    From frequent scrum photos of the president holding up signed executive orders in the Oval Office to images of the president energetically sitting behind the wheel of a Mack truck parked outside his back door, like previous administrations, visuals have been central to Donald Trump\u27s presidency. This chapter analyzes the visuals on White House social media accounts (i.e., Twitter and Facebook) in Trump\u27s first 100 days office and explores how his administration used visuals as an essential vehicle for storytelling, image building, and persuading. Building on previous research on the communicative functions of visual symbols in politics, the chapter finds that the Trump administration used socially-mediated images to build an image based on success, power, and leadership and to legitimize his new administration

    Assessing Stakeholder Preferences for Chesapeake Bay Restoration Options : a stated preference discrete choice-based assessment

    Chesapeake 2000 or C2K is a multi-jurisdictional agreement between the states of Virginia, Maryland, Pennsylvania, the District of Columbia, the Chesapeake Bay Commission and the U.S. Environmental Protection Agency, representing the federal government, to restore the health of the Chesapeake Bay’s ecosystem. This agreement commits the participants to achieve five major restoration goals, 22 sub-objectives or categories, and 102 specific commitments or restoration activities. The five major goals are the following: (1) restore and protect natural living resources; (2) restore and protect vital habitat; (3) restore and protect water quality; (4) promote sound land use; and (5) promote stewardship and community engagement. The sub-categories and specific commitments impose specific restoration requirements relative to each of the five major categories. In 2003, the Chesapeake Bay Commission, utilizing a panel of experts, estimated the cost of achieving all five major objectives equaled approximately 18.7billion,whichequalsapproximately18.7 billion, which equals approximately 21.0 billion in 2007 dollars. Unfortunately, all partners of C2K only committed 5.9billion(5.9 billion (6.6 billion in 2007 dollars) in funding to achieving the five major objectives. There is, thus, a deficit of 12.8billionor12.8 billion or 14.4 billion in 2007 dollars. The funding available to achieve the goals of C2K is of considerable concern because the single sub-objective of the category of reducing nutrients and sediments requires more than $12.0 billion in 2007 dollars, and this is a major requirement for restoring the health of the Bay’s ecosystem. The cost of restoring the Bay complicates the choices and levels of restoration options. Given the large deficit for achieving the goals and objectives of C2K, it is necessary to assess how restoration might proceed. The available level of funding is simply inadequate for achieving all the goals and objectives necessary to restore the Bay’s ecosystem.     Precision delivery of RAS-inhibiting siRNA to KRAS driven cancer via peptide-based nanoparticles

    Over 95% of pancreatic adenocarcinomas (PDACs), as well as a large fraction of other tumor types, such as colorectal adenocarcinoma, are driven by KRAS activation. However, no direct RAS inhibitors exist for cancer therapy. Furthermore, the delivery of therapeutic agents of any kind to PDAC in particular has been hindered by the extensive desmoplasia and resultant drug delivery challenges that accompanies these tumors. Small interfering RNA (siRNA) is a promising modality for anti-neoplastic therapy due to its precision and wide range of potential therapeutic targets. Unfortunately, siRNA therapy is limited by low serum half-life, vulnerability to intracellular digestion, and transient therapeutic effect. We assessed the ability of a peptide based, oligonucleotide condensing, endosomolytic nanoparticle (NP) system to deliver siRNA to KRAS-driven cancers.     Flooding Events Post Hurricane Harvey: Potential Liability for Dam and Reservoir Operators and Recommendations Moving Forward

    When Hurricane Harvey hit the Texas coast as a category 4 hurricane on August 25, 2017, it resulted in $125 billion in damage, rivaling only Hurricane Katrina in the amount of damage caused. It also resulted in the deaths of 88 people and destroyed or damaged 135,000 homes. Much of that devastation was the result of flooding. The storm dumped over 27 trillion gallons of rain over Texas in a matter of days. Some parts of Houston received over 50 inches of rainfall. The potential liability that dam and reservoir operators may face for decisions they make during storm and flooding events has now become a major concern for Texas citizens and its elected officials. Law suits have now been instituted against the federal government for its operation of two flood control reservoirs, as well as against the San Jacinto River Authority for its operation of a water supply reservoir. Moreover, the issues and concerns have been placed on the agenda of a number of committees preparing for the 2019 Texas legislative session. This report reviews current dam and reservoir operations in Texas and examines the potential liability that such operators may face for actions and decisions taken in response to storm and flooding events. In Section III, the report reviews dam gate operations and differentiates between water supply reservoirs and flood control reservoirs. It also considers pre-release options and explains why such actions are disfavored and not recommended. In Section IV, the report evaluates liabilities and defenses applicable to dam and reservoir operators. It explains how governmental immunity can limit the exposure of state and federally-run facilities to claims seeking monetary damages. It also discusses how such entities could be subject to claims of inverse condemnation, which generally are not subject to governmental immunity, under Texas law as well as under the Fifth Amendment to the U.S. Constitution. In addition, the Section discusses negligence and nuisance claims and concludes that plaintiffs asserting either or both of these claims will have difficulty presenting successful arguments for flooding-related damage and harm against operators who act reasonably in the face of storm-related precipitation. Finally, Section V offers recommendations that dam and reservoir operators might pursue in order to engage and educate the public and thereby reduce the potential for disputes and litigation. Specifically, the report highlights the need for expanded community outreach efforts to engage with municipalities, private land owners, and the business community in flood-prone neighborhoods both below and above a dam. It also recommends implementation of proactive flood notification procedures as a way of reaching and alerting as many people as possible of potential and imminent flooding events. Finally, the report proposes implementation of a dispute prevention and minimization mechanism and offers recommendations for the design and execution of such a program

    Personalised service? Changing the role of the government librarian

    Investigates the feasibility of personalised information service in a government department. A qualitative methodology explored stakeholder opinions on the remit, marketing, resourcing and measurement of the service. A questionnaire and interviews gathered experiences of personalised provision across the government sector. Potential users were similarly surveyed to discuss how the service could meet their needs. Data were analysed using coding techniques to identify emerging theory. Lessons learned from government librarians centred on clarifying requirements, balancing workloads and selective marketing. The user survey showed low usage and awareness of existing specialist services, but high levels of need and interest in services repackaged as a tailored offering. Fieldwork confirmed findings from the literature on the scope for adding value through information management advice, information skills training and substantive research assistance and the need to understand business processes and develop effective partnerships. Concluding recommendations focus on service definition, strategic marketing, resource utilisation and performance measurement

    Forest fine-root production and nitrogen use under elevated CO 2 : contrasting responses in evergreen and deciduous trees explained by a common principle

    Despite the importance of nitrogen (N) limitation of forest carbon (C) sequestration at rising atmospheric CO 2 concentration, the mechanisms responsible are not well understood. To elucidate the interactive effects of elevated CO 2 (eCO 2 ) and soil N availability on forest productivity and C allocation, we hypothesized that (1) trees maximize fitness by allocating N and C to maximize their net growth and (2) that N uptake is controlled by soil N availability and root exploration for soil N. We tested this model using data collected in Free-Air CO 2 Enrichment sites dominated by evergreen ( Pinus taeda ; Duke Forest) and deciduous [ Liquidambar styraciflua ; Oak Ridge National Laboratory (ORNL)] trees. The model explained 80–95% of variation in productivity and N-uptake data among eCO 2 , N fertilization and control treatments over 6 years. The model explains why fine-root production increased, and why N uptake increased despite reduced soil N availability under eCO 2 at ORNL and Duke. In agreement with observations at other sites, the model predicts that soil N availability reduced below a critical level diminishes all eCO 2 responses. At Duke, a negative feedback between reduced soil N availability and N uptake prevented progressive reduction in soil N availability at eCO 2 . At ORNL, soil N availability progressively decreased because it did not trigger reductions in N uptake; N uptake was maintained at ORNL through a large increase in the production of fast turnover fine roots. This implies that species with fast root turnover could be more prone to progressive N limitation of carbon sequestration in woody biomass than species with slow root turnover, such as evergreens. However, longer term data are necessary for a thorough evaluation of this hypothesis. The success of the model suggests that the principle of maximization of net growth to control growth and allocation could serve as a basis for simplification and generalization of larger scale forest and ecosystem models, for example by removing the need to specify parameters for relative foliage/stem/root allocation.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/73052/1/j.1365-2486.2008.01710.x.pd

    The clonal evolution of metastatic colorectal cancer

    Tumor heterogeneity and evolution drive treatment resistance in metastatic colorectal cancer (mCRC). Patient-derived xenografts (PDXs) can model mCRC biology; however, their ability to accurately mimic human tumor heterogeneity is unclear. Current genomic studies in mCRC have limited scope and lack matched PDXs. Therefore, the landscape of tumor heterogeneity and its impact on the evolution of metastasis and PDXs remain undefined. We performed whole-genome, deep exome, and targeted validation sequencing of multiple primary regions, matched distant metastases, and PDXs from 11 patients with mCRC. We observed intricate clonal heterogeneity and evolution affecting metastasis dissemination and PDX clonal selection. Metastasis formation followed both monoclonal and polyclonal seeding models. In four cases, metastasis-seeding clones were not identified in any primary region, consistent with a metastasis-seeding-metastasis model. PDXs underrepresented the subclonal heterogeneity of parental tumors. These suggest that single sample tumor sequencing and current PDX models may be insufficient to guide precision medicine

    A comparative study of four intensive care outcome prediction models in cardiac surgery patients

    <p>Abstract</p> <p>Background</p> <p>Outcome prediction scoring systems are increasingly used in intensive care medicine, but most were not developed for use in cardiac surgery patients. We compared the performance of four intensive care outcome prediction scoring systems (Acute Physiology and Chronic Health Evaluation II [APACHE II], Simplified Acute Physiology Score II [SAPS II], Sequential Organ Failure Assessment [SOFA], and Cardiac Surgery Score [CASUS]) in patients after open heart surgery.</p> <p>Methods</p> <p>We prospectively included all consecutive adult patients who underwent open heart surgery and were admitted to the intensive care unit (ICU) between January 1<sup>st </sup>2007 and December 31<sup>st </sup>2008. Scores were calculated daily from ICU admission until discharge. The outcome measure was ICU mortality. The performance of the four scores was assessed by calibration and discrimination statistics. Derived variables (Mean- and Max- scores) were also evaluated.</p> <p>Results</p> <p>During the study period, 2801 patients (29.6% female) were included. Mean age was 66.9 ± 10.7 years and the ICU mortality rate was 5.2%. Calibration tests for SOFA and CASUS were reliable throughout (p-value not < 0.05), but there were significant differences between predicted and observed outcome for SAPS II (days 1, 2, 3 and 5) and APACHE II (days 2 and 3). CASUS, and its mean- and maximum-derivatives, discriminated better between survivors and non-survivors than the other scores throughout the study (area under curve ≥ 0.90). In order of best discrimination, CASUS was followed by SOFA, then SAPS II, and finally APACHE II. SAPS II and APACHE II derivatives had discrimination results that were superior to those of the SOFA derivatives.</p> <p>Conclusions</p> <p>CASUS and SOFA are reliable ICU mortality risk stratification models for cardiac surgery patients. SAPS II and APACHE II did not perform well in terms of calibration and discrimination statistics.</p
