    Measuring fast electron spectra and laser absorption in relativistic laser-solid interactions using differential bremsstrahlung photon detectors

    A photon detector suitable for the measurement of bremsstrahlung spectra generated in relativistically-intense laser-solid interactions is described. The Monte Carlo techniques used to back-out the fast electron spectrum and laser energy absorbed into fast electrons are detailed. A relativistically-intense laser-solid experiment using frequency doubled laser light is used to demonstrate the effective operation of the detector. The experimental data was interpreted using the 3-spatial-dimension Monte Carlo code MCNPX (Pelowitz 2008), and the fast electron temperature found to be 125 keV

    The structure of Phocaeicola vulgatus sialic acid acetylesterase

    Sialic acids terminate many N- and O-glycans and are widely distributed on cell surfaces. There are a diverse range of enzymes which interact with these sugars throughout the tree of life. They can act as receptors for influenza and specific betacoronaviruses in viral binding and their cleavage is important in virion release. Sialic acids are also exploited by both commensal and pathogenic bacteria for nutrient acquisition. A common modification of sialic acid is 9-O-acetylation, which can limit the action of sialidases. Some bacteria, including human endosymbionts, employ esterases to overcome this modification. However, few bacterial sialic acid 9-O-acetylesterases (9-O-SAEs) have been structurally characterized. Here, the crystal structure of a 9-O-SAE from Phocaeicola vulgatus (PvSAE) is reported. The structure of PvSAE was determined to resolutions of 1.44 and 2.06 Å using crystals from two different crystallization conditions. Structural characterization revealed PvSAE to be a dimer with an SGNH fold, named after the conserved sequence motif of this family, and a Ser-His-Asp catalytic triad. These structures also reveal flexibility in the most N-terminal α-helix, which provides a barrier to active-site accessibility. Biochemical assays also show that PvSAE deacetylates both mucin and the acetylated chromophore para-nitrophenyl acetate. This structural and biochemical characterization of PvSAE furthers the understanding of 9-O-SAEs and may aid in the discovery of small molecules targeting this class of enzyme.Bio-organic Synthesi

    X-ray and Optical Study of Low Core Density Globular Clusters NGC6144 and E3

    We report on the Chandra X-ray Observatory and Hubble Space Telescope observation of two low core density globular clusters, NGC6144 and E3. By comparing the number of X-ray sources inside the half-mass radius to those outside, we found 6 X-ray sources within the half-mass radius of NGC6144, among which 4 are expected to be background sources; 3 X-ray sources are also found within the half-mass radius of E3, of which 3 is expected to be background source. Therefore, we cannot exclude that all our sources are background sources. However, combining the results from X-ray and optical observations, we found that 1-2 sources in NGC6144 and 1 source in E3 are likely to be cataclysmic variables and that 1 source in NGC6144 is an active binary, based on the X-ray and optical properties. The number of faint X-ray sources in NGC6144 and E3 found with Chandra and HST is higher than a prediction based on collision frequency, but is closer to that based on mass.     An R^4 non-renormalisation theorem in N=4 supergravity

    We consider the four-graviton amplitudes in CHL constructions providing four-dimensional N=4 models with various numbers of vector multiplets. We show that in these models the two-loop amplitude has a prefactor of d^2R^4. This implies a non-renormalisation theorem for the R^4 term, which forbids the appearance of a three-loop ultraviolet divergence in four dimensions in the four-graviton amplitude.     Fishing within offshore wind farms in the North Sea:Stakeholder perspectives for multi-use from Scotland and Germany

    Offshore wind power generation requires large areas of sea to accommodate its activities, with increasing claims for exclusive access. As a result, pressure is placed on other established maritime uses, such as commercial fisheries. The latter sector has often been taking a back seat in the thrust to move energy production offshore, thus leading to disagreements and conflicts among the different stakeholder groups. In recognition of the latter, there has been a growing international interest in exploring the combination of multiple maritime activities in the same area (multi-use; MU), including the re-instatement of fishing activities within, or in close proximity to, offshore wind farms (OWFs). We summarise local stakeholder perspectives from two sub-national case studies (East coast of Scotland and Germany's North Sea EEZ) to scope the feasibility of combining multiple uses of the sea, such as offshore wind farms and commercial fisheries. We combined a desk-based review with 15 semi-structured qualitative interviews with key knowledge holders from both industries, regulators, and academia to aggregate key results. Drivers, barriers and resulting effects (positive and negative) for potential multi-use of fisheries and OWFs are listed and ranked (57 factors in total). Factors are of economic, social, policy, legal, and technical nature. To date, in both case study areas, the offshore wind industry has shown little interest in multi-use solutions, unless clear added value is demonstrated and no risks to their operations are involved. In contrast, the commercial fishing sector is proactive towards multi-use projects and acts as a driving force for MU developments. We provide a range of management recommendations, based on stakeholder input, to support progress towards robust decision making in relation to multi-use solutions, including required policy and regulatory framework improvements, good practice guidance, empirical studies, capacity building of stakeholders and improvements of the consultation process. Our findings represent a comprehensive depiction of the current state and key stakeholder aspirations for multi-use solutions combining fisheries and OWFs. We believe that the pathways towards robust decision making in relation to multi-use solutions suggested here are transferable to other international locations

    A pyridinium derivative from Red Sea soft corals inhibited voltage-activated potassium conductances and increased excitability of rat cultured sensory neurones

    BACKGROUND: Whole cell patch clamp recording and intracellular Ca(2+ )imaging were carried out on rat cultured dorsal root ganglion (DRG) neurones to characterize the actions of crude extracts and purified samples from Red Sea soft corals. The aim of the project was to identify compounds that would alter the excitability of DRG neurones. RESULTS: Crude extracts of Sarcophyton glaucum and Lobophyton crassum attenuated spike frequency adaptation causing DRG neurones to switch from firing single action potentials to multiple firing. The increase in excitability was associated with enhanced KCl-evoked Ca(2+ )influx. The mechanism of action of the natural products in the samples from the soft corals involved inhibition of voltage-activated K(+ )currents. An active component of the crude marine samples was identified as 3-carboxy-1-methyl pyridinium (trigonelline). Application of synthetic 3-carboxy-1-methyl pyridinium at high concentration (0.1 mM) also induced multiple firing and reduced voltage-activated K(+ )current. The changes in excitability of DRG neurones induced by 3-carboxy-1-methyl pyridinium suggest that this compound contributes to the bioactivity produced by the crude extracts from two soft corals. CONCLUSION: Sarcophyton glaucum and Lobophyton crassum contain natural products including 3-carboxy-1-methyl pyridinium that increase the excitability of DRG neurones. We speculate that in addition to developmental control and osmoregulation these compounds may contribute to chemical defenses

    Organ-on-a-chip for studying immune cell adhesion to liver sinusoidal endothelial cells:the potential for testing immunotherapies and cell therapy trafficking

    Immunotherapy has changed the landscape of treatment options for patients with hepatocellular cancer. Checkpoint inhibitors are now standard of care for patients with advanced tumours, yet the majority remain resistant to this therapy and urgent approaches are needed to boost the efficacy of these agents. Targeting the liver endothelial cells, as the orchestrators of immune cell recruitment, within the tumour microenvironment of this highly vascular cancer could potentially boost immune cell infiltration. We demonstrate the successful culture of primary human liver endothelial cells in organ-on-a-chip technology followed by perfusion of peripheral blood mononuclear cells. We confirm, with confocal and multiphoton imaging, the capture and adhesion of immune cells in response to pro-inflammatory cytokines in this model. This multicellular platform sets the foundation for testing the efficacy of new therapies in promoting leukocyte infiltration across liver endothelium as well as a model for testing cell therapy, such as chimeric antigen receptor (CAR)-T cell, capture and migration across human liver endothelium

    The ATLAS3D project - XXV: Two-dimensional kinematic analysis of simulated galaxies and the cosmological origin of fast and slow rotators

    We present a detailed two-dimensional stellar dynamical analysis of as ample of 44 cosmological hydrodynamical simulations of individual central galaxies with stellar masses of 2 x 1010Msun ∌≀ Mstar ∌≀ 6x 1011Msun. Kinematic maps of the stellar line-of-sight velocity, velocity dispersion, and higher-order Gauss-Hermite moments h3 and h4 are constructed for each central galaxy and for the most massive satellites. The amount of rotation is quantified using the λR-parameter. The velocity, velocity dispersion, h3, and h4 fields of the simulated galaxies show a diversity similar to observed kinematic maps of early-type galaxies in the ATLAS3D survey. This includes fast (regular), slow, and misaligned rotation, hot spheroids with embedded cold disk components as well as galaxies with counter-rotating cores or central depressions in the velocity dispersion. We link the present-day kinematic properties to the individual cosmological formation histories of the galaxies. In general, major galaxy mergers have a significant influence on the rotation properties resulting in both a spin-down as well as a spin-up of the merger remnant. Lower mass galaxies with significant in-situ formation of stars, or with additional gas-rich major mergers - resulting in a spin-up - in their formation history, form elongated fast rotators with a clear anti-correlation of h3 and v/σ. An additional formation path for fast rotators includes gas-poor major mergers leading to a spin-up of the remnants. This formation path does not result in anti-correlated h3 and v/σ. The galaxies most consistent with the rare class of non-rotating round early-type galaxies grow by gas-poor minor mergers alone. In general, more massive galaxies have less in-situ star formation since z ∌ 2, rotate slower and have older stellar populations. (shortened)PostprintPeer reviewe
