    Experimental evidence of shock mitigation in a Hertzian tapered chain

    We present an experimental study of the mechanical impulse propagation through a horizontal alignment of elastic spheres of progressively decreasing diameter ϕn\phi_n, namely a tapered chain. Experimentally, the diameters of spheres which interact via the Hertz potential are selected to keep as close as possible to an exponential decrease, ϕn+1=(1q)ϕn\phi_{n+1}=(1-q)\phi_n, where the experimental tapering factor is either q15.60q_1\simeq5.60~% or q28.27q_2\simeq8.27~%. In agreement with recent numerical results, an impulse initiated in a monodisperse chain (a chain of identical beads) propagates without shape changes, and progressively transfer its energy and momentum to a propagating tail when it further travels in a tapered chain. As a result, the front pulse of this wave decreases in amplitude and accelerates. Both effects are satisfactorily described by the hard spheres approximation, and basically, the shock mitigation is due to partial transmissions, from one bead to the next, of momentum and energy of the front pulse. In addition when small dissipation is included, a better agreement with experiments is found. A close analysis of the loading part of the experimental pulses demonstrates that the front wave adopts itself a self similar solution as it propagates in the tapered chain. Finally, our results corroborate the capability of these chains to thermalize propagating impulses and thereby act as shock absorbing devices.Comment: ReVTeX, 7 pages with 6 eps, accepted for Phys.     Experimental evidence of solitary wave interaction in Hertzian chains

    We study experimentally the interaction between two solitary waves that approach one to another in a linear chain of spheres interacting via the Hertz potential. When these counter propagating waves collide, they cross each other and a phase shift respect to the noninteracting waves is introduced, as a result of the nonlinear interaction potential. This observation is well reproduced by our numerical simulations and it is shown to be independent of viscoelastic dissipation at the beads contact. In addition, when the collision of equal amplitude and synchronized counter propagating waves takes place, we observe that two secondary solitary waves emerge from the interacting region. The amplitude of secondary solitary waves is proportional to the amplitude of incident waves. However, secondary solitary waves are stronger when the collision occurs at the middle contact in chains with even number of beads. Although numerical simulations correctly predict the existence of these waves, experiments show that their respective amplitude are significantly larger than predicted. We attribute this discrepancy to the rolling friction at the beads contacts during solitary wave propagation

    PURPOSE: Mitochondrial disorders are a common cause of inherited metabolic disease and can be due to mutations affecting mitochondrial DNA or nuclear DNA. The current diagnostic approach involves the targeted resequencing of mitochondrial DNA and candidate nuclear genes, usually proceeds step by step, and is time consuming and costly. Recent evidence suggests that variations in mitochondrial DNA sequence can be obtained from whole-exome sequence data, raising the possibility of a comprehensive single diagnostic test to detect pathogenic point mutations. METHODS: We compared the mitochondrial DNA sequence derived from off-target exome reads with conventional mitochondrial DNA Sanger sequencing in 46 subjects. RESULTS: Mitochondrial DNA sequences can be reliably obtained using three different whole-exome sequence capture kits. Coverage correlates with the relative amount of mitochondrial DNA in the original genomic DNA sample, heteroplasmy levels can be determined using variant and total read depths, and-providing there is a minimum read depth of 20-fold-rare sequencing errors occur at a rate similar to that observed with conventional Sanger sequencing. CONCLUSION: This offers the prospect of using whole-exome sequence in a diagnostic setting to screen not only all protein coding nuclear genes but also all mitochondrial DNA genes for pathogenic mutations. Off-target mitochondrial DNA reads can also be used to assess quality control and maternal ancestry, inform on ethnic origin, and allow genetic disease association studies not previously anticipated with existing whole-exome data sets

    Acute febrile illness is associated with Rickettsia spp infection in dogs

    BACKGROUND: Rickettsia conorii is transmitted by Rhipicephalus sanguineus ticks and causes Mediterranean Spotted Fever (MSF) in humans. Although dogs are considered the natural host of the vector, the clinical and epidemiological significance of R. conorii infection in dogs remains unclear. The aim of this prospective study was to investigate whether Rickettsia infection causes febrile illness in dogs living in areas endemic for human MSF. METHODS: Dogs from southern Italy with acute fever (n = 99) were compared with case–control dogs with normal body temperatures (n = 72). Serology and real-time PCR were performed for Rickettsia spp., Ehrlichia canis, Anaplasma phagocytophilum/A. platys and Leishmania infantum. Conventional PCR was performed for Babesia spp. and Hepatozoon spp. Acute and convalescent antibodies to R. conorii, E. canis and A. phagocytophilum were determined. RESULTS: The seroprevalence rates at first visit for R. conorii, E. canis, A. phagocytophilum and L. infantum were 44.8%, 48.5%, 37.8% and 17.6%, respectively. The seroconversion rates for R. conorii, E. canis and A. phagocytophilum were 20.7%, 14.3% and 8.8%, respectively. The molecular positive rates at first visit for Rickettsia spp., E. canis, A. phagocytophilum, A. platys, L. infantum, Babesia spp. and Hepatozoon spp. were 1.8%, 4.1%, 0%, 2.3%, 11.1%, 2.3% and 0.6%, respectively. Positive PCR for E. canis (7%), Rickettsia spp. (3%), Babesia spp. (4.0%) and Hepatozoon spp. (1.0%) were found only in febrile dogs. The DNA sequences obtained from Rickettsia and Babesia PCRs positive samples were 100% identical to the R. conorii and Babesia vogeli sequences in GenBank®, respectively. Febrile illness was statistically associated with acute and convalescent positive R. conorii antibodies, seroconversion to R. conorii, E. canis positive PCR, and positivity to any tick pathogen PCRs. Fourteen febrile dogs (31.8%) were diagnosed with Rickettsia spp. infection based on seroconversion and/or PCR while only six afebrile dogs (12.5%) seroconverted (P = 0.0248). The most common clinical findings of dogs with Rickettsia infection diagnosed by seroconversion and/or PCR were fever, myalgia, lameness, elevation of C-reactive protein, thrombocytopenia and hypoalbuminemia. CONCLUSIONS: This study demonstrates acute febrile illness associated with Rickettsia infection in dogs living in endemic areas of human MSF based on seroconversion alone or in combination with PCR

    Rickettsia slovaca Infection: DEBONEL/TIBOLA

    Producción CientíficaThis study describes the epidemiological, clinical, and microbiological characteristics of a new tick-borne disease in Spain—Dermacentor-borne necrosis erythema lymphadenopathy (DEBONEL). The clinical presentations include an eschar at the site of the tick bite, surrounded by an erythema and painful regional lymphadenopathy. The disease appears during the colder months and its vector is Dermacentor marginatus (D. marginatus). From January 1990 to December 2004, 54 patients presented at Hospital of La Rioja with these clinical and epidemiological data. The ratio of females to males was 32/22. The average age was 37 years. In all cases tick bites were located on the upper body (90% on the scalp). The median incubation period was 4.7 days. Signs and symptoms were mild in all cases. Only a small number of patients presented mild and nonspecific abnormalities in a complete blood cell count and mild elevation of erythrocyte sedimentation rates and C-protein reactive and liver enzyme levels. Serological evidence of acute rickettsiosis was observed in 19 patients (61%). In 29% sera tested by polymerase chain reactions (PCRs) were positive. The sequence obtained from a PCR product revealed 98% identity with Rickettsia sp. strains RpA4, DnS14, and DnS28. All ticks removed from patients were PCR-positive. Sequencing showed 8 of them identified as R. slovaca and 2 as Rickettsia sp. strains RpA4, DnS14, and DnS28
