    Modern theoretical and technical approaches in plant morphology

    Morphology is deeply rooted in organismal biology, which in recent years has gone through a steady decline in interest both at research institutions and funding agencies. In parallel with this development, morphology as a discipline has been marginalized and nowadays many think of it as just a classical and largely obsolete field of research. However, this is far from the truth. Thanks to modern theoretical concepts and novel technical applications, plant morphology has much to contribute to modern botanical and evolutionary research. In our presentation, we will first outline the application of High Resolution X-Ray Computed Tomography (HRXCT) to the study of plant structure. The ideal way to describe the morphological phenotype of a given organism is to build a three dimensional (3D) model, which may then be interpreted with respect to other types of data, e.g., metabolite content or functional groups of pollinators. We have developed simple but efficient lab protocols using contrasting agents such as phosphotungstate and bismuth tartrate that allow for the streamlined acquisition of high resolution phenotypic data and 3D-representations even of soft plant tissues such as floral organs, ovules, and meristematic tissues. To illustrate this, we will outline selected ongoing studies in comparative plant science that make use of high resolution tomography. In the second part of our talk, we will present a project on the floral morphospace. A striking feature of morphological variation is that due to developmental, functional, and phylogenetic constraints, not all theoretically possible architectures have been explored during evolution. A modern approach to studying the evolution of realized forms among possible ones is to construct morphospaces, i.e. theoretical, mathematical spaces describing and relating organismal phenotypes. Although widely applied in zoology, morphospace analyses and related approaches have so far been largely disregarded in botany, with notable exceptions in the field of pollination biology. Here, we use a morphospace approach to describe and quantify the morphological diversity (disparity) of flowers in the asterid order Ericales. To do so, we have built a dataset containing 37 floral characters for more than 380 species (275 genera) representative of the entire order. We have used non-parametric representations and statistics methods based on distance matrices to build and analyze a morphospace, in which we compare the relative positions of the different ericalean families. We quantify and interpret the disparity among these groups in the light of their taxonomic diversity, their evolutionary history, and their ecology.     Energetic Demands of Immature Sea Otters From Birth to Weaning: Implications for Maternal Costs, Reproductive Behavior and Population-Level Trends

    Sea otters (Enhydra lutris) have the highest mass-specific metabolic rate of any marine mammal, which is superimposed on the inherently high costs of reproduction and lactation in adult females. These combined energetic demands have been implicated in the poor body condition and increased mortality of female sea otters nearing the end of lactation along the central California coast. However, the cost of lactation is unknown and currently cannot be directly measured for this marine species in the wild. Here, we quantified the energetic demands of immature sea otters across five developmental stages as a means of assessing the underlying energetic challenges associated with pup rearing that may contribute to poor maternal condition. Activity-specific metabolic rates, daily activity budgets and field metabolic rates (FMR) were determined for each developmental stage. Mean FMR of pre-molt pups was 2.29±0.81 MJ day−1 and increased to 6.16±2.46 and 7.41±3.17 MJ day−1 in post-molt pups and dependent immature animals, respectively. Consequently, daily energy demands of adult females increase 17% by 3 weeks postpartum and continue increasing to 96% above pre-pregnancy levels by the average age of weaning. Our results suggest that the energetics of pup rearing superimposed on small body size, marine living and limited on-board energetic reserves conspire to make female sea otters exceptionally vulnerable to energetic shortfalls. By controlling individual fitness, maternal behavior and pup provisioning strategies, this underlying metabolic challenge appears to be a major factor influencing current population trends in southern sea otters (Enhydra lutris nereis)

    Levodopa-induced dyskinesia in Parkinson's disease: sleep matters

    OBJECTIVE: The spectrum of clinical symptoms changes during the course of Parkinson's disease. Levodopa therapy, while offering remarkable control of classical motor symptoms, causes abnormal involuntary movements as the disease progresses. These levodopa-induced dyskinesias (LIDs) have been associated with abnormal cortical plasticity. Since slow wave activity (SWA) of nonrapid eye movement (NREM) sleep underlies adjustment of cortical excitability, we sought to elucidate the relationship between this physiological process and LIDs. METHODS: Thirty-six patients at different stages of Parkinson's disease (PD) underwent whole-night video polysomnography-high-density EEG (vPSG-hdEEG), preceded by 1 week of actigraphy. To represent the broad spectrum of the disease, patients were divided into three groups by disease stage, (i) de novo (DNV; n = 9), (ii) advanced (ADV; n = 13), and (iii) dyskinetic (DYS; n = 14) and were compared to an age-matched control group (CTL; n = 12). The SWA-NREM content of the PSG-hdEEG was then temporally divided into 10 equal parts, from T1 to T10, and power and source analyses were performed. T2-T3-T4 were considered early sleep and were compared to T7-T8-T9, representing late sleep. RESULTS: We found that all groups, except the DYS group, manifested a clear-cut SWA decrease between early and late sleep. INTERPRETATION: Our data demonstrate a strong pathophysiological association between sleep and PD. Given that SWA may be a surrogate for synaptic strength, our data suggest that DYS patients do not have adequate synaptic downscaling. Further analysis is needed to determine the effect of drugs that can enhance cortical SWA in LIDs

    Emergence of a floral colour polymorphism by pollinator-mediated overdominance.

    Maintenance of polymorphism by overdominance (heterozygote advantage) is a fundamental concept in evolutionary biology. In most examples known in nature, overdominance is a result of homozygotes suffering from deleterious effects. Here we show that overdominance maintains a non-deleterious polymorphism with black, red and white floral morphs in the Alpine orchid Gymnadenia rhellicani. Phenotypic, metabolomic and transcriptomic analyses reveal that the morphs differ solely in cyanidin pigments, which are linked to differential expression of an anthocyanidin synthase (ANS) gene. This expression difference is caused by a premature stop codon in an ANS-regulating R2R3-MYB transcription factor, which is heterozygous in the red colour morph. Furthermore, field observations show that bee and fly pollinators have opposite colour preferences; this results in higher fitness (seed set) of the heterozygous morph without deleterious effects in either homozygous morph. Together, these findings demonstrate that genuine overdominance exists in nature

    The Economic Value of Sea Otters and Recreational Tourism in a California Estuary

    The recovery of marine megafauna can lead to improved ecosystem function and services, but not all stakeholders may benefit equally. Quantifying the local economic value of a species’ presence may appeal to broader range of stakeholders when developing conservation strategies. This study aims to examine the economic effect recreational activities can have on a local region, and to determine what role the presence of southern sea otters (Enhydra lutris nereis) had on the value visitors placed on the visit and the area’s preservation. We surveyed visitors to Elkhorn Slough, a small estuary in California known for diverse wildlife, modeled direct and indirect economic contributions to local economics, and evaluated perceptions and value placed on the area and wildlife. Annually, visitors contributed approximately 3.2million(USD)indirectspending,withanadditional3.2 million (USD) in direct spending, with an additional 1.85 million in indirect economic gains, which could support over 300 full-time, part-time, and seasonal jobs to the region. Whether sea otters were observed during a trip influenced how visitors ranked their importance, and the perceived value of the estuary and sea otters. Combined, this study quantified what recreational visitors could contribute to local economies and that sea otters play a role in what visitor’s value about their visit. These results provide additional support to the benefits of species presence at a local scale. We discuss how these types of studies can be used as part of larger species and ecosystem management plans, particularly considering species recovery and range expansion

    Species recognition limits mating between hybridizing ant species

    Identifying mechanisms limiting hybridization is a central goal of speciation research. Here, we studied pre-mating and post-mating barriers to hybridization between two ant species, Formica selysi and Formica cinerea. These species hybridize in the Rhône valley in Switzerland, where they form a mosaic hybrid zone, with limited introgression from F. selysi into F. cinerea. There was no sign of temporal isolation between the two species in the production of queens and males. With choice experiments, we showed that queens and males strongly prefer to mate with conspecifics. Yet, we did not detect post-mating barriers caused by genetic incompatibilities. Specifically, hybrids of all sexes and castes were found in the field and F1 hybrid workers did not show reduced viability compared to non-hybrid workers. To gain insights into the cues involved in species recognition, we analyzed the cuticular hydrocarbons of queens, males and workers and staged dyadic encounters between workers. Cuticular hydrocarbon profiles differed markedly between species, but were similar in F. cinerea and hybrids. Accordingly, workers also discriminated species, but they did not discriminate F. cinerea and hybrids. We discuss how the CHC-based recognition system of ants may facilitate the establishment of pre-mating barriers to hybridization, independent of hybridization costs.     Histological and micro-CT evidence of stigmatic rostellum receptivity promoting auto-pollination in the Madagascan orchid Bulbophyllum bicoloratum

    Background: The rostellum, a projecting part of the gynostemium in orchid flowers, separates the anther(s) from the stigma and thus commonly prevents auto-pollination. Nonetheless, as a modified (usually distal) portion of the median stigma lobe, the rostellum has been frequently invoked of having re-gained a stigmatic function in rare cases of orchid auto-pollination. Here it is shown that a newly discovered selfing variant of Madagascan Bulbophyllum bicoloratum has evolved a modified rostellum allowing the penetration of pollen tubes from in situ pollinia. Methods: Gynostemium micro-morphology and anatomy of selfing and outcrossing variants of B. bicoloratum was studied by using light and scanning electron microscopy and histological sections. Pollen tube growth in the selfing variant was further observed via X-ray computed microtomography (micro-CT), providing 3D reconstructions of floral tissues at a micron scale. Findings: Selfing variants possess a suberect (‘displaced) rostellum rather than the conventional, erect type. Very early in anthesis, the pollinia of selfers are released from the anther and slide down onto the suberect rostellum, where pollen tube growth preferentially occurs through the non-vascularized, i.e. rear (adaxial) and (semi-) lateral parts. This penetrated tissue is comprised of a thin layer of elongate and loosely arranged cells, embedded in stigmatic exudates, as also observed in the stigmatic cavity of both selfing and outcrossing variants. Conclusions: Our results provide the first solid evidence of a stigmatic function for the rostellum in orchid flowers, thereby demonstrating for the first time the feasibility of the micro-CT technique for accurately visualizing pollen tube growth in flowering plants. Rostellum receptivity in B. bicoloratum probably uniquely evolved as an adaptation for reproductive assurance from an outcrossing ancestor possessing an erect (non-receptive) rostellum.     Neurologic complications of acute hepatitis E virus infection.

    To assess the prevalence and clinical features of neurologic involvement in patients with acute hepatitis E virus (HEV) infection in Southern Switzerland. Among 1,940 consecutive patients investigated for acute hepatitis E, we identified 141 cases of acute of HEV infection (anti-HEV immunoglobulin M and immunoglobulin G both reactive and/or HEV RNA positive) between June 2014 and September 2017. Neurologic cases were followed up for 6 months. We compared patients with and without neurologic symptoms. Neurologic symptoms occurred in 43 acute HEV cases (30.4%) and consisted of neuralgic amyotrophy (NA, n = 15, 10.6%) and myalgia (n = 28, 19.8%). All NA cases were immunocompetent. Men had higher odds (OR = 5.2, CI 1.12-24.0, p = 0.03) of developing NA after infection with HEV, and in 3 couples simultaneously infected with HEV, only men developed NA. Bilateral involvement of NA was predominant (2:1) and occurred only in men. Seven NA cases were viremic (all genotype 3), but HEV was undetectable in their CSF. In the acute phase of NA, 9 patients were treated with intravenous immunoglobulin and 4 with prednisone, reporting no side effects and improvement in pain and strength. Myalgia occurred both without (n = 16) or with (n = 12) concomitant elevated serum creatinine kinase. Seven cases with myalgia in the shoulder girdle did not have muscle weakness ("forme fruste" of NA). Neurologic symptoms occurred in one-third of acute HEV infections and consisted of NA and myalgia. NA seems to occur more frequently in men infected by HEV and has a predominant (but not exclusive) bilateral involvement

    Polyphenols act synergistically with doxorubicin and etoposide in leukaemia cell lines

    The study aimed to assess the effects of polyphenols when used in combination with doxorubicin and etoposide, and to determine whether polyphenols sensitised leukaemia cells, causing inhibition of cell proliferation, cell cycle arrest and induction of apoptosis. This study is based on findings in solid cancer tumours, which have shown that polyphenols can sensitize cells to chemotherapy, and induce apoptosis and/or cell-cycle arrest. This could enable a reduction of chemotherapy dose and off-target effects, whilst maintaining treatment efficacy. Quercetin, apigenin, emodin, rhein and cis-stilbene were investigated alone and in combination with etoposide and doxorubicin in two lymphoid and two myeloid leukaemia cells lines. Measurements were made of ATP levels (using CellTiter-Glo assay) as an indication of total cell number, cell cycle progression (using propidium iodide staining and flow cytometry) and apoptosis (NucView caspase 3 assay and Hoechst 33342/propidium iodide staining). Effects of combination treatments on caspases 3, 8 and 9 activity were determined using Glo luminescent assays, glutathione levels were measured using the GSH-Glo Glutathione Assay and DNA damage determined by anti-γH2AX staining. Doxorubicin and etoposide in combination with polyphenols synergistically reduced ATP levels, induced apoptosis and increased S and/or G2/M phase cell cycle arrest in lymphoid leukaemia cell lines. However, in the myeloid cell lines the effects of the combination treatments varied; doxorubicin had a synergistic or additive effect when combined with quercetin, apigenin, emodin, and cis-stilbene, but had an antagonistic effect when combined with rhein. Combination treatment caused a synergistic downregulation of glutathione levels and increased DNA damage, driving apoptosis via caspase 8 and 9 activation. However, in myeloid cells where antagonistic effects were observed, this was associated with increased glutathione levels and a reduction in DNA damage and apoptosis. This study has demonstrated that doxorubicin and etoposide activity were enhanced by polyphenols in lymphoid leukaemia cells, however, differential responses were seen in myeloid cells with antagonistic responses seen in some combination therapies
