A surprise for you and me?:The effect of surprise appeals and choices for close others on consumption experience
Surprise labels increase indulgent food portion size choice
Food package labels can significantly influence food portion size choice. In this research we investigate whether package labels featuring the word 'surprise' influence food portion size choice of hedonic snacks. Surprise appeals are used frequently by the food industry to encourage product choice (e.g., surprise menus at restaurants), but their effect on portion size choice is not yet well understood. In four experimental studies, we investigate the effect of surprise labels-without changing the level of information about the product-on food portion size choice. We consistently find that surprise labels (e.g., surprise chocolates box)-compared to conventional labels (e.g., chocolates box)-increase food portion size choice. We furthermore show that this effect is driven by an increase in anticipated enjoyment associated with the surprise-labeled snack. Theoretically, our findings contribute to literature on the implications of the halo effect in food portion size choice by showing that a surprise label triggers a positive halo effect and thereby increases portion size choice
Taking care of you and me: How choosing for others impacts self-indulgence within family caregiving relationships
Consumers frequently make choices for family members they take care of or from whom they receive care (e.g., their children or partner), yet marketing research has given little attention to how these other-oriented choices might impact the chooser’s self-indulgence. In this research we consider familial caregiving relationships as a relevant and ubiquitous context of other-oriented choices and identify the role of the chooser (i.e., caregiver versus care-receiver) as an important moderator that determines when virtuous other-oriented choices within caregiving relationships lead to licensing and when they encourage consistent virtuous consumption behaviors. Three studies demonstrate that making virtuous food choices for others affect the chooser’s subsequent self-regulatory behavior in two ways: After making a virtuous choice for a care-receiving other (e.g., a young child), caregivers (e.g., parents) are more likely to license, and thus to subsequently self-indulge (Study 1, 3). In contrast, care-receivers are more likely to act consistently with an initial virtuous choice for the caregiver and thus are less likely to self-indulge (Study 2, 3). Our findings extend research on moral licensing and consistency effects by demonstrating that—within familial caregiving relationships—the degree to which one receives and provides care may determine when choosers engage in licensing and when they act consistently with an initial virtuous other-oriented choice
Use of acid ceramidase and sphingosine kinase inhibitors as antiviral compounds against measles virus infection of lymphocytes in vitro
As structural membrane components and signaling effector molecules sphingolipids influence a plethora of host cell functions, and by doing so also the replication of viruses. Investigating the effects of various inhibitors of sphingolipid metabolism in primary human peripheral blood lymphocytes (PBL) and the human B cell line BJAB we found that not only the sphingosine kinase (SphK) inhibitor SKI-II, but also the acid ceramidase inhibitor ceranib-2 efficiently inhibited measles virus (MV) replication. Virus uptake into the target cells was not grossly altered by the two inhibitors, while titers of newly synthesized MV were reduced by approximately 1 log (90%) in PBL and 70–80% in BJAB cells. Lipidomic analyses revealed that in PBL SKI-II led to increased ceramide levels, whereas in BJAB cells ceranib-2 increased ceramides. SKI-II treatment decreased sphingosine-1-phosphate (S1P) levels in PBL and BJAB cells. Furthermore, we found that MV infection of lymphocytes induced a transient (0.5–6 h) increase in S1P, which was prevented by SKI-II. Investigating the effect of the inhibitors on the metabolic (mTORC1) activity we found that ceranib-2 reduced the phosphorylation of p70 S6K in PBL, and that both inhibitors, ceranib-2 and SKI-II, reduced the phosphorylation of p70 S6K in BJAB cells. As mTORC1 activity is required for efficient MV replication, this effect of the inhibitors is one possible antiviral mechanism. In addition, reduced intracellular S1P levels affect a number of signaling pathways and functions including Hsp90 activity, which was reported to be required for MV replication. Accordingly, we found that pharmacological inhibition of Hsp90 with the inhibitor 17-AAG strongly impaired MV replication in primary PBL. Thus, our data suggest that treatment of lymphocytes with both, acid ceramidase and SphK inhibitors, impair MV replication by affecting a number of cellular activities including mTORC1 and Hsp90, which alter the metabolic state of the cells causing a hostile environment for the virus
Argumentationsanalyse von Kommentaren in einem Forum der BBC zur Unabhängigkeit des Kosovo
Die vorliegende Arbeit geht hervor aus dem Hauptseminar „Argumentationstheorie“, das im Wintersemester 2008/09 am Institut für Linguistik der Universität zu Köln unter der Leitung von PD Dr. Leila Behrens abgehalten wurde. Ziel dieses Seminars war es, ausgehend von traditionellen Begriffen der Rhetorik, Dialektik und Logik, in die Terminologie sowie in zentrale Modelle der zeitgenössischen Argumentationsforschung einzuführen. Die dabei erworbenen Kenntnisse sollen im Folgenden bei der Analyse von Beiträgen eines Diskussionsforums im Internet angewendet werden. Hierbei handelt es sich um ein sogenanntes „newsforum“ der BBC mit dem Titel „Have Your Say“ (BBC 2008), in dem aktuelle Themen und Nachrichten von Internetnutzern weltweit diskutiert werden können. Im untersuchten Fall behandeln wir die Frage, wie mit der Unabhängigkeitserklärung des Kosovo vom 17. Februar 2008 umzugehen sei: „Should the world recognise an independent Kosovo?“ […]. Zu dieser Fragestellung wurden insgesamt 3195 Beiträge im Forum veröffentlicht, von denen hier 780 ausgewertet werden. Diese folgen chronologisch aufeinander und umfassen den Zeitraum zwischen 7:49 Uhr (mittlere Greenwich-Zeit) und 14:26 Uhr des 17. Februar 2008
Improving the Prognostic Ability through Better Use of Standard Clinical Data - The Nottingham Prognostic Index as an Example
Background Prognostic factors and prognostic models play a key role in medical
research and patient management. The Nottingham Prognostic Index (NPI) is a
well-established prognostic classification scheme for patients with breast
cancer. In a very simple way, it combines the information from tumor size,
lymph node stage and tumor grade. For the resulting index cutpoints are
proposed to classify it into three to six groups with different prognosis. As
not all prognostic information from the three and other standard factors is
used, we will consider improvement of the prognostic ability using suitable
analysis approaches. Methods and Findings Reanalyzing overall survival data of
1560 patients from a clinical database by using multivariable fractional
polynomials and further modern statistical methods we illustrate suitable
multivariable modelling and methods to derive and assess the prognostic
ability of an index. Using a REMARK type profile we summarize relevant steps
of the analysis. Adding the information from hormonal receptor status and
using the full information from the three NPI components, specifically
concerning the number of positive lymph nodes, an extended NPI with improved
prognostic ability is derived. Conclusions The prognostic ability of even one
of the best established prognostic index in medicine can be improved by using
suitable statistical methodology to extract the full information from standard
clinical data. This extended version of the NPI can serve as a benchmark to
assess the added value of new information, ranging from a new single clinical
marker to a derived index from omics data. An established benchmark would also
help to harmonize the statistical analyses of such studies and protect against
the propagation of many false promises concerning the prognostic value of new
measurements. Statistical methods used are generally available and can be used
for similar analyses in other diseases
Common germline polymorphisms associated with breast cancer-specific survival
Abstract
Introduction
Previous studies have identified common germline variants nominally associated with breast cancer survival. These associations have not been widely replicated in further studies. The purpose of this study was to evaluate the association of previously reported SNPs with breast cancer-specific survival using data from a pooled analysis of eight breast cancer survival genome-wide association studies (GWAS) from the Breast Cancer Association Consortium.
Methods
A literature review was conducted of all previously published associations between common germline variants and three survival outcomes: breast cancer-specific survival, overall survival and disease-free survival. All associations that reached the nominal significance level of P value <0.05 were included. Single nucleotide polymorphisms that had been previously reported as nominally associated with at least one survival outcome were evaluated in the pooled analysis of over 37,000 breast cancer cases for association with breast cancer-specific survival. Previous associations were evaluated using a one-sided test based on the reported direction of effect.
Results
Fifty-six variants from 45 previous publications were evaluated in the meta-analysis. Fifty-four of these were evaluated in the full set of 37,954 breast cancer cases with 2,900 events and the two additional variants were evaluated in a reduced sample size of 30,000 samples in order to ensure independence from the previously published studies. Five variants reached nominal significance (P <0.05) in the pooled GWAS data compared to 2.8 expected under the null hypothesis. Seven additional variants were associated (P <0.05) with ER-positive disease.
Conclusions
Although no variants reached genome-wide significance (P <5 x 10−8), these results suggest that there is some evidence of association between candidate common germline variants and breast cancer prognosis. Larger studies from multinational collaborations are necessary to increase the power to detect associations, between common variants and prognosis, at more stringent significance levels
