    Immune-Endocrine Interactions in the Fish Gonad during Infection: An Open Door to Vertical Transmission

    The interaction between the immune and endocrine systems has long been recognized in vertebrates. In fish, it is known that the prevalence and intensity of such infections are higher in males than in females and probably related to sex steroid hormone levels. In addition, the immune response in the fish gonad tissues is specifically regulated to prevent infertility. This condition is used by some pathogens to colonize the fish gonad, evade the systemic immune response, and so spread to the progeny. This review brings up to date our knowledge concerning fish gonad immunity and its regulation, immune-endocrine interactions, and how some pathogens use this tissue to spread to the progeny through vertical transmission. More specifically, we will look at the case of the European sea bass and nodavirus (NNV). Sea bass is a very susceptible fish species to NNV infections, and this virus has been associated to vertical transmission since it is detected in gonad fluids as well as in testicular gametes. In fact, sea bass immunity in the gonad is regulated in a very different way to other target tissues (brain and retina) or immune-relevant tissues (head-kidney or spleen).Versión del edito

    First report on the occurrence of an actinosporean stage (Myxozoa) in oligochaetes from Spanish freshwaters

    Oligochaetes living in the Mijares River close to the Sitjar reservoir, and polychaetes from the brackish and marine waters of a channel flowing into the Mediterranean Sea, both in the province of Castellon (Spain), were examined for the presence of actinosporea. An aurantiactinomyxon was isolated from 60 specimens of the oligochaete Branchiura sowerbyi collected from the river, but no actinospores were isolated from 160 polychaetes collected from the sea channel. The aurantiactinospores were detected by the cell-well plate method. The detected species are not identical with any of the aurantiactinomyxon forms hitherto described in the literature. This is the first report on the occurrence of an actinosporea in Spanish waters

    In vitro testing of alternative synthetic and natural antiparasitic compounds against the monogenean sparicotyle chrysophrii

    Gill monogenean Sparicotyle chrysophrii is considered the most detrimental fish parasite to the Mediterranean aquaculture. Treatment of sparicotylosis relies on frequent gill inspections correlated with the seasonal increase in seawater temperature, application of functional feeds, and treatments with formalin baths where permitted. While the latter is bound to be banned in Europe, other synthetic anthelminthics, such as praziquantel and ivermectin, are prone to induce resistance in the parasites. Therefore, we investigated, in vitro, 14 synthetic and natural compounds against adult S. chrysophrii, developing dose–response modelsm and estimated toxicity levels at 20%, 50%, and 80% parasite mortality. Bactericidal activity of target compounds was also tested in two important aquaculture bacteria; Vibrio harveyi and V. anguillarum, while their potential host toxicity was evaluated in gilthead seabream SAF-1 cell line. Synthetic compound bithionate sodium exerted the most potent toxicity against the monogenean, no host cytotoxicity, and a medium and high potency against two bacterial pathogens. In comparison, target natural compounds were approximately 20 (cedrol) or up to 154 times (camphor) less toxic for the monogenean. Rather than completely dismissing natural compounds, we suggest that their application in combination with synthetic drugs, especially if administered in the feed, might be useful in sparicotylosis treatment

    An overview of the reproductive cycle of cultured specimens of a potential candidate for Mediterranean aquaculture, Umbrina cirrosa

    The shi drum (Umbrina cirrosa), belonging to the Sciaenidae family of fish (Nelson et al., 2016) has attracted attention as it is an excellent diversification species in Mediterranean aquaculture to combat the low prices of the most cultured species due to the appearance of new producing countries in the Mediterranean area. In this manuscript we describe for the first time the reproductive cycle of captive shi drum specimens that are able to spontaneously spawn in captivity. Our data show that spermatogenesis started in one year old shi drum spe- cimens, although the fish were not mature enough to complete the reproductive cycle. Thus, three year old specimens showed multiple spawns with around 65% of the eggs fertilized. In mature males, levels of 11-ke- totestosterone (11KT), testosterone (T) and progesterone (P) were detected in serum and gonad, while 17β- estradiol (E2) was only detected in gonad. Testicular levels of E2 and P suggest that these hormones might have a role in spawning and in the subsequent spermatogonial proliferation. In mature females, however, P and E2 were detected in serum and gonad, while T and 11KT were only detected in gonad. The pattern shown by P in the ovary indicates a link between this hormone and ovarian renewal and a delay in vitellogenesis, which would explain why the vitellogenic stage of shi drum is quite short. The level of T and 11KT in the ovary suggests a role in oocyte maturation and spawning. The differences observed between sexes in the prevalence of some infection and the important economic losses produced by some vertically transmitted pathogens; have increase the at- tention to the interactions between the immune and reproductive systems. In this study we observed differences between sexes in the protease and bactericide activity levels. Thus, this manuscript represents the most complete overview to date of the shi drum reproductive cycle.Versión del editor2,04

    The impact of co-infections on fish: a review

    International audienceAbstractCo-infections are very common in nature and occur when hosts are infected by two or more different pathogens either by simultaneous or secondary infections so that two or more infectious agents are active together in the same host. Co-infections have a fundamental effect and can alter the course and the severity of different fish diseases. However, co-infection effect has still received limited scrutiny in aquatic animals like fish and available data on this subject is still scarce. The susceptibility of fish to different pathogens could be changed during mixed infections causing the appearance of sudden fish outbreaks. In this review, we focus on the synergistic and antagonistic interactions occurring during co-infections by homologous or heterologous pathogens. We present a concise summary about the present knowledge regarding co-infections in fish. More research is needed to better understand the immune response of fish during mixed infections as these could have an important impact on the development of new strategies for disease control programs and vaccination in fish
