    Recommandations pour une agriculture plus écologique respectant les besoins socio - écono-miques locaux, région du Menabe Central, côte ouest de Madagascar

    Dans le Menabe Central (côte ouest de Madagascar), les paysages forestiers deviennent toujours plus ouverts, le taux de déforestation avoisinant les 0,7 % . La déforestation étant notamment due à des défrichements pour l’agriculture qui est la principale activité de la région, une gestion agricole écologiquement durable apparaît comme une nécessité urgente. Afin de ne pas défavoriser les populations locales vivant dans des conditions socio-économiques difficiles (81,3 % de la population malgache vivait encore en - dessous du seuil de pauvreté en 2010), cet aménagement agricole doit également viser à proposer des  recommandations socio - économiquement rentables. Cette étude vise donc à confirmer l’hypothèse selon laquelle une agriculture plus écologique – permettant aux agriculteurs de se sédentariser sur leurs terres et diminuant les défrichements forestiers – qui fournirait des produits économiquement rentables sur le long terme répondrait aux nécessités socio-économiques et écologiques locales. La présente étude conclut en mettant en évidence quatre recommandations : i) La riziculture devrait être privilégiée ; ii) les techniques agro - forestières et les utilisations de fertilisants naturels devraient être encouragées ; iii) des alternatives permettant de diversifier le revenu des populations rurales, telle que la rizipisciculture, devraient être soutenues ; et iv) un aménagement des surfaces sylvicoles défrichées devrait être mis en place de manière participative.In the Central Menabe region on the west cost of Madagascar, traditional uses of forest resources create an increasingly open landscape. The current annual rate of loss of Malagasy dense dry forest, the natural forest type of the region, is 0.7 % . Agriculture represents the principal activity of people in Central Menabe, and the main reason for the decrease in forest cover. In the current difficult socio-economical context (81.3 % of the Malagasy population lived under the poverty line in 2010), where a threat to dry forests clearly exists, it is therefore urgent to propose scientifically-sound and participatory recommendations for ecologically sustainable and socio - economically profitable agricultural use of the Central Menabe landscape. The objective of this study is to confirm the hypothesis that a more sustainable form of agriculture – promoting farmers’ permanent use of cultivated fields – would generate high value economic products, and contribute to the socio - economic and environmental needs of the region. To meet this objective, the study answers three research questions: i) What are the principal products of the villager economy? ii) what is the role of traditional agriculture in deforestation?; and, iii) what is the potential for, and the expectations of the local populations towards, more sedentary agricultural techniques? The research uses methods from both social (scoring and questionnaires) and natural sciences (inventories and measures of clearings), and was carried out in six villages representative of the Central Menabe region. Two villages mainly practiced rice cultivation, two mainly carried out slash and burn cultivation (of maize, cassava and peanut), and two practiced both slash and burn and rice cultivation. Half of the villages were situated near a national road and have thus an easy access to regional markets; the three other villages were more remote. In total 120 inhabitants were involved in 72 different scoring exercises and 288 participated in a questionnaire survey. Mann-Whitney and chi - square tests were used to test for statistical significance in observed differences.Analyses confirm that rice is the main pillar of the villager economy in the region, and that – at the village scale – this product serves a strong commercial demand which is not always satisfied. Farmers take a weak interest in the consumption of products from slash and burn cultivation (low demand), although the majority of people that cultivate maize, cassava or peanut perceive slash and burn cultivation as a source of alimentary or financial security (subsistence or sale).     Energy-sensitive imaging detector applied to the dissociative recombination of D2H+

    We report on an energy-sensitive imaging detector for studying the fragmentation of polyatomic molecules in the dissociative recombination of fast molecular ions with electrons. The system is based on a large area (10 cm x 10 cm) position-sensitive, double-sided Si-strip detector with 128 horizontal and 128 vertical strips, whose pulse height information is read out individually. The setup allows to uniquely identify fragment masses and is thus capable of measuring branching ratios between different fragmentation channels, kinetic energy releases, as well as breakup geometries, as a function of the relative ion-electron energy. The properties of the detection system, which has been installed at the TSR storage ring facility of the Max-Planck Institute for Nuclear Physics in Heidelberg, is illustrated by an investigation of the dissociative recombination of the deuterated triatomic hydrogen cation D2H+. A huge isotope effect is observed when comparing the relative branching ratio between the D2+H and the HD+D channel; the ratio 2B(D2+H)/B(HD+D), which is measured to be 1.27 +/- 0.05 at relative electron-ion energies around 0 eV, is found to increase to 3.7 +/- 0.5 at ~5 eV.Comment: 11 pages, 12 figures, submitted to Physical Review

    Theory of visual attention's thalamic model for visual short-term memory capacity and top-down control: evidence from a thalamo-cortical structural connectivity analysis

    In the theory of visual attention (TVA), it is suggested that objects in a visual scene compete for representation in a visual short-term memory (vSTM) store. The race towards the store is assumed to be biased by top-down controlled weighting of the objects according to their task relevance. Only objects that reach the store before its capacity limitation is reached are represented consciously in a given instant. TVA-based computational modeling of participants' performance in whole- and partial-report tasks permits independent parameters of individual efficiency of top-down control α and vSTM storage capacity K to be extracted. The neural interpretation of the TVA proposes recurrent loops between the posterior thalamus and posterior visual cortices to be relevant for generating attentional weights for competing objects and for maintaining selected objects in vSTM. Accordingly, we tested whether structural connectivity between posterior thalamus and occipital cortices (PT-OC) is associated with estimates of top-down control and vSTM capacity. We applied whole- and partial-report tasks and probabilistic tractography in a sample of 37 healthy adults. We found vSTM capacity K to be associated with left PT-OC structural connectivity and a trend-wise relation between top-down control α and right PT-OC structural connectivity. These findings support the assumption of the relevance of thalamic structures and their connections to visual cortex for top-down control and vSTM capacity

    Impaired visual short-term memory capacity is distinctively associated with structural connectivity of the posterior thalamic radiation and the splenium of the corpus callosum in preterm-born adults

    Preterm birth is associated with an increased risk for lasting changes in both the cortico-thalamic system and attention; however, the link between cortico-thalamic and attention changes is as yet little understood. In preterm newborns, cortico-cortical and cortico-thalamic structural connectivity are distinctively altered, with increased local clustering for cortico-cortical and decreased integrity for cortico-thalamic connectivity. In preterm-born adults, among the various attention functions, visual short-term memory (vSTM) capacity is selectively impaired. We hypothesized distinct associations between vSTM capacity and the structural integrity of cortico-thalamic and cortico-cortical connections, respectively, in preterm-born adults. A whole-report paradigm of briefly presented letter arrays based on the computationally formalized Theory of Visual Attention (TVA) was used to quantify parameter vSTM capacity in 26 preterm- and 21 full-term-born adults. Fractional anisotropy (FA) of posterior thalamic radiations and the splenium of the corpus callosum obtained by diffusion tensor imaging were analyzed by tract-based spatial statistics and used as proxies for cortico-thalamic and cortico-cortical structural connectivity. The relationship between vSTM capacity and cortico-thalamic and cortico-cortical connectivity, respectively, was significantly modified by prematurity. In full-term-born adults, the higher FA in the right posterior thalamic radiation the higher vSTM capacity; in preterm-born adults this FA-vSTM-relationship was inversed. In the splenium, higher FA was correlated with higher vSTM capacity in preterm-born adults, whereas no significant relationship was evident in full-term-born adults. These results indicate distinct associations between cortico-thalamic and cortico-cortical integrity and vSTM capacity in preterm-and full-term-born adults.     Unsupervised, Efficient and Semantic Expertise Retrieval

    We introduce an unsupervised discriminative model for the task of retrieving experts in online document collections. We exclusively employ textual evidence and avoid explicit feature engineering by learning distributed word representations in an unsupervised way. We compare our model to state-of-the-art unsupervised statistical vector space and probabilistic generative approaches. Our proposed log-linear model achieves the retrieval performance levels of state-of-the-art document-centric methods with the low inference cost of so-called profile-centric approaches. It yields a statistically significant improved ranking over vector space and generative models in most cases, matching the performance of supervised methods on various benchmarks. That is, by using solely text we can do as well as methods that work with external evidence and/or relevance feedback. A contrastive analysis of rankings produced by discriminative and generative approaches shows that they have complementary strengths due to the ability of the unsupervised discriminative model to perform semantic matching.Comment: WWW2016, Proceedings of the 25th International Conference on World Wide Web. 201

    Transcriptional analysis of temporal gene expression in germinating Clostridium difficile 630 endospores.

    Clostridium difficile is the leading cause of hospital acquired diarrhoea in industrialised countries. Under conditions that are not favourable for growth, the pathogen produces metabolically dormant endospores via asymmetric cell division. These are extremely resistant to both chemical and physical stress and provide the mechanism by which C. difficile can evade the potentially fatal consequences of exposure to heat, oxygen, alcohol, and certain disinfectants. Spores are the primary infective agent and must germinate to allow for vegetative cell growth and toxin production. While spore germination in Bacillus is well understood, little is known about C. difficile germination and outgrowth. Here we use genome-wide transcriptional analysis to elucidate the temporal gene expression patterns in C. difficile 630 endospore germination. We have optimized methods for large scale production and purification of spores. The germination characteristics of purified spores have been characterized and RNA extraction protocols have been optimized. Gene expression was highly dynamic during germination and outgrowth, and was found to involve a large number of genes. Using this genome-wide, microarray approach we have identified 511 genes that are significantly up- or down-regulated during C. difficile germination (p≤0.01). A number of functional groups of genes appeared to be co-regulated. These included transport, protein synthesis and secretion, motility and chemotaxis as well as cell wall biogenesis. These data give insight into how C. difficile re-establishes its metabolism, re-builds the basic structures of the vegetative cell and resumes growth

    Evoked responses to rhythmic visual stimulation vary across sources of intrinsic alpha activity in humans

    Rhythmic flickering visual stimulation produces steady-state visually evoked potentials (SSVEPs) in electroencephalogram (EEG) recordings. Based on electrode-level analyses, two dichotomous models of the underpinning mechanisms leading to SSVEP generation have been proposed: entrainment or superposition, i.e., phase-alignment or independence of endogenous brain oscillations from flicker-induced oscillations, respectively. Electrode-level analyses, however, represent an averaged view of underlying ‘source-level’ activity, at which variability in SSVEPs may lie, possibly suggesting the co-existence of multiple mechanisms. To probe this idea, we investigated the variability of SSVEPs derived from the sources underpinning scalp EEG responses during presentation of a flickering radial checkerboard. Flicker was presented between 6 and 12 Hz in 1 Hz steps, and at individual alpha frequency (IAF i.e., the dominant frequency of endogenous alpha oscillatory activity). We tested whether sources of endogenous alpha activity could be dissociated according to evoked responses to different flicker frequencies relative to IAF. Occipitoparietal sources were identified by temporal independent component analysis, maximal resting-state alpha power at IAF and source localisation. The pattern of SSVEPs to rhythmic flicker relative to IAF was estimated by correlation coefficients, describing the correlation between the peak-to-peak amplitude of the SSVEP and the absolute distance of the flicker frequency from IAF across flicker conditions. We observed extreme variability in correlation coefficients across sources, ranging from −0.84 to 0.93, with sources showing largely different coefficients co-existing within subjects. This result demonstrates variation in evoked responses to flicker across sources of endogenous alpha oscillatory activity. Data support the idea of multiple SSVEP mechanisms

    CD69 is a TGF-β/1α,25-dihydroxyvitamin D3 target gene in monocytes

    CD69 is a transmembrane lectin that can be expressed on most hematopoietic cells. In monocytes, it has been functionally linked to the 5-lipoxygenase pathway in which the leukotrienes, a class of highly potent inflammatory mediators, are produced. However, regarding CD69 gene expression and its regulatory mechanisms in monocytes, only scarce data are available. Here, we report that CD69 mRNA expression, analogous to that of 5-lipoxygenase, is induced by the physiologic stimuli transforming growth factor-β (TGF-β) and 1α,25-dihydroxyvitamin D3 (1α,25(OH)2D3) in monocytic cells. Comparison with T- and B-cell lines showed that the effect was specific for monocytes. CD69 expression levels were increased in a concentration-dependent manner, and kinetic analysis revealed a rapid onset of mRNA expression, indicating that CD69 is a primary TGF-β/1α,25(OH)2D3 target gene. PCR analysis of different regions of the CD69 mRNA revealed that de novo transcription was initiated and proximal and distal parts were induced concomitantly. In common with 5-lipoxygenase, no activation of 0.7 kb or ~2.3 kb promoter fragments by TGF-β and 1α,25(OH)2D3 could be observed in transient reporter assays for CD69. Analysis of mRNA stability using a transcription inhibitor and a 3′UTR reporter construct showed that TGF-β and 1α,25(OH)2D3 do not influence CD69 mRNA stability. Functional knockdown of Smad3 clearly demonstrated that upregulation of CD69 mRNA, in contrast to 5-LO, depends on Smad3. Comparative studies with different inhibitors for mitogen activated protein kinases (MAPKs) revealed that MAPK signalling is involved in CD69 gene regulation, whereas 5-lipoxygenase gene expression was only partly affected. Mechanistically, we found evidence that CD69 gene upregulation depends on TAK1-mediated p38 activation. In summary, our data indicate that CD69 gene expression, conforming with 5-lipoxygenase, is regulated monocyte-specifically by the physiologic stimuli TGF-β and 1α,25(OH)2D3 on mRNA level, although different mechanisms account for the upregulation of each gene
