    The endemic vascular flora of Peloritani Mountains (NE Sicily): Plant functional traits and phytogeographical relationships in the most isolated and fragmentary micro-plate of the Alpine orogeny

    This study is aimed at (1) producing a complete and updated inventory of the endemic vascular flora of Peloritani Mountains, (2) defining the geographical limits of Peloritani, regarded here as a biogeographical district and (3) highlighting possible paleogeographic connections with other Mediterranean lands. The heterogeneity analysis of the endemic flora was performed by means of contingency tables, through the x 2 test. The endemic flora of this area consists of 129 specific and infraspecific taxa, of which 15 are restricted to the Peloritani Mountains. The analysis of habitats revealed that endemic taxa are most abundant on cliffs, rangelands, woods and garrigues. A large number of surveyed taxa are endemic to central-southern Italy and Sicily, while the number of endemic taxa in common with Calabria, Etna and Aeolian Islands turned out to be rather low, in spite of the geographical proximity. The endemic flora of Peloritani allows to emphasize palaeogeographical relationships not only with the neighbouring Mediterranean territories, but also with currently remote ones, such as southern Spain, Sardinia and Corsica. The phytogeographical framework substantiates the hypothesis that the Peloritani floristic district coincides with the limit given to Peloritani Mountains by structural geologists

    Opioid activity profiles of oversimplified peptides lacking in the protonable N-terminus

    Recently, we described cyclopeptide opioid agonists containing the D-Trp-Phe sequence. To expand the scope of this atypical pharmacophore, we tested the activity profiles of the linear peptides Ac-Xaa-Phe-Yaa (Xaa = L/D-Trp, D-His/Lys/Arg; Yaa = H, GlyNH2). Ac-D-Trp-PheNH2 appeared to be the minimal binding sequence, while Ac-D-Trp-Phe-GlyNH 2 emerged as the first noncationizable short peptide (partial) agonist with high \u3bc-opioid receptor affinity and selectivity.     Bioclimatology, structure, and conservation perspectives of Quercus pyrenaica, Acer opalus subsp. Granatensis, and Corylus avellana deciduous forests on Mediterranean bioclimate in the South-Central part of the Iberian Peninsula

    The plant variability in the southern Iberian Peninsula consists of around 3500 different taxa due to its high bioclimatic, geographic, and geological diversity. The deciduous forests in the southern Iberian Peninsula are located in regions with topographies and specific bioclimatic conditions that allow for the survival of taxa that are typical of cooler and wetter bioclimatic regions and therefore represent the relict evidence of colder and more humid paleoclimatic conditions. The floristic composition of 421 samples of deciduous forests in the south-central part of the Iberian Peninsula were analyzed. The ecological importance index (IVI) was calculated, where the most important tree species were Quercuspyrenaica, Aceropalus subsp. Granatensis, and Corylusavellana. These species are uncommon in the south-central part of the Iberian Peninsula, forming forests of little extension. An analysis of the vertical distribution of the species (stratum) shows that the majority of the species of stratum 3 (hemicriptophics, camephytes, geophites, and nanophanerophytes) are characteristic of deciduous forests, and their presence is positively correlated with high values of bioclimatic variables related to humidity and presence of water in the soil (nemoral environments), while they are negatively correlated with high values of bioclimatic variables related to high temperatures, evapotranspiration, and aridity. This work demonstrates that several characteristic deciduous forest taxa are more vulnerable to disappearance due to the loss of their nemoral conditions caused by gaps in the tree or shrub canopy.     Epilepsy-related cytoarchitectonic abnormalities along white matter pathways

    Objective Temporal lobe epilepsy (TLE) is one of the most common forms of epilepsy. Unfortunately, the clinical outcomes of TLE cannot be determined based only on current diagnostic modalities. A better understanding of white matter (WM) connectivity changes in TLE may aid the identification of network abnormalities associated with TLE and the phenotypic characterisation of the disease. Methods We implemented a novel approach for characterising microstructural changes along WM pathways using diffusional kurtosis imaging (DKI). Along-the-tract measures were compared for 32 subjects with left TLE and 36 age-matched and gender-matched controls along the left and right fimbria-fornix (FF), parahippocampal WM bundle (PWMB), arcuate fasciculus (AF), inferior longitudinal fasciculus (ILF), uncinate fasciculus (UF) and cingulum bundle (CB). Limbic pathways were investigated in relation to seizure burden and control with antiepileptic drugs. Results By evaluating measures along each tract, it was possible to identify abnormalities localised to specific tract subregions. Compared with healthy controls, subjects with TLE demonstrated pathological changes in circumscribed regions of the FF, PWMB, UF, AF and ILF. Several of these abnormalities were detected only by kurtosis-based and not by diffusivity-based measures. Structural WM changes correlated with seizure burden in the bilateral PWMB and cingulum. Conclusions DKI improves the characterisation of network abnormalities associated with TLE by revealing connectivity abnormalities that are not disclosed by other modalities. Since TLE is a neuronal network disorder, DKI may be well suited to fully assess structural network abnormalities related to epilepsy and thus serve as a tool for phenotypic characterisation of epilepsy

    Salvia ceratophylloides (Ard.) is an endemic and rare plant species recently rediscovered as very few individuals at two different Southern Italy sites. The study of within-plant variation is fundamental to understand the plant adaptation to the local conditions, especially in rare species, and consequently to preserve plant biodiversity. Here, we reported the variation of the morpho-ecophysiological and metabolic traits between the sessile and petiolate leaf of S. ceratophylloides plants at two different sites for understanding the adaptation strategies for surviving in these habitats. The S. ceratophylloides individuals exhibited different net photosynthetic rate, maximum quantum yield, light intensity for the saturation of the photosynthetic machinery, stomatal conductance, transpiration rate, leaf area, fractal dimension, and some volatile organic compounds (VOCs) between the different leaf types. This within-plant morpho-physiological and metabolic variation was dependent on the site. These results provide empirical evidence of sharply within-plant variation of the morpho-physiological traits and VOCs profiles in S. ceratophylloides, explaining the adaptation to the local conditions

    2023 Update on Sepsis and Septic Shock in Adult Patients: Management in the Emergency Department

    Background: Sepsis/septic shock is a life-threatening and time-dependent condition that requires timely management to reduce mortality. This review aims to update physicians with regard to the main pillars of treatment for this insidious condition. Methods: PubMed, Scopus, and EMBASE were searched from inception with special attention paid to November 2021–January 2023. Results: The management of sepsis/septic shock is challenging and involves different pathophysiological aspects, encompassing empirical antimicrobial treatment (which is promptly administered after microbial tests), fluid (crystalloids) replacement (to be established according to fluid tolerance and fluid responsiveness), and vasoactive agents (e.g., norepinephrine (NE)), which are employed to maintain mean arterial pressure above 65 mmHg and reduce the risk of fluid overload. In cases of refractory shock, vasopressin (rather than epinephrine) should be combined with NE to reach an acceptable level of pressure control. If mechanical ventilation is indicated, the tidal volume should be reduced from 10 to 6 mL/kg. Heparin is administered to prevent venous thromboembolism, and glycemic control is recommended. The efficacy of other treatments (e.g., proton-pump inhibitors, sodium bicarbonate, etc.) is largely debated, and such treatments might be used on a case-to-case basis. Conclusions: The management of sepsis/septic shock has significantly progressed in the last few years. Improving knowledge of the main therapeutic cornerstones of this challenging condition is crucial to achieve better patient outcomes

    Millstones as indicators of relative sea-level changes in northern Sicily and southern Calabria coast lines, Italy

    New data are presented for late Holocene relative sea-level changes in two coastal sites of Sicily and Calabria, southern Italy. Reconstructions are based on precise measurements of submerged archaeological remains that are valuable indicators of past sea-level position. The archaeological remains are millstone quarries carved on sandstone coastal rocks and nowadays partially submerged which, to the authors’ knowledge, are used for the first time as sea-level markers. Millstones of similar typology are located on the coast of Capo d’Orlando (northern Sicily) and Capo dell’Armi (southern Calabria). When the archeologically-based sea-level position is compared with the shoreline elevation provided by geological markers (Holocene beachrock, Late Pleistocene marine terraces), a refined understanding of relative sea-level changes and rates of vertical tectonic movements for these coastline locations is gained

    Analysis of the muscarinic receptor subtype mediating inhibition of the neurogenic contractions in rabbit isolated vas deferens by a series of polymethylene tetra-amines

The pharmacological characteristics of the presynaptic muscarinic receptor subtype, which mediates inhibition of the neurogenic contractions in the prostatic portion of rabbit vas deferens, have been investigated by using a series of polymethylene tetra-amines, which were selected for their ability to differentiate among muscarinic receptor subtypes. 2. It was found that all tetra-amines antagonized McN-A-343-induced inhibition in electrically stimulated rabbit vas deferens in a competitive manner and with affinity values (pA2) ranging between 6.27 ± 0.09 (spirotramine) and 8.51 ± 0.02 (AM170). 3. Competition radioligand binding studies, using native muscarinic receptors from rat tissues (M1, cortex; M2, heart; M3, submaxillary gland) or from NG 108-15 cells (M4) and human cloned muscarinic M1-M4 receptors expressed in CHO-K1 cells, were undertaken with the same tetraamines employed in functional assays. All antagonists indicated a one-site fit. 4. The affinity estimates (pKi) of tetra-amines calculated in binding assays using native receptors were similar to those obtained using cloned receptors. Among these compounds some displayed selectivity between muscarinic receptor subtypes, indicating that they may be valuable tools in receptor characterization. Spirotramine was selective for M1 receptors versus all other subtypes (pKi native: M1, 7.32 ± 0.10; M2, 6.50 ± 0.11; M3, 6.02 ± 0.13; M4, 6.28 ± 0.16; pKi cloned: M1, 7.69 ± 0.08; M2, 6.22 ± 0.14; M3, 6.11 ± 0.16; 6.35 ± 0.11) whereas CC8 is highly selective for M2 receptors versus the other subtypes (pKi native: M1 7.50 ± 0.04; M2, 9.01 ± 0.12; M3, 6.70 ± 0.08; M4, 7.56 ± 0.04; pKi cloned: M1, 7.90 ± 0.20; M2, 9.04 ± 0.08; M3, 6.40 ± 0.07; M4, 7.40 ± 0.04). Furthermore, particularly relevant for this investigation were tetra-amines dipitramine and AM172 for their ability to significantly differentiate M1 and M4 receptors. 5. The apparent affinity values (pA2) obtained for tetra-amines in functional studies using the prostatic portion of rabbit vas deferens correlated most closely with the values (pKi) obtained at either native or human recombinant muscarinic M4 receptors. This supports the view that the muscarinic receptor mediating inhibition of neurogenic contractions of rabbit vas deferens may not belong to the M1 type but rather appears to be of the M4 subtype
