    SSR assessment of Phytophthora infestans populations on tomato and potato in British gardens demonstrates high diversity but no evidence for host specialisation

    Phytophthora infestans populations can differ in composition as a result of host-specialisation on tomato and potato hosts. In Great Britain many amateur gardeners grow outdoor tomatoes but there is little or no commercial tomato production outdoors. This study analysed isolates of P. infestans from British gardens with twelve multiplexed SSR markers that are used to monitor the disease on commercial potato crops. Samples of P. infestans from tomato hosts were collected in three years and from potato in one year from across Great Britain. Seven previously unreported genotypes were detected in garden populations and higher frequencies of unique clonal lineages (28% to 40%) were present compared with populations from British commercial potato crops reported elsewhere. Garden populations had a lower proportion (11% to 48% less) of the most common lineages (13_A2 and 6_A1) which together made up at least 86% of the commercial potato populations during the sampling period. Host species accounted for only 2.6% of molecular variance detected between garden potato- and tomato-hosted samples. No significant difference in clonal lineage composition was found between host species in Great Britain and this could be due to the whole P. infestans population over-wintering on potato. British garden populations on both hosts were much more diverse than those on commercial potato crops; this finding may be influenced by less frequent fungicide use by gardeners and a higher diversity of un-sprayed susceptible potato cultivars enabling metalaxyl-sensitive and less virulent genotypes to survive in gardens

    Multilocus sequence typing of the pathogenic fungus Aspergillus fumigatus

    A multilocus sequence typing (MLST) scheme was devised for Aspergillus fumigatus. The system involved sequencing seven gene fragments and was applied to a panel of 100 isolates of A. fumigatus from diverse sources. Thirty different sequence types were found among the 100 isolates, and 93% of the isolates differed from the other isolates by only one allele sequence, forming a single clonal cluster as indicated by the eBURST algorithm. The discriminatory power of the MLST method was only 0.93. These results strongly indicate that A. fumigatus is a species of a relatively recent origin, with low levels of sequence dissimilarity. Typing methods based on variable numbers of tandem repeats offer higher levels of strain discrimination. Mating type data for the 100 isolates showed that 71 isolates were type MAT1-2 and 29 isolates were MAT1-1

    Modelling the effect of non-planarity on luminescence energy of conjugated polymers

    We present theoretical investigations of structural and electronic properties of ground-state and low-lying excited singlet states in isolated chains of conjugated polymers using a self-consistent quantum molecular dynamics method. With this approach, we have determined the energy of both states as function of the twist angle between two planar segments of the same polymer chain, for polymer chains with variable length. The conjugated polymers investigated here are poly(para-phenylene vinylene) (PPV) and polydiacetylene (PDA). Our results show that the energy of the excited-state increases more than that of the ground-state, as the twist angle increases up to 90º degrees. The change in the twist angle of both polymers leads to a blueshift in luminescence transition energy, the effect being stronger in PPV when the planar segments have similar sizes. The predicted blueshift in both polymers is dependent on the chain length, the effect being more pronounced for shorter-chains.Comunidade Europeia (CE).     Patient-Powered Research Networks of the Autoimmune Research Collaborative: Rationale, Capacity, and Future Directions

    Patient-Powered Research Networks (PPRNs) are US-based registry infrastructures co-created by advocacy groups, patient research partners, academic investigators, and other healthcare stakeholders. Patient-Powered Research Networks collect information directly from patients to conduct and disseminate the results of patient-centered/powered research that helps patients make more informed decisions about their healthcare. Patient-Powered Research Networks gather and utilize real-world data and patient-reported outcomes to conduct comparative effectiveness, safety, and other research, and leverage the Internet to accomplish this effectively and efficiently. Four PPRNs focused on autoimmune and immune-mediated conditions formed the Autoimmune Research Collaborative: ArthritisPower (rheumatoid arthritis, spondyloarthritis, and other rheumatic and musculoskeletal diseases), IBD Partners (inflammatory bowel disease), iConquerMS (multiple sclerosis), and the Vasculitis PPRN (vasculitis). The Autoimmune Research Collaborative aims to inform the healthcare decision making of patients, care partners, and other stakeholders, such as clinicians, regulators, and payers. Illustrated by practical applications from the Autoimmune Research Collaborative and its constituent PPRNs, this article discusses the shared capacities and challenges of the PPRN model, and the opportunities presented by collaborating across autoimmune conditions to design, conduct, and disseminate patient-centered outcomes research

    GS-2: A novel Broad-Spectrum agent for environmental microbial control

    The environmental control of microbial pathogens currently relies on compounds that do not exert long-lasting activity on surfaces, are impaired by soil, and contribute to the growing problem of antimicrobial resistance. This study presents the scientific development and characterization of GS-2, a novel, water-soluble ammonium carboxylate salt of capric acid and L-arginine that demonstrates activity against a range of bacteria (particularly Gram-negative bacteria), fungi, and viruses. In real-world surface testing, GS-2 was more effective than a benzalkonium chloride disinfectant at reducing the bacterial load on common touch-point surfaces in a high-traffic building (average 1.6 vs. 32.6 CFUs recovered from surfaces 90 min after application, respectively). Toxicology testing in rats confirmed GS-2 ingredients were rapidly cleared and posed no toxicities to humans or animals. To enhance the time-kill against Gram-positive bacteria, GS-2 was compounded at a specific ratio with a naturally occurring monoterpenoid, thymol, to produce a water-based antimicrobial solution. This GS-2 with thymol formulation could generate a bactericidal effect after five minutes of exposure and a viricidal effect after 10 min of exposure. Further testing of the GS-2 and thymol combination on glass slides demonstrated that the compound retained bactericidal activity for up to 60 days. Based on these results, GS-2 and GS-2 with thymol represent a novel antimicrobial solution that may have significant utility in the long-term reduction of environmental microbial pathogens in a variety of settings

    Influence of the feedstock characteristics on the microstructure and properties of Al2O3 TiO2 plasma-sprayed coatings

    [EN] Atmospheric plasma spraying (APS) is an interesting technique to obtain nanostructured coatings due to its versatility, simplicity and relatively low cost. However, nanometric powders cannot be fed into the plume using conventional feeding systems, due to their low mass and poor flowability, and must be adequately reconstituted into sprayable micrometric agglomerates. In this work, Al2O3–13 wt.%TiO2 nanostructured and submicron-nanostructured powders were deposited using APS. The feedstocks were obtained by spray drying from two starting suspensions, prepared by mixing two commercial nanosuspensions of Al2O3 and TiO2, or by adding nanosized TiO2 and submicron-sized Al2O3 powders to water. The spray-dried granules were heat-treated to reduce their porosity and the resultant powders were fully characterized. Optimization of the deposition conditions enabled the reconstituted powders to be successfully deposited, yielding coatings that were well bonded to the substrate. The coating microstructure, characterized by SEM, was formed by semi-molten feedstock agglomerates surrounded by fully molten particles that act as a binder. Moreover, microhardness, adhesion, and tribological behavior were determined, and the impact of the granule characteristics on these properties was studied.     Metric properties of discrete time exclusion type processes in continuum

    A new class of exclusion type processes acting in continuum with synchronous updating is introduced and studied. Ergodic averages of particle velocities are obtained and their connections to other statistical quantities, in particular to the particle density (the so called Fundamental Diagram) is analyzed rigorously. The main technical tool is a "dynamical" coupling applied in a nonstandard fashion: we do not prove the existence of the successful coupling (which even might not hold) but instead use its presence/absence as an important diagnostic tool. Despite that this approach cannot be applied to lattice systems directly, it allows to obtain new results for the lattice systems embedding them to the systems in continuum.
