Convergence and evaluation-complexity analysis of a regularized tensor-Newton method for solving nonlinear least-squares problems
Given a twice-continuously differentiable vector-valued function r(x), a local minimizer of ∥r(x)∥2 is sought. We propose and analyse tensor-Newton methods, in which r(x) is replaced locally by its second-order Taylor approximation. Convergence is controlled by regularization of various orders. We establish global convergence to a first-order critical point of ∥r(x)∥2, and provide function evaluation bounds that agree with the best-known bounds for methods
using second derivatives. Numerical experiments comparing tensor-Newton methods with regularized Gauss-Newton and Newton methods demonstrate the practical performance of
the newly proposed method
Near-optimal mean value estimates for multidimensional Weyl sums
We obtain sharp estimates for multidimensional generalisations of
Vinogradov's mean value theorem for arbitrary translation-dilation invariant
systems, achieving constraints on the number of variables approaching those
conjectured to be the best possible. Several applications of our bounds are
discussed
Coulomb Blockade in low mobility nanometer size Si:MOSFETs
We investigate coherent transport in Si:MOSFETs with nominal gate lengths 50
to 100nm and various widths at very low temperature. Independent of the
geometry, localized states appear when G=e^{2}/h and transport is dominated by
resonant tunnelling through a single quantum dot formed by an impurity
potential. We find that the typical size of the relevant impurity quantum dot
is comparable to the channel length and that the periodicity of the observed
Coulomb blockade oscillations is roughly inversely proportional to the channel
length. The spectrum of resonances and the nonlinear I-V curves allow to
measure the charging energy and the mean level energy spacing for electrons in
the localized state. Furthermore, we find that in the dielectric regime, the
variance var(lng) of the logarithmic conductance lng is proportional to its
average value consistent with one-electron scaling models.Comment: 4 pages, 4 figure
Detecting new microRNAs in human osteoarthritic chondrocytes identifies miR-3085 as a human, chondrocyte-selective, microRNA
Objective: To use deep sequencing to identify novel microRNAs in human osteoarthritic cartilage which have a functional role in chondrocyte phenotype or function. Design: A small RNA library was prepared from human osteoarthritic primary chondrocytes using in-house adaptors and analysed by Illumina sequencing. Novel candidate microRNAs were validated by northern blot and qRT-PCR. Expression was measured in cartilage models. Targets of novel candidates were identified by microarray and computational analysis, validated using 3’-UTR-luciferase reporter plasmids. Protein levels were assessed by western blot and functional analysis by cell adhesion. Results: We identified 990 known microRNAs and 1621 potential novel microRNAs in human osteoarthritic chondrocytes, 60 of the latter were expressed in all samples assayed. MicroRNA-140-3p was the most highly expressed microRNA in osteoarthritic cartilage. Sixteen novel candidate microRNAs were analysed further, of which 6 remained after northern blot analysis. Three novel microRNAs were regulated across models of chondrogenesis, chondrocyte differentiation or cartilage injury. One sequence (novel #11), annotated in rodents as microRNA-3085-3p, was preferentially expressed in cartilage, dependent on chondrocyte differentiation and, in man, is located in an intron of the cartilage-expressed gene CRTAC-1. This microRNA was shown to target the ITGA5 gene directly (which encodes integrin alpha5) and inhibited adhesion to fibronectin (dependent on alpha5beta1 integrin). Conclusion: Deep sequencing has uncovered many potential microRNA candidates expressed in human cartilage. At least three of these show potential functional interest in cartilage homeostasis and osteoarthritis. Particularly, novel #11 (microRNA-3085-3p) which has been identified for the first time in man
The effect of culture preservation techniques on patulin and citrinin production by Penicillium expansum Link
Aims: To study the influence of culture preservation methods and culture conditions on the
production of the mycotoxins patulin and citrinin by Penicillium expansum.
Methods and results: Ten strains of Penicillium expansum were preserved using subculture
and maintenance at 4 ºC, mineral oil, drying on silica gel and freeze-drying. Patulin and
citrinin production was assessed on yeast extract sucrose agar (YES) and grape juice agar (GJ),
using TLC before and after 0.5, 2–3, 6 and 12 months preservation. Citrinin was detected in all
cultures for all preservation techniques on YES. The patulin profiles obtained differed with
strain and culture media used.
Conclusions: Citrinin production seems to be a stable character for the tested strains. There is
a tendency for patulin detection with time apparently more consistent for silica gel storage and
freeze-drying, especially when the strains are grown on GJ.
Significance and Impact of the Study: Variability in the profiles of the mycotoxins tested
seems to be more strain-specific than dependent on the preservation technique used
Faking like a woman? Towards an interpretative theorization of sexual pleasure.
This article explores the possibility of developing a feminist approach to gendered and sexual embodiment which is rooted in the pragmatist/interactionist tradition derived from G.H. Mead, but which in turn develops this perspective by inflecting it through more recent feminist thinking. In so doing we seek to rebalance some of the rather abstract work on gender and embodiment by focusing on an instance of 'heterosexual' everyday/night life - the production of the female orgasm. Through engaging with feminist and interactionist work, we develop an approach to embodied sexual pleasure that emphasizes the sociality of sexual practices and of reflexive sexual selves. We argue that sexual practices and experiences must be understood in social context, taking account of the situatedness of sex as well as wider socio-cultural processes the production of sexual desire and sexual pleasure (or their non-production) always entails interpretive, interactional processes
Genetic determinants of thyroid function in children
OBJECTIVE:Genome-wide association studies in adults have identified 42 loci associated with thyroid stimulating hormone (TSH) and 21 loci associated with free thyroxine (FT4) concentrations. While biologically plausible, age-dependent effects have not been assessed. We aimed to study the association of previously identified genetic determinants of TSH and FT4 with TSH and FT4 concentrations in newborns and (pre)school children. METHODS: We selected participants from three population-based prospective cohorts with data on genetic variants and thyroid function: Generation R (N = 2169 children, mean age 6 years; N = 2388 neonates, the Netherlands), the Avon Longitudinal Study of Parents and Children (ALSPAC; N = 3382, age 7.5 years, United Kingdom), and the Brisbane Longitudinal Twin Study (BLTS; N = 1680, age 12.1 years, Australia). The association of single nucleotide polymorphisms (SNPs) with TSH and FT4 concentrations was studied with multivariable linear regression models. Weighted polygenic risk scores (PRSs) were defined to combine SNP effects.RESULTS:In childhood, 30/60 SNPs were associated with TSH and 11/31 SNPs with FT4 after multiple testing correction. The effect sizes for AADAT, GLIS3, TM4SF4, and VEGFA were notably larger than in adults. The TSH PRS explained 5.3%-8.4% of the variability in TSH concentrations; the FT4 PRS explained 1.5%-4.2% of the variability in FT4 concentrations. Five TSH SNPs and no FT4 SNPs were associated with thyroid function in neonates. CONCLUSIONS: The effects of many known thyroid function SNPs are already apparent in childhood and some might be notably larger in children as compared to adults. These findings provide new knowledge about genetic regulation of thyroid function in early life.</p
Multiple stressors in Southern Africa: the link between HIV/AIDS, food insecurity, poverty and children's vulnerability now and in the future
Several countries in Southern Africa now see large numbers of their population barely subsisting at poverty levels in years without shocks, and highly vulnerable to the vagaries of the weather, the economy and government policy. The combination of HIV/AIDS, food insecurity and a weakened capacity for governments to deliver basic social services has led to the region experiencing an acute phase of a long-term emergency. “Vulnerability” is a term commonly used by scientists and practitioners to describe these deteriorating conditions. There is particular concern about the “vulnerability” of children in this context and implications for children's future security. Through a review of literature and recent case studies, and using a widely accepted conceptualisation of vulnerability as a lens, we reflect on what the regional livelihoods crisis could mean for children's future wellbeing. We argue that an increase in factors determining the vulnerability of households — both through greater intensity and frequency of shocks and stresses (“external” vulnerability) and undermined resilience or ability to cope (“internal” vulnerability) — are threatening not only current welfare of children, but also their longer-term security. The two specific pathways we explore are (1) erosive coping strategies employed by families and individuals; and (2) their inability to plan for the future. We conclude that understanding and responding to this crisis requires looking at the complexity of these multiple stressors, to try to comprehend their interconnections and causal links. Policy and programme responses have, to date, largely failed to take into account the complex and multi-dimensional nature of this crisis. There is a misfit between the problem and the institutional response, as responses from national and international players have remained relatively static. Decisive, well-informed and holistic interventions are needed to break the potential negative cycle that threatens the future security of Southern Africa's children
Recent Developments in Understanding Two-dimensional Turbulence and the Nastrom-Gage Spectrum
Two-dimensional turbulence appears to be a more formidable problem than
three-dimensional turbulence despite the numerical advantage of working with
one less dimension. In the present paper we review recent numerical
investigations of the phenomenology of two-dimensional turbulence as well as
recent theoretical breakthroughs by various leading researchers. We also review
efforts to reconcile the observed energy spectrum of the atmosphere (the
spectrum) with the predictions of two-dimensional turbulence and
