRM tool that the stores could utilize as they begin to build the B2B segment in their stores. Because it is cloud based the stores can all participate with the same tool instead of incurring the cost of having to individually find and purchase off the shelf CRM software or have it custom built by a local developer. They will have to purchase cloud storage, administration fees, and maintenance costs but all those will be a fraction of buying Epicor’s offering and much less than off the shelf or local developer options.
The stores from the Houston retailer group that are participating are encouraged at the prospect of finally having a CRM solution even if it is in trial stages and are beginning to engage their B2B program more assertively. As the extranet is accepted integration with Epicor will have to occur at a later date in order to feed the CRM tool with the stores POS data currently being gathered so as to prevent double entry labor

Structural basis of HIV-1 Vpu-mediated BST2 antagonism via hijacking of the clathrin adaptor protein complex 1.
BST2/tetherin, an antiviral restriction factor, inhibits the release of enveloped viruses from the cell surface. Human immunodeficiency virus-1 (HIV-1) antagonizes BST2 through viral protein u (Vpu), which downregulates BST2 from the cell surface. We report the crystal structure of a protein complex containing Vpu and BST2 cytoplasmic domains and the core of the clathrin adaptor protein complex 1 (AP1). This, together with our biochemical and functional validations, reveals how Vpu hijacks the AP1-dependent membrane trafficking pathways to mistraffick BST2. Vpu mimics a canonical acidic dileucine-sorting motif to bind AP1 in the cytosol, while simultaneously interacting with BST2 in the membrane. These interactions enable Vpu to build on an intrinsic interaction between BST2 and AP1, presumably causing the observed retention of BST2 in juxtanuclear endosomes and stimulating its degradation in lysosomes. The ability of Vpu to hijack AP-dependent trafficking pathways suggests a potential common theme for Vpu-mediated downregulation of host proteins.DOI: http://dx.doi.org/10.7554/eLife.02362.001
Behavioral Outcomes of Home-Based Services for Children and Adolescents with Serious Emotional Disorders
The current study evaluates the effectiveness of an intensive home-based treatment program, Families First, on the behaviors of children and adolescents suffering from mental disorders and being at risk for out-ofi home placement. The sample included 85 youngsters and their families from a semi-rural community. The Diagnostic Interview for Children and Adolescents-Revised (DICA-R) was administered to the children, and the Child Behavior Checklist (CBCL) was completed by a parent at pretreatment and posttreatment. The families participated in a 4-6 week, intensive home intervention where crisis intervention, social support services, and needed psychological services were offered. The results indicated that both externalizing and internalizing behavior problems in youngsters with different diagnoses of mental disorders were significantly reduced at posttreatment as indicated by their CBCL scores. Furthermore, youngsters with a diagnosis of Oppositional Defiant Disorder seemed to benefit the most, as evidenced by the improved scores on most subscales of the CBCL. Youngsters with mood disorders and conduct disorders seemed to benefit in their most deficient areas, internalizing behavior problems and delinquent behaviors, respectively. Theoretical–Experimental Study of the Action of Trace Amounts of Formaldehyde, Propionaldehyde, and Butyraldehyde as Inhibitors of the Ziegler–Natta Catalyst and the Synthesis of an Ethylene–Propylene Copolymer
The copolymer synthesis process can be affected by failures in the production process or by contaminating compounds such as ketones, thiols, and gases, among others. These impurities act as an inhibiting agent of the Ziegler–Natta (ZN) catalyst affecting its productivity and disturbing the polymerization reaction. In this work, the effect of formaldehyde, propionaldehyde,and butyraldehyde on the ZN catalyst and the way in which it affects the final properties of the ethylene-propylene copolymer is presented by analyzing 30 samples with different concentrations of the mentioned aldehydes along with three control samples. It was determined that the presence of formaldehyde 26 ppm, propionaldehyde 65.2 ppm, and butyraldehyde 181.2 ppm considerably affect the productivity levels of the ZN catalyst; this effect increases as the concentration of aldehydes is higher in the process; likewise, these impurities affect the properties of the final product, such as the fluidity index (MFI), thermogravimetric analysis (TGA), bending, tension, and impact, which leads to a polymer with low-quality standards and less resistance to breakage. The computational analysis showed that the complexes formed by formaldehyde, propionaldehyde, and butyraldehyde with the active center of the catalyst are more stable than those obtained by the ethylene-Ti and propylene-Ti complexes, presenting values of −40.5, −47.22, −47.5, −5.2 and −1.3 kcal mol−1 respectively
Stratospheric ozone change and related climate impacts over 1850-2100 as modelled by the ACCMIP ensemble
Stratospheric ozone and associated climate impacts in the Atmospheric Chemistry and Climate Model Intercomparison Project (ACCMIP) simulations are evaluated in the recent past (1980–2000), and examined in the long-term (1850–2100) using the Representative Concentration Pathways (RCPs) low- and high-emission scenarios (RCP2.6 and RCP8.5, respectively) for the period 2000–2100. ACCMIP multi-model mean total column ozone (TCO) trends compare favourably, within uncertainty estimates, against observations. Particularly good agreement is seen in the Antarctic austral spring (−11.9 % dec−1 compared to observed  ∼  −13.9 ± 10.4 % dec−1), although larger deviations are found in the Arctic's boreal spring (−2.1 % dec−1 compared to observed  ∼  −5.3 ± 3.3 % dec−1). The simulated ozone hole has cooled the lower stratosphere during austral spring in the last few decades (−2.2 K dec−1). This cooling results in Southern Hemisphere summertime tropospheric circulation changes captured by an increase in the Southern Annular Mode (SAM) index (1.3 hPa dec−1). In the future, the interplay between the ozone hole recovery and greenhouse gases (GHGs) concentrations may result in the SAM index returning to pre-ozone hole levels or even with a more positive phase from around the second half of the century (−0.4 and 0.3 hPa dec−1 for the RCP2.6 and RCP8.5, respectively). By 2100, stratospheric ozone sensitivity to GHG concentrations is greatest in the Arctic and Northern Hemisphere midlatitudes (37.7 and 16.1 DU difference between the RCP2.6 and RCP8.5, respectively), and smallest over the tropics and Antarctica continent (2.5 and 8.1 DU respectively). Future TCO changes in the tropics are mainly determined by the upper stratospheric ozone sensitivity to GHG concentrations, due to a large compensation between tropospheric and lower stratospheric column ozone changes in the two RCP scenarios. These results demonstrate how changes in stratospheric ozone are tightly linked to climate and show the benefit of including the processes interactively in climate models
Cross-stiffened continuous fiber structures
Under NASA's Novel Composites for Wing and Fuselage Applications (NCWFA) program, Contract NAS1-18784, Grumman is evaluating the structural efficiency of graphite/epoxy cross-stiffened panel elements fabricated using innovative textile preforms and cost effective Resin Transfer Molding (RTM) and Resin Film Infusion (RFI) processes. Two three-dimensional woven preform assembly concepts have been defined for application to a representative window belt design typically found in a commercial transport airframe. The 3D woven architecture for each of these concepts is different; one is vertically woven in the plane of the window belt geometry and the other is loom woven in a compressed state similar to an unfolded eggcrate. The feasibility of both designs has been demonstrated in the fabrication of small test element assemblies. These elements and the final window belt assemblies will be structurally tested, and results compared
Regional perturbation of gene transcription is associated with intrachromosomal rearrangements and gene fusion transcripts in high grade ovarian cancer.
Genomic rearrangements are a hallmark of cancer biology and progression, allowing cells to rapidly transform through alterations in regulatory structures, changes in expression patterns, reprogramming of signaling pathways, and creation of novel transcripts via gene fusion events. Though functional gene fusions encoding oncogenic proteins are the most dramatic outcomes of genomic rearrangements, we investigated the relationship between rearrangements evidenced by fusion transcripts and local expression changes in cancer using transcriptome data alone. 9,953 gene fusion predictions from 418 primary serious ovarian cancer tumors were analyzed, identifying depletions of gene fusion breakpoints within coding regions of fused genes as well as an N-terminal enrichment of breakpoints within fused genes. We identified 48 genes with significant fusion-associated upregulation and furthermore demonstrate that significant regional overexpression of intact genes in patient transcriptomes occurs within 1 megabase of 78 novel gene fusions that function as central markers of these regions. We reveal that cancer transcriptomes select for gene fusions that preserve protein and protein domain coding potential. The association of gene fusion transcripts with neighboring gene overexpression supports rearrangements as mechanism through which cancer cells remodel their transcriptomes and identifies a new way to utilize gene fusions as indicators of regional expression changes in diseased cells with only transcriptomic data
