    The Internet Implementation of the Hierarchical Aggregate Assessment Process with the ā€œClusterā€ Wi-Fi E-Learning and EAssessment Application ā€” A Particular Case of Teamwork Assessment

    A Wi-Fi e-learning and e-assessment Internet application named ā€œClusterā€ was developed in the context of a research project concerning the implementation of a teamwork assessment mobile application able to assess teams with several levels of hierarchy. Usually, teamwork assessment software and Internet applications for several hierarchy level teams are included in the field of Management Information Systems (MIS). However, some assessment tasks in teams with several levels of hierarchy and assessment may be performed in an educational context, and the existing applications for the assessment and evaluation of teams with several levels of hierarchy are not applications dedicated to the assessment of students in an educational context. The ā€œClusterā€ application is able to present the course material, to train the students in teams as well as to present individual and team assessment tasks. The applicationā€™s special functionalities enable it to assess the teams at several levels of hierarchy, which constitute the hierarchical aggregate assessment process. In effect, the members of the teams may have appointments of team member, team leader and team administrator that supervises team leaders. This application can therefore evaluate simultaneously different knowledge and skills in the same assessment task based on the hierarchical position of the team member. The summative evaluation of the application consists of work to submit as well as objective examinations in HTML format, while the formative evaluation is composed of assessment grid computer forms of self-assessment and peer assessment. The application contains two mutually exclusive modes, the assessor mode and the student mode. The assessor mode allows the teacher to create courses, manage students, form the teams and also assess the students and the teams in a summative manner. The student mode allows the students to follow courses, write exams, submit homework, perform in teams and submit self- and peers formative assessment. The theoretical consideration of the project establishes the link between hierarchical aggregate assessment applications and management information systems (MIS). The application is an electronic portfolio (e-portfolio) management system in the competency-based learning and an Internet test administration system in the mastery learning approach. The aim of the chapter is to introduce the reader to the field of hierarchical aggregate assessment and to show how to implement complex assessment tasks with several levels of hierarchy into an Internet software application

    A refined radio-telemetry technique to monitor right ventricle or pulmonary artery pressures in rats: a useful tool in pulmonary hypertension research

    Implantable radio-telemetry methodology, allowing for continuous recording of pulmonary haemodynamics, has previously been used to assess effects of therapy on development and treatment of pulmonary hypertension. In the original procedure, rats were subjected to invasive thoracic surgery, which imposes significant stress that may disturb critical aspects of the cardiovascular system and delay recovery. In the present study, we describe and compare the original trans-thoracic approach with a new, simpler trans-diaphragm approach for catheter placement, which avoids the need for surgical invasion of the thorax. Satisfactory overall success rates up to 75% were achieved in both approaches, and right ventricular pressures and heart and respiratory rates normalised within 2Ā weeks. However, recovery was significantly faster in trans-diaphragm than in trans-thoracic operated animals (6.4ā€‰Ā±ā€‰0.5 vs 9.5ā€‰Ā±ā€‰1.1Ā days, respectively; pā€‰<ā€‰0.05). Stable right ventricular pressures were recorded for more than 4Ā months, and pressure changes, induced by monocrotaline or pulmonary embolisms, were readily detected. The data demonstrate that right ventricular telemetry is a practicable procedure and a useful tool in pulmonary hypertension research in rats, especially when used in combination with echocardiography. We conclude that the described trans-diaphragm approach should be considered as the method of choice, for it is less invasive and simpler to perform

    Inhibition of SOC/Ca2+/NFAT pathway is involved in the anti-proliferative effect of sildenafil on pulmonary artery smooth muscle cells

    <p>Abstract</p> <p>Background</p> <p>Sildenafil, a potent phosphodiesterase type 5 (PDE5) inhibitor, has been proposed as a treatment for pulmonary arterial hypertension (PAH). The mechanism of its anti-proliferative effect on pulmonary artery smooth muscle cells (PASMC) is unclear. Nuclear translocation of nuclear factor of activated T-cells (NFAT) is thought to be involved in PASMC proliferation and PAH. Increase in cytosolic free [Ca<sup>2+</sup>] ([Ca<sup>2+</sup>]<sub>i</sub>) is a prerequisite for NFAT nuclear translocation. Elevated [Ca<sup>2+</sup>]<sub>i </sub>in PASMC of PAH patients has been demonstrated through up-regulation of store-operated Ca<sup>2+ </sup>channels (SOC) which is encoded by the transient receptor potential (TRP) channel protein. Thus we investigated if: 1) up-regulation of TRPC1 channel expression which induces enhancement of SOC-mediated Ca<sup>2+ </sup>influx and increase in [Ca<sup>2+</sup>]<sub>i </sub>is involved in hypoxia-induced PASMC proliferation; 2) hypoxia-induced promotion of [Ca<sup>2+</sup>]<sub>i </sub>leads to nuclear translocation of NFAT and regulates PASMC proliferation and TRPC1 expression; 3) the anti-proliferative effect of sildenafil is mediated by inhibition of this SOC/Ca<sup>2+</sup>/NFAT pathway.</p> <p>Methods</p> <p>Human PASMC were cultured under hypoxia (3% O<sub>2</sub>) with or without sildenafil treatment for 72 h. Cell number and cell viability were determined with a hemocytometer and MTT assay respectively. [Ca<sup>2+</sup>]<sub>i </sub>was measured with a dynamic digital Ca<sup>2+ </sup>imaging system by loading PASMC with fura 2-AM. TRPC1 mRNA and protein level were detected by RT-PCR and Western blotting respectively. Nuclear translocation of NFAT was determined by immunofluoresence microscopy.</p> <p>Results</p> <p>Hypoxia induced PASMC proliferation with increases in basal [Ca<sup>2+</sup>]<sub>i </sub>and Ca<sup>2+ </sup>entry via SOC (SOCE). These were accompanied by up-regulation of TRPC1 gene and protein expression in PASMC. NFAT nuclear translocation was significantly enhanced by hypoxia, which was dependent on SOCE and sensitive to SOC inhibitor SKF96365 (SKF), as well as cGMP analogue, 8-brom-cGMP. Hypoxia-induced PASMC proliferation and TRPC1 up-regulation were inhibited by SKF and NFAT blocker (VIVIT and Cyclosporin A). Sildenafil treatment ameliorated hypoxia-induced PASMC proliferation and attenuated hypoxia-induced enhancement of basal [Ca<sup>2+</sup>]<sub>i</sub>, SOCE, up-regulation of TRPC1 expression, and NFAT nuclear translocation.</p> <p>Conclusion</p> <p>The SOC/Ca<sup>2+</sup>/NFAT pathway is, at least in part, a downstream mediator for the anti-proliferative effect of sildenafil, and may have therapeutic potential for PAH treatment.</p

    Elevated Uptake of Plasma Macromolecules by Regions of Arterial Wall Predisposed to Plaque Instability in a Mouse Model

    Atherosclerosis may be triggered by an elevated net transport of lipid-carrying macromolecules from plasma into the arterial wall. We hypothesised that whether lesions are of the thin-cap fibroatheroma (TCFA) type or are less fatty and more fibrous depends on the degree of elevation of transport, with greater uptake leading to the former. We further hypothesised that the degree of elevation can depend on haemodynamic wall shear stress characteristics and nitric oxide synthesis. Placing a tapered cuff around the carotid artery of apolipoprotein E -/- mice modifies patterns of shear stress and eNOS expression, and triggers lesion development at the upstream and downstream cuff margins; upstream but not downstream lesions resemble the TCFA. We measured wall uptake of a macromolecular tracer in the carotid artery of C57bl/6 mice after cuff placement. Uptake was elevated in the regions that develop lesions in hyperlipidaemic mice and was significantly more elevated where plaques of the TCFA type develop. Computational simulations and effects of reversing the cuff orientation indicated a role for solid as well as fluid mechanical stresses. Inhibiting NO synthesis abolished the difference in uptake between the upstream and downstream sites. The data support the hypothesis that excessively elevated wall uptake of plasma macromolecules initiates the development of the TCFA, suggest that such uptake can result from solid and fluid mechanical stresses, and are consistent with a role for NO synthesis. Modification of wall transport properties might form the basis of novel methods for reducing plaque rupture

    New perspectives for the treatment of pulmonary hypertension

    Pulmonary hypertension (PH) is a debilitating disease with a poor prognosis. Therapeutic options remain limited despite the introduction of prostacyclin analogues, endothelin receptor antagonists and phosphodiesterase 5 inhibitors within the last 15 years; these interventions address predominantly the endothelial and vascular dysfunctionS associated with the condition, but simply delay progression of the disease rather than offer a cure. In an attempt to improve efficacy, emerging approaches have focused on targeting the pro-proliferative phenotype that underpins the pulmonary vascular remodelling in the lung and contributes to the impaired circulation and right heart failure. Many novel targets have been investigated and validated in animal models of PH, including modulation of guanylate cyclases, phosphodiesterases, tyrosine kinases, Rho kinase, bone morphogenetic proteins signalling, 5-HT, peroxisome proliferator activator receptors and ion channels. In addition, there is hope that combinations of such treatments, harnessing and optimizing vasodilator and anti-proliferative properties, will provide a further, possibly synergistic, increase in efficacy; therapies directed at the right heart may also offer an additional benefit. This overview highlights current therapeutic options, promising new therapies, and provides the rationale for a combination approach to treat the disease

    A study of efficiency droop of green light emitting diodes grown by metalorganic chemical vapor deposition

    The objective of this thesis is to discuss the solutions investigated by AMDG (Advanced Materials and Devices Group) to reduce the "efficiency droop" effect that occurs in III-Nitrides Light Emitting Diodes (LEDs) when driven at high injection current densities. The efficiency droop refers to a decrease of the LED light emission efficiency when increases the current density from low values ~10 A/cm2 to higher values >100A/cm2. Many scientific papers have been written about the possible reasons for this phenomenon. Therefore, this thesis will discuss the different effects suspected to contribute to the droop, and discuss LED structure modifications studied by Dr. Dupuis' research group to reduce their impact. In addition to a description of a conventional LED structure, a discussion of the device fabrication process will be provided including the solutions investigated in our group to improve LED performance. Because measurement is critical to our studies, a description of the equipment used by the AMDG will be provided, e.g., the Electroluminescence (EL) and Photoluminescence (PL) test stations, Atomic Force Microscopy (AFM) for surface topology, TLM for metallic contact resistivity, X-Ray diffraction for crystal quality and epitaxial layer structure, and Hall-Effect measurement for doping concentration characterization and material resistivity. Because the IQE gives us a direct assessment of the active region's crystal quality, the setup and operation of a new Temperature-Dependent PL (TD-PL) system to measure the Internal Quantum Efficiency (IQE) was the main focus of this research. The External Quantum Efficiency (EQE) is measured using electroluminescence measurements. The EL measurements involve the acquisition of the emitted light spectrum along with different processed data such as the Full-Width at Half Maximum (FWHM) of the spectral intensity, the peak wavelength, output power, etc., which allows a comparison of the different LED structure performances. Within this work, a new LabVIEWĀ© program (called QuickTest 2.0) has been developed in order to automate the instrumentation setup and improve both the speed and accuracy of EL acquisition. A brief description of the G language used by the LabVIEWĀ© software will be provided along with the objective and motivation for upgrading the program, the general features of the program, and a comparison of spectrum acquisition and processed data results.     A novel wireless tongue tracking system for speech applications

    The technological innovation of MagTrack is its ability to track the position of a magnetic tracer wirelessly, and with an accuracy of less than 2 mm, by implementing a novel permanent magnet localization method. Combining its wireless and high-accuracy tracking capability with the millimetric dimensions of the tracer, MagTrack is very well-suited for tongue tracking. Compared to commercially available tongue tracking systems, MagTrack has many of their advantages but without most of their shortcomings. For instance, MagTrack can be embedded in a headset to be portable, is affordable by using off-the-shelf components and custom-designed parts that are 3D-printed in our lab, and the tongue can be tracked wirelessly, thus enabling it to move without any hindrance. Having access to tongue motion is important for many applications. Chief among them is the treatment of speech sound disorders in which the errant placement of the tongue is responsible for the decreased speech intelligibility. A preliminary study was conducted to assess the feasibility of MagTrack to be used in gamified visual feedback that would help patients better practice correct placement of their tongue by providing objective and quantitative measures of speech performance based on the error between their tongue placement and carefully-selected visual targets. Another promising application for MagTrack is as the input of a silent speech interface that can recognize speech solely from tongue motion to generate synthesized voice for people that cannot produce sounds. Pilot human studies were conducted to obtain a preliminary assessment of speech recognition accuracy and the quality of synthesized speech from tongue motion recorded on hundreds of utterances by MagTrack. Finally, since MagTrack is an improved version of the Tongue Drive System, it has the potential to be a practical assistive technology to help people with quadriplegia to regain autonomy for some tasks of their daily life such as driving their powered wheelchair, or even control their computer and mobile devices without the help of their caregivers.Ph.D
