Hepatoma cell density promotes claudin-1 and scavenger receptor BI expression and hepatitis C virus internalization.
Hepatitis C virus (HCV) entry occurs via a pH- and clathrin-dependent endocytic pathway and requires a number of cellular factors, including CD81, the tight-junction proteins claudin 1 (CLDN1) and occludin, and scavenger receptor class B member I (SR-BI). HCV tropism is restricted to the liver, where hepatocytes are tightly packed. Here, we demonstrate that SR-BI and CLDN1 expression is modulated in confluent human hepatoma cells, with both receptors being enriched at cell-cell junctions. Cellular contact increased HCV pseudoparticle (HCVpp) and HCV particle (HCVcc) infection and accelerated the internalization of cell-bound HCVcc, suggesting that the cell contact modulation of receptor levels may facilitate the assembly of receptor complexes required for virus internalization. CLDN1 overexpression in subconfluent cells was unable to recapitulate this effect, whereas increased SR-BI expression enhanced HCVpp entry and HCVcc internalization, demonstrating a rate-limiting role for SR-BI in HCV internalization
Der spätantike Hochrhein-Limes - Zwischenbilanz und Forschungsperspektiven
La (re)construction de nouvelles fortifications sur le haut Rhin (« Hochrhein ») sous Valentinien a servi à signaler et à contrôler la frontière nord de la provincia Maxima Sequanorum . L'épine dorsale de ce « Hochrhein-Limes » est formée par les fortifications déjà en place de Basilia (Bâle), Tenedo (Bad Zurzach), Tasgetium (Stein am Rhein) et le Castrum Rauracense (Kaiseraugst). Les têtes de pont des quatre castra , les bâtiments de stockage à Mumpf et Sisseln et les quelque 50 tours de guet sont caractéristiques de ce tronçon de la frontière de l'Empire romain. Deux tours peuvent être datées de l'an 371 sur la base des inscriptions. La composition de l' opus caementicium suggère que les autres tours de guet, non datées précisément, sont également de la période valentinienne. Dans le cas de Bâle, des sources écrites attestent que Valentinien s'est installé dans un camp de campagne fortifié ( munimentum ) en 374 prope Basiliam . Le quadriburgus situé dans l'actuel quartier de Kleinbasel en faisait aussi partie. Le nom Robur qui désignait ce camp impérial est un toponyme encore utilisé aujourd'hui par la population locale pour Kleinbasel. Rückgrat des Hochrhein-Limes bildeten die bereits bestehenden castra in Basilia (Basel), Tenedo (Bad Zurzach), Tasgetium (Stein am Rhein) sowie das Castrum Rauracense (Kaiseraugst). Charakteristisch für den Hochrhein-Limes sind die Brückenköpfe der vier castra , die 'Magazinbauten' in Mumpf und Sisseln sowie die etwa 50 Wachttürme. Zwei können anhand von Inschriften in das Jahr 371 datiert werden. Die Zusammensetzung des opus caementicium legt zudem nahe, dass auch die nicht genauer datierbaren Wachttürme aus valentinianischer Zeit stammen. Im Fall von Basel bezeugen die Schriftquellen, dass Valentinian im Jahr 374 prope Basiliam ein befestigtes Feldlager ( munimentum ) bezog. Zu diesem gehörte unter anderem der in Kleinbasel nachgewiesene quadriburgus . Namensgebend für das kaiserliche Feldlager wurde Robur , ein älteres und von den Einheimischen verwendetes Toponym für das heutige Kleinbasel
Density Waves Inside Inner Lindblad Resonance: Nuclear Spirals in Disk Galaxies
We analyze formation of grand-design two-arm spiral structure in the nuclear
regions of disk galaxies. Such morphology has been recently detected in a
number of objects using high-resolution near-infrared observations. Motivated
by the observed (1) continuity between the nuclear and kpc-scale spiral
structures, and by (2) low arm-interarm contrast, we apply the density wave
theory to explain the basic properties of the spiral nuclear morphology. In
particular, we address the mechanism for the formation, maintenance and the
detailed shape of nuclear spirals. We find, that the latter depends mostly on
the shape of the underlying gravitational potential and the sound speed in the
gas. Detection of nuclear spiral arms provides diagnostics of mass distribution
within the central kpc of disk galaxies. Our results are supported by 2D
numerical simulations of gas response to the background gravitational potential
of a barred stellar disk. We investigate the parameter space allowed for the
formation of nuclear spirals using a new method for constructing a
gravitational potential in a barred galaxy, where positions of resonances are
