Quantification and confocal imaging of protein specific molecularly imprinted polymers
We have employed FITC-albumin as the protein template molecule in an aqueous phase molecular imprinted polymer (HydroMIP) strategy. For the first time, the use of a fluorescently labelled template is reported, with subsequent characterisation of the smart material to show that the HydroMIP possess a significant molecular memory in comparison to that of the nonimprinted control polymer (HydroNIP). The imaging of the FITC-albumin imprinted HydroMIP using confocal microscopy is described, with the in situ removal of imprinted protein displayed in terms of observed changes in the fluorescence of the imprinted polymer, both before and after template elution (using a 10% SDS/10% AcOH (w/v) solution). We also report the imaging of a bovine haemoglobin (BHb) imprinted HydroMIP using two-photon confocal microscopy, and describe the effects of template elution upon protein autofluorescence. The findings further contribute to the understanding of aqueous phase molecular imprinting protocols, and document the use of fluorescence as a useful tool in template labelling/detection and novel imaging strategies
Novel critical point drying (CPD) based preparation and transmission electron microscopy (TEM) imaging of protein specific molecularly imprinted polymers (HydroMIPs)
We report the transmission electron microscopy (TEM) imaging of a hydrogel-based molecularly imprinted polymer (HydroMIP) specific to the template molecule bovine haemoglobin (BHb). A novel critical point drying based sample preparation technique was employed to prepare the molecularly imprinted polymer (MIP) samples in a manner that would facilitate the use of TEM to image the imprinted cavities, and provide an appropriate degree of both magnification and resolution to image polymer architecture in the <10 nm range. For the first time, polymer structure has been detailed that clearly displays molecularly imprinted cavities, ranging from 5-50 nm in size, that correlate (in terms of size) with the protein molecule employed as the imprinting template. The modified critical point drying sample preparation technique used may potentially play a key role in the imaging of all molecularly imprinted polymers, particularly those prepared in the aqueous phase

Topographic controls on the development of contemporaneous but contrasting basin-floor depositional architectures
Sediment-laden gravity-driven-flow deposits on the basin floor are typically considered to form either discrete lobes that stack compensationally, or packages of laterally extensive beds, commonly termed âsheets.â These end-member stacking patterns are documented in several basinfills. However, whether they can coexist in a single basin, or there are intermediate or transitional stacking patterns, is poorly understood. An analysis of depositional architecture and stacking patterns along a 70 km dip-oriented transect in the Upper Broto Turbidite System (Jaca Basin, south-central Pyrenees, Spain), which displays disparate stacking patterns in contemporaneous strata, is presented. Proximal and medial deposits are characterized by discrete packages of clean sandstones with sharp bed tops which exhibit predictable lateral and longitudinal facies changes, and are interpreted as lobes. Distal deposits comprise both relatively clean sandstones and hybrid beds that do not stack to form lobes. Instead, localized relatively thick hybrid beds are inferred to have inhibited the development of lobes. Hybrid beds developed under flows which were deflected and entrained carbonate mud substrate off a carbonate slope that bounded the basin to the south; evidence for this interpretation includes: 1) divergent paleoflow indicators and hummock-like features in individual beds; 2) a decrease in hybrid-bed thickness and abundance away from the lateral confining slope; 3) a carbonate-rich upper division, not seen in more proximal turbidites. The study demonstrates the co-occurrence of different styles of basin-floor stacking patterns in the same stratigraphic interval, and suggests that characterization of deep-water systems as either lobes or sheets is a false dichotomy

It is possible to learn a great deal about exoplanet atmospheres even when we
cannot spatially resolve the planets from their host stars. In this chapter, we
overview the basic techniques used to characterize transiting exoplanets -
transmission spectroscopy, emission and reflection spectroscopy, and full-orbit
phase curve observations. We discuss practical considerations, including
current and future observing facilities and best practices for measuring
precise spectra. We also highlight major observational results on the

Composition of lower urinary tract stones in canines in Mexico City

The purpose of this study was to determine the chemical composition of uroliths obtained from the low urinary tract of dogs in Mexico City. One hundred and five cases of urolithiasis were studied in which stones were surgically obtained from the low urinary tracts of dogs treated in different hospitals. The chemical composition of the uroliths was quantita- tively and qualitatively determined by stereoscopic microscopy, IR-spectroscopy, scanning electron micros- copy and X-ray microanalysis. Age of animals ranged from 4 months to 14 years, with a median of 5 years. Compo- sition and distribution of the uroliths were struvite 38.1%,calcium oxalate 26.7%, silica 13.3%, urate 7.6%, mixed 11.4%, compounds 1.9%, and cystine 1%. Most uroliths were found in pure breed dogs (75.2%); 23 different breeds were identified, and more than half of the submissions were from breeds of small size. In our study, the frequency of struvite, calcium oxalate, cystine, urates, mixed and com- pounds stones are in agreement with papers that report on dog populations in America and Europe, but a higher fre- quency of silica uroliths was observed in Mexico City dogs.This work has been partially supported by a project of Waltham Foundation in Mexico
Exploring pig trade patterns to inform the design of risk-based disease surveillance and control strategies
An understanding of the patterns of animal contact networks provides essential information for the design of risk-based animal disease surveillance and control strategies. This study characterises pig movements throughout England and Wales between 2009 and 2013 with a view to characterising spatial and temporal patterns, network topology and trade communities. Data were extracted from the Animal and Plant Health Agency (APHA)âs RADAR (Rapid Analysis and Detection of Animal-related Risks) database, and analysed using descriptive and network approaches. A total of 61,937,855 pigs were moved through 872,493 movements of batches in England and Wales during the 5-year study period. Results show that the network exhibited scale-free and small-world topologies, indicating the potential for diseases to quickly spread within the pig industry. The findings also provide suggestions for how risk-based surveillance strategies could be optimised in the country by taking account of highly connected holdings, geographical regions and time periods with the greatest number of movements and pigs moved, as these are likely to be at higher risk for disease introduction. This study is also the first attempt to identify trade communities in the country, information which could be used to facilitate the pig trade and maintain disease-free status across the country in the event of an outbreak

Vascular disrupting agent for neovascular age related macular degeneration: a pilot study of the safety and efficacy of intravenous combretastatin A-4 phosphate
BACKGROUND:
This study was designed to assess the safety, tolerability, and efficacy of intravenous infusion of CA4P in patients with neovascular age-related macular degeneration (AMD). METHODS:
Prospective, interventional, dose-escalation clinical trial. Eight patients with neovascular AMD refractory to at least 2 sessions of photodynamic therapy received CA4P at a dose of 27 or 36 mg/m2 as weekly intravenous infusion for 4 consecutive weeks. Safety was monitored by vital signs, ocular and physical examinations, electrocardiogram, routine laboratory tests, and collection of adverse events. Efficacy was assessed using retinal fluorescein angiography, optical coherence tomography, and best corrected visual acuity (BCVA). RESULTS:
The most common adverse events were elevated blood pressure (46.7%), QTc prolongation (23.3%), elevated temperature (13.3%), and headache (10%), followed by nausea and eye injection (6.7%). There were no adverse events that were considered severe in intensity and none resulted in discontinuation of treatment. There was reduction of the excess foveal thickness by 24.15% at end of treatment period and by 43.75% at end of the two-month follow-up (pâ=â0.674 and 0.161, respectively). BCVA remained stable throughout the treatment and follow-up periods. CONCLUSIONS:
The safety profile of intravenous CA4P was consistent with that reported in oncology trials of CA4P and with the class effects of vascular disruptive agents; however, the frequency of adverse events was different. There are evidences to suggest potential efficacy of CA4P in neovascular AMD. However, the level of systemic safety and efficacy indicates that systemic CA4P may not be suitable as an alternative monotherapy to current standard-of-care therapy
