    Rethinking Leadership: The Changing Role of Principal Supervisors

    In the fall of 2012, the Council of the Great City Schools launched a two-part study of the ways principal supervisors are selected, supported, and evaluated in major school districts across the country. The first part involved a survey administered to district staff serving as principal supervisors in the fall of 2012. The second part of the study involved site visits to the six districts participating in The Wallace Foundation's Principal Pipeline Initiative -- Charlotte-Mecklenburg Schools, Denver Public Schools, Gwinnett County Public Schools, Hillsborough County Public Schools, the New York City Department of Education, and Prince George's County Public Schools. This report provides a summary of findings from both the survey and site visits. Part I presents a description of the organizational structure and general features of the various principal supervisory systems, including the roles, selection, deployment, staffing, professional development, and evaluation of principal supervisors, as well as the preparation, selection, support, and evaluation of principals. Part II provides recommendations for building more effective principal supervisory systems. Based on the survey results and observations from the site visits, these recommendations identify those structures and practices that are most likely to result in stronger school leaders and higher student achievement

    An open-data, agent-based model of alcohol related crime

    The allocation of resources to challenge city centre violent crime traditionally relies on historical data to identify hot-spots. The usefulness of such data-driven approaches is limited when historical data is scarce or unavailable (e.g. planning of a new city) or insufficiently representative (e.g. does not account for novel events, such as Olympic Games). In some cities, crime data is not systematically accumulated at all. We present a graph-constrained agent based simulation model of alcohol-related violent crime that is capable of predicting areas of likely violent crime without requiring any historical data. The only inputs to our simulation are publicly available geographical data, which makes our method immediately applicable to a wide range of tasks, such as optimal city planning, police patrol optimisation, devising alcohol licensing policies. In experiments, we evaluate our model and demonstrate agreement of our model's predictions on where and when violence will occur with real-world violent crime data. Analyses indicate that our agent based model may be able to make a significant contribution to attempts to prevent violence through deterrence or by design

    Association of violence with urban points of interest

    The association between alcohol outlets and violence has long been recognised, and is commonly used to inform policing and licensing policies (such as staggered closing times and zoning). Less investigated, however, is the association between violent crime and other urban points of interest, which while associated with the city centre alcohol consumption economy, are not explicitly alcohol outlets. Here, machine learning (specifically, LASSO regression) is used to model the distribution of violent crime for the central 9 km2 of ten large UK cities. Densities of 620 different Point of Interest types (sourced from Ordnance Survey) are used as predictors, with the 10 most explanatory variables being automatically selected for each city. Cross validation is used to test generalisability of each model. Results show that the inclusion of additional point of interest types produces a more accurate model, with significant increases in performance over a baseline univariate alcohol-outlet only model. Analysis of chosen variables for city-specific models shows potential candidates for new strategies on a per-city basis, with combined-model variables showing the general trend in POI/violence association across the UK. Although alcohol outlets remain the best individual predictor of violence, other points of interest should also be considered when modelling the distribution of violence in city centres. The presented method could be used to develop targeted, city-specific initiatives that go beyond alcohol outlets and also consider other locations

    Aging and obesity prime the methylome and transcriptome of adipose stem cells for disease and dysfunction

    The epigenome of stem cells occupies a critical interface between genes and environment, serving to regulate expression through modification by intrinsic and extrinsic factors. We hypothesized that aging and obesity, which represent major risk factors for a variety of diseases, synergistically modify the epigenome of adult adipose stem cells (ASCs). Using integrated RNA- and targeted bisulfite-sequencing in murine ASCs from lean and obese mice at 5- and 12-months of age, we identified global DNA hypomethylation with either aging or obesity, and a synergistic effect of aging combined with obesity. The transcriptome of ASCs in lean mice was relatively stable to the effects of age, but this was not true in obese mice. Functional pathway analyses identified a subset of genes with critical roles in progenitors and in diseases of obesity and aging. Specifically, Mapt, Nr3c2, App, and Ctnnb1 emerged as potential hypomethylated upstream regulators in both aging and obesity (AL vs. YL and AO vs. YO), and App, Ctnnb1, Hipk2, Id2, and Tp53 exhibited additional effects of aging in obese animals. Furthermore, Foxo3 and Ccnd1 were potential hypermethylated upstream regulators of healthy aging (AL vs. YL), and of the effects of obesity in young animals (YO vs. YL), suggesting that these factors could play a role in accelerated aging with obesity. Finally, we identified candidate driver genes that appeared recurrently in all analyses and comparisons undertaken. Further mechanistic studies are needed to validate the roles of these genes capable of priming ASCs for dysfunction in aging- and obesity-associated pathologies

    Clinical and Echocardiographic Findings in an Aged Population of Cavalier King Charles Spaniels

    Myxomatous mitral valve disease (MMVD) is the most common cardiac disease in dogs. It varies from dogs without clinical signs to those developing left-sided congestive heart failure, leading to death. Cavalier King Charles Spaniels (CKCSs) are particularly susceptible to MMVD. We hypothesised that within the elderly CKCS population, there is a sub-cohort of MMVD-affected dogs that do not have cardiac remodelling. The objectives of the present study were (i) to determine the prevalence and the degree of cardiac remodelling associated with MMVD; and (ii) assess the effect of age, gender, and body weight on echocardiographic status in a population of aged CKCSs. A total of 126 CKCSs ≄ 8 years old were prospectively included. They all had a physical and echocardiographic examination. A systolic murmur was detected in 89% of dogs; the presence of clinical signs was reported in 19% of them; and echocardiographic evidence of MMVD was described in 100%. Despite the high prevalence, 44.4% of the dogs were clear of echocardiographic signs of cardiac remodelling. Age was significantly associated with the presence and severity of cardiac remodelling and mitral valve prolapse. Our results showed that a proportion of elderly CKCS with confirmed MMVD did not undergo advanced stages of this pathology.This research was funded by the British CKCS Breed Club and The Roslin Institute, University of Edinburgh

    Pax3 expression enhances PDGF-B-induced brainstem gliomagenesis and characterizes a subset of brainstem glioma

    High-grade Brainstem Glioma (BSG), also known as Diffuse Intrinsic Pontine Glioma (DIPG), is an incurable pediatric brain cancer. Increasing evidence supports the existence of regional differences in gliomagenesis such that BSG is considered a distinct disease from glioma of the cerebral cortex (CG). In an effort to elucidate unique characteristics of BSG, we conducted expression analysis of mouse PDGF-B-driven BSG and CG initiated in Nestin progenitor cells and identified a short list of expression changes specific to the brainstem gliomagenesis process, including abnormal upregulation of paired box 3 (Pax3). In the neonatal mouse brain, Pax3 expression marks a subset of brainstem progenitor cells, while it is absent from the cerebral cortex, mirroring its regional expression in glioma. Ectopic expression of Pax3 in normal brainstem progenitors in vitro shows that Pax3 inhibits apoptosis. Pax3-induced inhibition of apoptosis is p53-dependent, however, and in the absence of p53, Pax3 promotes proliferation of brainstem progenitors. In vivo, Pax3 enhances PDGF-B-driven gliomagenesis by shortening tumor latency and increasing tumor penetrance and grade, in a region-specific manner, while loss of Pax3 function extends survival of PDGF-B-driven;p53-deficient BSG-bearing mice by 33%. Importantly, Pax3 is regionally expressed in human glioma as well, with high PAX3 mRNA characterizing 40% of human BSG, revealing a subset of tumors that significantly associates with PDGFRA alterations, amplifications of cell cycle regulatory genes, and is exclusive of ACVR1 mutations. Collectively, these data suggest that regional Pax3 expression not only marks a novel subset of BSG but also contributes to PDGF-B-induced brainstem gliomagenesis

    A dozen colliding wind X-ray binaries in the star cluster R136 in the 30Doradus region

    We analyzed archival Chandra X-ray observations of the central portion of the 30 Doradus region in the Large Magellanic Cloud. The image contains 20 X-ray point sources with luminosities between 5×10325 \times 10^{32} and 2×10352 \times 10^{35} erg s−1^{-1} (0.2 -- 3.5 keV). A dozen sources have bright WN Wolf-Rayet or spectral type O stars as optical counterparts. Nine of these are within ∌3.4\sim 3.4pc of R136, the central star cluster of NGC2070. We derive an empirical relation between the X-ray luminosity and the parameters for the stellar wind of the optical counterpart. The relation gives good agreement for known colliding wind binaries in the Milky Way Galaxy and for the identified X-ray sources in NGC2070. We conclude that probably all identified X-ray sources in NGC2070 are colliding wind binaries and that they are not associated with compact objects. This conclusion contradicts Wang (1995) who argued, using ROSAT data, that two earlier discovered X-ray sources are accreting black-hole binaries. Five of the eighteen brightest stars in R136 are not visible in our X-ray observations. These stars are either single, have low mass companions or very wide orbits.     Close binary companions of the HAeBe stars LkHa 198, Elias 1, HK Ori and V380 Ori

    We present diffraction-limited bispectrum speckle interferometry observations of four well-known Herbig Ae/Be (HAeBe) stars, LkHa 198, Elias 1, HK Ori and V380 Ori. For two of these, LkHa 198 and Elias 1, we present the first unambiguous detection of close companions. The plane of the orbit of the new LkHa 198 companion appears to be significantly inclined to the plane of the circumprimary disk, as inferred from the orientation of the outflow. We show that the Elias 1 companion may be a convective star, and suggest that it could therefore be the true origin of the X-ray emission from this object. In the cases of HK Ori and V380 Ori, we present new measurements of the relative positions of already-known companions, indicating orbital motion. For HK Ori, photometric measurements of the brightness of the individual components in four bands allowed us to decompose the system spectral energy distribution (SED) into the two separate component SEDs. The primary exhibits a strong infrared excess which suggests the presence of circumstellar material, whereas the companion can be modelled as a naked photosphere. The infrared excess of HK Ori A was found to contribute around two thirds of the total emission from this component, suggesting that accretion power contributes significantly to the flux. Submillimetre constraints mean that the circumstellar disk cannot be particularly massive, whilst the near-infrared data indicates a high accretion rate.
