Student perceptions of computer-mediated communication tools in online learning: Helpfulness and effects on teaching, social, and cognitive presence
Distance learning continues to be used in the context of teacher training in special education. Distance learning is experienced through computer-mediated communication (CMC) tools via the Internet, or online learning. Little research has been done to evaluate student perceptions of the helpfulness of CMC tools in online learning. This quantitative study utilizes an online survey of graduate and certification teachers in training for special education. The survey evaluates student perceptions of CMC tool helpfulness, social presence, teaching presence and cognitive presence. Descriptive and inferential statistical analyses are used to evaluate mean differences for different combinations of CMC tool usage and for possible relationships between tool helpfulness and social or teaching presence. Additional demographic variables of gender, level of study, registration status, concentration of study, prior online courses taken and years of teaching experience are also evaluated for possible relationships with social, teaching, and cognitive presence.
Results suggest that participants were comfortable with using CMC tools and perceived e-mail, discussion forums, news forums, web-conferencing, and text chat as helpful tools for social and teaching presence communications. Participants using discussion forums perceived higher teaching and cognitive presence than those using only web-conferencing. Participants using both discussion forums and web-conferencing perceived higher cognitive presence than those using only web-conferencing. Results of standard multiple regressions indicated that 31% of the variance in teaching presence
(24% of variance in social presence) was accounted for by knowing helpfulness scores on e-mail, discussion forums, and news forums. Discussion forums were most helpful for promoting teaching presence. E-mail was most helpful for promoting social presence.
There were no statistically significant mean differences among groups based on gender, level of study, registration status, concentration of study, or years of teaching experience. Results suggest that perceptions of teaching and cognitive presence may decline as the number of online courses taken increases
Factors Affecting the Uptake of COVID-19 Vaccine amongDubai Airport's Professionals
Aim: Comprehending the elements that influence COVID-19 vaccination acceptability and recognizing expediters for vaccination decisions are critical components of developing effective ways to increase vaccine coverage in the general population. This study aims to investigate the main factors affecting COVID-19 vaccination uptake among Dubai 'Airport's employees. In addition, it seeks to explore the main signs and symptoms that appeared on vaccinated employees after taking the COVID-19 vaccination, hence, track the vaccine's safety.
Methods: Employees at Dubai's airport in the United Arab Emirates (UAE), mainly in Dubai, provided data. To gather data online utilising the Google Forms platform, a questionnaire was used as the main quantitative tool. As 2000 questionnaires got distributed, 1007 employees participated in the survey, yielding a 50.4% response rate.
Results: The results show that employees overwhelmingly agree with the assertion that the factors of accessibility and affordability have a significant effect on their decision to receive the COVID-19 vaccine, followed by a trust in vaccine, knowledge, vaccine safety, advice and information, and beliefs on the vaccine. In this study, the agreement level on factors affecting the COVID-19 vaccine uptake was found significantly to be higher in females (88.6%) who were married (91.6%) and those aged over 60 years (89.2%) at P <.05. In addition, the results show that 53.7% of vaccinated staff was found to have one or more side effects of the vaccine, where none of them was hospitalized after immunization. The binary logistic regression analysis in this study shows that females were two times more likely to have 'vaccine's symptoms after vaccination than males (Exp (B): 1.6; 95%CI: 1.127 - 2.351, P< .01). It further reveals that participants in the age group over 50 were three times more likely to have 'vaccine's symptoms after vaccination than participants in the age group 20-29 (Exp (B): 2.9; 95%CI: 2.497-9.681, P< .001). Finally, it indicates that individuals with previous SARS-CoV-2 infection were 2 times more likely to have 'vaccine's symptoms after vaccination than those without known past infection (Exp (B): 1.9; 95%CI: 1.272 - 2.542, P< .01).
Conclusion: There are several factors that playing a significant role in population’s decision to receive the COVID-19 vaccine, where the accessibility and affordability factors were found to have the greatest effect on their decision to uptake the vaccine. The current study concluded that COVID-19 vaccination is safe and that adverse effects from a vaccine are usually modest and affected by several factors such as age, gender, and COVID-19 infection history.
 
Automatic Autism Spectrum Disorder Detection Using Artificial Intelligence Methods with MRI Neuroimaging: A Review
Autism spectrum disorder (ASD) is a brain condition characterized by diverse
signs and symptoms that appear in early childhood. ASD is also associated with
communication deficits and repetitive behavior in affected individuals. Various
ASD detection methods have been developed, including neuroimaging modalities
and psychological tests. Among these methods, magnetic resonance imaging (MRI)
imaging modalities are of paramount importance to physicians. Clinicians rely
on MRI modalities to diagnose ASD accurately. The MRI modalities are
non-invasive methods that include functional (fMRI) and structural (sMRI)
neuroimaging methods. However, the process of diagnosing ASD with fMRI and sMRI
for specialists is often laborious and time-consuming; therefore, several
computer-aided design systems (CADS) based on artificial intelligence (AI) have
been developed to assist the specialist physicians. Conventional machine
learning (ML) and deep learning (DL) are the most popular schemes of AI used
for diagnosing ASD. This study aims to review the automated detection of ASD
using AI. We review several CADS that have been developed using ML techniques
for the automated diagnosis of ASD using MRI modalities. There has been very
limited work on the use of DL techniques to develop automated diagnostic models
for ASD. A summary of the studies developed using DL is provided in the
appendix. Then, the challenges encountered during the automated diagnosis of
ASD using MRI and AI techniques are described in detail. Additionally, a
graphical comparison of studies using ML and DL to diagnose ASD automatically
is discussed. We conclude by suggesting future approaches to detecting ASDs
using AI techniques and MRI neuroimaging
Identification of potential transcription factors that enhance human iPSC generation
Although many factors have been identified and used to enhance the iPSC reprogramming process, its efficiency remains quite low. In addition, reprogramming efficacy has been evidenced to be affected by disease mutations that are present in patient samples. In this study, using RNA-seq platform we have identified and validated the differential gene expression of five transcription factors (TFs) (GBX2, NANOGP8, SP8, PEG3, and ZIC1) that were associated with a remarkable increase in the number of iPSC colonies generated from a patient with Parkinson's disease. We have applied different bioinformatics tools (Gene ontology, protein–protein interaction, and signaling pathways analyses) to investigate the possible roles of these TFs in pluripotency and developmental process. Interestingly, GBX2, NANOGP8, SP8, PEG3, and ZIC1 were found to play a role in maintaining pluripotency, regulating self-renewal stages, and interacting with other factors that are involved in pluripotency regulation including OCT4, SOX2, NANOG, and KLF4. Therefore, the TFs identified in this study could be used as additional transcription factors that enhance reprogramming efficiency to boost iPSC generation technology.This study was supported by QBRI internal grant (QB16) and the Qatar University Student grant (QUST-2-CMED-2019-1)
Hyperosmotic Stress Induces a Specific Pattern for Stress Granule Formation in Human-Induced Pluripotent Stem Cells
Stress granules (SGs) are assemblies of selective messenger RNAs (mRNAs), translation factors, and RNA-binding proteins in small untranslated messenger ribonucleoprotein (mRNP) complexes in the cytoplasm. Evidence indicates that different types of cells have shown different mechanisms to respond to stress and the formation of SGs. In the present work, we investigated how human-induced pluripotent stem cells (hiPSCs/IMR90-1) overcome hyperosmotic stress compared to a cell line that does not harbor pluripotent characteristics (SH-SY5Y cell line). Gradient concentrations of NaCl showed a different pattern of SG formation between hiPSCs/IMR90-1 and the nonpluripotent cell line SH-SY5Y. Other pluripotent stem cell lines (hiPSCs/CRTD5 and hESCs/H9 (human embryonic stem cell line)) as well as nonpluripotent cell lines (BHK-21 and MCF-7) were used to confirm this phenomenon. Moreover, the formation of hyperosmotic SGs in hiPSCs/IMR90-1 was independent of eIF2α phosphorylation and was associated with low apoptosis levels. In addition, a comprehensive proteomics analysis was performed to identify proteins involved in regulating this specific pattern of hyperosmotic SG formation in hiPSCs/IMR90-1. We found possible implications of microtubule organization on the response to hyperosmotic stress in hiPSCs/IMR90-1. We have also unveiled a reduced expression of tubulin that may protect cells against hyperosmolarity stress while inhibiting SG formation without affecting stem cell self-renewal and pluripotency. Our observations may provide a possible cellular mechanism to better understand SG dynamics in pluripotent stem cells
Fundamentos e aplicações da metodologia de ensaios não destrutivos com células bacterianas
Os Ensaios Não Destrutivos (END) são determinantes para a fiabilidade de materiais cuja integridade é de extrema importância. A técnica de Ensaios Não Destrutivos com células bacterianas (CB) tem demonstrado viabilidade para deteção de defeitos superficiais, com espessuras e profundidades inferiores a 5 μm em vários materiais de engenharia. O conhecimento adquirido sobre esta técnica já é significativo mas alguns aspetos necessitam de mais desenvolvimentos, como a interação bactéria-defeito e a viabilidade da técnica para condições de superfÃcie diferentes das já ensaiadas.
O objetivo deste trabalho é alargar a técnica a uma maior gama de materiais de engenharia com condições de superfÃcie diferentes, assim como, desenvolver o conhecimento sobre a interação bactéria-defeito.
A bactéria Rhodococcus erythropolis foi usada na inspeção de vários materiais como AlumÃnio Liga 1100, Estanho, Ouro, Prata, INCONEL 9095, Aço revestido com Nickel, Cobre revestido com Ouro, AlumÃnio revestido com Cobre, PolÃmero com nano tubos de carbono, entre outros, e com condições de superfÃcie diferentes como superfÃcies anodizadas e revestidas. Foram também caracterizados os campos magnéticos de dois equipamentos desenvolvidos para esta técnica de Ensaios Não Destrutivos.
Os resultados experimentais mostraram que a utilização de campos magnéticos contribui positivamente para a deteção de defeitos e que provetes com revestimentos superficiais diferentes revelam resultados diferentes apesar de terem o mesmo material base
CCN3, POSTN, and PTHLH as potential key regulators of genomic integrity and cellular survival in iPSCs
Reprogramming human somatic cells into a pluripotent state, achieved through the activation of well-defined transcriptional factors known as OSKM factors, offers significant potential for regenerative medicine. While OSKM factors are a robust reprogramming method, efficiency remains a challenge, with only a fraction of cells undergoing successful reprogramming. To address this, we explored genes related to genomic integrity and cellular survival, focusing on iPSCs (A53T-PD1) that displayed enhanced colony stability. Our investigation had revealed three candidate genes CCN3, POSTN, and PTHLH that exhibited differential expression levels and potential roles in iPSC stability. Subsequent analyses identified various protein interactions for these candidate genes. POSTN, significantly upregulated in A53T-PD1 iPSC line, showed interactions with extracellular matrix components and potential involvement in Wnt signaling. CCN3, also highly upregulated, demonstrated interactions with TP53, CDKN1A, and factors related to apoptosis and proliferation. PTHLH, while upregulated, exhibited interactions with CDK2 and genes involved in cell cycle regulation. RT-qPCR validation confirmed elevated CCN3 and PTHLH expression in A53T-PD1 iPSCs, aligning with RNA-seq findings. These genes' roles in preserving pluripotency and cellular stability require further exploration. In conclusion, we identified CCN3, POSTN, and PTHLH as potential contributors to genomic integrity and pluripotency maintenance in iPSCs. Their roles in DNA repair, apoptosis evasion, and signaling pathways could offer valuable insights for enhancing reprogramming efficiency and sustaining pluripotency. Excessive substance use in bipolar disorder is associated with impaired functioning rather than clinical characteristics, a descriptive study
<p>Abstract</p> <p>Background</p> <p>There is a strong association between bipolar disorder (BD) and substance use disorder (SUD). The clinical and functional correlates of SUD in BD are still unclear and little is known about the role of excessive substance use that does not meet SUD criteria. Thus, the aims of the current study were to investigate lifetime rates of illicit substance use in BD relative to the normal population and if there are differences in clinical and functional features between BD patients with and without excessive substance use.</p> <p>Methods</p> <p>125 consecutively recruited BD in- and outpatients from the Oslo University Hospitals and 327 persons randomly drawn from the population in Oslo, Norway participated. Clinical and functional variables were assessed. Excessive substance use was defined as DSM-IV SUD and/or excessive use according to predefined criteria.</p> <p>Results</p> <p>The rate of lifetime illicit substance use was significantly higher among patients compared to the reference population (OR = 3.03, CI = 1.9-4.8, p < .001). Patients with excessive substance use (45% of total) had poorer educational level, occupational status, GAF-scores and medication compliance, with a trend towards higher suicidality rates, compared to patients without. There were no significant group differences in current symptom levels or disease course between groups.</p> <p>Conclusion</p> <p>The percentage of patients with BD that had tried illicit substances was significantly higher than in the normal population. BD patients with excessive substance use clearly had impaired functioning, but not a worse course of illness compared to patients without excessive substance use. An assessment of substance use beyond SUD criteria in BD is clinically relevant.</p
Multisite evaluation of institutional processes and implementation determinants for pharmacogenetic testing to guide antidepressant therapy.
There is growing interest in utilizing pharmacogenetic (PGx) testing to guide antidepressant use, but there is lack of clarity on how to implement testing into clinical practice. We administered two surveys at 17 sites that had implemented or were in the process of implementing PGx testing for antidepressants. Survey 1 collected data on the process and logistics of testing. Survey 2 asked sites to rank the importance of Consolidated Framework for Implementation Research (CFIR) constructs using best-worst scaling choice experiments. Of the 17 sites, 13 had implemented testing and four were in the planning stage. Thirteen offered testing in the outpatient setting, and nine in both outpatient/inpatient settings. PGx tests were mainly ordered by psychiatry (92%) and primary care (69%) providers. CYP2C19 and CYP2D6 were the most commonly tested genes. The justification for antidepressants selected for PGx guidance was based on Clinical Pharmacogenetics Implementation Consortium guidelines (94%) and US Food and Drug Administration (FDA; 75.6%) guidance. Both institutional (53%) and commercial laboratories (53%) were used for testing. Sites varied on the methods for returning results to providers and patients. Sites were consistent in ranking CFIR constructs and identified patient needs/resources, leadership engagement, intervention knowledge/beliefs, evidence strength and quality, and the identification of champions as most important for implementation. Sites deployed similar implementation strategies and measured similar outcomes. The process of implementing PGx testing to guide antidepressant therapy varied across sites, but key drivers for successful implementation were similar and may help guide other institutions interested in providing PGx-guided pharmacotherapy for antidepressant management
