METHODOLOGY/PRINCIPAL FINDINGS: A cross-sectional survey was performed between 15 July 2020 and 23 July 2020 among individuals who visited 4 medical laboratories or 5 health centers for routine screening or clinical management, with the exception of symptomatic suggestive cases of covid-19. Samples were screened for the presence of anti-SARS-CoV-2 IgG directed against domain S1 of the SARS-CoV-2 spike protein using the anti-SARS-CoV-2 enzyme-linked immunosorbent assay (ELISA) from Euroimmun. CONCLUSIONS/SIGNIFICANCE: The overall seroprevalence was 15.4% [9.3%-24.4%] among 480 participants, ranging from 4.0% to 25.5% across the different municipalities. The seroprevalence did not differ according to gender (p = 0.19) or age (p = 0.51). Among SARS-CoV-2 positive individuals, we found that 24.6% [11.5%-45.2%] reported symptoms consistent with COVID-19. Our findings revealed high levels of infection across the territory but a low number of resulting deaths, which can be explained by French Guiana's young population structure

    Fehlgefaltete sekretorische Proteine werden aus dem Lumen des Endoplasmatischen Retikulums (ER) in das Cytoplasma transloziert, wo sie von dem Ubiquitin-Proteasom-System degradiert werden (ER-assoziierte Degradation, ERAD). Wie genau die Proteine aus dem ER transportiert werden, ist noch nicht geklĂ€rt. Verschiedene Studien, die eine einzige „leaky“ sec63-Mutante verwendeten, lassen vermuten, dass Sec63p beim Proteinexport beteiligt sein könnte. Um diesen Sachverhalt genauer zu erforschen, habe ich einen Screen verwendet, bei dem willkĂŒrliche Punktmutationen in das SEC63-Gen eingefĂŒgt wurden. Ich habe die Mutanten auf Akkumulation des ERAD-Substrats CPY* getestet, welche auf einen Defekt bei dessen Degradation hindeutet. Ich habe sechs Mutanten isoliert, wovon nur sec63-402 eine deutliche Akkumulation von CPY* aufwies. Diese Mutante trĂ€gt eine Mutation in dem Interaktionsbereich mit dem Chaperon Kar2p, der sogenannten J-DomĂ€ne. Mutanten mit Punktmutationen in der Transmembran-, Brl- oder sauren DomĂ€ne wiesen keine Degradationsdefekte auf. Durch Untersuchung von anderen ERAD-Substraten zeigte sich, dass sich der Degradationsdefekt in sec63-mutierten Hefezellen auf lösliche Proteine beschrĂ€nkt. Meine Ergebnisse deuten darauf hin, dass Sec63p durch seine Interaktion mit Kar2p an der Degradation von löslichen ERAD-Substraten beteiligt ist.Misfolded secretory proteins are translocated from the lumen of the endoplasmic reticulum (ER) to the cytoplasm for degradation by the ubiquitin-proteasome system (ER-associated degradation, ERAD). How exactly this export from the ER is accomplished is still not understood. Several studies using a single leaky sec63 mutant suggested that Sec63p might be involved in protein export. To examine this issue further, I used a screen in which random point mutations were inserted into the SEC63 gene. I tested the mutants for accumulation of the ERAD substrate CPY*. Accumulation of CPY* in cells indicates a degradation defect. I isolated six sec63 mutants of which only sec63-402 showed a strong accumulation of CPY*. This mutant carries a mutation in the interaction area with the chaperone Kar2p, the so called J-domain. Mutants with point mutations in the transmembrane, the Brl or the acidic domain caused no detectable ERAD defect for CPY*. The examination of other ERAD substrates showed that sec63-402 affected only degradation of soluble substrates. This demonstrates that in yeast the interaction of Sec63p with Kar2p is important for the specific degradation of soluble ERAD substrates
