Zinc is required for the activity of many enzymes and plays an essential role in gene regulation and redox homeostasis. In Anabaena (Nostoc) sp. PCC7120, the genes involved in zinc uptake and transport are controlled by the metalloregulator Zur (FurB). Comparative transcriptomics of a zur mutant (Δzur) with the parent strain unveiled unexpected links between zinc homeostasis and other metabolic pathways. A notable increase in the transcription of numerous desiccation tolerance-related genes, including genes involved in the synthesis of trehalose and the transference of saccharide moieties, among many others, was detected. Biofilm formation analysis under static conditions revealed a reduced capacity of Δzur filaments to form biofilms compared to the parent strain, and such capacity was enhanced when Zur was overexpressed. Furthermore, microscopy analysis revealed that zur expression is required for the correct formation of the envelope polysaccharide layer in the heterocyst, as Δzur cells showed reduced staining with alcian blue compared to Anabaena sp. PCC7120. We suggest that Zur is an important regulator of the enzymes involved in the synthesis and transport of the envelope polysaccharide layer, influencing heterocyst development and biofilm formation, both relevant processes for cell division and interaction with substrates in its ecological niche

Differences in metabolic and physiological responses between local and widespread grapevine cultivars underwater deficit stress

Climate change forecasts suggest temperature increases and lower rainfall rates, both of which challenge viticulture, particularly in semi-arid areas where water availability is critical. In this scenario, tlx-use of tlx-genetic variability in grapevine varieties reported around the world represents an important strategy for the selection of climate-resilient cultivars. In this work, physiological and metabolomics analyses were conducted to Compare the water deficit stress (VVIJS) responses of red and white, local and widespread grapevines cultivars. Leaf gas exchange, water use efficiency (WUK) and water relation parameters were determined in plants under well-watered and WDS conditions alongside assessment of the levels of foliar primary metabolites using gas-chromatography coupled to mass-spectrometry. Results denote that red and white local cultivars displayed more adapted physiological performance under WDS as Compared to the widely-distributed ones. Multivariate analyses and specific changes in leaf primary metabolites indicate genotype-specific responses of local cultivars as Compared to widespread ones. Differences in ascorbate-related and shikimate/phenylpropanoid metabolism could explain the better physiological performance under WDS in red local as Compared to widespread cultivars. On the other hand, coordinated changes in respiratory- A nd stress-related sugars and amino acids Could underlie the better WUK under WDS in tlx-white local Cultivar. All these results suggest several metabolic targets that Could be useful as metabolic markers or for metabolic engineering in grapevine breeding programs to improve drought tolerance

Finançament de: Fundación Dexeus Salud de la Mujer; CardiolinkTo compare the effectiveness and safety of polypropylene (PP) and polyvinylidene fluoride (PVDF) transobturator tapes (TOT) for the treatment of female stress urinary incontinence (SUI). This is a multicentre randomized trial. Women with SUI or stress-predominant mixed urinary incontinence and scheduled for a TOT procedure were randomized to PP or PVDF slings. The primary outcome was 1-year cure or improvement rate using composite criteria. Complications were also compared. Relationships with outcomes were analyzed using multivariable logistic regressions models. From April 2016 to January 2018 285 participants were randomized. PP and PVDF slings showed similar high cure or improvement rate (91.0% vs. 95.6%, p = .138). Improvement in validated questionnaires was also similar. PVDF slings were associated with a lower rate of de novo urgency incontinence (adjusted odds ratio = 0.35; 95% confidence interval = 0.15-0.80). We found no statistical differences in complications rates, although a higher incidence of long-term pain events were observed in the PP group. The study is underpowered to find differences in specific complications owing to the low number of events. PP and PVDF TOTs are equally effective, although PVDF is associated with fewer cases of de novo urgency incontinence. Further studies are needed to give robust conclusions on safety profiles

Proinflammatory cytokine levels in fibromyalgia patients are independent of body mass index

Abstract

Background

Fibromyalgia (FM) is characterized by chronic, widespread muscular pain and tenderness and is generally associated with other somatic and psychological symptoms. Further, circulatory levels of proinflammatory cytokines (IL-1β, TNF-α, and IL-6) may be altered in FM patients, possibly in association with their symptoms. Recently, rises in BMI have been suggested to contribute to increased circulating levels of proinflammatory cytokines in FM patients. Our aim was to measure the circulatory levels of proinflammatory cytokines to determine the influence of BMI on these levels in FM patients and healthy volunteers (HVs). In Spanish FM patients (n = 64) and HVs (n = 25), we measured BMI and serum concentrations of proinflammatory cytokines by capture ELISA.

Findings

There were significant differences in BMI levels between FM patients (26.40 ± 4.46) and HVs (23.64 ± 3.45) and significant increase in IL-6 in FM patients (16.28 ± 8.13 vs 0.92 ± 0.32 pg/ml) (P < 0.001). IL-1β and TNF-α decreased in FM patients compared with HVs. By ANCOVA, there was no significant association between BMI and TNF-α (F = 0.098, p = 0.75) or IL-6 (F = 0.221, p = 0.63) levels in FM patients.

Conclusions

Our analysis in FM patients of BMI as a covariate of proinflammatory cytokines levels showed that serum TNF-α and IL-6 levels are independent of BMI. Further studies are necessary to dissect these findings and their implication in future therapeutic approaches for FM patients.</p

Women's employment and the adult caring burden

Demographic ageing is increasing pensions, health and social services spending and threatening the future balance of public budgets. Providing home care can help to curb health expenditure and it may improve elderly welfare also, but EU states have chosen different policies in providing home are. Main differences are related with source of financing and eligibility criteria but also with the kind of benefits (benefits in cash or in kind). How these different options affect welfare and carers’ employment opportunities is the core of this research. Home care growth is going to be more efficient as far as it pro motes employment and, public revenues consequently. Using microdata from the European Community Household Panel, British and Spanish means tested programs are compared with German and Austrian ‘in cash’ benefits, and with Danish ‘in kind’ benefits also. The results show that Danish policies are the most efficient and equitable while the British and Spanish ones are the worst

    Get PDF
The report describes trends in drug injection in the EU Member States, Norway and Turkey between 2000 and 2011, through an analysis of data on those admitted to drug treatment. Results indicate that current drug injection, including its incidence, has declined in recent years in Europe. While heroin remains the drug most frequently involved in drug injection, data show how drug injectors are now more likely to be using stimulants than in the past
