Crossroads between peripheral atherosclerosis, western-type diet and skeletal muscle pathophysiology: emphasis on apolipoprotein E deficiency and peripheral arterial disease
Atherosclerosis is a chronic inflammatory process that, in the presence of hyperlipidaemia, promotes the formation of atheromatous plaques in large vessels of the cardiovascular system. It also affects peripheral arteries with major implications for a number of other non-vascular tissues such as the skeletal muscle, the liver and the kidney. The aim of this review is to critically discuss and assimilate current knowledge on the impact of peripheral atherosclerosis and its implications on skeletal muscle homeostasis. Accumulating data suggests that manifestations of peripheral atherosclerosis in skeletal muscle originates in a combination of increased i)-oxidative stress, ii)-inflammation, iii)-mitochondrial deficits, iv)-altered myofibre morphology and fibrosis, v)-chronic ischemia followed by impaired oxygen supply, vi)-reduced capillary density, vii)- proteolysis and viii)-apoptosis. These structural, biochemical and pathophysiological alterations impact on skeletal muscle metabolic and physiologic homeostasis and its capacity to generate force, which further affects the individual’s quality of life. Particular emphasis is given on two major areas representing basic and applied science respectively: a)-the abundant evidence from a well-recognised atherogenic model; the Apolipoprotein E deficient mouse and the role of a western-type diet and b)-on skeletal myopathy and oxidative stress-induced myofibre damage from human studies on peripheral arterial disease. A significant source of reactive oxygen species production and oxidative stress in cardiovascular disease is the family of NADPH oxidases that contribute to several pathologies. Finally, strategies targeting NADPH oxidases in skeletal muscle in an attempt to attenuate cellular oxidative stress are highlighted, providing a better understanding of the crossroads between peripheral atherosclerosis and skeletal muscle pathophysiology

Platelet releasate promotes skeletal myogenesis by increasing muscle stem cell commitment to differentiation and accelerates muscle regeneration following acute injury
Aim: The use of platelets as biomaterials has gained intense research interest. However,
the mechanisms regarding platelet-mediated skeletal myogenesis remain to be established.
The aim of this study was to determine the role of platelet releasate in skeletal myogenesis
and muscle stem cell fate in vitro and ex vivo respectively.
Methods: We analysed the effect of platelet releasate on proliferation and differentiation of
C2C12 myoblasts by means of cell proliferation assays, immunohistochemistry, gene
expression and cell bioenergetics. We expanded in vitro findings on single muscle fibres by
determining the effect of platelet releasate on murine skeletal muscle stem cells using
protein expression profiles for key myogenic regulatory factors.
Results: TRAP6 and collagen used for releasate preparation had a more pronounced effect
on myoblast proliferation versus thrombin and sonicated platelets (P<0.05). In addition,
platelet concentration positively correlated with myoblast proliferation. Platelet releasate
increased myoblast and muscle stem cell proliferation in a dose-dependent manner, which
was mitigated by VEGFR and PDGFR inhibition. Inhibition of VEGFR and PDGFR ablated
MyoD expression on proliferating muscle stem cells, compromising their commitment to
differentiation in muscle fibres (P<0.001). Platelet releasate was detrimental for myoblast
fusion and affected differentiation of myoblasts in a temporal manner. Most importantly we
show that platelet releasate promotes skeletal myogenesis through the PDGF/VEGF-Cyclin
D1-MyoD-Scrib-Myogenin axis and accelerates skeletal muscle regeneration after acute
injury.
Conclusion: This study provides novel mechanistic insights on the role of platelet releasate
in skeletal myogenesis and set the physiological basis for exploiting platelets as biomaterials
in regenerative medicine

Mechanisms underpinning the permanent muscle damage induced by snake venom metalloprotease
Snakebite is a major neglected tropical health issue that affects over 5 million people worldwide resulting in around 1.8 million envenomations and 100,000 deaths each year. Snakebite envenomation also causes innumerable morbidities specifically loss of limbs as a result of excessive tissue/muscle damage. Snake venom metalloproteases (SVMPs) are a predominant component of viper venoms, and are involved in the degradation of basement membrane proteins (particularly collagen) surrounding the tissues around the bite site. Although their collagenolytic properties have been established, the molecular mechanisms through which SVMPs induce permanent muscle damage are poorly understood. Here, we demonstrate the purification and characterisation of an SVMP from a viper (Crotalus atrox) venom. Mass spectrometry analysis confirmed that this protein is most likely to be a group III metalloprotease (showing high similarity to VAP2A) and has been referred to as CAMP (Crotalus atrox metalloprotease). CAMP displays both collagenolytic and fibrinogenolytic activities and inhibits CRP-XL-induced platelet aggregation. To determine its effects on muscle damage, CAMP was administered into the tibialis anterior muscle of mice and its actions were compared with cardiotoxin I (a three-finger toxin) from an elapid snake (Naja pallida) venom. Extensive immunohistochemistry analyses revealed that CAMP significantly damages skeletal muscles by attacking the collagen scaffold and other important basement membrane proteins, and prevents their regeneration through disrupting the functions of satellite cells. In contrast, cardiotoxin I destroys skeletal muscle by damaging the plasma membrane, but does not impact regeneration due to its inability to affect the extracellular matrix. Overall, this study provides novel insights into the mechanisms through which SVMPs induce permanent muscle damage

Attenuation of oxidative stress-induced lesions in skeletal muscle in a mouse model of obesity-independent hyperlipidaemia and atherosclerosis through the inhibition of Nox2 activity
Obesity leading to hyperlipidaemia and atherosclerosis is recognised to induce
morphological and metabolic changes in many tissues. However, both hyperlipidaemia and
atherosclerosis can occur in the absence of obesity. The impact of the latter scenario on
skeletal muscle and liver is not understood sufficiently. In this regard, we used the
Apolipoprotein E-deficient (ApoE-/-) mouse model, an established model of hyperlipidaemia
and atherosclerosis, that does not become obese when subjected to a high-fat diet, to
determine the impact of Western-type diet (WD) and ApoE deficiency on skeletal muscle
morphological, metabolic and biochemical properties. To establish the potential of
therapeutic targets, we further examined the impact of Nox2 pharmacological inhibition on
skeletal muscle redox biology. We found ectopic lipid accumulation in skeletal muscle and
the liver, and altered skeletal muscle morphology and intramuscular triacylglycerol fatty acid
composition. WD and ApoE deficiency had a detrimental impact in muscle metabolome,
followed by perturbed gene expression for fatty acid uptake and oxidation. Importantly, there
was enhanced oxidative stress in the skeletal muscle and development of liver steatosis,
inflammation and oxidative protein modifications. Pharmacological inhibition of Nox2
decreased reactive oxygen species production and protein oxidative modifications in the
muscle of ApoE-/- mice subjected to a Western-type diet. This study provides key evidence to
better understand the pathophysiology of skeletal muscle in the context of hyperlipidaemia
and atherosclerosis and identifies Nox2 as a potential target for attenuating oxidative stress
in skeletal muscle in a mouse model of obesity-independent hyperlipidaemia
Humidity resistant fabrication of CH3NH3PbI3 perovskite solar cells and modules
A humidity resistant and versatile fabrication method for the production of very high quality, organic-inorganic perovskite films, solar cells and solar modules is presented. By using ethyl acetate as an anti-solvent during deposition, perovskite solar cells with power conversion efficiencies (PCEs) up to 15% were fabricated in a 75% relative humidity (RH) environment. Ethyl acetate acts as a moisture absorber during spin-coating, protecting sensitive perovskite intermediate phases from airborne water during film formation and annealing. We have demonstrated the manufacture of 50 mm x 50 mm series interconnected modules with PCEs in excess of 10% for 13.5 cm2 devices processed in air at 75%RH and 11.8% at 50%RH. To the best of our knowledge, these results represent the highest efficiency for perovskite solar modules processed under high humidity ambient conditions. This new deposition protocol allows for low-cost, efficient and consistent device fabrication in humid climates and uncontrolled laboratories

Platelet releasate normalises the compromised muscle regeneration in a mouse model of hyperlipidaemia
Muscle satellite cells are important stem cells for skeletal muscle regeneration
and repair after injury. ApoE deficient mice, an established mouse model of
hyperlipidaemia and atherosclerosis, show evidence of oxidative stress-induced
lessions and fat infiltration in skeletal muscle followed by impaired repair after injury. However, the mechanisms underpinning attenuated muscle regeneration remain to be fully defined. Key to addressing the latter is to understand the properties of muscle stem cells from ApoE deficient mice and their myogenic potential. Muscle stem cells from ApoE deficient mice were cultured both ex vivo (on single fibres) and in vitro (primary myoblasts) and their myogenic
capacity was determined. Skeletal muscle regeneration was studied on days 5 and 10 after cardiotoxin injury. ApoE deficient muscle stem cells showed delayed activation and differentiation on single muscle fibres ex vivo. Impaired
proliferation and differentiation profiles were also evident on isolated primary muscle stem cells in culture. ApoE deficient mice displayed impaired skeletal muscle regeneration after acute injury in vivo. Administration of platelet releasate in ApoE deficient mice reversed the deficits of muscle regeneration after acute injury to wild-type levels. These findings indicate that muscle stem
cell myogenic potential is perturbed in skeletal muscle of a mouse model of hyperlipidaemia. We propose that platelet-releasate could be a therapeutic intervention for conditions with associated myopathy such as peripheral arterial
disease

Optimising platelet secretomes to deliver robust tissue specific regeneration
Promoting cell proliferation is the cornerstone of most tissue regeneration therapies. As platelet‐based applications promote cell division and can be customised for tissue‐specific efficacy, this makes them strong candidates for developing novel regenerative therapies. Therefore, the aim of this study was to determine if platelet releasate could be optimised to promote cellular proliferation and differentiation of specific tissues. Growth factors in platelet releasate were profiled for physiological and supra‐physiological platelet concentrations. We analysed the effect of physiological and supra‐physiological releasate on C2C12 skeletal myoblasts, H9C2 rat cardiomyocytes, human dermal fibroblasts (HDF), HaCaT keratinocytes and chondrocytes. Cellular proliferation and differentiation were assessed through proliferation assays, mRNA and protein expression. We show that supra‐physiological releasate is not simply a concentrated version of physiological releasate. Physiological releasate promoted C2C12, HDF and chondrocyte proliferation with no effect on H9C2 or HaCaT cells. Supra‐physiological releasate induced stronger proliferation in C2C12 and HDF cells compared to physiological releasate. Importantly, supra‐physiological releasate induced proliferation of H9C2 cells. The proliferative effects of skeletal and cardiac muscle cells were in part driven by VEGFα. Furthermore, supra‐physiological releasate induced differentiation of H9C2 and C2C12, HDF and keratinocyte differentiation. This study provides insights into the ability of releasate to promote muscle, heart, skin and cartilage cell proliferation and differentiation and highlights the importance of optimising releasate composition for tissue‐specific regeneration
Innovative materials for nanacrystalline solar cells
This PhD dissertation examines the possibility of converting light energy into electricity by taking advantage of the properties of third generation solid state solar cells. With the purpose of reducing manufacturing costs, third generation solar cells are constructed by mild chemical processes under ambient conditions and they can be distinguished into a few sub-categories: photosensitized solar cells using liquid electrolyte, photosensitized solar cells in which the liquid electrolyte is replaced by a hole transporting material, quantum dot solar cells, organic solar cells and perovskite solar cells. During the present study, only solid state solar cells using hole transporting materials were examined, more specifically, quantum dot solar cells and perovskite solar cells.Solid state solar cells are constructed by depositing successive mesoporous layers on a transparent conductive substrate. The active films of this type of solar cells are a compact layer of titanium dioxide, a mesoporous titanium dioxide film, a layer of a photosensitizer and a layer of a hole transporting material. In order to collect the photocurrent, a metal electrode is grown on the top of the above mentioned layers. When the photosensitive material is exposed to light, electrons move from the valence band to the conduction band of the sensitizer resulting in an accumulation of holes in its valence band. The charge carriers move towards opposite directions-paths through the bulk of the materials of the various substrates. Thus the non recombined charges end up in the metal electrodes. Solar cells generally function by the separation of photogenerated electrons and holes. In the present case separation is achieved through the injection of electrons and holes into the n-type semiconductor (i.e. titania) and the hole transporting material, respectively. The hole transporting material should have the appropriate potential in order to ensure that the holes can be injected in it. In this study two types of sensitizers and their ability to be combined with various hole transporting materials were examined.Firstly, quantum dot solid state solar cells where constructed by using as sensitizers CdSe and ZnSe both individually and mixed. Advantages that derive from the combination of different quantum dots result into more efficient solar cells compared to the individual use of each one. Moreover, quantum dot solar cells were constructed by using Sb2S3 as sensitizer, synthesized through two different techniques in order to examine the more effective one. In all tested quantum dot solar cells, P3HT was used as the organic p type semiconductor.Perovskites are materials of great interest due to their ability to absorb light in the whole range of visible spectrum. The perovskite structure that was synthesized and studied in this work was CH3NH3PbI3-xCl3. Following the optimization of the synthesis and deposition parameters of the perovskite layer, various hole transporting materials were used in order to increase the efficiency and decrease the cost. The typical representative of hole transporting materials is Spiro-OMeTAD. Although the efficiency of CH3NH3PbI3-xCl3/ Spiro-OMeTAD was high enough, the high cost of this material lead to the need of turning into other low cost materials. Several commercial or synthesized phthalocyanines were tested for this purpose with the perovskites. These structures have offered satisfactory efficiency.Overall in this dissertation, synthesis, characterization and deposition processes of innovative materials in the form of thin films were studied. In addition, construction and characterization of devices and integrated modules based on the appropriate combination of these materials has been studied.Η παρούσα διδακτορική διατριβή εξετάζει τη δυνατότητα μετατροπής της φωτεινής ενέργειας σε ηλεκτρική μέσω εκμετάλλευσης των ιδιοτήτων ευαισθητοποιημένων ηλιακών κυψελίδων τρίτης γενιάς, στερεάς μορφής.
Epidemiological study of dermatological diseases in swimmers
The skin is the largest organ of the human body which has lipids that differ from those of aquatic animals and make it susceptible to the harmful effects of maceration, pollution, allergy, insect bites and extreme temperatures. Τhe special conditions that prevail in swimming pools, the exposure to humidity, heat, cold, wind, sun and the chemical products used for the maintenance of water have an important role in the manifestation of cutaneous diseases. For the purpose of this study, these skin diseases were classified into categories according to their type - allergic diseases and dermatitis, viral, fungal and microbial infections, as well as other cutaneous diseases and injuries. The purpose of this study was the recording of the prevalence of cutaneous diseases that occur in junior (9-12 years old) and age group swimming categories (> 13 years old) within Greek territory. This cross-sectional, non-interventional study was conducted with the approval of the Ethics Committee of the University of Western Attica (pro. no. 52645-20/07/2020) and the Hellenic Swimming Federation, which participated in sending the relevant e-questionnaire (google forms) via e-mail to the swimming clubs. The questionnaire was divided into two parts. The first part concerned general information such as demographics, training routine, behavior in the swimming pool area and general information about skin health. The second part consisted of questions related to multiple infections, location of infections, the season of manifestation, absence from the training program because of the infections and dermatological treatment. In case of recurrent infections participants were asked to report on their last episode of the infection as it was easier to recall it with accuracy. The categorical variables are presented as absolute (n) and related (%) frequencies. The chi-square test (X2)—Fisher’s exact test was used to investigate the existence of a relationship between two categorical variables. To investigate the existence of a relationship between a categorical and an ordinal variable the chi-square trend test was used. In addition, in order to calculate the probability of a skin disease occurrence while considering the effect of some independent variables that appear statistically significant in the initial investigation of the analysis, multivariate logistic regression was applied and the odds ratios, the 95% confidence intervals and the p values are presented. The two-sided level of statistical significance was set at 0.05. Data analysis was estimate with SPSS 25.0.1047 male and female swimmers were enrolled in this study, representing 9.2% of all junior and age group swimming categories. Female swimmers represented 55.1% of the sample. 60.8% used an open facility, 44.3% trained after 17:00 PM and most participants had been swimming for 7-9 years. Additionally, they did more than six training sessions per week and had been swimming for 2 hours per day. 48.8% of the participants came from swimming clubs in Attica county and 18.1% from swimming clubs in Thessaloniki county. The cutaneous diseases with the most manifestations were xerosis with a percentage of 38,6%, warts (35,4%), acne (16,3%), Tinea pedis (16%), urticaria (15,3%), pityriasis versicolor (13%), dermatitis from chemical pool disinfectants (11,3%), dermatitis from bathing suits (11,2%) and impetigo (10,5%). In allergies and dermatitis categories there was a definite ascendance of female swimmers in the manifestation of symptoms compared to male swimmers. The factors which were mainly associated with the occurrence of pool-related allergies and dermatitis were pre-existing allergies such as seasonal and skin allergy. Accordingly, the majority of occurrences of viral, fungal and bacterial infections are not affected by gender, while adult swimmers were those who showed the highest prevalence due to higher swimming frequency. The factors associated with the occurrence of these cutaneous diseases are related to a pre-existing skin or seasonal allergy, habits such as walking without flip flops in pool areas, placing the bathrobe on communal benches and behaviors such as sharing swimming equipment. In the category of other cutaneous diseases, sunburn (p13 ετών) στην ελληνική επικράτεια.
