    Nitro-fatty acid formation and metabolism

    Nitro-fatty acids (NO 2 -FA) are pleiotropic modulators of redox signaling pathways. Their effects on inflammatory signaling have been studied in great detail in cell, animal and clinical models primarily using exogenously administered nitro-oleic acid. While we know a considerable amount regarding NO 2 -FA signaling, endogenous formation and metabolism is relatively unexplored. This review will cover what is currently known regarding the proposed mechanisms of NO 2 -FA formation, dietary modulation of endogenous NO 2 -FA levels, pathways of NO 2 -FA metabolism and the detection of NO 2 -FA and corresponding metabolites.Fil: Buchan, Gregory J..     European Union legislation overview about used vegetable oils recycling: the spanish and italian case studies

    The employment of used vegetable oils (UVOs) as raw materials in key sectors as energy production or bio-lubricant synthesis represents one of the most relevant priorities in the European Union (EU) normative context. In many countries, the development of new production processes based on the circular economy model, as well as the definition of future energy and production targets, involve the utilization of wastes as raw material. In this context, the main currently applied EU regulations are presented and discussed. As in the EU, the general legislative process consists of the definition in each State Member of specific legislation, which transposes the EU indications. Two relevant countries are herein considered: Italy and Spain. Through the analysis of the conditions required in both countries for UVOs’ collection, disposal, storage, and recycling, a wide panorama of the current situation is provided

    Medical therapy for patients with subclinical and clinical carotid atherosclerosis

    The management of carotid artery disease includes both modifications in life style as well treatment of vascular risk factors. However, strict risk factor modification, including improved antihypertensive therapy, lipid management, smoking cessation, and antiplatelet therapy, promise for reducing the vascular event rate in patients with carotid atherosclerosis. The best medical management for stroke prevention was highlighted in clinical practice guidelines issued jointly in 2006 by the American Heart Association and the American Stroke Association, and co-sponsored by the Council on Cardiovascular Radiology and Intervention and the American Academy of Neurology. Lowering blood pressure to a target below 120/80 mm Hg by life style interventions and antihypertensive treatment. Glucose control to near-normoglycemic levels (target hemoglobin A1C ≤7%) is recommended among diabetics to reduce micro-vascular complications and, with lesser certainty, macrovascular complications. The primary objective of this review is to summarize the current evidence and standards for the advanced diagnostic and management strategies used in asymptomatic and symptomatic patients with carotid atherosclerosis

    Ordered mesoporous silica functionalized with beta-cyclodextrin derivative for stereoisomer separation of flavanones and flavanone glycosides by nano-liquid chromatography and capillary electrochromatography

    In this paper a chiral stationary phase (CSP) was prepared by the immobilization of a beta-CD derivative (3,5-dimethylphenylcarbamoylated beta-CD) onto the surface of amino-functionalized spherical ordered mesoporous silica (denoted as SM) via a urea linkage using the Staudinger reaction. The CSP was packed into fused silica capillaries 100 mu m I.D. and evaluated by means of nano-liquid chromatography (nano-LC) and capillary electrochromatography (CEC) using model compounds for the enantio- and the diastereomeric separation. The compounds flavanone, 2'-hydroxyflavanone, 4'-hydroxyflavanone, 6hydroxyflavanone, 4'-methoxyflavanone, 7-methoxyfiavanone, hesperetin, hesperidin, naringenin, and naringin were studied using reversed and polar organic elution modes. Baseline stereoisomer resolution and good results in terms of peak efficiency and short analysis time of all studied flavonoids and flavanones glycosides were achieved in reversed phase mode, using as mobile phase a mixture of MeOH/H2O, 10 mM ammonium acetate pH 4.5 at different ratios. For the polar organic mode using 100% of MeOH as mobile phase, the CSP showed better performances and the baseline chiral separation of several studied compounds occurred in an analysis time of less than 10 min. Good results were also achieved by CEC employing two different mobile phases. The use of MeOH/H2O, 5 mM ammonium acetate buffer pH 6.0 (90/10, v/v) was very effective for the chiral resolution of flavanone and its methoxy and hydroxy derivatives. (C) 2017 Elsevier B.V.     Exogenous Nitro-Oleic Acid Treatment Inhibits Primary Root Growth by Reducing the Mitosis in the Meristem in Arabidopsis thaliana

    Nitric oxide (NO) is a second messenger that regulates a broad range of physiological processes in plants. NO-derived molecules called reactive nitrogen species (RNS) can react with unsaturated fatty acids generating nitrated fatty acids (NO2-FA). NO2-FA work as signaling molecules in mammals where production and targets have been described under different stress conditions. Recently, NO2-FAs were detected in plants, however their role(s) on plant physiological processes is still poorly known. Although in this work NO2-OA has not been detected in any Arabidopsis seedling tissue, here we show that exogenous application of nitro-oleic acid (NO2-OA) inhibits Arabidopsis primary root growth; this inhibition is not likely due to nitric oxide (NO) production or impaired auxin or cytokinin root responses. Deep analyses showed that roots incubated with NO2-OA had a lower cell number in the division area. Although this NO2-FA did not affect the hormonal signaling mechanisms maintaining the stem cell niche, plants incubated with NO2-OA showed a reduction of cell division in the meristematic area. Therefore, this work shows that the exogenous application of NO2-OA inhibits mitotic processes subsequently reducing primary root growth.Fil: Di Fino, Luciano Martin.     Metrics for monitoring patients progress in a rehabilitation context: a case study based on wearable inertial sensors

    Inertial data can represent a rich source of clinically relevant information which can provide details on motor assessment in subjects undertaking a rehabilitation process. Indeed, in clinical and sport settings, motor assessment is generally conducted through simple subjective measures such as a visual assessment or questionnaire given by caregivers. As part of a mobile health application, wireless sensors such as inertial measurement units and associated data sets can help provide an objective and empirical measure of a patient’s progress through rehabilitation using on body sensors. In this publication, several metrics in different domains have been considered and extrapolated from the 3D accelerometer and angular rate data sets collected on an impaired subject with knee injury, via a wearable sensing system developed at the Tyndall National Institute. These data sets were collected for different activities performed across a number of sessions as the subject progressed through the rehabilitation process. Using these data sets, a novel and effective method has been investigated in order to define a single score indicator which can provide accurate quantitative analysis of the improvement of the subject throughout their rehabilitation. The indicator compares impaired and unimpaired limb motor performance. The present work proves that the defined score indicator can be taken into account by clinicians to study the overall patients’ condition and provide accurate clinical feedback as to their rehabilitative progress
