    Rat Monoclonal Antibodies Specific for LST1 Proteins

    The LST1 gene is located in the human MHC class III region and encodes transmembrane and soluble isoforms that have been suggested to play a role in the regulation of the immune response and are associated with inflammatory diseases such as rheumatoid arthritis. Here we describe the generation and characterization of the first monoclonal antibodies against LST1. Two hybridoma lines secreting monoclonal antibodies designated 7E2 and 8D12 were established. The 7E2 antibody detects recombinant and endogenous LST1 by Western blot analysis while 8D12 reacts with recombinant and endogenous LST1 in immunoprecipitation and flow cytometry procedures. The newly established antibodies were used to survey LST1 protein expression in human cell lines, which was found to be tightly regulated, allowing the expression of transmembrane isoforms but suppressing soluble isoforms

    Simulating personal carbon trading (Pct) with an agent-based model (abm): Investigating adaptive reduction rates and path dependence

    A decade after the publication of seminal papers on personal carbon trading (PCT), few empirical studies on its implementation exist. Investigating how to design, set up and implement a PCT scheme for a community or country raises several difficulties. For instance, it is unclear how to introduce a reduction rate of CO2 allowances to ensure a steady decrease in CO2 emissions from households. Computational approaches have been introduced to address these challenges of PCT by providing an opportunity to test counterfactual scenarios. Among the benefits of an agent-based modeling approach (ABM) is the potential to directly address dynamic developments and introduce counterfactual situations. In this paper, we review existing modeling approaches and present an ABM for PCT. With simulations of an artificial population of 1000 and 30,000 agents, we address questions on the price and reduction rate of allowances. A key contribution of our model is the inclusion of an adaptive reduction rate, which reduces the yearly allocated amount of allowances depending on a set CO2 abatement target. The results confirm that increased emissions targets are related to higher allowance prices and a higher proportion of buying households. Our analysis also suggests a significant path dependence in the dynamics of allowance prices and availability, but that adaptive reduction rates have little impact on outcomes other than the price. We discuss data availability and computational challenges to modeling a PCT scheme with an ABM. Ideal data to populate an ABM on PCT are not available due to the lack of real-world implementations of a PCT. Nonetheless, meaningful insights about the dynamics and the focal variables in a PCT scheme can be generated by the exploratory use of an ABM. © 2021 by the authors.     Voltage-Controlled Optics of a Quantum Dot

    We show how the optical properties of a single semiconductor quantum dot can be controlled with a small dc voltage applied to a gate electrode. We find that the transmission spectrum of the neutral exciton exhibits two narrow lines with 2\sim 2 μ\mueV linewidth. The splitting into two linearly polarized components arises through an exchange interaction within the exciton. The exchange interaction can be turned off by choosing a gate voltage where the dot is occupied with an additional electron. Saturation spectroscopy demonstrates that the neutral exciton behaves as a two-level system. Our experiments show that the remaining problem for manipulating excitonic quantum states in this system is spectral fluctuation on a μ\mueV energy scale.Comment: 4 pages, 4 figures; content as publishe

    Single-molecule real-time sequencing combined with optical mapping yields completely finished fungal genome

    Next-generation sequencing (NGS) technologies have increased the scalability, speed, and resolution of genomic sequencing and, thus, have revolutionized genomic studies. However, eukaryotic genome sequencing initiatives typically yield considerably fragmented genome assemblies. Here, we assessed various state-of-the-art sequencing and assembly strategies in order to produce a contiguous and complete eukaryotic genome assembly, focusing on the filamentous fungus Verticillium dahliae. Compared with Illumina-based assemblies of the V. dahliae genome, hybrid assemblies that also include PacBio- generated long reads establish superior contiguity. Intriguingly, provided that sufficient sequence depth is reached, assemblies solely based on PacBio reads outperform hybrid assemblies and even result in fully assembled chromosomes. Furthermore, the addition of optical map data allowed us to produce a gapless and complete V. dahliae genome assembly of the expected eight chromosomes from telomere to telomere. Consequently, we can now study genomic regions that were previously not assembled or poorly assembled, including regions that are populated by repetitive sequences, such as transposons, allowing us to fully appreciate an organism’s biological complexity. Our data show that a combination of PacBio-generated long reads and optical mapping can be used to generate complete and gapless assemblies of fungal genomes. IMPORTANCE Studying whole-genome sequences has become an important aspect of biological research. The advent of nextgeneration sequencing (NGS) technologies has nowadays brought genomic science within reach of most research laboratories, including those that study nonmodel organisms. However, most genome sequencing initiatives typically yield (highly) fragmented genome assemblies. Nevertheless, considerable relevant information related to genome structure and evolution is likely hidden in those nonassembled regions. Here, we investigated a diverse set of strategies to obtain gapless genome assemblies, using the genome of a typical ascomycete fungus as the template. Eventually, we were able to show that a combination of PacBiogenerated long reads and optical mapping yields a gapless telomere-to-telomere genome assembly, allowing in-depth genome sanalyses to facilitate functional studies into an organism’s biology

    Optical detection of single electron spin resonance in a quantum dot

    We demonstrate optically detected spin resonance of a single electron confined to a self-assembled quantum dot. The dot is rendered dark by resonant optical pumping of the spin with a coherent laser. Contrast is restored by applying a radio frequency (rf) magnetic field at the spin resonance. The scheme is sensitive even to rf fields of just a few micro-T. In one case, the spin resonance behaves exactly as a driven 3-level quantum system (a lambda-system) with weak damping. In another, the dot exhibits remarkably strong (67% signal recovery) and narrow (0.34 MHz) spin resonances with fluctuating resonant positions, evidence of unusual dynamic processes of non-Markovian character.Comment: 4 pages, 5 figure

    Modeling Social-Ecological Feedback Effects in the Implementation of Payments for Environmental Services in Pasture-Woodlands

    International audienceAn effective implementation of payment for environmental services (PES) must allow for complex interactions of coupled social-ecological systems. We present an integrative study of the pasture-woodland landscape of the Swiss Jura Mountains combining methods from natural and social sciences to explore feedback between vegetation dynamics on paddock level, farm-based decision making, and policy decisions on the national political level. Our modeling results show that concomitant climatic and socioeconomic changes advance the loss of open grassland in silvopastoral landscapes. This would, in the longer term, deteriorate the historical wooded pastures in the region, which fulfill important functions for biodiversity and are widely considered as landscapes that deserve protection. Payment for environmental services could counteract this development while respecting historical land-use and ecological boundary conditions. The assessed policy feedback process reveals that current policy processes may hinder the implementation of PES, even though a payment for the upkeep of wooded pasture would generally enjoy the backing of the relevant policy network. To effectively support the upkeep of the wooded pastures in the Jura, concomitant policy changes, such as market deregulation, must also be taken into account
