    Adjuvant Radiotherapy in Patients with Squamous Cell Carcinoma of the Oral Cavity or Oropharynx and Solitary Ipsilateral Lymph Node Metastasis (pN1) : A Prospective Multicentric Cohort Study

    (1) Background: Evaluation of impact of adjuvant radiation therapy (RT) in patients with oral squamous cell carcinoma of the oral cavity/oropharynx (OSCC) of up to 4 cm (pT1/pT2) and solitary ipsilateral lymph node metastasis (pN1). A non-irradiated group with clinical follow-up was chosen for control, and survival and quality of life (QL) were compared; (2) Methods: This prospective multicentric comprehensive cohort study included patients with resected OSCC (pT1/pT2, pN1, and cM0) who were allocated into adjuvant radiation therapy (RT) or observation. The primary endpoint was overall survival. Secondary endpoints were progression-free survival and QL after surgery; (3) Results: Out of 27 centers, 209 patients were enrolled with a median follow-up of 3.4 years. An amount of 137 patients were in the observation arm, and 72 received adjuvant irradiation. Overall survival did not differ between groups (hazard ratio (HR) 0.98 [0.55–1.73], p = 0.94). There were fewer neck metastases (HR 0.34 [0.15–0.77]; p = 0.01), as well as fewer local recurrences (HR 0.41 [0.19–0.89]; p = 0.02) under adjuvant RT. For QL, irradiated patients showed higher values for the symptom scale pain after 0.5, two, and three years (all p < 0.05). After six months and three years, irradiated patients reported higher symptom burdens (impaired swallowing, speech, as well as teeth-related problems (all p < 0.05)). Patients in the RT group had significantly more problems with mouth opening after six months, one, and two years (p < 0.05); (4) Conclusions: Adjuvant RT in patients with early SCC of the oral cavity and oropharynx does not seem to influence overall survival, but it positively affects progression-free survival. However, irradiated patients report a significantly decreased QL up to three years after therapy compared to the observation group

    Clioquinol and pyrrolidine dithiocarbamate complex with copper to form proteasome inhibitors and apoptosis inducers in human breast cancer cells

    INTRODUCTION: A physiological feature of many tumor tissues and cells is the tendency to accumulate high concentrations of copper. While the precise role of copper in tumors is cryptic, copper, but not other trace metals, is required for angiogenesis. We have recently reported that organic copper-containing compounds, including 8-hydroxyquinoline-copper(II) and 5,7-dichloro-8-hydroxyquinoline-copper(II), comprise a novel class of proteasome inhibitors and tumor cell apoptosis inducers. In the current study, we investigate whether clioquinol (CQ), an analog of 8-hydroxyquinoline and an Alzheimer's disease drug, and pyrrolidine dithiocarbamate (PDTC), a known copper-binding compound and antioxidant, can interact with copper to form cancer-specific proteasome inhibitors and apoptosis inducers in human breast cancer cells. Tetrathiomolybdate (TM), a strong copper chelator currently being tested in clinical trials, is used as a comparison. METHODS: Breast cell lines, normal, immortalized MCF-10A, premalignant MCF10AT1K.cl2, and malignant MCF10DCIS.com and MDA-MB-231, were treated with CQ or PDTC with or without prior interaction with copper, followed by measurement of proteasome inhibition and cell death. Inhibition of the proteasome was determined by levels of the proteasomal chymotrypsin-like activity and ubiquitinated proteins in protein extracts of the treated cells. Apoptotic cell death was measured by morphological changes, Hoechst staining, and poly(ADP-ribose) polymerase cleavage. RESULTS: When in complex with copper, both CQ and PDTC, but not TM, can inhibit the proteasome chymotrypsin-like activity, block proliferation, and induce apoptotic cell death preferentially in breast cancer cells, less in premalignant breast cells, but are non-toxic to normal/non-transformed breast cells at the concentrations tested. In contrast, CQ, PDTC, TM or copper alone had no effects on any of the cells. Breast premalignant or cancer cells that contain copper at concentrations similar to those found in patients, when treated with just CQ or PDTC alone, but not TM, undergo proteasome inhibition and apoptosis. CONCLUSION: The feature of breast cancer cells and tissues to accumulate copper can be used as a targeting method for anticancer therapy through treatment with novel compounds such as CQ and PDTC that become active proteasome inhibitors and breast cancer cell killers in the presence of copper

    Both the Caspase CSP-1 and a Caspase-Independent Pathway Promote Programmed Cell Death in Parallel to the Canonical Pathway for Apoptosis in Caenorhabditis elegans

    Caspases are cysteine proteases that can drive apoptosis in metazoans and have critical functions in the elimination of cells during development, the maintenance of tissue homeostasis, and responses to cellular damage. Although a growing body of research suggests that programmed cell death can occur in the absence of caspases, mammalian studies of caspase-independent apoptosis are confounded by the existence of at least seven caspase homologs that can function redundantly to promote cell death. Caspase-independent programmed cell death is also thought to occur in the invertebrate nematode Caenorhabditis elegans. The C. elegans genome contains four caspase genes (ced-3, csp-1, csp-2, and csp-3), of which only ced-3 has been demonstrated to promote apoptosis. Here, we show that CSP-1 is a pro-apoptotic caspase that promotes programmed cell death in a subset of cells fated to die during C. elegans embryogenesis. csp-1 is expressed robustly in late pachytene nuclei of the germline and is required maternally for its role in embryonic programmed cell deaths. Unlike CED-3, CSP-1 is not regulated by the APAF-1 homolog CED-4 or the BCL-2 homolog CED-9, revealing that csp-1 functions independently of the canonical genetic pathway for apoptosis. Previously we demonstrated that embryos lacking all four caspases can eliminate cells through an extrusion mechanism and that these cells are apoptotic. Extruded cells differ from cells that normally undergo programmed cell death not only by being extruded but also by not being engulfed by neighboring cells. In this study, we identify in csp-3; csp-1; csp-2 ced-3 quadruple mutants apoptotic cell corpses that fully resemble wild-type cell corpses: these caspase-deficient cell corpses are morphologically apoptotic, are not extruded, and are internalized by engulfing cells. We conclude that both caspase-dependent and caspase-independent pathways promote apoptotic programmed cell death and the phagocytosis of cell corpses in parallel to the canonical apoptosis pathway involving CED-3 activation.Howard Hughes Medical InstituteDamon Runyon Cancer Research FoundationCharles A.     TOI-836 : a super-Earth and mini-Neptune transiting a nearby K-dwarf

We obtain photometric follow-up observations with a variety of facilities, and we use these data-sets to determine that the inner planet, TOI-836 b, is a 1.70 ± 0.07 R⊕ super-Earth in a 3.82 day orbit, placing it directly within the so-called ‘radius valley’. The outer planet, TOI-836 c, is a 2.59 ± 0.09 R⊕ mini-Neptune in an 8.60 day orbit. Radial velocity measurements reveal that TOI-836 b has a mass of 4.5 ± 0.9 M⊕, while TOI-836 c has a mass of 9.6 ± 2.6 M⊕. Photometric observations show Transit Timing Variations (TTVs) on the order of 20 minutes for TOI-836 c, although there are no detectable TTVs for TOI-836 b.     We present the discovery of two exoplanets transiting TOI-836 (TIC 440887364) using data from TESS Sector 11 and Sector 38. TOI-836 is a bright (T=8.5T = 8.5 mag), high proper motion (∌ 200\sim\,200 mas yr−1^{-1}), low metallicity ([Fe/H]≈ −0.28\approx\,-0.28) K-dwarf with a mass of 0.68±0.050.68\pm0.05 M⊙_{\odot} and a radius of 0.67±0.010.67\pm0.01 R⊙_{\odot}. We obtain photometric follow-up observations with a variety of facilities, and we use these data-sets to determine that the inner planet, TOI-836 b, is a 1.70±0.071.70\pm0.07 R⊕_{\oplus} super-Earth in a 3.82 day orbit, placing it directly within the so-called 'radius valley'.     Untersuchungen zum Einsatz von Antiseptika in der Behandlung akuter Wunden anhand einer evidenzbasierten Literaturanalyse, Auswertung eines Fragebogens und internationaler Symposiumdiskussion

    Ziel dieser Arbeit war es anhand einer evidenzbasierten Literaturanalyse, einer Umfrage sowie eines Expertentreffens herauszufinden, welche WundspĂŒllösung fĂŒr die Behandlung von akuten Weichgewebewunden mit/ ohne Knochenbeteiligung am geeignetsten ist und ob eine starke Evidenz fĂŒr Handlungsempfehlungen besteht. Des Weiteren sollte anhand der vorliegenden Arbeit untersucht werden, inwieweit ein Konsens bezĂŒglich der Therapie akuter Wunden besteht. Die Umfrageergebnisse als auch die Expertensitzung bestĂ€tigten, dass in der Anwendung von Antiseptika zur Behandlung akuter Wunden weiterer Diskussionsbedarf besteht, da noch viele Fragen ungeklĂ€rt sind. Basierend auf den Erkenntnissen dieser evidenzbasierten Literaturauswertung konnten folgende Empfehlungen mit höherem Empfehlungsgrad (A und B) formuliert werden: Zur Behandlung von akuten Wunden mit Knochenbeteiligung sollen eine physiologische warme, sterile Kochsalzlösung oder Trinkwasser nach ausreichendem DĂ©bridement eingesetzt werden. (A) Auf antibiotische und antiseptische Zusatzstoffe sollte solange keine Infektion vorliegt, aufgrund der ungenĂŒgenden Wirkungsnachweise und möglichen Risiken verzichtet werden. (B) Gentamicin-Kollagen Implantate fĂŒhren zu einer Verringerung von sternalen Wundinfekten in der Herzchirurgie und sollten zum Einsatz kommen. (B) Zur Versorgung von Verbrennungswunden 2. Grades sollte Polyhexanid zum Einsatz kommen. (B) Zu diskutieren bleibt, dass ein Einsatz von Trinkwasser nicht bedenkenlos empfohlen werden kann, da die TrinkwasserqualitĂ€t von Region zu Region unterschiedlich ist. Bei Verbrennungswunden ist die Anwendung von Polyhexanid als Antiseptikum der Wahl zu empfehlen, wie in der vorliegenden Arbeit gezeigt werden konnte. Zur Beantwortung der bestehenden offenen Fragen sind weitere Studien in hochwertiger QualitĂ€t notwendig

    <div><p>Endocytosis is a multi-step process involving a large number of proteins, both general factors, such as clathrin and adaptor protein complexes, and unique proteins, which modulate specialized endocytic processes, like the EHD proteins. EHDs are a family of <u>E</u>ps15 <u>H</u>omology <u>D</u>omain containing proteins that consists of four mammalian homologs, one <i>C</i>. <i>elegans</i>, one <i>Drosophila melanogaster</i> and two plants orthologs. These membrane-associated proteins are involved in different steps of endocytic trafficking pathways. We have previously shown that the <i>Drosophila</i> EHD ortholog, PAST1, associates predominantly with the plasma membrane. Mutations in <i>Past1</i> result in defects in endocytosis, male sterility, temperature sensitivity and premature death of the flies. Also, <i>Past1</i> genetically interacts with <i>Notch</i>. In the present study, we investigated the role of PAST1 in the developing fly eye. In mutant flies lacking PAST1, abnormal differentiation of photoreceptors R1, R6 and R7 was evident, with partial penetrance. Likewise, five cone cells were present instead of four. Expression of transgenic PAST1 resulted in a dominant negative effect, with a phenotype similar to that of the deletion mutant, and appearance of additional inter-ommatidial pigment cells. Our results strongly suggest a role for PAST1 in differentiation of photoreceptors R1/R6/R7 and cone cells of the fly ommatidia.</p></div
