    Leishmaniasis of the eyelid mimicking an infundibular cyst and review of the literature on ocular leishmaniasis

    Abstract Cutaneous leishmaniasis is an infection caused by protozoa belonging to the genus Leishmania . The disease is transmitted by sandflies. Reservoirs are represented by dogs, mice, rats, and wild rodents. Cutaneous leishmaniasis is usually characterized by a single, polymorphous lesion located in an uncovered area. We report a case of cutaneous leishmaniasis localized on the left upper eyelid in a 36-year-old woman. The disease was characterized by a single, asymptomatic nodule, which was clinically diagnosed as an inflammatory infundibular cyst. The lesion was excised surgically. Histopathological examination showed an inflammatory infiltrate consisting of lymphocytes, histiocytes, and plasma cells. Several Leishmania spp amastigotes were observed in the cytoplasm of macrophages. Culture examination on Novy–MacNeal–Nicolle medium was positive for Leishmania spp . PCR was positive for Leishmania infantum . No relapses were observed during follow-up (17 months).     Aroma precursors of Grignolino grapes (Vitis vinifera L.) and their modulation by vintage in a climate change scenario

    Current climatic conditions may cause significant changes in grapevine phenology and maturity dynamics linked often with changes to ecoclimatic indicators. The influence exerted by different meteorological conditions during four consecutive years on the aromatic potential of Grignolino grapes was investigated for the first time. The samples were collected from three vineyards characterized by different microclimatic conditions mainly related to the vineyard exposure and by a different age of the plants. Important differences as far as temperature and rainfall patterns are concerned during ripening were observed among the 4 years. Grape responses to abiotic stress, with particular emphasis on aromatic precursors, were evaluated using gas chromatography coupled to mass spectrometry. The results highlighted significant differences among the vintages for each vineyard in terms of the berry weight and the aromatic precursor concentration. For the grapes of the younger-vine vineyard, the content of aroma compounds showed a different variability among the vintages if compared to the old-vine vineyards. Optimal conditions in terms of temperature and rainfall during the green phase followed by a warm and dry post-veraison period until harvest favored all classes of compounds especially terpenoids mainly in the grapes of the old vines. High-temperature (>30°C) and low-rainfall pattern before veraison led to high benzenoid contents and increased differences among vineyards such as berry weight, whereas cooler conditions favored the terpenoid levels in grapes from southeast-oriented vineyards. In a hilly environment, lack of rainfall and high temperature that lately characterize the second part of berry development seem to favor the grape quality of Grignolino, a cultivar of medium-late ripening, by limiting the differences on bunch ripening, allowing a greater accumulation of secondary metabolites but maintaining at the same time an optimum balance sugar/acidity

    Fatty Liver Is Associated with Transcriptional Downregulation of Stearoyl-CoA Desaturase and Impaired Protein Dimerization

    AIMS AND METHODS: We evaluated the modulation of liver stearoyl-CoA desaturase-1 (Scd1) by dietary factors and insulin resistance (IR) in two experimental models of high-fat diet (HFD)-induced nonalcoholic fatty liver disease (NAFLD). The first model included Sprague Dawley (SD) rats that developed NAFLD without IR, and the second one included a rat model of genetic IR and cardiovascular disease, the spontaneously hypertensive rats (SHR) and its normotensive, insulin-sensitive control Wistar-Kyoto (WKY). The adult rats were given standard chow diet (CD) or HFD for 10 weeks. In all the animals, we explored the hepatic Scd1 transcriptional activity and protein levels. RESULTS: HFD-fed rats of both strains developed severe NAFLD. Liver abundance of Scd1 mRNA was significantly decreased in HFD-fed rats regardless of the strain; SD-CD: 235±195 vs. SD-HFD 4.5±2.9, p<0.0004, and SHR-CD: 75.6±10.8 vs. SHR-HFD: 4.48±17.4, and WKY-CD: 168.7±17.4 vs. WKY-HFD: 12.9±17.4, p<0.000001 (mean±SE, ANCOVA adjusted by HOMA). Analysis of liver Scd1 protein expression showed a particular pattern in the HFD groups, characterized by the presence of high levels of a monomeric protein band (32.2-36.6 Kda, p<0.003) and decreased levels of a dimeric protein band (61.9-66.1 Kda, p<0.02) regardless of the rat strain. Pharmacologic intervention with the peroxisome proliferator-activated receptor α agonist clofibrate reverted the liver phenotype and significantly modified the hepatic Scd1 transcriptional activity and protein expression.     Telomere length in the two extremes of abnormal fetal growth and the programming effect of maternal arterial hypertension

    We tested the hypothesis that leukocyte telomere length (LTL) is associated with birth weight in both extremes of abnormal fetal growth: small (SGA) and large for gestational age newborns (LGA). Clinical and laboratory variables of the mothers and the neonates were explored; 45 newborns with appropriate weight for gestational age (AGA), 12 SGA and 12 LGA were included. Whether the differences might be explained by variation in OBFC1 (rs9419958) and CTC1 (rs3027234) genes associated with LTL was determined. A significant association between birth weight and LTL was observed; LTL was significantly shorter in LGA newborns (1.01 ± 0.12) compared with SGA (1.73 ± 0.19) p < 0.005, mean ± SE. Maternal (Spearman R = −0.6, p = 0.03) and neonatal LTL (R = −0.25, p = 0.03) were significantly and inversely correlated with maternal history of arterial hypertension in previous gestations. Neonatal LTL was not significantly associated with either rs9419950 or rs3027234, suggesting that the association between neonatal LTL and birth weight is not influenced by genetic variation in genes that modify the interindividual LTL. In conclusion, telomere biology seems to be modulated by abnormal fetal growth; modifications in telomere length might be programmed by an adverse environment in utero.Fil: Tellechea, Mariana Lorena.     Beam test of ALPIDE Sensor

    The Alice Pixel Detector (ALPIDE) is developed for the upgrade of the Inner Tracking System of the ALICE experiment at CERN, which will take place during second Long Shutdown in 2019-2020. ALPIDE is a Monolithic Active Pixel Sensor (MAPS), manufactured in a 180 nm CMOS Imaging Process of TowerJazz. Forecoming tracking detectors, based on this technology, will see strong advantages with the application of these sensors as they provide the highest capabilities in spatial resolution and utmost potential for being thin.     In Vivo Effects of Einkorn Wheat (Triticum monococcum) Bread on the Intestinal Microbiota, Metabolome, and on the Glycemic and Insulinemic Response in the Pig Model

    Einkorn wheat (Triticum monococcum) is characterized by high content of proteins, bioactive compounds, such as polyunsaturated fatty acids, fructans, tocols, carotenoids, alkylresorcinols, and phytosterols, and lower α-, β-amylase and lipoxygenase activities compared to polyploid wheat. These features make einkorn flour a good candidate to provide healthier foods. In the present study, we investigated the effects of einkorn bread (EB) on the intestinal physiology and metabolism of the pig model by characterizing the glycemic and insulinemic response, and the microbiota and metabolome profiles. Sixteen commercial hybrid pigs were enrolled in the study; four pigs were used to characterize postprandial glycemic and insulinemic responses and twelve pigs underwent a 30-day dietary intervention to assess microbiota and metabolome changes after EB or standard wheat bread (WB) consumption. The postprandial insulin rise after an EB meal was characterized by a lower absolute level, and, as also observed for glucose, by a biphasic shape in contrast to that in response to a WB meal. The consumption of EB led to enrichment in short-chain fatty acid producers (e.g., Blautia, Faecalibacterium, and Oscillospira) in the gut microbiota and to higher metabolic diversity with lower content of succinate, probably related to improved absorption and therefore promoting intestinal gluconeogenesis. The observed changes, at both a compositional and metabolic scale, strongly suggest that EB consumption may support a health-promoting configuration of the intestinal ecosystem

    Epigenetic Modifications in the Biology of Nonalcoholic Fatty Liver Disease: The Role of DNA-Hydroxymethylation and TET Proteins

    The 5-Hydroxymethylcytosine (5-hmC) is an epigenetic modification whose role in the pathogenesis of metabolic-related complex diseases remains unexplored; 5-hmC appears to be prevalent in the mitochondrial genome. The Ten-Eleven-Translocation (TET) family of proteins is responsible for catalyzing the conversion of 5-methylcytosine to 5-hmC. We hypothesized that epigenetic editing by 5-hmC might be a novel mechanism through which nonalcoholic fatty liver disease (NAFLD)-associated molecular traits could be explained. Hence, we performed an observational study to explore global levels of 5-hmC in fresh liver samples of patients with NAFLD and controls (n = 90) using an enzyme-linked-immunosorbent serologic assay and immunohistochemistry. We also screened for genetic variation in TET 1–3 loci by next generation sequencing to explore its contribution to the disease biology. The study was conducted in 2 stages (discovery and replication) and included 476 participants. We observed that the amount of 5-hmC in the liver of both NAFLD patients and controls was relatively low (up to 0.1%); a significant association was found with liver mitochondrial DNA copy number (R = 0.50, P = 0.000382) and PPARGC1A-mRNA levels (R = −0.57, P = 0.04). We did not observe any significant difference in the 5-hmC nuclear immunostaining score between NAFLD patients and controls; nevertheless, we found that patients with NAFLD (0.4 ± 0.5) had significantly lower nonnuclear-5-hmC staining compared with controls (1.8 ± 0.8), means ± standard deviation, P = 0.028. The missense p.Ile1123Met variant (TET1-rs3998860) was significantly associated with serum levels of caspase-generated CK-18 fragment-cell death biomarker in the discovery and replication stage, and the disease severity (odds ratio: 1.47, 95% confidence interval: 1.10–1.97; P = 0.005). The p.Ile1762Val substitution (TET2-rs2454206) was associated with liver PPARGC1A-methylation and transcriptional levels, and Type 2 diabetes. Our results suggest that 5-hmC might be involved in the pathogenesis of NAFLD by regulating liver mitochondrial biogenesis and PPARGC1A expression. Genetic diversity at TET loci suggests an “epigenetic” regulation of programmed liver-cell death and a TET-mediated fine-tuning of the liver PPARGC1A-transcriptional program.Fil: Pirola, Carlos José.     Circulating microRNA signature in non-alcoholic fatty liver disease: from serum non-coding RNAs to liver histology and disease pathogenesis

    OBJECTIVES: We used a screening strategy of global serum microRNA (miRNA) profiling, followed by a second stage of independent replication and exploration of liver expression of selected miRNAs to study: (1) the circulating miRNA signature associated with non-alcoholic fatty liver disease (NAFLD) progression and predictive power, (2) the role of miRNAs in disease biology and (3) the association between circulating miRNAs and features of the metabolic syndrome. METHODS: The study used a case-control design and included patients with NAFLD proven through biopsy and healthy controls. RESULTS: Among 84 circulating miRNAs analysed, miR-122, miR-192, miR-19a and miR-19b, miR-125b, and miR-375 were upregulated >2-fold (p<0.05) either in simple steatosis (SS) or non-alcoholic steatohepatitis (NASH). The most dramatic and significant fold changes were observed in the serum levels of miR-122 (7.2-fold change in NASH vs controls and 3.1-fold change in NASH vs SS) and miR-192 (4.4-fold change in NASH vs controls); these results were replicated in the validation set. The majority of serum miR-122 circulate in argonaute2-free forms. Circulating miR-19a/b and miR-125b were correlated with biomarkers of atherosclerosis. Liver miR-122 expression was 10-fold (p<0.03) downregulated in NASH compared with SS and was preferentially expressed at the edge of lipid-laden hepatocytes. In vitro exploration showed that overexpression of miR-122 enhances alanine aminotransferase activity. CONCLUSIONS: miR-122 plays a role of physiological significance in the biology of NAFLD; circulating miRNAs mirror the histological and molecular events occurring in the liver. NAFLD has a distinguishing circulating miRNA profile associated with a global dysmetabolic disease state and cardiovascular risk.Fil: Pirola, Carlos Jose.     Effect of cold plasma generated with different gas mixtures on safety, quality and nutritional aspects of fresh sea bream fillets

    Cold atmospheric plasma (CAP) is a non-thermal technology, successfully used to decontaminate and extend the shelf-life of various foods. However, since CAP is highly oxidative, it can cause quality deterioration in sensitive matrices, such as fish products. This research aimed to evaluate the effect of CAP treatment with a surface dielectric barrier discharge (SDBD) with different gas mixtures (80% Ar/20% O2, or 80% N2/20% O2) on the decontamination of inoculated pathogens (E.coli and L.innocua; log 4 CFU/g inoculum) and endogenous spoilage microbiota and on the main quality indices of seabream (Spaurus aurata) fillets. For selected appropriate treatments, the impact on the nutritional value of the products was investigated through in vitro digestion, bioaccessibility of fatty acids and the degree of protein hydrolysis. The use of CAP resulted in a decrease in the bacterial load in the fresh sea bream fillets up to 1 log CFU/g obtained with Ar/O2 gas mixture for 20 min, affected by the treatment duration, but not by the gas mixture. Although a slight increase in lipid oxidation was observed (from 0.5 mg MDA/kg to a maximum of 4 mg MDA/kg), the digestibility of the products was not affected. Industrial relevance From an industrial point of view, increasing shelf-life of perishable products such as fish fillets with an environmentally friendly and non-thermal technology could represent a great advantage; however, maintaining quality is of paramount importance for the industrial use of this novel processing technology. The results of the present study show negligible effects on the nutritional quality of seabream fillets, which encourages further research.publishedVersio
