    Análise in vivo do selamento do canal protéico com diferentes materais adesivos

    This in vivo study analyzed the sealing ability of two adhesives in post spaces, cyanoacrylate (Super Bonder® - Henkel Loctite Adesivos Ltda., Itapevi, SP, Brazil) and ScotchbondTM Multi-Purpose (3M Dental Products, St. Paul, MN, USA), testing the hypothesis that their use would result in a decreased leakage through the remaining filling material. Forty extracted premolars (80 root canals) of dogs were used. The root canals were cleaned, shaped and filled by the lateral condensation technique using Sealer 26TM. The post space was created removing two thirds of the filling material within the root canal. The canals were randomly divided in three groups, which were treated as follows: Group A received the ScotchbondTM Multi-Purpose adhesive system; for Group B the cyanoacrylate adhesive, Super BonderTM, was employed; and no adhesive was applied into the post space for Group C (control group). A glass ionomer provisional restoration was placed allowing the sealer to set for 72 hours. Then the restoration was removed and the root canal was exposed to the oral environment for 45 days. The dogs were then killed and their jaws were removed. The post spaces were filled with India ink and the teeth were restored for 96 hours. Afterwards, the teeth were extracted and the roots were sectioned at the furcation for allocation to their specific groups. The teeth were turned transparent and the quantitative analysis of leakage was performed using light microscopy. The results showed no significant differences between groups, rejecting the initial hypothesis. Leakage occurred in a great extent in all specimens. Therefore, sealing post spaces with the adhesives used in this study was not an effective method to prevent microleakage.Esse estudo in vivo analisou a capacidade de selamento do canal protético com dois adesivos, o Super Bonder® (Henkel Loctite Adesivos Ltda., Itapevi, SP) e o Scotchbond® Multi-Purpose (3M Dental Products, St. Paul, MN, EUA), testando a hipótese de que a aplicação desses adesivos resulta em menor infiltração de corante no material obturador remanescente. Quarenta dentes pré-molares (80 canais) de cães foram obturados pela técnica da condensação lateral usando o cimento endodôntico Sealer 26®. O canal protético foi preparado removendo-se 2/3 da obturação do canal radicular. Os canais preparados foram distribuídos, aleatoriamente, em três grupos: no grupo A, foi realizado o selamento do canal protético com o adesivo dentinário; no grupo B, esta impermeabilização foi realizada com o Super Bonder®; e no grupo C, controle, não foi aplicado qualquer material às paredes do canal protético. Os dentes foram selados, provisoriamente, com cimento de ionômero de vidro por 72 horas, objetivando a presa do cimento endodôntico. Removido o selamento coronário, os canais ficaram expostos ao meio bucal por 45 dias, quando os cães foram mortos e os maxilares removidos. Tinta nanquim foi colocada no canal protético e os dentes foram selados por 96 horas. Depois de extraídos, os dentes foram seccionados na região da furca e cada raiz alocada em seu respectivo grupo. Feita a diafanização, foi realizada a análise quantitativa da infiltração do corante com lupa esteroscópica. Os resultados mostraram não haver diferenças entre os três grupos experimentais, negando a hipótese inicial. Houve infiltração do corante no remanescente obturador de todas as raízes analisadas

    Climatologia da função frontogenética sobre a América do Sul em um cenário de clima presente e futuro

    Os campos climatológicos da Função Frontogenética de Petterssen (FF) foram calculados com a partir do modelo global HadGEM2-ES para uma série 1975-2005. Os primeiros resultados mostram-se concordantes com aqueles obtidos anteriormente\ud por outros autores, indicando uma maior atividade de sistemas frontais durante o inverno e início da primavera.     Pathogenic SPTBN1 variants cause an autosomal dominant neurodevelopmental syndrome

    SPTBN1 mutations cause a neurodevelopmental syndrome characterized by intellectual disability, language and motor delays, autism, seizures and other features. The variants disrupt beta II-spectrin function and disturb cytoskeletal organization and dynamics. SPTBN1 encodes beta II-spectrin, the ubiquitously expressed beta-spectrin that forms micrometer-scale networks associated with plasma membranes. Mice deficient in neuronal beta II-spectrin have defects in cortical organization, developmental delay and behavioral deficiencies. These phenotypes, while less severe, are observed in haploinsufficient animals, suggesting that individuals carrying heterozygous SPTBN1 variants may also show measurable compromise of neural development and function. Here we identify heterozygous SPTBN1 variants in 29 individuals with developmental, language and motor delays;mild to severe intellectual disability;autistic features;seizures;behavioral and movement abnormalities;hypotonia;and variable dysmorphic facial features. We show that these SPTBN1 variants lead to effects that affect beta II-spectrin stability, disrupt binding to key molecular partners, and disturb cytoskeleton organization and dynamics. Our studies define SPTBN1 variants as the genetic basis of a neurodevelopmental syndrome, expand the set of spectrinopathies affecting the brain and underscore the critical role of beta II-spectrin in the central nervous system

    Value of computed tomography texture analysis for prediction of perioperative complications during laparoscopic partial nephrectomy in patients with renal cell carcinoma.

    Tumorous texture is a marker for tumor tissue inhomogeneity. Based on this assumption, this study aims to evaluate the value of computed tomography texture analysis for imaging-based prediction of perioperative complications during laparoscopic partial tumor nephrectomy.A total of 106 patients with histologically confirmed renal cell carcinoma and pre-operative CT were included and volumetric texture analysis of the tumors was performed by two readers. Texture analysis parameter ratios and differences were calculated using the kidney parenchyma as reference ("reference-corrected"). Regression analysis was performed, regarding the value of the texture analysis parameters, for assessment of the tumor nuclear grade and the prediction of peri- and postoperative complications and approximated blood loss. Moreover, the inter-rater agreement in terms of the intra-class correlation coefficient (ICC) was calculated.Regarding the reference-corrected values, the predictive value of texture analysis parameters for severe perioperative complications was highest for the standard deviation of the mean attenuation (Area under curve/AUC, .615; sensitivity, 93.8%, specificity, 30.0%), followed by the uniformity (AUC, .599; sensitivity, 62.5%, specificity, 60.0%), and the uniformity of distribution of positive pixels (AUC, .597; sensitivity, 62.5%; specificity, 61.1%). Regarding the blood loss, the uniformity of positive pixel values (UPP; AUC, 0.638), uniformity (AUC, 0.635), and entropy (AUC, 0.633) yielded the best predictive values, whilst the tumor grade was a weaker predictor (AUC, 0.574). The applied texture analysis parameters did not correlate with the time of surgery or the warm ischemic time. All measured parameters were better predictors for complications than the tumor diameter alone.     Comparação de Metodologias de Preenchimento de Falhas em Dados Meteorológicos para Estações no Sul do Brasil

    Resumo A rede de estações meteorológicas no Brasil é muito recente, com poucas estações no país com mais de 100 anos de dados. Além disso, muitas das séries oriundas destas estações apresentam falhas, seja por falta de medição (no caso das estações convencionais), seja por falha nos equipamentos (no caso das estações automáticas). Encontram-se diferentes métodos na literatura para o preenchimento destas falhas. Neste trabalho são comparados seis métodos, os quais foram aplicados a séries de dados mensais conhecidas de temperatura média compensada e precipitação para estações meteorológicas localizadas no estado do Rio Grande do Sul, região sul do Brasil. Os resultados para temperatura média compensada indicaram os métodos de regressão linear múltipla e ponderação regional como os mais adequados para estimativas de dados faltantes enquanto que para precipitação não houve um método a se sobressair

    Virulence Profiles of Vibrio vulnificus in German Coastal Waters, a Comparison of North Sea and Baltic Sea Isolates

    Vibrio vulnificus is a halophilic bacterium of coastal environments known for sporadically causing severe foodborne or wound infections. Global warming is expected to lead to a rising occurrence of V. vulnificus and an increasing incidence of human infections in Northern Europe. So far, infections in Germany were exclusively documented for the Baltic Sea coast, while no cases from the North Sea region have been reported. Regional variations in the prevalence of infections may be influenced by differences in the pathogenicity of V. vulnificus populations in both areas. This study aimed to compare the distribution of virulence-associated traits and genotypes among 101 V. vulnificus isolates from the Baltic Sea and North Sea in order to assess their pathogenicity potential. Furthermore, genetic relationships were examined by multilocus sequence typing (MLST). A high diversity of MLST sequences (74 sequence types) and differences regarding the presence of six potential pathogenicity markers were observed in the V. vulnificus populations of both areas. Strains with genotypes and markers associated with pathogenicity are not restricted to a particular geographic region. This indicates that lack of reported cases in the North Sea region is not caused by the absence of potentially pathogenic strains
