    A feasibility study with survival in swine model

    Transrectal access still has some unsolved issues such as spatial orientation, infection, access and site closure. This study presents a simple technique to perform transcolonic access with survival in a swine model series. A new technique for NOTES perirectal access to perform retroperitoneoscopy, peritoneoscopy, liver and lymphnode biopsies was performed in 6 pigs, using Totally NOTES technique. The specimens were extracted transanally. The flexible endoscope was inserted through a posterior transmural incision and the retrorectal space. Cultures of bacteria were documented for the retroperitoneal space and intra abdominal cavity after 14 days. Rectal site was closed using non-absorbable sutures. There was no bowel cleansing, nor preoperative fasting. The procedures were performed in 6 pigs through transcolonic natural orifice access using available endoscopic flexible instruments. All animals survived 14 days without complications, and cultures were negative. Histopathologic examination of the rectal closure site showed adequate healing of suture line and no micro abscesses. The results of feasibility and safety of experimental Transcolonic NOTES potentially brings new frontiers and future wider applications for minimally invasive surgery.     Purification and Preliminary Crystallographic Analysis of a New Lys49-PLA2 from B. Jararacussu

    BjVIII is a new myotoxic Lys49-PLA2 isolated from Bothrops jararacussu venom that exhibits atypical effects on human platelet aggregation. To better understand the mode of action of BjVIII, crystallographic studies were initiated. Two crystal forms were obtained, both containing two molecules in the asymmetric unit (ASU). Synchrotron radiation diffraction data were collected to 2.0 Å resolution and 1.9 Å resolution for crystals belonging to the space group P212121 (a = 48.4 Å, b = 65.3 Å, c = 84.3 Å) and space group P3121 (a = b = 55.7 Å, c = 127.9 Å), respectively. Refinement is currently in progress and the refined structures are expected to shed light on the unusual platelet aggregation activity observed for BjVIII

    Bulbos úmidos a partir da irrigação por gotejamento subsuperficial com água de abastecimento e efluente de esgoto tratado

    The use of treated sewage effluent (TSE) combined with the subsurface drip irrigation (SDI) method in agriculture can decrease the costs of agricultural production, in attempts to fertigate crops more efficiently. In this study it was compared the dimensions of the wet bulb formed by the application of TSE and municipal water supply (MWS) in an Oxisoil. We have evaluated the effect of water quality and discharge between drippers used in sugarcane crop. Three trenches were opened and 21 three-rod TDR probes were setup in a mesh and a dripper was buried at 0.30 m, for each constant discharge of 1.0 L h-1and 1.6 L h-1. Comparing results from different wetted soil profiles it was observed that the vertical and horizontal dimensions of the wet bulb are similar for both MWS and TSE, being peculiars according to the discharges used and volume applied. Regardless the water quality, an increase of 60% in discharge decreased the deepest infiltration.352242253O uso de efluente de esgoto tratado (EET), combinado com o método de irrigação por gotejamento subsuperfical (IGS) na agricultura, pode reduzir os custos de produção agrícola, fertirrigando de maneira eficiente as culturas. Neste artigo, compararam-se as dimensões do bulbo úmido formado pela aplicação de EET e água de abastecimento municipal (AAM), em um Latossolo Vermelho distroférrico. Avaliou-se o efeito da qualidade da água e da vazão entre gotejadores utilizados na cultura da cana-de-açúcar. Três trincheiras foram abertas e 21 sondas de TDR com três hastes foram instaladas em malha, e um gotejador foi enterrado a 0,30 m, para cada vazão constante de 1,0 Lh-1 e 1,6 Lh-1. Comparando os resultados de diferentes perfis de umidade do solo, sugere-se que as dimensões vertical e horizontal do bulbo úmido sejam similares para a AAM e para o EET, sendo peculiares de acordo com as vazões utilizadas e o volume aplicado. Independentemente da qualidade da água, o aumento de 60% na vazão reduziu a infiltração em profundidade

    Guillain-Barré syndrome in temporal association with influenza A vaccine

    OBJECTIVE: To report a case of Guillain-Barré syndrome following influenza A (H1N1) 2009 vaccine. CASE DESCRIPTION: A four-year-old boy presented right thigh pain and ascending muscular weakness 15 days after the second dose of influenza A (H1N1) 2009 vaccine. The neurological examination revealed tetraparesis and areflexia. Electroneuromyography showed lower velocity and conduction blockage with small secondary axonal loss. Treated with intravenous immunoglobulin, the patient reached a plateau in the 4th day, followed by progressive muscular strength improvement. COMMENTS: The employment of large-scale influenza A (H1N1) 2009 vaccination and the preliminary reports from the American Surveillance Program suggest a significant association between Guillain-Barré syndrome and influenza A H1N1 2009 vaccination. All suspected cases of this association should be published for further evaluation. Vaccination remains the most effective method to prevent serious illness and death related to influenza.OBJETIVO: Descrever um caso de síndrome de Guillain-Barré em associação temporal com a vacina influenza A (H1N1) 2009. DESCRIÇAO DO CASO: Menino de quatro anos com queixa inicial de dor em coxa direita e perda de força muscular ascendente 15 dias após a segunda dose da vacina influenza A (H1N1) 2009. Ao exame neurológico apresentava tetraparesia e arreflexia, com predomínio em membros inferiores. A eletroneuromiografia evidenciou redução da velocidade e bloqueio de condução neuronal, com discreta perda axonal secundária. Foi tratado com imunoglobulina por via intravenosa, atingiu platô no quarto dia de evolução da doença e, depois, houve melhora progressiva da força muscular. COMENTÁRIOS: Com o emprego em larga escala da vacina influenza A (H1N1) 2009 em nosso meio e os dados preliminares do sistema de vigilância norte-americano mostrando associação temporal significante com a síndrome de Guillain-Barré, recomenda-se a descrição dos casos suspeitos dessa associação.     The AD(EO)D process - a new concept

    In this study, cooking of the wood had been investigated until kappa numbers 17 and 14 for a process kinetically modified, elimination of oxygen delignification from the fiber line, and application of AD(EO)D and AD(PO)D techniques for pulp of kappa 17 and AD(EO)D and DHT(EPO)DP for pulp of kappa 14 for the bleaching at 90% ISO. As references, cookings had been carried out until kappa number 17 and 14, oxygen delignification in simple stage and conventional bleaching with sequences A/D(EO)D, A/D(EPO)D, A/D(EO)DP and A/D(EPO)DP for the kappa number 17 pulp and oxygen delignification in simple stage and conventional bleaching sequence DHT(EPO)DP for pulp of kappa 14. For the pulp of kappa 17, one concluded that processes AD(EO)D and AD(PO)D produced pulp of brightness 90% ISO with good viscosity and brightness stability, using TAC\ub4s (totally active chloro) relatively low, and it\u2019s not necessary the oxygen delignification. Amongst these two processes, the first one revealed to be more attractive to reach the level of 90% ISO brightness. However, comparatively to the reference processes, with oxygen delignification, for example O-A/D(EPO)DP, process AD(EO)D results in increase of the operational cost of the order of US78/odt,includingwoodandreagents.Forthekappanumber14pulp,processAD(EO)Dalsorevealedtobeattractive,butthesequenceDHT(EPO)DPwastheonethatusedlowerTACs;however,whencomparedwiththereference(ODHT(EPO)DP),itresultedinanincreaseofoperationalcostoftheorderofUS7-8/odt, including wood and reagents. For the kappa number 14 pulp, process AD(EO)D also revealed to be attractive, but the sequence DHT(EPO)DP was the one that used lower TAC`s; however, when compared with the reference (O-DHT(EPO)DP), it resulted in an increase of operational cost of the order of US5/odt. Therefore, it can be concluded that the alternative process, without the deslignification with oxygen, is viable only for pulps of lower kappa number.Nesse estudo, investigaram-se cozimentos da madeira at\ue9 n\ufameros kappa 17 e 14 por um processo cineticamente modificado, elimina\ue7\ue3o da deslignifica\ue7\ue3o com oxig\ueanio da linha de fibras, e aplica\ue7\ue3o das t\ue9cnicas AD(EO)D e AD(PO)D para polpa de kappa 17 e AD(EO)D e DHT(EPO)DP para polpa de kappa 14 para o branqueamento at\ue9 90% ISO. Para a polpa de kappa 17, concluiu-se que os processos AD(EO)D e AD(PO)D produzem polpa de alvura 90% ISO com boa viscosidade e estabilidade de alvura, utilizando TAC\ub4s (Cloro Ativo Total) relativamente baixos, e dispensam completamente a deslignifica\ue7\ue3o com oxig\ueanio. Dentre esses dois processos, o primeiro se mostrou mais atrativo para atingir o n\uedvel de alvura de 90% ISO. Por\ue9m, comparativamente aos processos de refer\ueancia, contendo a deslignifica\ue7\ue3o com oxig\ueanio, ex. OA/ D(EPO)DP, o processo AD(EO)D resulta em aumento do custo operacional da ordem de US7-8/tas, inclusos madeira e reagentes. Para a polpa de n\ufamero kappa 14, o processo AD(EO)D tamb\ue9m se mostrou atrativo, sendo que a seq\ufc\ueancia DHT(EPO)DP foi a que utilizou o menor TAC`s, por\ue9m quando comparada com a refer\ueancia (O-DHT(EPO)DP) resultou num aumento de custo operacional da ordem de US4/tas. Portanto, pode se concluir que o processo alternativo, sem a deslignifica\ue7\ue3o com oxig\ueanio, \ue9 vi\ue1vel somente para polpas de n\ufameros kappa mais baixos

    Constructal design method dealing with stiffened plates and symmetry boundaries

    A new computational procedure for modelling the structural behavior of stiffened plates with symmetry boundary conditions is here presented. It uses two-dimensional finite elements as a way to decrease computational time without losing precision thanks to a relatively small number of elements applied for analyzing out-of-plane displacements (deflections) and stresses. Adding, the constructal design method was included in the procedure, together with the exhaustive search technique, with the scope to optimize the stress/strain status of stiffened plates by design changes. For the purpose, a reference plate without stiffeners was initially design and used as starting point. Part of the volume was reshaped into stiffeners: thickness was reduced maintaining unchanged weight, length and width. The main goal was to minimize strains and stresses by geometric changes. Results demonstrated that, thanks to this design procedure, it is always possible to find an adequate geometry transformation from reference plate into stiffeners, allowing significant improvements in mechanical behavior

    The Genus Caesalpinia L. (Caesalpiniaceae): Phytochemical and Pharmacological Characteristics

    The genus Caesalpinia (Caesalpiniaceae) has more than 500 species, many of which have not yet been investigated for potential pharmacological activity. Several classes of chemical compounds, such as flavonoids, diterpenes, and steroids, have been isolated from various species of the genus Caesalpinia.     Distribuição do sistema radicular da cana-de-açúcar irrigada com efluente de esgoto doméstico por gotejamento subsuperficial

    Irrigation with domestic sewage effluent (DSE) has been recommended by subsurface dripping, as it can obtain a high rate of irrigation efficiency and faster use of salts in comparison with other irrigation methods. The study aimed at evaluating the area, the length and the effective depth of the root system of sugarcane irrigated with DSE by subsurface drip system and with different irrigation rates at depths of 0.00-0.20, 0.20-0.40, 0.40-0.60 and 0.60-0.80m. The experiment was carried out in the municipality of Piracicaba, in the state of São Paulo (SP), Brazil, in a sugarcane area irrigated with DSE in a completely randomized blocks set up in furrows, with three replications and four treatments, which are: one area without irrigation (AWI) and three irrigated areas meeting 50% (T50%), 100% (T100%) and 200% (T200%) of the crop's water need between each round of irrigation. T100% and T200% provided smaller areas and lengths of roots in the two deepest layers, as compared to AWI and T50%, which stimulated the development of deeper roots due to the water stress. TWI, T100% and T200% presented 80% of the roots up to a depth of 0.40m and T50% treatment presented 76.43% of roots total.A irrigação com efluente de esgoto doméstico (EED) tem sido recomendada para aplicação por gotejamento subsuperficial, podendo-se obter elevados índices de eficiência de irrigação e mais rápido aproveitamento dos sais em comparação com outros métodos de irrigação. O trabalho objetivou avaliar a área de raiz, o comprimento e a profundidade efetiva do sistema radicular da cana-de-açúcar irrigada com EED aplicado por gotejamento subsuperficial e com diferentes lâminas de irrigação, nas camadas de solo de 0,00-0,20; 0,20-0,40; 0,40-0,60 e 0,60-0,80 m. O experimento foi realizado em Piracicaba-SP, numa área de cana-de-açúcar irrigada com EED, em um delineamento de blocos casualizados dispostos no esquema de faixas, com três repetições e quatro tratamentos, sendo estes: uma área sem irrigação (TSI) e três áreas irrigadas, atendendo a 50% (T50%), 100% (T100%) e 200% (T200%) da necessidade hídrica da cultura entre cada turno de rega. O T100% e o T200% proporcionaram menores áreas e comprimento de raízes nas duas camadas mais profundas, quando comparados ao TSI e ao T50%, que devido ao estresse hídrico, proporcionado à cultura, estimularam o desenvolvimento de raízes mais profundas. O TSI, T100% e T200% apresentaram 80% das raízes até 0,40 m e, nessa mesma profundidade, o tratamento T50% apresentou 76,43% do total de raízes.64765

    Chemical Chaperones Curcumin and 4-Phenylbutyric Acid Improve Secretion of Mutant Factor H R127H by Fibroblasts from a Factor H-Deficient Patient

    Factor H (FH) is one of the most important regulatory proteins of the alternative pathway of the complement system. Patients with FH deficiency have a higher risk for development of infections and kidney diseases because of the uncontrolled activation and subsequent depletion of the central regulatory component C3 of the complement system. In this study, we investigated the consequences of the Arg(127)His mutation in FH (FHR127H) previously described in an FH-deficient patient, on the secretion of this protein by skin fibroblasts in vitro. We observed that, although the patient cells stimulated with IFN-gamma were able to synthesize FHR127H, the mutant protein was largely retained within the endoplasmic reticulum (ER), whereas normal human fibroblasts stimulated with IFN-gamma secrete FH without retention in the ER. Moreover, the retention of FHR127H provoked enlargement of ER cisterns after treatment with IFN-gamma. A similar ER retention was observed in Cos-7 cells expressing the mutant FHR127H protein. Despite this deficiency in secretion, we show that the FHR127H mutant is capable of functioning as a cofactor in the Factor I-mediated cleavage of C3. We then evaluated whether a treatment could increase the secretion of FH, and observed that the patient's fibroblasts treated with the chemical chaperones 4-phenylbutiric acid or curcumin increased the secretion rate of FH. We propose that these chemical chaperones could be used as alternative therapeutic agents to increase FH plasma levels in FH-deficient patients caused by secretion delay of this regulatory protein.
