    Prediction of the acoustic insulation of a prefabricated wooden-based system for collective buildings

    Using wood, a renewable, carbon sink material with outstanding thermal properties associated with the prefabrication process and its benefits, a construction system for multi-storey multifamily buildings based on prefabricated wooden panels was developed to act as a tool for meeting the national carbon neutrality targets. It will contribute to the promotion of increasingly sustainable cities that optimise the use of materials and rely on highly energy-efficient buildings. This sustainable alternative to conventional construction materials was designed to comply with the Portuguese needs, regulations, and regulatory requirements regarding construction, structural, functional, and logistics demands. As the acoustic behaviour of wooden buildings is a sensitive subject due to the wood's lightweight and poor insulation performance for low-frequency sounds, this work aims to analyse the panels regarding their acoustic behaviour through INSUL software.     Antimicrobial activity, phenolic profile and role in the inflammation of propolis

    Nowadays a great amount of information regarding chemical and biological aspects of bee products is available in the literature, but few data on its therapeutic uses are found. The aim of this study was to evaluate the phenolic profile, the in vitro antimicrobial activity and effect in the hyaluronidase enzyme (widely related with the inflammation process) of propolis harvested in Portugal. The efficacy of three extracts (hydro-alcoholic, methanolic and aqueous) was also compared. It was chosen the hydroalcoholic extract, because this was the most effective for extracting phenolic compounds. The antimicrobial activity was accessed in Gram-positive and Gram-negative bacteria and yeasts, isolated from different biological fluids and the results were then compared with the obtained for reference microorganisms. The propolis from Bragança was the one that possessed the highest polyphenols’ content. The sample from Beja showed the less significant inhibition of the hyaluronidase enzyme. Concerning the antimicrobial activity, Candida albicans was the most resistant and Staphylococcus aureus the most sensitive. The reference microorganisms were more sensitive than the ones isolated from biological fluids

    An integrative assessment to determine the genotoxic hazard of estuarine sediments: combining cell and whole-organism responses

    The application of the Comet assay in environmental monitoring remains challenging in face of the complexity of environmental stressors, e.g., when dealing with estuarine sediments, that hampers the drawing of cause-effect relationships. Although the in vitro Comet assay may circumvent confounding factors, its application in environmental risk assessment (ERA) still needs validation. As such, the present work aims at integrating genotoxicity and oxidative DNA damage induced by sediment-bound toxicants in HepG2 cells with oxidative stress-related effects observed in three species collected from an impacted estuary. Distinct patterns were observed in cells exposed to crude mixtures of sediment contaminants from the urban/industrial area comparatively to the ones from the rural/riverine area of the estuary, with respect to oxidative DNA damage and oxidative DNA damage. The extracts obtained with the most polar solvent and the crude extracts caused the most significant oxidative DNA damage in HepG2 cells, as measured by the formamidopyrimidine-DNA glycosylase (FPG)-modified Comet assay. This observation suggests that metals and unknown toxicants more hydrophilic than polycyclic aromatic hydrocarbons may be important causative agents, especially in samples from the rural part of the estuary, where oxidative DNA damage was the most significant. Clams, sole, and cuttlefish responded differentially to environmental agents triggering oxidative stress, albeit yielding results accordant with the oxidative DNA damage observed in HepG2 cells. Overall, the integration of in vivo biomarker responses and Comet assay data in HepG2 cells yielded a comparable pattern, indicating that the in vitro FPG-modified Comet assay may be an effective and complementary line-of-evidence in ERA even in particularly challenging, natural, scenarios such as estuarine environments

    Influence of estimated training status on anti and pro-oxidant activity, nitrite concentration, and blood pressure in middle-aged and older women

    The purpose of this study was to compare the association between anti and pro-oxidant activity, nitrite concentration, and blood pressure (BP) in middle-aged and older women with different levels of estimated training status (TS). The sample consisted of 155 females (50-84 years) who were submitted to a physical examination to evaluate estimated TS through the "Functional Fitness Battery Test," BP measurements, and plasma blood samples to evaluate pro-oxidant and antioxidant activity and nitrite concentrations. Participants were separated by age into a middle-aged group (< 65 years) and an older (≥65 years) group and then subdivided in each group according to TS. Blood biochemistry was similar between groups. On the other hand, protein oxidation was lower in participants with higher TS, independent of age. Older females with higher TS presented higher nitrite concentrations, lower lipoperoxidation, and lower values of BP compared with those with lower TS. Lower GPx activity was observed in participants with higher TS compared with middle-aged with lower TS. Thus, our results suggest that good levels of TS may be associated with lower oxidative stress and higher nitrite concentration and may contribute to maintain normal or reduced blood pressure values.Fil: Jacomini, André M..     Sett Use, Density and Breeding Phenology of Badgers in Mediterranean Agro-Sylvo-Pastoral Systems

    Carnivores social organization varies widely, from strongly social to solitary predators. European badgers are facultative social carnivores that also shows a geographical variation in social structure. These patterns derive mainly from central/west European regions, with an underrepresentation of Mediterranean populations that face different conservation challenges, especially regarding group composition, sett use patterns and breeding phenology. We addressed these traits topics for a population inhabiting a Portuguese agro-silvo-pastoral system. Based on monthly monitoring of 34 setts and continuous camera-trapping surveys of 12, we showed that setts surrounded by diversified vegetation and located in sandy sites are more used, a pattern probably linked to food availability and ease of sett excavation and maintenance, respectively. Badgers followed a general pattern regarding group size (2–4 adults), but showed an intermediate population density (0.49–0.73 badgers/km2), with values higher than those estimated for other Mediterranean environments, but lower than for central-western populations. This, together with the breeding (November/January) and cub emergence (1.8 cubs/sett; March/April) periods, indicates an ecological adaptation to the landscape context, where human-related resources and mild environmental conditions allow badger to reach higher densities than in many southern populations, and to reproduce earlier than their northern counterparts.Understanding carnivores social structure variation is pivotal for properly addressing conservation challenges and solutions. The European badgers is a social carnivore for which most of the available information regarding how this species is socially organized derives from central west populations. This article describes the group composition, den use patterns and breeding phenology of a Mediterranean population of badgers. We showed that badger live in low density, in relatively small groups, composed by 2–4 adult animals and ca. 2 cubs, born in winter.     Conservation of the Beira Railway Station in Mozambique: Lessons from a 20th-Century Heritage Case Study in Africa

    The conservation of built cultural heritage is a fundamental task for modern societies. Specialized expertise is necessary to define a proper strategy to preserve a heritage that is accumulating damage due to the deterioration of materials, repeated loading, and exceptional events. This paper provides part of the work developed for the definition of the Conservation Management Plan of a 20th-century building in Africa. The monumental building, the Beira railway station, is in Mozambique and was considered among the one hundred more important 20th-century Portuguese engineering buildings. The work presented focuses on the analysis of damage, material characterization, and assessment of the comfort conditions of this concrete building—fundamental tasks for the understanding of the current condition of the building and the definition of appropriate remedial actions and maintenance tasks. The work provides a detailed definition of different approaches and techniques that can be implemented for this type of building to obtain reliable information to define measures and actions that are more in line with the reality of the building.     H3K56me3 is a novel, conserved heterochromatic mark that largely but not completely overlaps with H3K9me3 in both regulation and localization.

    Histone lysine (K) methylation has been shown to play a fundamental role in modulating chromatin architecture and regulation of gene expression. Here we report on the identification of histone H3K56, located at the pivotal, nucleosome DNA entry/exit point, as a novel methylation site that is evolutionary conserved. We identify trimethylation of H3K56 (H3K56me3) as a modification that is present during all cell cycle phases, with the exception of S-phase, where it is underrepresented on chromatin. H3K56me3 is a novel heterochromatin mark, since it is enriched at pericentromeres but not telomeres and is thereby similar, but not identical, to the localization of H3K9me3 and H4K20me3. Possibly due to H3 sequence similarities, Suv39h enzymes, responsible for trimethylation of H3K9, also affect methylation of H3K56. Similarly, we demonstrate that trimethylation of H3K56 is removed by members of the JMJD2 family of demethylases that also target H3K9me3. Furthermore, we identify and characterize mouse mJmjd2E and its human homolog hKDM4L as novel, functionally active enzymes that catalyze the removal of two methyl groups from trimethylated H3K9 and K56. H3K56me3 is also found in C. elegans, where it co-localizes with H3K9me3 in most, but not all, tissues. Taken together, our findings raise interesting questions regarding how methylation of H3K9 and H3K56 is regulated in different organisms and their functional roles in heterochromatin formation and/or maintenance

    Environmental risk assessment in a contaminated estuary: an integrated weight of evidence approach as a decision support tool

    Environmental risk assessment of complex ecosystems such as estuaries is a challenge, where innovative and integrated approaches are needed. The present work aimed at developing an innovative integrative methodology to evaluate in an impacted estuary (the Sado, in Portugal, was taken as case study), the adverse effects onto both ecosystem and human health. For the purpose, new standardized lines of evidence based on multiple quantitative data were integrated into a weight of evidence according to a best expert judgment approach. The best professional judgment for a weight of evidence approach in the present study was based on the following lines of evidence: i) human contamination pathways; ii) human health effects: chronic disease; iii) human health effects: reproductive health; iv) human health effects: health care; v) human exposure through consumption of local agriculture produce; vi) exposure to contaminated of water wells and agriculture soils; vii) contamination of the estuarine sedimentary environment (metal and organic contaminants); viii) effects on benthic organisms with commercial value; and ix) genotoxic potential of sediments. Each line of evidence was then ordinally ranked by levels of ecological or human health risk, according to a tabular decision matrix and expert judgment. Fifteen experts scored two fishing areas of the Sado estuary and a control estuarine area, in a scale of increasing environmental risk and management actions to be taken. The integrated assessment allowed concluding that the estuary should not be regarded as impacted by a specific toxicant, such as metals and organic compounds hitherto measured, but by the cumulative risk of a complex mixture of contaminants. The proven adverse effects on species with commercial value may be used to witness the environmental quality of the estuarine ecosystem. This method argues in favor of expert judgment and qualitative assessment as a decision support tool to the integrative management of estuaries. Namely it allows communicating environmental risk and proposing mitigation measures to local authorities and population under a holistic perspective as an alternative to narrow single line of evidence approaches, which is mandatory to understand cause and effect relationships in complex areas like estuaries.info:eu-repo/semantics/publishedVersio
