    A study of the effect of movement of outdoor pigs to a new site on Salmonella prevalence

    Outdoor breeding of pigs in Great Britain constitutes a substantial proportion of the British pig industry, with at least 40% of the breeding herd kept outdoors. Outdoor production is viewed as more welfare friendly and forms a break between cereal crops. Outdoor pig production has a tendency for high Salmonella seroprevalence at slaughter, with evidence for a wide diversity of resident and transient infections on farm, often showing some overlap with local environmental and wildlife isolates. Outdoor pigs may be at increased risk of infection due to: the lack of a controlled environment that can be cleaned and disinfected between batches of pigs; less control over exposure of pigs to factors such as cold and heat stress; and increased exposure to Salmonella through difficulties in applying biosecurity for personnel and vehicles as well as increased contact with the environment and wildlife. Anecdotal information has shown that moving outdoor pigs to new land is usually followed by an improvement in herd health and productivity. It is believed that outdoor herds in the UK typically move site every 2-3 years. The aim of this longitudinal study was to investigate the effect on the occurrence of Salmonella of moving pigs to new land and the sustainability of any improvements over a one year period

    Maternal vaccination as an effective Salmonella reduction strategy

    Salmonella is a widespread pathogen that can infect a variety of animals, including man. Pork is considered, after eggs, the major source of infection in humans in the EU, with S. Typhimurium (ST), including monophasic strains (mST; S. 1,4,[5],12:i- and S. 1,4,12:i-) being frequently implicated. Reducing the prevalence of infected pigs on-farm contributes to minimising contamination of pig meat and edible offal for human consumption, as the slaughter process alone cannot cope with high levels of contamination. The persistent and frequently asymptomatic nature of porcine Salmonella infection, the organism’s colonisation of farm pests, such as rodents and wild birds, and ability to survive in the environment, means that effective control generally requires multiple measures. It is generally accepted that vaccination can play a role in reducing the prevalence of Salmonella in pigs and could assist other on-farm control measures by helping to prevent Salmonella colonizing the gut and reducing the subsequent shedding and development of a carrier state

    Salmonella is the second most commonly reported zoonotic gastrointestinal pathogen in the European Union. Although the majority of foodborne outbreaks have been linked to the consumption of eggs and egg products (44.0%), a substantial proportion originate from pork and pork products (9.3%). Biosecurity measures correctly implemented on farm are important to reducing Salmonella carriage in live pigs and consequently the number of Salmonella contaminated carcasses entering the food chain

    Weapon injuries in the crusader mass graves from a 13th century attack on the port city of Sidon (Lebanon).

    Archaeological excavations close to St Louis' castle in Sidon, Lebanon have revealed two mass grave deposits containing partially articulated and disarticulated human skeletal remains. A minimum of 25 male individuals have been recovered, with no females or young children. Radiocarbon dating of the human remains, a crusader coin, and the design of Frankish belt buckles strongly indicate they belong to a single event in the mid-13th century CE. The skeletal remains demonstrate a high prevalence of unhealed sharp force, penetrating force and blunt force trauma consistent with medieval weaponry. Higher numbers of wounds on the back of individuals than the front suggests some were attacked from behind, possibly as they fled. The concentration of blade wounds to the back of the neck of others would be compatible with execution by decapitation following their capture. Taphonomic changes indicate the skeletal remains were left exposed for some weeks prior to being collected together and re-deposited in the defensive ditch by a fortified gateway within the town wall. Charring on some bones provides evidence of burning of the bodies. The findings imply the systematic clearance of partially decomposed corpses following an attack on the city, where adult and teenage males died as a result of weapon related trauma. The skeletons date from the second half of the Crusader period, when Christian-held Sidon came under direct assault from both the Mamluk Sultanate (1253 CE) and the Ilkhanate Mongols (1260 CE). It is likely that those in the mass graves died during one of these assaults

    Prioritization of pig farm biosecurity for control of Salmonella and hepatitis E virus infections: results of a European expert opinion elicitation

    BackgroundIn the literature, there is absent or weak evidence on the effectiveness of biosecurity measures to the control of Salmonella spp. and hepatitis E virus (HEV) on pig farms. Therefore, the present study aimed to collect, weigh, and compare opinions from experts on the relevance of several biosecurity measures. An online questionnaire was submitted to selected experts, from multiple European countries, knowledgeable on either HEV or Salmonella spp., in either indoor or outdoor pig farming systems (settings). The experts ranked the relevance of eight biosecurity categories with regards to effectiveness in reducing the two pathogens separately, by assigning a score from a total of 80, and within each biosecurity category they scored the relevance of specific biosecurity measures (scale 1-5). Agreement among experts was analysed across pathogens and across settings.ResultsAfter filtering for completeness and expertise, 46 responses were analysed, with 52% of the experts identified as researchers/scientists, whereas the remaining 48% consisted of non-researchers, veterinary practitioners and advisors, governmental staff, and consultant/industrial experts. The experts self-declared their level of knowledge but neither Multidimensional Scaling nor k-means cluster analyses produced evidence of an association between expertise and the biosecurity answers, and so all experts' responses were analysed together without weighting or adaptation. Overall, the top-ranked biosecurity categories were pig mixing; cleaning and disinfection; feed, water and bedding; and purchase of pigs or semen, while the lowest ranked categories were transport, equipment, animals (other than pigs and including wildlife) and humans. Cleaning and disinfection was ranked highest for both pathogens in the indoor setting, whereas pig mixing was highest for outdoor settings. Several (94/222, 42.3%) measures across all four settings were considered highly relevant. Measures with high disagreement between the respondents were uncommon (21/222, 9.6%), but more frequent for HEV compared to Salmonella spp.ConclusionsThe implementation of measures from multiple biosecurity categories was considered important to control Salmonella spp. and HEV on farms, and pig mixing activities, as well as cleaning and disinfection practices, were perceived as consistently more important than others. Similarities and differences in the prioritised biosecurity measures were identified between indoor and outdoor systems and pathogens. The study identified the need for further research especially for control of HEV and for biosecurity in outdoor farming

    FAIR v1.3: A simple emissions-based impulse response and carbon cycle model

    Simple climate models can be valuable if they are able to replicate aspects of complex fully coupled earth system models. Larger ensembles can be produced, enabling a probabilistic view of future climate change. A simple emissions-based climate model, FAIR, is presented, which calculates atmospheric concentrations of greenhouse gases and effective radiative forcing (ERF) from greenhouse gases, aerosols, ozone and other agents. Model runs are constrained to observed temperature change from 1880 to 2016 and produce a range of future projections under the Representative Concentration Pathway (RCP) scenarios. The constrained estimates of equilibrium climate sensitivity (ECS), transient climate response (TCR) and transient climate response to cumulative CO₂ emissions (TCRE) are 2.86 (2.01 to 4.22) K, 1.53 (1.05 to 2.41) K and 1.40 (0.96 to 2.23) K (1000 GtC)¯¹ (median and 5–95 % credible intervals). These are in good agreement with the likely Intergovernmental Panel on Climate Change (IPCC) Fifth Assessment Report (AR5) range, noting that AR5 estimates were derived from a combination of climate models, observations and expert judgement. The ranges of future projections of temperature and ranges of estimates of ECS, TCR and TCRE are somewhat sensitive to the prior distributions of ECS∕TCR parameters but less sensitive to the ERF from a doubling of CO₂ or the observational temperature dataset used to constrain the ensemble. Taking these sensitivities into account, there is no evidence to suggest that the median and credible range of observationally constrained TCR or ECS differ from climate model-derived estimates. The range of temperature projections under RCP8.5 for 2081–2100 in the constrained FAIR model ensemble is lower than the emissions-based estimate reported in AR5 by half a degree, owing to differences in forcing assumptions and ECS∕TCR distributions

    Cross-sectional study of hepatitis E virus (HEV) circulation in Italian pig farms

    OHEJP Project: BIOPIGEE Foodborne transmission is considered the main way of spreading zoonotic hepatitis E virus (HEV) infection in Europe. In recent years, the human cases of hepatitis E in subjects without history of travel in endemic areas have raised, suggesting that domestic HEV transmission is increasing. Pork products with or without liver, are often indicated as the source of many human foodborne HEV cases as well as small outbreaks. Pigs are recognized as the main reservoir of the zoonotic HEV-3 genotype, the most frequently detected in human cases in the EU. In the absence of a harmonized surveillance of HEV circulation, data on prevalence are heterogeneous but confirm a widespread circulation of HEV-3 in pig herds across EU. HEV-3 can pass through the food chain from farm to fork when infected animals are slaughtered. In Italy, several studies reported the circulation of HEV-3 in pig farms, but results are heterogeneous due to dierent methodologies applied. In the present study, we performed a survey over 51 pig herds belonging to three main types of farms: breeding, fattening and farrow-to- finish. HEV-RNA was analyzed by broad range Real-time RT-PCR on 20 samples for each farm, obtained by pooling together feces from 10 individuals. Overall, HEV RNA was confirmed on 150 fecal pooled samples out of 1,032 (14.5%). At least one positive pooled sample was detected from 18 farms out of 51 tested (35.3%). By lowering the number of infected pigs at primary production, the risk of HEV-3 entering into the food chain can be reduced. Hence, information on HEV circulation in herds is highly relevant for choosing preventive measures and deserves development of a monitoring program and further investigations

    Evaluation of an enhanced cleaning and disinfection protocol in Salmonella contaminated pig holdings in the United Kingdom

    Salmonella is the second most commonly reported zoonotic gastrointestinal pathogen in the European Union, and a significant proportion of the cases are linked to the consumption of contaminated pork. Reduction of Salmonella at the farm level helps to mininimise the contamination pressure at the slaughterhouse, and therefore the number of Salmonella bacteria entering the food chain. Cleaning and disinfection (C&D) between batches of pigs is an intervention measure that has potential to reduce the transmission of Salmonella contamination within farms. In this study, two pig finisher buildings in each of 10 Salmonella positive farms were sampled pre-C&D, post-C&D, post-restocking with the following batch of pigs, and shortly before these pigs were sent to slaughter. The incoming batch of pigs was also sampled before it reached the study building (pre-restocking). At each visit, pooled and individual faecal samples were collected and Salmonella isolation was carried out according to an ISO 6579:2002 Annex D-based method. One building on each farm (intervention) was cleaned and disinfected according to a rigorous protocol consisting of several steps and a Defra-approved disinfectant used at the General Orders concentration, whilst the other building (control) was cleaned and disinfected as per normal farm routine. At the post-C&D visit, Enterobacteriaceae and total bacterial counts were determined to evaluate residual faecal contamination and general hygiene levels. Rodent specialists visited the farms before and after C&D and rodent carcasses were collected for Salmonella testing. The intervention buildings were significantly less likely (p = 0.004) to be positive for Salmonella after C&D. The pre-restocking pigs had the highest likelihood (p<0.001) of being Salmonella positive (often with multiple serovars) and there was no significant difference between intervention and control buildings in Salmonella prevalence at the post-restocking visit (p = 0.199). However, the pigs housed in the intervention buildings were significantly less likely (p = 0.004) to be positive for Salmonella at slaughter age. Multivariable analysis suggested that cleaning all fixtures of buildings, leaving the pens empty for 2-3 days and using an effective disinfectant are factors significantly improving the likelihood of removing Salmonella contamination during C&D. Signs of rodents were recorded in all farms, but rodent activity and harbourage availability decreased between visits. All the rats tested were Salmonella negative. S. Typhimurium or its monophasic variants were isolated from 6 mouse carcasses in 3 farms where the same serovars were isolated from pigs. This study demonstrates that an appropriate C&D programme significantly reduces the likelihood of residual contamination in Salmonella positive pig buildings, and suggests a significant reduction in the prevalence of Salmonella in the pigs in appropriately cleaned and disinfected buildings when sampled before slaughter. Due to a high prevalence of infection in replacement pigs, control of Salmonella in pig farms is challenging. Rodents may also contribute to the carry-over of infection between batches. C&D is a useful measure to help reduce the number of infected pigs going to the slaughterhouse, but should be supplemented by other control measures along the pig breeding and production chain

    A Phase I Study of Pegylated Arginine Deiminase (Pegargiminase), Cisplatin, and Pemetrexed in Argininosuccinate Synthetase 1-Deficient Recurrent High-grade Glioma.

    PURPOSE: Patients with recurrent high-grade gliomas (HGG) are usually managed with alkylating chemotherapy ± bevacizumab. However, prognosis remains very poor. Preclinically, we showed that HGGs are a target for arginine depletion with pegargiminase (ADI-PEG20) due to epimutations of argininosuccinate synthetase (ASS1) and/or argininosuccinate lyase (ASL). Moreover, ADI-PEG20 disrupts pyrimidine pools in ASS1-deficient HGGs, thereby impacting sensitivity to the antifolate, pemetrexed. PATIENTS AND METHODS: We expanded a phase I trial of ADI-PEG20 with pemetrexed and cisplatin (ADIPEMCIS) to patients with ASS1-deficient recurrent HGGs (NCT02029690). Patients were enrolled (01/16-06/17) to receive weekly ADI-PEG20 36 mg/m2 intramuscularly plus pemetrexed 500 mg/m2 and cisplatin 75 mg/m2 intravenously once every 3 weeks for up to 6 cycles. Patients with disease control were allowed ADI-PEG20 maintenance. The primary endpoints were safety, tolerability, and preliminary estimates of efficacy. RESULTS: Ten ASS1-deficient heavily pretreated patients were treated with ADIPEMCIS therapy. Treatment was well tolerated with the majority of adverse events being Common Terminology Criteria for Adverse Events v4.03 grade 1-2. The best overall response was stable disease in 8 patients (80%). Plasma arginine was suppressed significantly below baseline with a reciprocal increase in citrulline during the sampling period. The anti-ADI-PEG20 antibody titer rose during the first 4 weeks of treatment before reaching a plateau. Median progression-free survival (PFS) was 5.2 months (95% confidence interval (CI), 2.5-20.8) and overall survival was 6.3 months (95% CI, 1.8-9.7). CONCLUSIONS: In this recurrent HGG study, ADIPEMCIS was well tolerated and compares favorably to historical controls. Additional trials of ADI-PEG20 in HGG are planned
