    CHEMICAL CHARACTERIZATION AND ANTIMICROBIAL ACTIVITY OF HYDROETHANOLIC CRUDE EXTRACT OF EUGENIA FLORIDA DC (MYRTACEAE) LEAVES

    Objective: The present study aimed to characterize and quantify the total phenolics, flavonoids and tannins in a hydroethanolic crude extract (70% (v/v) (EB)) of the leaves of E. florida DC, as well as to evaluate the antimicrobial activity of the extract against different species of micro-organisms.Methods: EB was characterized using a mass spectrometer equipped with a direct insertion device for in-stream injection (FIA). Quantitative analyses of major compounds were carried out by spectrophotometry. In addition, we evaluated the sensitivity profiles of different strains of yeast and bacteria against different concentrations of EB.Results: The classes found were in agreement with those described in the literature: flavonoids, tannins, phenolic acids and saponins. EB showed levels of phenolic compounds, flavonoids and tannins equal to 25.82 mg gallic acid equivalents per gram of extract (EAG/g), 8.42 mg quercetin equivalents per gram of extract (EQ/g) and 7.30 mg tannic acid equivalents per gram of extract (AT/g), respectively. In the analysis of antimicrobial activity, EB was more active against yeasts but was not effective against the bacteria used in the test.Conclusion: We can conclude that E. florida DC has antimicrobial potential, due to the presence of bioactive secondary metabolites.Keywords: Phenolics, Micro-organisms, Mass spectrometer, Yeasts, Bacteria, PotentialÂ

    The electroencephalogram in the parkinsonian syndrome

    The electroencephalograms of 50 parkinsonian patients with no convulsive antecedents were studied. EEG abnormalities were found in 20 patients (40% of the cases). The incidence of abnormal records was larger in the bilateral parkinsonian syndrome than in the unilateral one (52% and 25%, respectively). Paroxysmal abnormalities characterized mainly by bursts of theta waves were the most frequent ones (65% of the cases). Sharp activity found in the EEG patterns of three patients is emphasized. Alterations of the base rhythm were recorded in 30% of the abnormal EEG cases. The patient's age doesn't seem to have an important role in the genesis of the electroencephalographic abnormalities.Foram estudados os eletrencefalogramas de 50 pacientes parkinsonianos livres de qualquer antecedente convulsivo, sendo registrados traçados anormais em 40% dos casos. A incidência de traçados anormais foi maior na síndrome parkinsoniana bilateral do que na unilateral (52% e 25%, respectivamente). Entre as anormalidades assinaladas foram as paroxísticas as mais freqüentes (65% dos casos), caracterizando-se predominantemente por surtos de ondas teta. O registro de anormalidades paroxísticas do tipo irritativo encontrado em três casos é salientado. Alterações da atividade de base foram assinaladas em 35% dos pacientes.     Ochna serrulata (Ochnaceae) is an ornamental plant introduced into Brazil from Asia and Africa. Species of the Ochna genus are rich in phenolic compounds, mainly flavonoids. The biological screening of extracts and fractions showed that this plant exhibited a significant antioxidant activity, when evaluated by the DPPH and reducing potential assays. Ochna serrulata also demonstrated slight toxic activity against Artemia salina and a potential inhibitory allelopathic activity, when evaluated using the Lactuca sativa seed germination test.     Caracterização química e avaliação das atividades antibacteriana, antifúngica, antimicobacteriana e citotóxica de Talinum paniculatum

    In this study, the bioactivity of Talinum paniculatum was evaluated, a plant widely used in folk medicine. The extract from the T. paniculatum leaves (LE) was obtained by percolation with ethanol-water and then subjecting it to liquid-liquid partitions, yielding hexane (HX), ethyl acetate (EtOAc), butanol (BuOH), and aqueous (Aq) fractions. Screening for antimicrobial activity of the LE and its fractions was evaluated in vitro through broth microdilution method, against thirteen pathogenic and non-pathogenic microorganisms, and the antimycobacterial activity was performed through agar diffusion assay. The cytotoxic concentrations (CC90) for LE, HX, and EtOAc were obtained on BHK-21 cells by using MTT reduction assay. The LE showed activity against Serratia marcescens and Staphylococcus aureus, with Minimum Inhibitory Concentration (MIC) values of 250 and 500 µg/mL, respectively. Furthermore, HX demonstrated outstanding activity against Micrococcus luteus and Candida albicans with a MIC of 31.2 µg/mL in both cases. The MIC for EtOAc also was 31.2 µg/mL against Escherichia coli. Conversely, BuOH and Aq were inactive against all tested microorganisms and LE proved inactive against Mycobacterium tuberculosisand Mycobacterium bovisas well. Campesterol, stigmasterol, and sitosterol were the proposed structures as main compounds present in the EF and HX/EtOAc fractions, evidenced by mass spectrometry. Therefore, LE, HX, and EtOAc from T. paniculatumshowed potential as possible sources of antimicrobial compounds, mainly HX, for presenting low toxicity on BHK-21 cells with excellent Selectivity Index (SI = CC90/MIC) of 17.72 against C. albicans.Neste estudo foi avaliada a bioatividade de Talinum paniculatum, planta amplamente utilizada na medicina popular. O extrato das folhas (EF) de T. paniculatum foi obtido por percolação com etanol-água e, em seguida, submetido à partição líquido-líquido, obtendo-se as frações hexânica (HX), acetato-etílica (AcOEt), butanólica (BuOH) e aquosa (Aq). A triagem para a atividade antimicrobiana do EF e de suas frações foram avaliadas in vitro através do método de microdiluição em caldo contra treze micro-organismos patogênicos e não-patogênicos e, a atividade antimicobacteriana, foi avaliada através do teste de difusão em ágar. As concentrações citotóxicas (CC90) do EF e das frações HX e AcOEt foram obtidas sobre células da linhagem BHK-21 através do ensaio de redução do MTT. O EF mostrou atividade contra Serratia marcescens e Staphylococcus aureus, com valores de concentração inibitória mínima (CIM) de 250 e 500 µg/mL, respectivamente. Além disso, HX demonstrou excelente atividade contra Micrococcus luteus e Candida albicans com uma CIM de 31,2 µg/mL, em ambos os casos. Contra Escherichia coli, a CIM para AcOEt foi também de 31,2 µg/mL. Por outro lado, as frações BuOH e Aq foram inativas contra todos os micro-organismos testados, assim como o EF contra Mycobacterium tuberculosis e Mycobacterium bovis. Campesterol, estigmasterol e sitosterol foram as estruturas propostas como principais compostos presentes no EF e nas frações HX e AcOEt, evidenciadas através de espectrometria de massas. Portanto, o extrato da folha e as frações HX e AcOEt provenientes de T. paniculatum apresentaram potencial como possíveis fontes de compostos antimicrobianos, HX principalmente, por ter apresentado uma baixa toxicidade sobre células BHK-21 com um bom índice de seletividade (IS = CC90/MIC) de 17,72 contra C. albicans

    Candida auris: a literature review

    Background: Emerging pathogen Candida auris has been associated with nosocomial outbreaks demonstrating widespread antifungal resistance. This microorganism is associated with systemic infections with a high mortality rate, and studies that contribute to a better understanding of this agent are important. Objectives: The present article aimed to carry out bibliographic research on Candida auris. Therefore, a literature search was carried out between January 2018 and January 2022, applying the following: C. auris; Candida auris and Infection, in Portuguese, English and Spanish. This review considers the available literature on C. auris and highlights key unknowns, which will provide guidance for future work in this field. Results: The data collected in the present review allowed us to divide the theme into topics on Pathophysiology, Epidemiology/clinical and drug resistance mechanisms, so that the understanding of Candida auris can be better detailed and understood. Conclusion: The predilection of C. auris by the most vulnerable and immunosuppressed patients or with comorbidities and with the potential to cause invasive and bloodstream infections with a propensity to cause outbreaks and concerns regarding resistance to antifungal agents, the fungus Candida auris, is of increasing clinical relevance. The increase in the number of detected cases and the occurrence of strains with multiple drug resistance prove to be worrisome, and applied research is essential to contribute to the knowledge of this strain and effective therapy

    Assessing carriers' logistical performance indicator by shippers in the São Paulo Metropolitan Area

    Article Info Abstract Keywords: road freight transportation, logistical performance indicator, non-parametric tests, sustainable practices, urban freight transportation This paper aims to assess the logistics services provided by carriers in the São Paulo Metropolitan Area (SPMA) considering two perspectives: the efficiency of the operations and the adoption of sustainable measures. A survey was applied to the 15 main shippers working with the selected carrier in order to assess logistics performance indicators (on-time-in-full, relationship, lead time, service quality, etc.) as well as sustainability demands (disposal of oil, post-use tires, spare parts, etc.). A second survey was applied to 21 other shippers that hire similar services provided by other carriers in the SPMA. The results indicate that a good relationship between shippers and carriers positively impacts the evaluation of the carriers. Approximately 64% of the informants claim that gas emissions followed by disposal of lubricant, tires and spare parts are the major unsustainable practices to be avoided. This study makes two contributions: to show the most relevant sustainable and logistics aspects that carriers must consider for their services from the viewpoint of shippers and to analyse the influence of the relationship between the companies on the performance indicators
