Freeze/thaw stress induces organelle remodeling and membrane recycling in cryopreserved human mature oocytes

Purpose: Our aim was to evaluate the ultrastructure of human metaphase II oocytes subjected to slow freezing and fixed after thawing at different intervals during post-thaw rehydration. Methods: Samples were studied by light and transmission electron microscopy. Results: We found that vacuolization was present in all cryopreserved oocytes, reaching a maximum in the intermediate stage of rehydration. Mitochondria-smooth endoplasmic reticulum (M-SER) aggregates decreased following thawing, particularly in the first and intermediate stages of rehydration, whereas mitochondria-vesicle (MV) complexes augmented in the same stages. At the end of rehydration, vacuoles and MV complexes both diminished and M-SER aggregates increased again. Cortical granules (CGs) were scarce in all cryopreserved oocytes, gradually diminishing as rehydration progressed. Conclusions: This study also shows that such a membrane remodeling is mainly represented by a dynamic process of transition between M-SER aggregates and MV complexes, both able of transforming into each other. Vacuoles and CG membranes may take part in the membrane recycling mechanism

Induced Reproduction of Aphanius fasciatus by Ecophysiological Conditioning and Hormonal Treatment in Fresh and Marine Water

The Mediterranean toothcarp (Aphanius fasciatus) can be used to biologically control mosquito larvae. The reproductive performance of 234 Mediterranean toothcarp (180 females and 54 males) was investigated. Reproductive factors were determined in fish kept in fresh or saline water (males and females together), and in fish kept separately by sex in saline water and hormonally-treated with carp pituitary extract (CPE). In the hormonally-treated group, the combined effect of temperature, photoperiod, and hormonal treatment induced the best ovary maturation and larvae production rates. The rate of reproduction was very high (average eggs/female: 5.9-10.5) compared to natural reproduction in the wild (average eggs/female: 2-4). Additionally, the hatching rate was 97-100%. The results of this study show that reproduction of Mediterranean toothcarp can be controlled in an artificial environment: the lack of mortality in adults during acclimation and conditioning in fresh and marine waters indicates good domestication and plasticity in reproductive parameters

The first new species of European Ascocotyle Looss, 1899 (Digenea: Heterophyidae) described in more than half a century

Ascocotyle (Phagicola) trentinii n. sp. is described based on adults from experimentally infected ducklings (Anas platyrhynchos domesticus) fed with metacercariae from the visceral serosa of the Mediterranean banded killifish, Aphanius fasciatus (Cyprinodontiformes: Aphaniidae), from coastal lagoons in northeastern Italy (Emilia-Romagna Region). The new species is placed into the subgenus Phagicola because of the presence of a single row of circumoral spines, vitelline follicles being confined between the ventral sucker and testes, and uterine loops not reaching anterior to the ventral sucker. Ascocotyle (P.) trentinii n. sp. differs from other members of the subgenus Phagicola, as well as other species of Ascocotyle, by the number (27–33) of circumoral spines which are 13.5–17 μm long and 3.5–5 μm wide, and by the morphology of a gonotyl which is composed of about 8 large refractile pockets. The occurrence of metacercariae in A. fasciatus indicates that the life cycle of the new species is completed in brackish water lagoons. It is the fourth species of Ascocotyle described in Europe and may be endemic to the Mediterranean region because its second (fish) intermediate host is endemic to this region

First case of traumatic myiasis caused by Calliphora vicina in a crested porcupine Hystrix cristata L. in Italy

Calliphora vicina (Diptera: Calliphoridae) it is a facultative ectoparasite responsible for traumatic myiasis in humans and warmblooded vertebrates in the world. In this work one case of traumatic myasis caused by C. vicina (Diptera Calliphoirdae) is reported for the first time in a vulnerable crested porcupine (Hystrix cristata Linnaeus, 1758). A total of 30 larvae located in the posterior-dorsal area of the animal were removed from inside the lesion and either preserved in ethanol or reared to the adult stage. This report shows the great ability of C. vicina to use many organic matter for the food source's offspring, including many live vertebrates

Retreating to nature : rethinking 'therapeutic landscapes'

    Get PDF
There is a long history of removing oneself from ‘society’ in order to recuperate or repair. This paper considers a yoga and massage retreat in Southern Spain, and what opportunities this retreat experience might offer for recuperation and the creation of healthy bodies. The paper positions ‘nature’ as an active participant, and as ‘enrolled’ in the experiences of the retreat as a ‘therapeutic landscape’, and questions how and what particular aspects of yoga practice (in intimate relation with place) give rise to therapeutic experiences

This report describes the preparation and characterisation of a set of three plasmid solutions, ERM-AD482a, ERM-AD482b and ERM-AD482c. The material was produced in accordance with ISO Guide 34:2009 [1]. A DNA fragment specific for the identification of ruminant meat was cloned into a pUC18 vector to construct the pIRMM-0103 plasmid. The nucleic acid sequence of the entire pIRMM-0103 plasmid was determined by dye terminator cycle sequencing applying the primer walking method on the entire plasmid. The plasmid was put into a solution and its concentration was measured by ultraviolet (UV) spectrophotometry. Afterwards this solution was gravimetrically diluted to obtain three different plasmid concentration levels. The plasmid copy number concentration of the three concentration levels were certified by digital quantitative polymerase chain reaction methods (dPCR). In addition, between-unit homogeneity, as well as short-term long-term and freeze-thaw stability was assessed in accordance with ISO Guide 35:2006 [2]. The materials are intended for the determination of a cut-off value to discriminate positive samples (containing the ruminant target sequence) from negative samples by quantitative PCR as defined in the Standard Operating Procedure of the EU Reference Laboratory for animal proteins (EURL-AP) according to Commission Regulation No 51/2013 [3, 4]. As any certified reference material (CRM), the materials can also be used for control charts or validation studies. The CRM is available as a set of three vials each containing at least 1 mL of plasmid solution. The minimum amount of sample to be used is 4 µL.JRC.D.2-Standards for Innovation and sustainable Developmen

Objects on sale. What do the newspapers tell us? This work is dedicated to the presentation of datas and analysis made from the identification in printed newspapers that broadcasted in Santa Catarina between 1908 and 1921, from news related to school objects selling, which were atributed to school Law, it offers indicatives of a material composition of the school. It was found a way of doing some reflexions, refered to the trade that is formed with the expansion of the primary school. A consume of objects was required from the State, with the argument of being essencial for the teaching and learning from the student. The emphasys here, are news of school materials, destinated to an organization of the classroom: maps, blackboards. Inkstand, etc. It is understood that daily newspapers printed and destinated to the general public, are made of sources of great relevance and it contains informations that help to compose the chart which it builds itself, its relations with economic market. From 47 newspapers consulted, it were analysed news located in a group of 13: O Chatarinense de São Bento, A Comarca, O Commércio de Joinville, a Gazeta de Joinville, o Município de Joinville e a Gazeta do Comércio de Joinville, A revista de São Francisco do Sul, O Pharol de Itajaí, A Época, O Estado, O dia, O Correio do Povo e Republica de Florianópolis. It´s a big part of newspapers of regional circulation that reverberates in locations of economic relations in large scale. Keywords: School material, Cultural material, newspapers, school market
