    The concept of constitutional supercooling (CS) including the term itself was first described and discussed qualitatively by Rutter and Chalmers in order to understand the formation of cellular structures during the solidification of tin, and then quantified by Tiller et al. On that basis, Winegard and Chalmers further considered 'supercooling and dendritic freezing of alloys' where they described how CS promotes the heterogeneous nucleation of new crystals and the formation of an equiaxed zone. Since then the importance of CS in promoting the formation of equiaxed microstructures in both grain refined and unrefined alloys has been clearly revealed and quantified. This paper describes our current understanding of the role of CS in promoting nucleation and grain formation. It also highlights that CS, on the one hand, develops a nucleation-free zone surrounding each nucleated and growing grain and, on the other hand, protects this grain from readily remelting when temperature fluctuations occur due to convection. Further, due to the importance of the diffusion field that generates CS, recent analytical models are evaluated and compared with a numerical model. A comprehensive description of the mechanisms affecting nucleation and grain formation and the prediction of grain size is presented with reference to the influence of the casting conditions applied during the practical casting of an alloy

    Rapid Evaporative Ionisation Mass Spectrometry of Electrosurgical Vapours for the Identification of Breast Pathology: Towards an Intelligent Knife for Breast Cancer Surgery

    Background: Re-operation for positive resection margins following breast-conserving surgery occurs frequently (average = 20–25%), is cost-inefficient, and leads to physical and psychological morbidity. Current margin assessment techniques are slow and labour intensive. Rapid evaporative ionisation mass spectrometry (REIMS) rapidly identifies dissected tissues by determination of tissue structural lipid profiles through on-line chemical analysis of electrosurgical aerosol toward real-time margin assessment. Methods: Electrosurgical aerosol produced from ex-vivo and in-vivo breast samples was aspirated into a mass spectrometer (MS) using a monopolar hand-piece. Tissue identification results obtained by multivariate statistical analysis of MS data were validated by histopathology. Ex-vivo classification models were constructed from a mass spectral database of normal and tumour breast samples. Univariate and tandem MS analysis of significant peaks was conducted to identify biochemical differences between normal and cancerous tissues. An ex-vivo classification model was used in combination with bespoke recognition software, as an intelligent knife (iKnife), to predict the diagnosis for an ex-vivo validation set. Intraoperative REIMS data were acquired during breast surgery and time-synchronized to operative videos. Results: A classification model using histologically validated spectral data acquired from 932 sampling points in normal tissue and 226 in tumour tissue provided 93.4% sensitivity and 94.9% specificity. Tandem MS identified 63 phospholipids and 6 triglyceride species responsible for 24 spectral differences between tissue types. iKnife recognition accuracy with 260 newly acquired fresh and frozen breast tissue specimens (normal n = 161, tumour n = 99) provided sensitivity of 90.9% and specificity of 98.8%. The ex-vivo and intra-operative method produced visually comparable high intensity spectra. iKnife interpretation of intra-operative electrosurgical vapours, including data acquisition and analysis was possible within a mean of 1.80 seconds (SD ±0.40). Conclusions: The REIMS method has been optimised for real-time iKnife analysis of heterogeneous breast tissues based on subtle changes in lipid metabolism, and the results suggest spectral analysis is both accurate and rapid. Proof-of-concept data demonstrate the iKnife method is capable of online intraoperative data collection and analysis. Further validation studies are required to determine the accuracy of intra-operative REIMS for oncological margin assessment

    Recent advances in grain refinement of light metals and alloys

    Grain refinement leads, in general, to a decreased tendency to hot tearing, a more dispersed and refined porosity distribution, and improved directional feeding characteristics during solidification. Reduced as-cast grain size can also lead to improved mechanical properties and wrought processing by reducing the recrystallized grain size and achieving a fully recrystallized microstructure. It is now well established that the two key factors controlling grain refinement are the nucleant particles including their potency, size distribution and particle number density, and the rate of development of growth restriction, Q, generated by the alloy chemistry which establishes the undercooling needed to trigger nucleation events and facilitates their survival. The theories underpinning our current understanding of nucleation and grain formation are presented. The application of the latest theories to the light alloys of Al, Mg and Ti is explored as well as their applicability to a range of casting and solidification environments. In addition, processing by the application of physical processes such as external fields and additive manufacturing is discussed. To conclude, the current challenges for the development of reliable grain refining technologies for difficult to refine alloy systems are presented

    LIPOGENESIS IN ADIPOSE TISSUE FROM OVARIECTOMIZED AND INTACT HEIFERS IMMUNIZED AGAINST ESTRADIOL AND(OR) IMPLANTED WITH TRENBOLONE ACETATE

    Forty-two heifers were allotted randomly to six treatment groups: 1) intact controls, 2) intact heifers implanted with trenbolone acetate, 3) ovariectomized heifers, 4) ovariectomized heifers implanted with trenbolone acetate, 5) intact heifers immunized against estradiol and 6) intact heifers immunized against estradiol and implanted with trenbolone acetate. Blood titers of estradiol-17β were increased over lO0-fold in heifers immunized against estradiol in Freund\u27s complete adjuvant or saline:squalene/arlacel containing Mycobacterium. Lipogenic enzyme activities and acetate incorporation into fatty acids were increased in subcutaneous adipose tissue obtained at slaughter from heifers receiving immunization or the combination of immunization and trenbolone acetate. The increased lipogenic capacity was not reflected in either cell diameter or cells per gram adipose tissue. Ovariectomy in combination with trenbolone acetate caused the lowest activities for all enzymes measured. This treatments also caused the greatest decrease in cell diameter, which resulted in the largest number of cells per gram of adipose tissue. Trenbotone acetate alone had no detectable effect on lipogenesis in the intact heifer, but the combination of ovariectomy and trenbolone acetate caused substantial decreases in enzyme activities, in most cases a significant decrease as compared with ovariectomized heifers. The data suggest that trenbolone acetate is able to depress lipogenesis only when not competing with the effects of circulating estradiol

    Edge-based nonlinear diffusion for finite element approximations of convection–diffusion equations and its relation to algebraic flux-correction schemes

    The final publication is available at Springer via http://dx.doi.org/10.1007/s00211-016-0808-zFor the case of approximation of convection–diffusion equations using piecewise affine continuous finite elements a new edge-based nonlinear diffusion operator is proposed that makes the scheme satisfy a discrete maximum principle. The diffusion operator is shown to be Lipschitz continuous and linearity preserving. Using these properties we provide a full stability and error analysis, which, in the diffusion dominated regime, shows existence, uniqueness and optimal convergence. Then the algebraic flux correction method is recalled and we show that the present method can be interpreted as an algebraic flux correction method for a particular definition of the flux limiters. The performance of the method is illustrated on some numerical test cases in two space dimensions
