    Digital Inclusion of Low-Literate Adults: Challenging the Sequential Underpinnings of the Digital Divide

    Contemporary models of digital inclusion and the digital divide assume that developing the digital literacy that enables individuals to participate in society is a sequential and linear process that is more or less similar for all individuals in all contexts and requires basic linguistic skills. This paper challenges these understandings, arguing that such a technical, normative perspective excludes marginalized and disadvantaged publics, such as low-(digital) literate citizens. Based on a longitudinal ethnographic study of low-literate Dutch adults, we show that the often-described causal relation between (digital) literacies, (digital) participation, and (digital) inclusion is not as evident as it seems and neglects the important socio-cultural contexts through which (digital) literacies are often gained and enacted in everyday practice. Consequently, we argue that current conceptualizations of (digital) inclusion and (digital) participation need to be rethought in terms of the limitations, potential, and capabilities of low-literate people

    Digital Literacies as Socially Situated Pedagogical Processes: Genealogically Understanding Media, Information, and Digital Literacies

    Despite the increasing importance of digital literacies for citizens to be able to participate in society, there is little scholarly agreement over what digital literacies entail. This conceptual ambiguity hinders the translation of digital literacies into educational programs and policies that foster citizens' digital literacies and inclusion. While various authors have attempted to define digital literacy separately and in relation to other concepts, such as information literacy and media literacy, little attention has been paid to the historical backdrop of these concepts. By tracing the historical development of three literacies (media-, information-, and digital literacy), we reflect on how societal demands shaped conceptual frameworks of these literacies and how these conceptualizations are situated within the broader pedagogical systems that aim to enable participation in digital societies. Using a genealogical approach, we explore and describe the changes in definition, understanding, and enactment of the three literacies, which illustrate how these concepts have developed towards the conceptual frameworks we employ today. Based on this analysis, we argue that digital literacies must be flexible to anticipate challenges that result from the rise of new technologies and need to be appropriated within different socio-cultural contexts. We pledge for an understanding of digital literacies as socially situated pedagogical processes aimed at the way citizens appropriate digital practices within their daily lives. This implies shifting away from formulating one-size-fits-all understandings based upon generic uses of digital technologies. Instead, we must appropriate the understandings of digital literacies based upon their socio-technical, cultural, political, economic, and material dimensions

    Abundance of correctly folded RNA motifs in sequence space, calculated on computational grids

    Although functional RNA molecules are known to be biased in overall composition, the effects of background composition on the probability of finding a particular active site by chance has received little attention. The probability of finding a particular motif has important implications both for understanding the distribution of functional RNAs in ancient and modern organisms with varying genome compositions and for tuning SELEX pools to optimize the chance of finding specific functions. Here we develop a new method for calculating the probability of finding a modular motif containing base-paired regions, and use a computational grid to fold several hundred million random RNA sequences containing the core elements of the isoleucine aptamer and the hammerhead ribozyme to estimate the probability that a sequence containing these structural elements will fold correctly when isolated from background sequences of different compositions. We find that the two motifs are most likely to be found in distinct regions of compositional space, and that the regions of greatest abundance are influenced by the probability of finding the conserved bases, finding the flanking helices, and folding, in that order of importance. Additionally, we can refine our estimates of the number of random sequences required for a 50% probability of finding an example of each site in unbiased random pools of length 100 to 4.1 × 10(9) for the isoleucine aptamer and 1.6 × 10(10) for the hammerhead ribozyme. These figures are consistent with the facile recovery of these motifs from SELEX experiments

    The self-referential method combined with thermodynamic integration

    The self-referential method [M. B. Sweatman, Phys. Rev. E 72, 016711 (2005)] for calculating the free energy of crystalline solids via molecular simulation is combined with thermodynamic integration to produce a technique that is convenient and efficient. Results are presented for the chemical potential of hard sphere and Lennard-Jones face centered cubic crystals that agree well with this previous work. For the small system sizes studied, this technique is about 100 times more efficient than the parameter hopping technique used previously

    Human and cervid osseous materials used for barbed point manufacture in Mesolithic Doggerland

    Barbed bone points originally deposited in Doggerland are regularly collected from the shores of the Netherlands. Their typology and direct 14C dating suggest they are of Mesolithic age. However, the species of which the barbed points were made cannot be identified based on morphological criteria. The bones used to produce the barbed points have been intensively modified during manufacture, use, and post-depositional processes. Here, we taxonomically assess ten barbed points found on the Dutch shore using mass spectrometry and collagen peptide mass fingerprinting alongside newly acquired 14C ages and δ13C and δ15N measurements. Our results demonstrate a sufficient preservation of unmodified collagen for mass spectrometry-based taxonomic identifications of bone and antler artefacts which have been preserved in marine environments since the beginning of the Holocene. We show that Homo sapiens bones as well as Cervus elaphus bones and antlers were transformed into barbed points. The 14C dating of nine barbed points yielded uncalibrated ages between 9.5 and 7.3 ka 14C BP. The δ13C and δ15N values of the seven cervid bone points fall within the range of herbivores, recovered from the North Sea, whereas the two human bone points indicate a freshwater and/or terrestrial fauna diet. The wide-scale application of ZooMS is a critical next step towards revealing the selection of species for osseous-tool manufacture in the context of Mesolithic Doggerland, but also further afield. The selection of Cervus elaphus and human bone for manufacturing barbed points in Mesolithic Doggerland is unlikely to have been opportunistic and instead seems to be strategic in nature. Further, the occurrence of Homo sapiens and Cervus elaphus bones in our random and limited dataset suggests that the selection of these species for barbed point production was non-random and subject to specific criteria. By highlighting the transformation of human bones into barbed points – possibly used as weapons – our study provides additional evidence for the complex manipulation of human remains during the Mesolithic, now also evidenced in Doggerland

    Plasma lipidomics and coronary plaque changes: a substudy of the SMARTool clinical trial

We aimed to prospectively determine the association between lipid species quantified by a plasma lipidomic analysis and coronary plaque changes according to composition assessed by a quantitative serial analysis of coronary computed tomography angiography (CTA). Methods and results Patients with suspected coronary artery disease (CAD) undergoing baseline coronary CTA were prospectively enrolled by seven EU centres in the SMARTool study and submitted to clinical, molecular, and coronary CTA re-evaluation at follow-up (an inter-scan period of 6.39 ± 1.17 years). Out of 202 patients who were analysed in the SMARTool main clinical study, a lipidomic analysis was performed in 154 patients before the baseline coronary CTA, and this group was included in the present study. A quantitative CTA analysis was performed by using a separate core laboratory blinded from clinical data. In the univariable analysis, it was found that no lipid species were significantly associated with annual total and calcified plaque changes. After adjusting for clinical variables at baseline and statin use, it was found that three lipid species were significantly associated with non-calcified plaque progression. In detail, cholesteryl ester(20:3), sphingomyelin (SM)(40:3), and SM(41:1) were found to be positively related to non-calcified plaque progression (Bonferroni-adjusted P-values = 0.005, 0.016, and 0.004, respectively). Conclusion The current study showed an independent relationship between specific lipid species determined by a plasma lipidomic analysis and non-calcified coronary plaque progression assessed by a serial, quantitative coronary CTA analysis.This work is part-funded by the European Commission SMARTool (Simulation Modelling of coronary ARTery disease: a tool for clinical decision support—GA 689068).     Prevention of major depression in complex medically ill patients: preliminary results from a randomized, controlled trial

    Background: Depression is highly prevalent in patients with physical illness and is associated with a diminished quality of life and poorer medical outcomes. Objective: The authors evaluated whether a multifaceted intervention conducted by a psychiatric consultation-liaison nurse could reduce the incidence of major depression in rheumatology inpatients and diabetes outpatients with a high level of case complexity. Method: Of 247 randomized patients, the authors identified 100 patients with a high level of case complexity at baseline and without major depression (65 rheumatology and 35 diabetes patients). Patients were randomized to usual care (N = 53) or to a nurse-led intervention (N = 47). Main outcomes were the incidence of major depression and severity of depressive symptoms during a 1-year follow-up, based on quarterly assessments with standardized psychiatric interviews. Results: The incidence of major depression was 63% in usual-care patients and 36% in the intervention group. Effects of intervention on depressive symptoms were observed in outpatients with diabetes but not in rheumatology inpatients. Conclusion: These preliminary results based on subgroup analysis suggest that a multifaceted nurse-led intervention may prevent the occurrence of major depression in complex medically ill patients and reduce depressive symptoms in diabetes outpatients.     Antifibrinolytics attenuate inflammatory gene expression after cardiac surgery

    ObjectivesAnti-inflammatory effects of tranexamic acid and aprotinin, used to abate perioperative blood loss, are reported and might be of substantial clinical relevance. The study of messenger ribonucleic acid synthesis provides a valuable asset in evaluating the inflammatory pathways involved.MethodsWhole-blood messenger ribonucleic acid expression of 114 inflammatory genes was compared pre- and postoperatively in 35 patients randomized to receive either placebo, tranexamic acid, or aprotinin. These results were further confirmed by reverse transcription–polymerase chain reaction.ResultsOf the 23 genes exhibiting independently altered postoperative gene expression levels, 8 were restricted to the aprotinin group only (growth differentiation factor 3, interleukin 19, interleukin 1 family member 7, transforming growth factor α, tumor necrosis factor superfamily 10, tumor necrosis factor superfamily 12, tumor necrosis factor superfamily 13B, vascular endothelial growth factor α), whereas both aprotinin and tranexamic acid altered gene expression of 3 genes as compared with placebo (FMS-related tyrosine kinase 3 ligand, growth differentiation factor 5, interferon-α8). In general, less upregulation of pro-inflammatory, and more upregulation of anti-inflammatory, genes was observed for patients treated with antifibrinolytics. Gene expression affected by aprotinin coded mostly for proteins that function through serine proteases.ConclusionsThis study demonstrates that the use of tranexamic acid and aprotinin results in altered inflammatory pathways on the genomic expression level. We further demonstrate that the use of aprotinin leads to significant attenuation of the immune response, with several inhibitory effects restricted to the use of aprotinin only. The results aid in a better understanding of the targets of these drugs, and add to the discussion on which antifibrinolytic can best be used in the cardiac surgical patient

    Experimental avian paramyxovirus serotype-3 infection in chickens and turkeys

    Avian paramyxoviruses (APMV) are divided into nine serotypes. Newcastle disease virus (APMV-1) is the most extensively characterized, while relatively little information is available for the other APMV serotypes. In the present study, we examined the pathogenicity of two divergent strains of APMV-3, Netherlands and Wisconsin, in (i) 9-day-old embryonated chicken eggs, (ii) 1-day-old specific pathogen free (SPF) chicks and turkeys, and (iii) 2-week-old SPF chickens and turkeys. The mean death time in 9-day-old embryonated chicken eggs was 112 h for APMV-3 strain Netherlands and > 168 h for strain Wisconsin. The intracerebral pathogenicity index in 1-day-old chicks for strain Netherlands was 0.39 and for strain Wisconsin was zero. Thus, both strains are lentogenic. Both the strains replicated well in brain tissue when inoculated intracerebrally in 1-day-old SPF chicks, but without causing death. Mild respiratory disease signs were observed in 1-day-old chickens and turkeys when inoculated through oculonasal route with either strain. There were no overt signs of illness in 2-weeks-old chickens and turkeys by either strain, although all the birds seroconverted after infection. The viruses were isolated predominantly from brain, lungs, spleens, trachea, pancreas and kidney. Immunohistochemistry studies also showed the presence of large amount of viral antigens in both epithelial and sub-epithelial lining of respiratory and alimentary tracts. Our result suggests systemic spread of APMV-3 even though the viral fusion glycoprotein does not contain the canonical furin proteases cleavage site. Furthermore, there was little or no disease despite systemic viral spread and abundant viral replication in all the tissues tested
