    This paper presents some details from an observational evaluation of a computer assisted detection tool in mammography. The use of the tool, its strengths and weaknesses, are documented and its impact on reader's 'professional vision' (Goodwin 1994) considered. The paper suggests issues for the design, use and, importantly, evaluation of new technologies in everyday medical work, pointing to general issues concerning trust – users’ perception of the dependability of the evidence generated by such tools and suggesting that evaluations require an emphasis on the complex issue of what technologies afford their users in everyday work

    Abstractions, accounts and grid usability

    The vision of the Grid is one of seamless, virtual and constantly changing resources where users need not concern themselves about details, such as exactly where an application is running or where their data is being stored. However, seamless and virtual often imply a lack of control that users may be wary of, or even opposed to. Drawing upon our studies of HCI and of collaborative work, this paper examines whether the Grid development community should be taking this vision literally and argues for the need for accountability of systems ‘in interaction’. We give examples of an alternative approach that seeks to provide ways in which administrators, technical support and user communities can make sense of the behaviour of the complex socio-technical ensembles that are the reality of Grids

    A biphasic model of lifespan in nematode Caenorhabditis elegans worm

    Ageing research focuses on identifying lifespan modifiers and understanding and appropriately interpreting their effects. One of the most relevant quantities being studied is the shape of the survival curve that can reveal crucial information on the mechanism of action. Here, we introduce a bilogistic model to describe the shape of the lifespan curves of Caenorhabditis elegans populations. Using the corrected Akaike information criterion and the RMSE as goodness-of-fit tests, we show that the bilogistic model provides a better fit to the experimental data from nematode worms than other mathematical models and can identify and confirm biphasic lifespan data. Our parametric model offers a method to interpret replicate experiments data in terms of the shape parameters of the lifespan curve and enables robust statistical analysis of intra- and inter-group variance. We apply the model to novel lifespan data from C. elegans and Drosophila melanogaster and provide a rational statistical analysis of lifespan modifiers such as temperature and daf-16/FOXO mutation

    Characterization of a Bacillus anthracis spore coat-surface protein that influences coat-surface morphology

    Bacterial spores are encased in a multilayered proteinaceous shell, called the coat. In many Bacillus spp., the coat protects against environmental assault and facilitates germination. In Bacillus anthracis , the spore is the etiological agent of anthrax, and the functions of the coat likely contribute to virulence. Here, we characterize a B. anthracis spore protein, called CotΒ, which is encoded only in the genomes of the Bacillus cereus group. We found that CotΒ is synthesized specifically during sporulation and is assembled onto the spore coat surface. Our analysis of a cotΒ null mutant in the Sterne strain reveals that CotΒ has a role in determining coat-surface morphology but does not detectably affect germination.     The GridWise Architecture Council was formed by the U.S. Department of Energy to promote and enable interoperability among the many entities that interact with the electric power system. This balanced team of industry representatives proposes principles for the development of interoperability concepts and standards. The Council provides industry guidance and tools that make it an available resource for smart grid implementations. In the spirit of advancing interoperability of an ecosystem of smart grid devices and systems, this document presents a model for evaluating the maturity of the artifacts and processes that specify the agreement of parties to collaborate across an information exchange interface. You are expected to have a solid understanding of large, complex system integration concepts and experience in dealing with software component interoperation. Those without this technical background should read the Executive Summary for a description of the purpose and contents of the document.     Comparative analysis of the transcriptome across distant species

    The transcriptome is the readout of the genome. Identifying common features in it across distant species can reveal fundamental principles. To this end, the ENCODE and modENCODE consortia have generated large amounts of matched RNA-sequencing data for human, worm and fly. Uniform processing and comprehensive annotation of these data allow comparison across metazoan phyla, extending beyond earlier within-phylum transcriptome comparisons and revealing ancient, conserved features. Specifically, we discover co-expression modules shared across animals, many of which are enriched in developmental genes. Moreover, we use expression patterns to align the stages in worm and fly development and find a novel pairing between worm embryo and fly pupae, in addition to the embryo-to-embryo and larvae-to-larvae pairings. Furthermore, we find that the extent of non-canonical, non-coding transcription is similar in each organism, per base pair. Finally, we find in all three organisms that the gene-expression levels, both coding and non-coding, can be quantitatively predicted from chromatin features at the promoter using a 'universal model' based on a single set of organism-independent parameters

    SARS-CoV-2-specific nasal IgA wanes 9 months after hospitalisation with COVID-19 and is not induced by subsequent vaccination

    BACKGROUND: Most studies of immunity to SARS-CoV-2 focus on circulating antibody, giving limited insights into mucosal defences that prevent viral replication and onward transmission. We studied nasal and plasma antibody responses one year after hospitalisation for COVID-19, including a period when SARS-CoV-2 vaccination was introduced. METHODS: In this follow up study, plasma and nasosorption samples were prospectively collected from 446 adults hospitalised for COVID-19 between February 2020 and March 2021 via the ISARIC4C and PHOSP-COVID consortia. IgA and IgG responses to NP and S of ancestral SARS-CoV-2, Delta and Omicron (BA.1) variants were measured by electrochemiluminescence and compared with plasma neutralisation data. FINDINGS: Strong and consistent nasal anti-NP and anti-S IgA responses were demonstrated, which remained elevated for nine months (p < 0.0001). Nasal and plasma anti-S IgG remained elevated for at least 12 months (p < 0.0001) with plasma neutralising titres that were raised against all variants compared to controls (p < 0.0001). Of 323 with complete data, 307 were vaccinated between 6 and 12 months; coinciding with rises in nasal and plasma IgA and IgG anti-S titres for all SARS-CoV-2 variants, although the change in nasal IgA was minimal (1.46-fold change after 10 months, p = 0.011) and the median remained below the positive threshold determined by pre-pandemic controls. Samples 12 months after admission showed no association between nasal IgA and plasma IgG anti-S responses (R = 0.05, p = 0.18), indicating that nasal IgA responses are distinct from those in plasma and minimally boosted by vaccination. INTERPRETATION: The decline in nasal IgA responses 9 months after infection and minimal impact of subsequent vaccination may explain the lack of long-lasting nasal defence against reinfection and the limited effects of vaccination on transmission. These findings highlight the need to develop vaccines that enhance nasal immunity. FUNDING: This study has been supported by ISARIC4C and PHOSP-COVID consortia.
