    On the Sensitivity of a Ground-Based Tropospheric Lidar to Aitken Mode Particles in the Upper Troposphere

    Airborne observations have shown high concentrations of ultrafine aerosols in the Amazon upper troposphere (UT), which are key for replenishing the planetary boundary layer (PBL) with cloud condensation nuclei that sustain the “green ocean” clouds. Given their climatic relevance, longterm observations are needed, but aircraft measurements are only available in short-term campaigns. Alternatively, continuous observations of the aerosol vertical structure could be performed by a lidar (acronym for “light detection and ranging”) system in long-term campaigns. Here we assess whether a ground-based tropospheric lidar system could detect these ultrafine UT aerosols. To this aim, we simulated the lidar signal of a real instrument and then varied the instrument’s efficiency and the UT-particle concentration to determine under which conditions the detection is possible. Optical properties were computed with a Mie code based on the size distributions and numerical concentration profiles measured by the aircraft, and on the refractive indexes inverted from AERONET measurements. The aerosol optical depth (AOD) was retrieved by inverting the elastic lidar signal, and a statistical test was applied to evaluate the detection of the UT-aerosol layer. Our results indicate that, for the instrument we simulated, a 55-fold increase in the signal-to-noise ratio (SNR) is required for a 100% detection rate. This could be achieved by simultaneously time averaging over 30 min and spatially averaging to vertical bin lengths of 375 m, or by modifying the hardware. We repeated the analysis for under- and overestimated aerosol lidar ratio (Laer), and found that possible systematic errors did not affect the detection rate. Further studies are necessary to assess whether such longtime averages are feasible in the Amazon region (given the very high cloud cover), and to design a hardware upgrade. Although simulations and analyses here were based on a particular instrument and for the presence of new organic particles in the Amazonian upper troposphere, our methodology and results are general and applicable to other instruments and sites.Conselho Nacional de Desenvolvimento Cientifico e Tecnologico (CNPQ) 132402/2020-3 308682/2017-

    Temporal control of epigenetic centromere specification

    All living organisms require accurate mechanisms to faithfully inherit their genetic material during cell division. The centromere is a unique locus on each chromosome that supports a multiprotein structure called the kinetochore. During mitosis, the kinetochore is responsible for connecting chromosomes to spindle microtubules, allowing faithful segregation of the duplicated genome. In most organisms, centromere position and function is not defined by the local DNA sequence context but rather by an epigenetic chromatin-based mechanism. Centromere protein A (CENP-A) is central to this process, as chromatin assembled from this histone H3 variant is essential for assembly of the centromere complex, as well as for its epigenetic maintenance. As a major determinant of centromere function, CENP-A assembly requires tight control, both in its specificity for the centromere and in timing of assembly. In the last few years, there have been several new insights into the molecular mechanism that allow this process to occur.     A barrier for further approximating Sorting By Transpositions

    The Transposition Distance Problem (TDP) is a classical problem in genome rearrangements which seeks to determine the minimum number of transpositions needed to transform a linear chromosome into another represented by the permutations π\pi and σ\sigma, respectively. This paper focuses on the equivalent problem of Sorting By Transpositions (SBT), where σ\sigma is the identity permutation ι\iota. Specifically, we investigate palisades, a family of permutations that are "hard" to sort, as they require numerous transpositions above the celebrated lower bound devised by Bafna and Pevzner. By determining the transposition distance of palisades, we were able to provide the exact transposition diameter for 33-permutations (TD3), a special subset of the Symmetric Group SnS_n, essential for the study of approximate solutions for SBT using the simplification technique. The exact value for TD3 has remained unknown since Elias and Hartman showed an upper bound for it. Another consequence of determining the transposition distance of palisades is that, using as lower bound the one by Bafna and Pevzner, it is impossible to guarantee approximation ratios lower than 1.3751.375 when approximating SBT. This finding has significant implications for the study of SBT, as this problem has been subject of intense research efforts for the past 25 years

    Adhesion of an endodontic sealer to dentin and gutta-percha: shear and push-out bond strength measurements and SEM analysis

    The use of an adequate method for evaluation of the adhesion of root canal filling materials provides more reliable results to allow comparison of the materials and substantiate their clinical choice. The aims of this study were to compare the shear bond strength (SBS) test and push-out test for evaluation of the adhesion of an epoxy-based endodontic sealer (AH Plus) to dentin and gutta-percha, and to assess the failure modes on the debonded surfaces by means of scanning electron microscopy (SEM). Three groups were established (n=7): in group 1, root cylinders obtained from human canines were embedded in acrylic resin and had their canals prepared and filled with sealer; in group 2, longitudinal sections of dentin cylinders were embedded in resin with the canal surface smoothed and turned upwards; in group 3, gutta-percha cylinders were embedded in resin. Polyethylene tubes filled with sealer were positioned on the polished surface of the specimens (groups 2 and 3). The push-out test (group 1) and the SBS test (groups 2 and 3) were performed in an Instron universal testing machine running at crosshead speed of 1 mm/min. Means (±SD) in MPa were: G1 (8.8±1.13), G2 (5.9±1.05) and G3 (3.8±0.55). Statistical analysis by ANOVA and Student's t-test (a=0.05) revealed statistically significant differences (p<0.01) among the groups. SEM analysis showed a predominance of adhesive and mixed failures of AH Plus sealer. The tested surface affected significantly the results with the sealer reaching higher bond strength to dentin than to gutta-percha with the SBS test. The comparison of the employed methodologies showed that the SBS test produced significantly lower bond strength values than the push-out test, was skilful in determining the adhesion of AH Plus sealer to dentin and gutta-percha, and required specimens that could be easily prepared for SEM, presenting as a viable alternative for further experiments

    On the Two Spectra Inverse Problem for Semi-Infinite Jacobi Matrices

    We present results on the unique reconstruction of a semi-infinite Jacobi operator from the spectra of the operator with two different boundary conditions. This is the discrete analogue of the Borg-Marchenko theorem for Schr{\"o}dinger operators in the half-line.
