    “Been there---ah, haven’t tried it that way”: A Professional Effort to Differentiate Instruction

    It goes without saying that the most critical component of preparing educators lies in their ability to competently teach. Differentiation provides a framework to develop classrooms where realities of genuine student variance can be addressed with curricular realities. The author describes a professional development project that differentiated a series of teacher workshops that were designed to increase teachers’ perceived competency to differentiate instruction. The purpose here is to describe a collaboratively created and implemented professional development program designed to train staff in ways to differentiate instruction for all learners. Sample training activities, along with perceptions from participants and suggestions for realistic extensions are shared

    The Long-Baseline Neutrino Experiment: Exploring Fundamental Symmetries of the Universe

    The preponderance of matter over antimatter in the early Universe, the dynamics of the supernova bursts that produced the heavy elements necessary for life and whether protons eventually decay --- these mysteries at the forefront of particle physics and astrophysics are key to understanding the early evolution of our Universe, its current state and its eventual fate. The Long-Baseline Neutrino Experiment (LBNE) represents an extensively developed plan for a world-class experiment dedicated to addressing these questions. LBNE is conceived around three central components: (1) a new, high-intensity neutrino source generated from a megawatt-class proton accelerator at Fermi National Accelerator Laboratory, (2) a near neutrino detector just downstream of the source, and (3) a massive liquid argon time-projection chamber deployed as a far detector deep underground at the Sanford Underground Research Facility. This facility, located at the site of the former Homestake Mine in Lead, South Dakota, is approximately 1,300 km from the neutrino source at Fermilab -- a distance (baseline) that delivers optimal sensitivity to neutrino charge-parity symmetry violation and mass ordering effects. This ambitious yet cost-effective design incorporates scalability and flexibility and can accommodate a variety of upgrades and contributions. With its exceptional combination of experimental configuration, technical capabilities, and potential for transformative discoveries, LBNE promises to be a vital facility for the field of particle physics worldwide, providing physicists from around the globe with opportunities to collaborate in a twenty to thirty year program of exciting science. In this document we provide a comprehensive overview of LBNE's scientific objectives, its place in the landscape of neutrino physics worldwide, the technologies it will incorporate and the capabilities it will possess.Comment: Major update of previous version.     A framework for human microbiome research

    A variety of microbial communities and their genes (the microbiome) exist throughout the human body, with fundamental roles in human health and disease. The National Institutes of Health (NIH)-funded Human Microbiome Project Consortium has established a population-scale framework to develop metagenomic protocols, resulting in a broad range of quality-controlled resources and data including standardized methods for creating, processing and interpreting distinct types of high-throughput metagenomic data available to the scientific community. Here we present resources from a population of 242 healthy adults sampled at 15 or 18 body sites up to three times, which have generated 5,177 microbial taxonomic profiles from 16S ribosomal RNA genes and over 3.5 terabases of metagenomic sequence so far. In parallel, approximately 800 reference strains isolated from the human body have been sequenced. Collectively, these data represent the largest resource describing the abundance and variety of the human microbiome, while providing a framework for current and future studies

    Structure, function and diversity of the healthy human microbiome

The definitive version was published in Nature 486 (2012): 207-214, doi:10.1038/nature11234.Studies of the human microbiome have revealed that even healthy individuals differ remarkably in the microbes that occupy habitats such as the gut, skin and vagina. Much of this diversity remains unexplained, although diet, environment, host genetics and early microbial exposure have all been implicated. Accordingly, to characterize the ecology of human-associated microbial communities, the Human Microbiome Project has analysed the largest cohort and set of distinct, clinically relevant body habitats so far. We found the diversity and abundance of each habitat’s signature microbes to vary widely even among healthy subjects, with strong niche specialization both within and among individuals. The project encountered an estimated 81–99% of the genera, enzyme families and community configurations occupied by the healthy Western microbiome. Metagenomic carriage of metabolic pathways was stable among individuals despite variation in community structure, and ethnic/racial background proved to be one of the strongest associations of both pathways and microbes with clinical metadata.     Missing in Action: Research on the Accountability of Multicultural, Inclusive Teacher Education

    While a substantial amount of research has been conducted on the effects of various teacher education programs to prepare teachers for multicultural, multilingual, inclusive classrooms, very little of it examines the impact of multicultural, inclusive teacher education on how pre- and inservice teachers actually teach children in the classroom. Few researchers have followed teachers into the classroom to find out if or what “carry over” exists from multicultural teacher education (Sleeter, 2001). Even if teachers show growth through their course work and learning experiences in multicultural education, what evidence is there that they are or are becoming strong teachers in culturally diverse classrooms? This article reports on a study “in progress” that is part of a longitudinal investigation of preservice and novice teachers’ classroom practices and behaviors to address the diverse needs of students whose backgrounds and abilities differ from their own. Specifically, the present study examines the impact of multicultural, inclusive teacher education on how novice teachers actually instruct children in multicultural, multilingual, and inclusive classroom contexts

    Tissue Slice Grafts : An in Vivo Model of Human Prostate Androgen Signaling

    We developed a tissue slice graft (TSG) model by implanting thin, precision-cut tissue slices derived from fresh primary prostatic adenocarcinomas under the renal capsule of immunodeficient mice. This new in vivo model not only allows analysis of approximately all of the cell types present in prostate cancer within an intact tissue microenvironment, but also provides a more accurate assessment of the effects of interventions when tissues from the same specimen with similar cell composition and histology are used as control and experimental samples. The thinness of the slices ensures that sufficient samples can be obtained for large experiments as well as permits optimal exchange of nutrients, oxygen, and drugs between the grafted tissue and the host. Both benign and cancer tissues displayed characteristic histology and expression of cell-type specific markers for up to 3 months. Moreover, androgen-regulated protein expression diminished in TSGs after androgen ablation of the host and was restored after androgen repletion. Finally, many normal secretory epithelial cells and cancer cells in TSGs remained viable 2 months after androgen ablation, consistent with similar observations in postprostatectomy specimens following neoadjuvant androgen ablation. Among these were putative Nkx3.1+ stem cells. Our novel TSG model has the appropriate characteristics to serve as a useful tool to model all stages of disease, including normal tissue, premalignant lesions, well-differentiated cancer, and poorly differentiated cancer

    Redesigning Personnel Preparation

    Changing legal requirements, economic demands, accountability, and student demographics focus the need for reform among personnel preparation programs. Although redesign efforts in general and special education teacher preparation programs have increased, little evidence from research describes enhancement initiatives. In this article, the authors report the findings of an investigation of contextual variables related to reforming teacher preparation as reported by project personnel at Institutions of Higher Education (IHEs) who were awarded 325T priority grants by the Office of Special Education Programs (OSEP) during a 5-year period. This study employed a sequential mixed-methods design, which included (a) the administration of a survey to all project directors and (b) interviews with a purposive sample of project directors.
