    Neutrophils are a specialized subset of white blood cells, which have the ability to store pre-formed mediators in their cytoplasmic granules. Neutrophils are well-known effector cells involved in host protection against pathogens through diverse mechanisms such as phagocytosis, degranulation, extracellular traps, and oxidative burst. In this study, we provide evidence highlighting the significance of the SNARE proteins syntaxin-4 and synaptosomal-associated protein (SNAP) 23 in the release of azurophilic granules, specific granules, and the production of reactive oxygen species in human neutrophils. In contrast, the specific blockade of either syntaxin-4 or SNAP23 did not prevent the release of mitochondrial dsDNA in the process of neutrophil extracellular trap (NET) formation. These findings imply that degranulation and the release of mitochondrial dsDNA involve at least partially distinct molecular pathways in neutrophils

    Gene expression modulation by paraquat-induced oxidative stress conditions in Paracoccidioides brasiliensis

    Paracoccidioides brasiliensis is a thermodimorphic fungus associated with paracoccidioidomycosis (PCM), the most common systemic mycosis in Latin America. the infection is initiated by inhalation of environmentally dispersed conidia produced by the saprophytic phase of the fungus. in the lungs, P. brasiliensis assumes the parasitic yeast form and must cope with the adverse conditions imposed by cells of the host immune system, which includes a harsh environment, highly concentrated in reactive oxygen species (ROS). in this work, we used the ROS-generating agent paraquat to experimentally simulate oxidative stress conditions in order to evaluate the stress-induced modulation of gene expression in cultured P. brasiliensis yeast cells, using a microarray hybridization approach. the large-scale evaluation inherent to microarray-based analyses identified 2070 genes differentially transcribed in response to paraquat exposure, allowing an integrated visualization of the major metabolic changes that constitute the systemic defense mechanism used by the fungus to overcome the deleterious effects of ROS. These include overexpression of detoxifying agents, as well as of molecular scavengers and genes involved in maintenance of the intracellular redox potential. Particularly noteworthy was to verify that the oxidative stress resistance mechanism of P. brasiliensis also involves coordinated overexpression of a series of genes responsible for chitin-biosynthesis, suggesting that this pathway may constitute a specific regulon. Further analyses aiming at confirming and understanding the mechanisms that control such regulon may provide interesting new targets for chemotherapeutic approaches against P. brasiliensis and other pathogenic fungi.     Evaluation of an International Entrepreneur Exchange Program: Impacts, Lessons Learned, and Implications for Agricultural Development

    This study evaluated a two-way, visitor exchange project for entrepreneurship development between three African countries and the United States. The study’s purpose was to determine outcomes, understand lessons learned, and derive implications for international agricultural development. Findings of the study confirm visiting African Entrepreneur Fellows (AEFs) developed entrepreneurial knowledge, gained business skills, and acquired positive attitudes toward U.S. business and culture. The majority of AEFs had applied acquired knowledge and skills to improve their businesses and promoted open economic ideals, business ethics, and human rights in their businesses. As a result, AEFs were able to expand their business into new ventures, improve customer services, establish communication networks, and serve their communities. Visitor exchange, entrepreneurship-building programs are effective strategies in contributing to development efforts in developing countries. Paying due attention to the selection and matching of U.S. mentors with the business interests and learning needs of international fellows is necessary to ensure their learning expectations are met. It is important to assign international participants with suitable mentors for longer periods of time to increase the likelihood of receiving more in-depth learning experiences and develop lasting professional relationships to further collaboration. Realization of the potential of entrepreneurship-focused, visitor exchange programs between nations as a strategy for international agricultural development is the major implication of this study

    Scaling of Star Polymers with one to 80 Arms

    We present large statistics simulations of 3-dimensional star polymers with up to f=80f=80 arms, and with up to 4000 monomers per arm for small values of ff. They were done for the Domb-Joyce model on the simple cubic lattice. This is a model with soft core exclusion which allows multiple occupancy of sites but punishes each same-site pair of monomers with a Boltzmann factor v<1v<1. We use this to allow all arms to be attached at the central site, and we use the `magic' value v=0.6v=0.6 to minimize corrections to scaling. The simulations are made with a very efficient chain growth algorithm with resampling, PERM, modified to allow simultaneous growth of all arms. This allows us to measure not only the swelling (as observed from the center-to-end distances), but also the partition sum. The latter gives very precise estimates of the critical exponents γf\gamma_f. For completeness we made also extensive simulations of linear (unbranched) polymers which give the best estimates for the exponent γ\gamma.Comment: 7 pages, 7 figure

    Analysis of mTOR Inhibition-Involved Pathway in Ovarian Clear Cell Adenocarcinoma

    This study was designed to clarify the mechanism of the mammalian target of rapamycin (mTOR)-hypoxia inducible factor-1 (HIF-1) pathway using the cultured cell strain derived from human ovarian clear cell adenocarcinoma (CCA). Everolimus (a derivative of rapamycin)-treated cells and non-treated cells did not show any difference in mTOR expression. But, phosphorylated-mTOR (p-mTOR) expression significantly decreased in the treated cells, and mTOR-related factors such as phosphorylated-4E-BP1 (p-4E-BP1), HIF-1α, and vascular endothelial growth factor (VEGF) in the downstream region of mTOR revealed a marked decrease in expression. The analysis of influences of the drug on the HIF-1α degradation system showed an increase in von-Hippel Lindau (VHL) expression in the treated cells. Increase of cleaved caspase-3, one of key factors involved in apoptosis, was also shown in the treated cells. In the next step, using nude mice implanted with RMG-1 cells, a decrease in tumor size was demonstrated in 4 of the 7 mice which were orally administered with everolimus. As a result, it was suggested that everolimus administration would be helpful as an anti-tumor therapy for CCA not only via down-regulation of p-mTOR but also degradation of HIF-1α by VHL and induction of apoptosis by cleaved caspase-3
