    The effects of supplementation with sunflower and soybean oils on the fatty acid profile of milk fat from grazing dairy cows

    Copyright © INRA, EDP Sciences, 2005.The objective of this study was to observe the effect of supplementation with vegetable oils (VO) on the fatty acid profiles and conjugated linoleic acid (CLA) content of milk fat. Twelve dairy cows in mid lactation fed on pasture were blocked by body weight, milk production and days in lactation and randomly allocated from blocks to 3 treatments repeated in a Latin square design with periods of 28 day duration. The treatments were as follows: cows on pasture supplemented with 5 kg concentrate per head per day (C), supplemented with 4.5 kg concentrate + 0.5 kg sunflower oil per head per day (SFO) and supplemented with 4.5 kg concentrate + 0.5 kg soybean oil per head per day (SBO). The animals were grazed as a group and were stocked at 2.5 heads per hectare. The treatments had no effect on milk yield or protein yield and content, but decreased milk fat yield and content (P < 0.05). Milk fat from the cows supplemented with VO had a lower concentration of short and medium chain fatty acids (P < 0.05) and a higher concentration of long chain fatty acids (P < 0.05). The addition of VO to the diet also resulted in a reduction in saturated and an increase in unsaturated fatty acids of milk fat (P < 0.05). The hypercholesterolemic fatty acids in milk fat (i.e. C12:0, C14:0 and C16:0) decreased while the concentration of oleic and linoleic acids increased with VO (P < 0.05). There was no effect on linolenic acid. Finally, the inclusion of VO in the diet increased (P < 0.05) the contents in milk fat of the various cis/trans isomers of oleic acid (including trans-vacenic, TVA) and of CLA which increased by 61%

    Analysis of predictive biomarkers in patients with lung adenocarcinoma from Southern Brazil reveals a distinct profile from other regions of the country

    PURPOSE Adenocarcinoma is the most common histologic subtype of non–small-cell lung cancer, representing 40% of all diagnoses. Several biomarkers are currently used to determine patient eligibility for targeted treatments, including analysis of molecular alterations in EGFR and ALK, as well as programmed death-ligand 1 (PD-L1) protein expression. Epidemiologic data reporting the frequency of these biomarkers in Brazilian patients with lung adenocarcinoma (LUAD) are limited, and existing studies predominantly included patients from the southeast region of the country. MATERIALS AND METHODS The goal of this study was to investigate the frequency of somatic mutations in the EGFR, KRAS, NRAS, and BRAF genes, ALK, and PD-L1 expression in a series of Brazilian patients diagnosed with LUAD predominantly recruited from centers in southern Brazil. Molecular analysis of the EGFR, KRAS, NRAS, and BRAF genes was performed by next-generation sequencing using DNA extracted from tumor tissue. Immunohistochemistry was used to detect ALK and PD-L1 expression. RESULTS Analysis of 619 tumors identified KRAS mutations in 189 (30.2%), EGFR mutations in 120 (19.16%), and BRAF mutations in 19 (3%). Immunohistochemistry demonstrated ALK and PD-L1 expression in 4% and 35.1% of patients, respectively. CONCLUSION To our knowledge, this is the first study investigating the molecular epidemiology of patients with LUAD from southern Brazil and the largest assessing the frequency of multiple predictive biomarkers for this tumor in the country. The study also reveals a distinct mutation profile compared with data originating from other regions of Brazil

    Constitutive deficiency of the neurogenic hippocampal modulator AP2γ promotes anxiety-like behavior and cumulative memory deficits in mice from juvenile to adult periods

    The transcription factor activating protein two gamma (AP2γ) is an important regulator of neurogenesis both during embryonic development as well as in the postnatal brain, but its role for neurophysiology and behavior at distinct postnatal periods is still unclear. In this work, we explored the neurogenic, behavioral, and functional impact of a constitutive and heterozygous AP2γ deletion in mice from early postnatal development until adulthood. AP2γ deficiency promotes downregulation of hippocampal glutamatergic neurogenesis, altering the ontogeny of emotional and memory behaviors associated with hippocampus formation. The impairments induced by AP2γ constitutive deletion since early development leads to an anxious-like phenotype and memory impairments as early as the juvenile phase. These behavioral impairments either persist from the juvenile phase to adulthood or emerge in adult mice with deficits in behavioral flexibility and object location recognition. Collectively, we observed a progressive and cumulative impact of constitutive AP2γ deficiency on the hippocampal glutamatergic neurogenic process, as well as alterations on limbic-cortical connectivity, together with functional behavioral impairments. The results herein presented demonstrate the modulatory role exerted by the AP2γ transcription factor and the relevance of hippocampal neurogenesis in the development of emotional states and memory processes.H2020 -“la Caixa” Foundation(101003187

    Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.
