    Comparison of bilateral vs. unilateral-biased strength and power training interventions on measures of physical performance in elite youth soccer players

    The aim of the present study was to compare the effects of bilateral and unilateral-biased strength and power training programs on measures of physical performance in male youth soccer players. Twenty-three elite youth players (age: 17.6 1.2 years) were randomly assigned to either a unilateral (n = 11) or a bilateral (n = 12) group, who completed a strength and power intervention, twice per week for 6-weeks. The unilateral group completed rear foot elevated split squats (RFESS), single leg countermovement jumps (SLCMJ), single leg drop jumps (SLDJ) and single leg broad jumps (SLBJ). The bilateral group intervention performed back squats, countermovement jumps (CMJ), drop jumps (DJ) and broad jumps (BJ). A 2 x 2 repeated measures ANOVA showed no between-group differences. However, within-group differences were evident. The bilateral training group showed significant (p < 0.05) improvements in back squat strength (d = 1.27; %Δ = 26.01), RFESS strength (d = 1.64; %Δ = 23.34), BJ (d = 0.76; %Δ = 5.12), 10 m (d = -1.17; %Δ = 4.29) and 30 m (d = -0.88; %Δ = 2.10) performance. The unilateral group showed significant (p < 0.05) improvements in RFESS strength (d = 1.40; %Δ = 33.29), SLCMJ on the left leg (d = 0.76; %Δ = 9.84), SLBJ on the left leg (d = 0.97; %Δ = 6.50), 10 m (d = -1.50; %Δ = 5.20), and 505 on the right leg (d = -0.78; %Δ = 2.80). Standardised mean differences showed that bilateral training favoured improvements in back squat strength and unilateral training favoured improvements in RFESS strength, SLDJ on the right leg and 505 on the right leg. These results show that although both training interventions demonstrated trivial to large improvements in physical performance, the notion of training specificity was evident with unilateral training showing greater improvements in unilateral test measures

    Implementation of Innovative Technology for Evaluating High-speed Rail Passenger Transportation

    The size and content of consumer value (CV) of the service for the transportation of passengers by the railways ofUkrainein long-distance communication by nighttime and daytime trains is determined. A comparison is made of the results of two approaches to researching the attitude of customers to the service – traditional (point-based) and new, taking into account the specific requirements of the total management of the customer value of the service. The directions of the managerial impact of CV on the passenger transportation in long-distance communication by the railways ofUkrainebased on the attitude of customers to the disadvantages and advantages of this service are determined.In three focus groups, discussions were held about travel by Ukrainian railways by daytime and nighttime trains. Based on the results, a list of the disadvantages and advantages that form the consumer value of such a transport service is formed. The list of 19 positive and negative statements reflected all stages of the trip by rail. Behind it, the Likert scale and the corresponding form for conducting an interview were developed. The number of respondents is 823. The sample is random.The results of the polls confidently prove that the prejudiced attitude of passengers towards daytime transportation has been overcome. The ratio of passengers of the railways ofUkrainein daytime transportation in long-distance traffic is better than in nighttime.Only one fifth of those traveling daytime and nighttime are fully satisfied with the level of advantages and disadvantages, which is not enough for most customers to make repeat purchases. However, such a hypothesis needs to be verified in future studies.The feeling of satisfaction among passengers of daytime trains with a positive value of consumer value does not arise in 29 %, which is a threat to the railway operator company. Women are generally worse off on transportation services than men.Personal experience using the train in general does not affect the attitude. However, the experience of using a nighttime train significantly reduces the consumer value of the transport service.The main conclusion is that when several customer segments are served simultaneously in a common space, the total management of the customer value of the service and the corresponding research methods are productive

    Intercultural Communication and Values-Based Orientations of Youth: Social Mobility and Problems of Self-Realisation

    The ability to interact at the level of interpersonal communication allows to say that young people not only perceive society as an environment for their self-realisation, but also use it to gain new experience, that can determine the development of an individual. The relevance of the study is determined by the fact that each act of communication must be implemented for the purposes of development under certain conditions that affect not only the communication environment itself, but also other forms of interaction at the level of the individual, communality or community. The novelty of the subject matter is determined by the fact that the formation of intercultural communication is possible only if programs of social mobility and, accordingly, cultural exchange are implemented. The authors show that such progress is most likely to be achieved in the external environment, which is determined by the presence of external influences. The authors of the paper primarily refer to this effect as the environment of an educational institution. The leading research method is a sociological survey and the method of analysis, which make it possible to comprehensively consider the value priorities and preferences that are characteristic of modern student youth. The practical significance of the study is determined by structuring and factor analysis, which allows to distribute the cultural layer into various social groups and ensure youth intercultural exchange both in the context of local student communication and in the context of an increase in the level of academic mobility

    Reforme energetskog sektora i nove poslovne mogućnosti nametnule su potrebu stjecanja i prenošenja znanja i vještina svim sudionicima koji preuzimaju nove uloge i odgovornosti u energetskom sektoru. Stoga su Fakultet elektrotehnike i računarstva Sveučilišta u Zagrebu (FER), Hrvatska elektroprivreda d.d. (HEP d.d.) i norveška tvrtka Technor Energy AS pokrenule projekt uspostave Trening centra za trgovanje energijom (Training Centre for Energy Trading TCET). U članku je predstavljen rezultat ove suradnje – pisani materijali koji služe kao podloga za učenje i simulator tržišta koji omogućava različitim tržišnim sudionicima, iz raznih zemalja i s različitim tržišnim interesima, da kroz igru iskušaju tržišno natjecanje. Ovo je prvi trening centar takve vrste u Europi, a projekt njegove uspostave financiralo je Ministarstvo vanjskih poslova Kraljevine Norveške i HEP d.d.The current reforms in the energy sector and the resulting new business opportunities have imposed a need for knowledge and skills to be acquired by or imparted to all those who will assume new roles and responsibilities in the energy sector. To this end the Faculty of Electrical Engineering and Computering of the University of Zagreb (FER), Hrvatska elektroprivreda d.d. – The Croatian Electricity Company (HEP d.d.) and the Norwegian company Technor Energy AS launched a project to establish the Training Centre for Energy Trading (TCET). This article presents the results of this co-operation – printed materials to serve as teaching aid and a market simulator to help various market participants from different countries and with different market interests to try their hand at market competition by playing games. This is the first training centre project of this kind in Europe, co-financed by the Ministry for Foreign Affairs of the Kingdom of Norway and HEP d.d

    The alpha 7 nicotinic receptor agonist PHA-543613 hydrochloride inhibits <i>Porphyromonas gingivalis</i>-induced expression of interleukin-8 by oral keratinocytes

    Objective: The alpha 7 nicotinic receptor (α7nAChR) is expressed by oral keratinocytes. α7nAChR activation mediates anti-inflammatory responses. The objective of this study was to determine if α7nAChR activation inhibited pathogen-induced interleukin-8 (IL-8) expression by oral keratinocytes.&lt;p&gt;&lt;/p&gt; Materials and methods: Periodontal tissue expression of α7nAChR was determined by real-time PCR. OKF6/TERT-2 oral keratinocytes were exposed to &lt;i&gt;Porphyromonas gingivalis&lt;/i&gt; in the presence and absence of a α7nAChR agonist (PHA-543613 hydrochloride) alone or after pre-exposure to a specific α7nAChR antagonist (α-bungarotoxin). Interleukin-8 (IL-8) expression was measured by ELISA and real-time PCR. Phosphorylation of the NF-κB p65 subunit was determined using an NF-κB p65 profiler assay and STAT-3 activation by STAT-3 in-cell ELISA. The release of ACh from oral keratinocytes in response to &lt;i&gt;P. gingivalis&lt;/i&gt; lipopolysaccharide was determined using a GeneBLAzer M3 CHO-K1-blacell reporter assay.&lt;p&gt;&lt;/p&gt; Results: Expression of α7nAChR mRNA was elevated in diseased periodontal tissue. PHA-543613 hydrochloride inhibited &lt;i&gt;P. Gingivalis&lt;/i&gt;-induced expression of IL-8 at the transcriptional level. This effect was abolished when cells were pre-exposed to a specific α7nAChR antagonist, α-bungarotoxin. PHA-543613 hydrochloride downregulated NF-κB signalling through reduced phosphorylation of the NF-κB p65-subunit. In addition, PHA-543613 hydrochloride promoted STAT-3 signalling by maintenance of phosphorylation. Furthermore, oral keratinocytes upregulated ACh release in response to &lt;i&gt;P. Gingivalis&lt;/i&gt; lipopolysaccharide.&lt;p&gt;&lt;/p&gt; Conclusion: These data suggest that α7nAChR plays a role in regulating the innate immune responses of oral keratinocytes.&lt;p&gt;&lt;/p&gt

    Lineage-specific compaction of Tcrb requires a chromatin barrier to protect the function of a long-range tethering element

    Gene regulation relies on dynamic changes in three-dimensional chromatin conformation, which are shaped by composite regulatory and architectural elements. However, mechanisms that govern such conformational switches within chromosomal domains remain unknown. We identify a novel mechanism by which cis-elements promote long-range interactions, inducing conformational changes critical for diversification of the TCRβ antigen receptor locus (Tcrb). Association between distal Vβ gene segments and the highly expressed DβJβ clusters, termed the recombination center (RC), is independent of enhancer function and recruitment of V(D)J recombinase. Instead, we find that tissue-specific folding of Tcrb relies on two distinct architectural elements located upstream of the RC. The first, a CTCF-containing element, directly tethers distal portions of the Vβ array to the RC. The second element is a chromatin barrier that protects the tether from hyperactive RC chromatin. When the second element is removed, active RC chromatin spreads upstream, forcing the tether to serve as a new barrier. Acquisition of barrier function by the CTCF element disrupts contacts between distal Vβ gene segments and significantly alters Tcrb repertoires. Our findings reveal a separation of function for RC-flanking regions, in which anchors for long-range recombination must be cordoned off from hyperactive RC landscapes by chromatin barriers
