    Effects of Dehydration on Resistance Training: An Ongoing Study

    Introduction: Dehydration via hot bath has been shown to impair resistance training performance when performed the same day. The performance decrements in this instance could potentially be due to a combination of the dehydration and residual effects of the heat exposure and not of the dehydration alone. To date, no research has examined the effects of previous night dehydration on resistance training performance. Purpose: The purpose of this ongoing study was to determine the effects of previous night dehydration on performance and perceptual measures during a full-body resistance training protocol. Methods: Healthy, resistance trained males (n = 4) completed two bouts of a full body resistance training protocol (three sets to failure for bench press, lat pull, overhead press, bicep curl, triceps push down, and leg press), dehydrated (~3% body weight) (DT), or heat exposed with fluid replacement (HT). Heart rate (HR) was taken after every set, and participants estimated ratings of perceived exertion (RPE) after each exercise. Session RPE (SRPE) was estimated 5 minutes following completion of the protocol and estimations for feelings of recovery (PRS), perceived readiness to exercise (PR), thirst, and sleep quality. Results: Analysis revealed a significantly less (p = \u3c 0.001) total reps were completed for DT (163.0 ± 4.8) compared to HT (191.3 ± 10.3), though no differences were found for reps per exercise. No significant differences were found for HR (DT vs HT) at any time point. RPE was significantly higher for DT following bicep curl (p = 0.03) (DT: 7.8 ± 0.96, HT: 6.0 ± 0.82) and triceps push down (p = 0.03) (DT: 7.5 ± 0.58, HT: 6.0 ± 0.82). SRPE was significantly higher (p = 0.003) for DT (8.0 ± 0.0) vs HT (6.3 ± 0.5). Significant differences for PRS (p = 0.004) (DT: 5.0 ± 1.4, HT: 7.5 ± 0.58) and PR (p = \u3c 0.001) (DT: 5.0 ± 0.0, HT: 2.5 ± 0.58) indicate participants expected impaired performance during DT. Feelings of thirst were significantly higher (p = 0.001) for DT (7.98 ± 0.94) vs HT (2.73 ± 1.11). No differences were found for estimations of sleep quality (p = 0.59). Conclusion: Even though only preliminary data from a study presently ongoing, the present results suggest that previous night dehydration has a negative influence on both performance and perceptual measures

    The Effects of Short-Term Detraining and Subsequent Retraining on Body Composition and Muscle Performance in Males Consuming a Whey Protein or Carbohydrate Supplement

    An acute bout of resistance exercise (RE) can up-regulate processes that stimulate muscle protein synthesis (MPS). Additionally, nutritional strategies involving carbohydrate (CHO) and whey protein (WP) supplementation can augment MPS. However, resistance training (RT) induced muscle anabolism during the early phases of training can attenuate over time. The use of a short-term cessation of training (detraining; DT) can potentially restore the attenuated muscular anabolic adaptive responses. Therefore, the purpose of this study was to explore the effects of a successive cycle of detraining and retraining (ReT) in humans on body composition and muscle performance. Resistance-trained males (age 20.95 ± 1.23 y; n=20) were recruited and randomized into one of two groups (WP or CHO; 25 grams) in a double-blind fashion. Both groups followed a standardized 4 days per week resistance-training program for 4 weeks, carried out 2 weeks of DT and continued the resistance-training program for another 4 weeks of ReT. Participants were instructed to consume their respective supplement only on workout days during RT, but every day during DT. Research visits were conducted at baseline, 4 weeks (post-RT), 6 weeks (post-2-week-DT), and after 10 weeks (post-ReT). Each visit consisted of body composition assessments and muscular strength and endurance testing using the bench press and angled leg press exercises. Four-day diet records, workout logs, and supplement compliance forms were utilized. Factorial 2x4 (group by time) ANOVAs with repeated measures were conducted using SPSS (version 20.0) with a probability level of ≤ .05. There were no significant group by time interactions for lean or fat mass changes throughout the study (p \u3e .05). However, both groups were able to retain lean mass following 2 weeks of DT. The WP group appeared to have an elevation in lean mass (+1.58kg on average) by the end of ReT in comparison to baseline, even though it was not statistically significant (p \u3e .05). Leg press strength (LPS) increased throughout the study (p=.003), and neither group showed a decrease in LPS following DT. There were no group-by-time interactions or group differences between WP and CHO for bench press strength (BPS), bench press endurance (BPE), leg press endurance (LPE), or any dietary variables (p \u3e .05). Interestingly, the WP group presented a non-significant overall increase in lean mass compared to the CHO group by the end of 10 weeks. LPS and BPS were also elevated and retained respectfully following DT. In summary, a short-term 2 week cycle of DT in resistance trained males maintained both muscle mass and muscular strength, which potentially reinforces the importance of recovery

    Small molecule inhibitors of Late SV40 Factor (LSF) abrogate hepatocellular carcinoma (HCC): evaluation using an endogenous HCC model

    Hepatocellular carcinoma (HCC) is a lethal malignancy with high mortality and poor prognosis. Oncogenic transcription factor Late SV40 Factor (LSF) plays an important role in promoting HCC. A small molecule inhibitor of LSF, Factor Quinolinone Inhibitor 1 (FQI1), significantly inhibited human HCC xenografts in nude mice without harming normal cells. Here we evaluated the efficacy of FQI1 and another inhibitor, FQI2, in inhibiting endogenous hepatocarcinogenesis. HCC was induced in a transgenic mouse with hepatocyte-specific overexpression of c-myc (Alb/c-myc) by injecting N-nitrosodiethylamine (DEN) followed by FQI1 or FQI2 treatment after tumor development. LSF inhibitors markedly decreased tumor burden in Alb/c-myc mice with a corresponding decrease in proliferation and angiogenesis. Interestingly, in vitro treatment of human HCC cells with LSF inhibitors resulted in mitotic arrest with an accompanying increase in CyclinB1. Inhibition of CyclinB1 induction by Cycloheximide or CDK1 activity by Roscovitine significantly prevented FQI-induced mitotic arrest. A significant induction of apoptosis was also observed upon treatment with FQI. These effects of LSF inhibition, mitotic arrest and induction of apoptosis by FQI1s provide multiple avenues by which these inhibitors eliminate HCC cells. LSF inhibitors might be highly potent and effective therapeutics for HCC either alone or in combination with currently existing therapies.The present study was supported in part by grants from The James S.     Negative parental responses to coming out and family functioning in a sample of lesbian and gay young adults

    Parental responses to youths' coming out (CO) are crucial to the subsequent adjustment of children and family. The present study investigated the negative parental reaction to the disclosure of same-sex attraction and the differences between maternal and paternal responses, as reported by their homosexual daughters and sons. Participants' perceptions of their parents' reactions (evaluated through the Perceived Parental Reactions Scale, PPRS), age at coming out, gender, parental political orientation, and religiosity involvement, the family functioning (assessed through the Family Adaptability and Cohesion Evaluation Scales, FACES IV), were assessed in 164 Italian gay and lesbian young adults. Pearson correlation coefficients were calculated to assess the relation between family functioning and parental reaction to CO. The paired sample t-test was used to compare mothers and fathers' scores on the PPRS. Hierarchical multiple regression was conducted to analyze the relevance of each variable. No differences were found between mothers and fathers in their reaction to the disclosure. The analysis showed that a negative reaction to coming out was predicted by parents' right-wing political conservatism, strong religious beliefs, and higher scores in the scales Rigid and Enmeshed. Findings confirm that a negative parental reaction is the result of poor family resources to face a stressful situation and a strong belief in traditional values. These results have important implications in both clinical and social fields

    Comparison of Eight Methods for the Extraction of Bacillus atrophaeus Spore DNA from Eleven Common Interferents and a Common Swab

    Eight DNA extraction products or methods (Applied Biosystems PrepFiler Forensic DNA Extraction Kit; Bio-Rad Instagene Only, Bio-Rad Instagene & Spin Column Purification; EpiCentre MasterPure DNA & RNA Kit; FujiFilm QuickGene Mini80; Idaho Technologies 1-2-3 Q-Flow Kit; MoBio UltraClean Microbial DNA Isolation Kit; Sigma Extract-N-Amp Plant and Seed Kit) were adapted to facilitate extraction of DNA under BSL3 containment conditions. DNA was extracted from 12 common interferents or sample types, spiked with spores of Bacillus atropheaus. Resulting extracts were tested by real-time PCR. No one method was the best, in terms of DNA extraction, across all sample types. Statistical analysis indicated that the PrepFiler method was the best method from six dry powders (baking, biological washing, milk, plain flour, filler and talcum) and one solid (Underarm deodorant), the UltraClean method was the best from four liquids (aftershave, cola, nutrient broth, vinegar), and the MasterPure method was the best from the swab sample type. The best overall method, in terms of DNA extraction, across all sample types evaluated was the UltraClean method

    Factors Associated with Bovine Neonatal Pancytopenia (BNP) in Calves: A Case-Control Study

    Bovine neonatal pancytopenia (BNP; previously known as idiopathic haemorrhagic diathesis and commonly known as bleeding calf syndrome) is a novel haemorrhagic disease of young calves which has emerged in a number of European countries during recent years. Data were retrospectively collected during June to November 2010 for 56 case calves diagnosed with BNP between 17 March and 7 June of the same year. These were compared with 58 control calves randomly recruited from herds with no history of BNP. Multivariable logistic regression analysis showed that increased odds of a calf being a BNP case were associated with its dam having received PregSure® BVD (Pfizer Animal Health) vaccination prior to the birth of the calf (odds ratio (OR) 40.78, p<0.001) and its herd of origin being located in Scotland (OR 9.71, p = 0.006). Decreased odds of a calf being a BNP case were associated with the calf having been kept outside (OR 0.11, p = 0.006). The longer that a cattle herd had been established on the farm was also associated with decreased odds of a calf in that herd being a BNP case (OR 0.97, p = 0.011)

    Attenuated Auditory Event-Related Potentials and Associations with Atypical Sensory Response Patterns in Children with Autism

    Neurobiological underpinnings of unusual sensory features in individuals with autism are unknown. Event-related potentials (ERPs) elicited by task-irrelevant sounds were used to elucidate neural correlates of auditory processing and associations with three common sensory response patterns (hyperresponsiveness; hyporesponsiveness; sensory seeking). Twenty-eight children with autism and 39 typically developing children (4–12 year-olds) completed an auditory oddball paradigm. Results revealed marginally attenuated P1 and N2 to standard tones and attenuated P3a to novel sounds in autism versus controls. Exploratory analyses suggested that within the autism group, attenuated N2 and P3a amplitudes were associated with greater sensory seeking behaviors for specific ranges of P1 responses. Findings suggest that attenuated early sensory as well as later attention-orienting neural responses to stimuli may underlie selective sensory features via complex mechanisms

    Genetic risk and a primary role for cell-mediated immune mechanisms in multiple sclerosis.

    Multiple sclerosis is a common disease of the central nervous system in which the interplay between inflammatory and neurodegenerative processes typically results in intermittent neurological disturbance followed by progressive accumulation of disability. Epidemiological studies have shown that genetic factors are primarily responsible for the substantially increased frequency of the disease seen in the relatives of affected individuals, and systematic attempts to identify linkage in multiplex families have confirmed that variation within the major histocompatibility complex (MHC) exerts the greatest individual effect on risk. Modestly powered genome-wide association studies (GWAS) have enabled more than 20 additional risk loci to be identified and have shown that multiple variants exerting modest individual effects have a key role in disease susceptibility. Most of the genetic architecture underlying susceptibility to the disease remains to be defined and is anticipated to require the analysis of sample sizes that are beyond the numbers currently available to individual research groups. In a collaborative GWAS involving 9,772 cases of European descent collected by 23 research groups working in 15 different countries, we have replicated almost all of the previously suggested associations and identified at least a further 29 novel susceptibility loci. Within the MHC we have refined the identity of the HLA-DRB1 risk alleles and confirmed that variation in the HLA-A gene underlies the independent protective effect attributable to the class I region. Immunologically relevant genes are significantly overrepresented among those mapping close to the identified loci and particularly implicate T-helper-cell differentiation in the pathogenesis of multiple sclerosis

    Educational attainment, health outcomes and mortality: a within-sibship Mendelian randomization study

    BACKGROUND: Previous Mendelian randomization (MR) studies using population samples (population MR) have provided evidence for beneficial effects of educational attainment on health outcomes in adulthood. However, estimates from these studies may have been susceptible to bias from population stratification, assortative mating and indirect genetic effects due to unadjusted parental genotypes. MR using genetic association estimates derived from within-sibship models (within-sibship MR) can avoid these potential biases because genetic differences between siblings are due to random segregation at meiosis. METHODS: Applying both population and within-sibship MR, we estimated the effects of genetic liability to educational attainment on body mass index (BMI), cigarette smoking, systolic blood pressure (SBP) and all-cause mortality. MR analyses used individual-level data on 72 932 siblings from UK Biobank and the Norwegian HUNT study, and summary-level data from a within-sibship Genome-wide Association Study including >140 000 individuals. RESULTS: Both population and within-sibship MR estimates provided evidence that educational attainment decreased BMI, cigarette smoking and SBP. Genetic variant-outcome associations attenuated in the within-sibship model, but genetic variant-educational attainment associations also attenuated to a similar extent. Thus, within-sibship and population MR estimates were largely consistent. The within-sibship MR estimate of education on mortality was imprecise but consistent with a putative effect. CONCLUSIONS: These results provide evidence of beneficial individual-level effects of education (or liability to education) on adulthood health, independently of potential demographic and family-level confounders
