    It is shown that for the Calogero-Cohn type upper bounds on the number of bound states of a negative spherically symmetric potential V(r)V(r), in each angular momentum state, that is, bounds containing only the integral ∫0∞∣V(r)∣1/2dr\int^\infty_0 |V(r)|^{1/2}dr, the condition Vâ€Č(r)≄0V'(r) \geq 0 is not necessary, and can be replaced by the less stringent condition (d/dr)[r1−2p(−V)1−p]≀0,1/2≀p<1(d/dr)[r^{1-2p}(-V)^{1-p}] \leq 0, 1/2 \leq p < 1, which allows oscillations in the potential.     Detection of water molecules on the radical transfer pathway of ribonucleotide reductase by 17O electron–nuclear double resonance spectroscopy

    The role of water in biological proton-coupled electron transfer (PCET) is emerging as a key for understanding mechanistic details at atomic resolution. Here we demonstrate 17O high-frequency electron–nuclear double resonance (ENDOR) in conjunction with H217O-labeled protein buffer to establish the presence of ordered water molecules at three radical intermediates in an active enzyme complex, the α2ÎČ2E. coli ribonucleotide reductase. Our data give unambiguous evidence that all three, individually trapped, intermediates are hyperfine coupled to one water molecule with Tyr-O···17O distances in the range 2.8–3.1 Å. The availability of this structural information will allow for quantitative models of PCET in this prototype enzyme.     ac-field-induced fluid pumping in microsystems with asymmetric temperature gradients

    We present two different designs of electrohydrodynamic micropumps for microfluidic systems. The micropumps have no movable parts, and their simple design allows for fabrication by microsystems technology. The pumps are operated by ac voltages from 1to60V and were tested with aqueous solutions in the conductivity range of 1–112mSm−1. The pump effect is induced by an ac electric field across a fluid medium with an inhomogeneous temperature distribution. It is constant over a wide range of the ac field frequency with a conductivity-dependent drop-off at high frequencies. The temperature-dependent conductivity and permittivity distributions in the fluid induce space charges that interact with the electric field and induce fluid motion. The temperature distribution can be generated either by Joule heating in the medium or by external heating. We present experimental results obtained with two prototypes featuring Joule heating and external heating by a heating filament.     Growth and Localization of Polyhydroxybutyrate Granules in Ralstonia eutropha

    The bacterium Ralstonia eutropha forms cytoplasmic granules of polyhydroxybutyrate that are a source of biodegradable thermoplastic. While much is known about the biochemistry of polyhydroxybutyrate production, the cell biology of granule formation and growth remains unclear. Previous studies have suggested that granules form either in the inner membrane, on a central scaffold, or in the cytoplasm. Here we used electron cryotomography to monitor granule genesis and development in 3 dimensions (3-D) in a near-native, “frozen-hydrated” state in intact Ralstonia eutropha cells. Neither nascent granules within the cell membrane nor scaffolds were seen. Instead, granules of all sizes resided toward the center of the cytoplasm along the length of the cell and exhibited a discontinuous surface layer more consistent with a partial protein coating than either a lipid mono- or bilayer. Putatively fusing granules were also seen, suggesting that small granules are continually generated and then grow and merge. Together, these observations support a model of biogenesis wherein granules form in the cytoplasm coated not by phospholipid but by protein. Previous thin-section electron microscopy (EM), fluorescence microscopy, and atomic force microscopy (AFM) results to the contrary may reflect both differences in nucleoid condensation and specimen preparation-induced artifacts

    A Hilbert space HH induces a formal context, the Hilbert formal context H‟\overline H, whose associated concept lattice is isomorphic to the lattice of closed subspaces of HH. This set of closed subspaces, denoted C(H)\mathcal C(H), is important in the development of quantum logic and, as an algebraic structure, corresponds to a so-called ``propositional system'', that is, a complete, atomistic, orthomodular lattice which satisfies the covering law.     A Ferredoxin Disulfide Reductase Delivers Electrons to the to the Methanosarcina barkeri Class III Ribonucleotide Reductase

    Two subtypes of class III anaerobic ribonucleotide reductases (RNRs) studied so far couple the reduction of ribonucleotides to the oxidation of formate, or the oxidation of NADPH via thioredoxin and thioredoxin reductase. Certain methanogenic archaea contain a phylogenetically distinct third subtype of class III RNR, with distinct active-site residues. Here we report the cloning and recombinant expression of the Methanosarcina barkeri class III RNR and show that the electrons required for ribonucleotide reduction can be delivered by a [4Fe-4S] protein ferredoxin disulfide reductase, and a conserved thioredoxin-like protein NrdH present in the RNR operon. The diversity of class III RNRs reflects the diversity of electron carriers used in anaerobic metabolism.Singapore.
