    Investigating the Dynamics and Density Evolution of Returning Plasma Blobs from the 2011 June 7 Eruption

    This work examines infalling matter following an enormous Coronal Mass Ejection (CME) on 2011 June 7. The material formed discrete concentrations, or blobs, in the corona and fell back to the surface, appearing as dark clouds against the bright corona. In this work we examined the density and dynamic evolution of these blobs in order to formally assess the intriguing morphology displayed throughout their descent. The blobs were studied in five wavelengths (94, 131, 171, 193 and 211 \AA) using the Solar Dynamics Observatory Atmospheric Imaging Assembly (SDO/AIA), comparing background emission to attenuated emission as a function of wavelength to calculate column densities across the descent of four separate blobs. We found the material to have a column density of hydrogen of approximately 2 ×\times 1019^{19} cm−2^{-2}, which is comparable with typical pre-eruption filament column densities. Repeated splitting of the returning material is seen in a manner consistent with the Rayleigh-Taylor instability. Furthermore, the observed distribution of density and its evolution are also a signature of this instability. By approximating the three-dimensional geometry (with data from STEREO-A), volumetric densities were found to be approximately 2 ×\times 10−14^{-14} g cm−3^{-3}, and this, along with observed dominant length-scales of the instability, was used to infer a magnetic field of the order 1 G associated with the descending blobs.Comment: 9 pages, 13 figures, accepted for publication in Ap

    Predicting the stability of biotherapeutics in formulation

    Biotherapeutics, especially monoclonal antibodies (mAbs), is one of the fastest growing groups of pharmaceuticals, corresponding to a treatment option to life-threatening conditions, such as cancer, autoimmune diseases and cardiovascular diseases. A key barrier in the production of these pharmaceuticals is the formation of protein aggregates, which can lead to increased production costs, loss of the biological function, and immunogenic responses from patients. To detect the presence of aggregates and ensure protein stability upon storage, efficient formulation screening methods are required. Self-Interaction Chromatography (SIC) has previously been shown to be an accurate, automated and high-throughput technique to predict protein aggregation [1]. This study is adapting SIC into Interaction Chromatography (IC), which takes the interactions between two different proteins or species into account. The aim of the first case study was to analyse different formulations containing histidine buffer, phosphate-citrate buffer and sodium citrate buffer at 25 mM, and using a therapeutic mAb. Additionally, the stabilising effects of NaCl, polysorbate and L-arginine were examined in the range of 0-250 mM, 0-0.1% v/v and 0-200 mM, respectively. The protein-protein interactions for each formulation condition were quantified by calculating the osmotic second virial coefficient (B22), a fundamental physicochemical property that describes protein-protein interactions in solution. These results were also compared with aggregation data and aggregation kinetics using size-exclusion chromatography and dynamic light scattering. In a second case study bovine serum albumin (BSA) was mixed in formulation buffers in combination with sucrose and trehalose, where the protein-protein interactions were studied by determining B22 and the aggregation propensity measured. Later these conditions were lyophilised, which is a common way to formulate many biotherapeutics. The aim was to see if the results from the initial screenings also could predict the stability of the freeze-dried cakes. The first case study showed that IC was a suitable technique to detect small changes in formulation conditions as it managed to predict the increase in stability of the mAb with the ionic strength and arginine concentration, but with no major effect with the addition of polysorbate. The second case study showed that BSA exhibited repulsive protein-protein interactions in all the range of concentrations analysed, as expected due to its high stability in solution, which could be increased with the addition of stabilisers, especially trehalose. Even in the freeze-dried cakes higher concentration of trehalose led to an increased Young’s modulus, and therefore higher mechanical stability. Based on these results it could be seen that a protein-protein interaction technique such as IC could be used as an early predictor for formulation behaviour both in liquid and solid states.     Regional Differences in Presence of Shiga toxin-producing Escherichia coli Virulence-Associated Genes in the Environment in the North West and East Anglian regions of England

    Shiga toxin-producing Escherichia coli is carried in the intestine of ruminant animals, and outbreaks have occurred after contact with ruminant animals or their environment. The presence of STEC virulence genes in the environment was investigated along recreational walking paths in the North West and East Anglia regions of England. In all, 720 boot sock samples from walkers’ shoes were collected between April 2013 and July 2014. Multiplex PCR was used to detect E. coli based on the amplification of the uidA gene and investigate STEC-associated virulence genes eaeA, stx1 and stx2. The eaeA virulence gene was detected in 45·5% of the samples, where stx1 and/or stx2 was detected in 12·4% of samples. There was a difference between the two regions sampled, with the North West exhibiting a higher proportion of positive boot socks for stx compared to East Anglia. In univariate analysis, ground conditions, river flow and temperature were associated with positive boot socks. The detection of stx genes in the soil samples suggests that STEC is present in the English countryside and individuals may be at risk for infection after outdoor activities even if there is no direct contact with animals. Significance and Impact of the Study: Several outbreaks within the UK have highlighted the danger of contracting Shiga toxin-producing Escherichia coli from contact with areas recently vacated by livestock. This is more likely to occur for STEC infections compared to other zoonotic bacteria given the low infectious dose required. While studies have determined the prevalence of STEC within farms and petting zoos, determining the risk to individuals enjoying recreational outdoor activities that occur near where livestock may be present is less researched.     Allometric length-weight relationships for benthic prey of aquatic wildlife in coastal marine habitats

    We developed models to estimate the soft tissue content of benthic marine invertebrates that are prey for aquatic wildlife. Allometric regression models of tissue wet weight with shell length for 10 species of benthic invertebrates had r2 values ranging from 0.29 for hermit crabs Pagurus longicarpus to 0.98 for green crabs Carcinus maenas. As a class, bivalves had the highest r2 values (0.84) and crustaceans the lowest (0.48). Energy and nutrient content of soft tissue is also presented for the 10 benthic species. The energy content was lowest in crabs, and ranged within 2.20–4.71 kcal g-1 dry weight. Fat content was highly variable (range: 3.5–16.0%), and protein content ranged within 43.1–68.1% and was highest for shrimp Palaemonetes pugio. Comparison between classes of organisms of the amount of soft tissue per unit shell length showed that crustaceans yield five times more soft tissue per unit shell length than bivalves, and four times more than gastropods. The models we present use simple measures, such as the length of shell or wet weight of the entire animal, to quantitatively estimate the amount of available soft tissue in benthic prey that are usually consumed in total (with shell and soft tissue intact) but for which only the soft tissue is used for nutritional gain. This information can be combined with energy and nutrient content data to calculate energy or nutrient based carrying capacities that can help assess available resources for shorebirds, waterfowl and marine mammals

    Ornamental marine species culture in the coral triangle: seahorse demonstration project in the Spermonde Islands, Sulawesi, Indonesia.

    Ornamental marine species ('OMS') provide valuable income for developing nations in the Indo-Pacific Coral Triangle, from which most of the specimens are exported. OMS culture can help diversify livelihoods in the region, in support of management and conservation efforts to reduce destructive fishing and collection practices that threaten coral reef and seagrass ecosystems. Adoption of OMS culture depends on demonstrating its success as a livelihood, yet few studies of OMS culture exist in the region. We present a case study of a land-based culture project for an endangered seahorse (Hippocampus barbouri) in the Spermonde Islands, Sulawesi, Indonesia. The business model demonstrated that culturing can increase family income by seven times. A Strengths Weaknesses Opportunities Threats (SWOT) analysis indicated good collaboration among diverse stakeholders and opportunities for culturing non-endangered species and for offshoot projects, but complicated permitting was an issue as were threats of market flooding and production declines. The OMS international market is strong, Indonesian exporters expressed great interest in cultured product, and Indonesia is the largest exporting country for H. barbouri. Yet, a comparison of Indonesia ornamental marine fish exports to fish abundance in a single local market indicated that OMS culture cannot replace fishing livelihoods. Nevertheless, seahorse and other OMS culture can play a role in management and conservation by supplementing and diversifying the fishing and collecting livelihoods in the developing nations that provide the majority of the global OMS

    Experiences of Middle-Level Students, Teachers, and Parents in the Do the Write Thing Violence Prevention Program

    We examined experiences of participants in Do the Write Thing national violence prevention program for middle-level students. Using mixed methods, we conducted surveys and focus groups with students, parents, and teachers who attended the program’s National Recognition Week in Washington, DC. Results revealed important affective, behavioral, and cognitive impacts on participants, including improved relationships, increased understanding of violence, and commitment to reduce violence. Participants from cities where insufficient time and resources were devoted to the project did not experience significant change. Teachers reported developing greater empathy for their students and making substantial changes in their teaching, providing support for students and infusing activities addressing violence into their curriculum. Recommendations are made for increased program support and future research

    Avoiding the Major Causes of Death: Does Childhood Misfortune Reduce the Likelihood of Being Disease Free in Later Life?

    Objectives: Although previous research reveals the detrimental effects of early misfortune on the development of chronic diseases in later life, few studies have investigated its effects on remaining disease free. This study draws on cumulative inequality theory to investigate whether experiencing childhood misfortune reduces the likelihood of remaining disease free over time. Method: This study utilizes five waves of data from the Health and Retirement Study to test whether five domains of childhood misfortune predict being disease free at baseline (2004) and developing disease over time (2004–2012). Results: Respondents reporting risky parental behaviors during childhood were less likely to be disease free at baseline and had an increased risk of disease onset over time, the latter driven by having a guardian who smoked in combination with more pack-years smoked in adulthood. Furthermore, we find that adult resources, that is wealth, help to mitigate the noxious effects of other misfortunes, notably poor socioeconomic conditions. Discussion: Consistent with cumulative inequality theory, these findings reveal that experiencing multiple types of misfortune during childhood decreases the likelihood of remaining disease free in later life, but engaging in health behaviors, such as physical activity, can help to ameliorate some of the noxious effects of early misfortune

    In Texas, the legislature meets every 2 years and at the end of a regular legislative session, hundreds of passed bills will have been sent to the governor for approval. The large number of bills and the wide range of topics they cover can make it difficult to gain an understanding of all the new laws that were passed. At the close of each legislative session the Earl Carl Institute publishes, for the benefit of its constituents, highlights from the session in a bi-annual legislative report. In this year’s publication entitled Report on the Texas Legislature, 85th Session: An Urban Perspective the Institute attempted to cover matters that it believes to be of concern to the urban community, however, many of the highlights cover issues of particular concern to other traditionally disenfranchised communities as well. The legislation covered in these reports generally falls under such issues as Election, Criminal Justice (Human Trafficking, Criminal Procedure, Wrongful Convictions, Domestic Violence), Juvenile Justice, Family Law, Property, Education, Healthcare, Wills, Estate and Probate, Wealth and Litigation.
